Relationships among MTHFR a1298c Gene Polymorphisms and Methylation Status of Dact1 Gene in Transitional Cell Carcinomas

  • Objectives: The purpose of this study was to determine the relationship between methylation status of the Dact1 gene and MTHFR a1298c polymorphic forms in transitional cell carcinoma tissues in a Chinese population. Methods: Polymorphisms of folate metabolism enzyme gene MTHFR were assessed by restrictive fragment length polymorphism (RFLP) methods and PCR-based DNA methylation analysis was used to determine the CpG island methylation status of the Dact1 gene. Associations between the methylation status of the Dact1 gene and clinical characteristics, as well as MTHFR a1298c polymorphisms, were analyzed. Results: aberrant methylation of the Dact1 gene was found in 68.3% of cancer tissues and 12.4% of normal tissues,. The methylation rate of the Dact1 gene in cancer tissues was significantly higher in patients with lymph node metastasis than in those without lymph node metastasis (46.3% vs. 17.2%, P = 0.018). No association was found between aberrant DNA methylation and selected factors including sex, age, tobacco smoking, alcohol consumption and green tea consumption. After adjusting for potential confounding variables, variant allele of MTHFR a1298c was found to be associated with methylation of the Dact1 gene. Compared with wild type CC, the odds ratio was 4.33 (95% CI: 1.06-10.59) for AC and 4.95 (95% CI: 1.18-12.74) for AA. The N stage in TNM staging and the occurrence of lymph node metastasis were associated with an MTHFR 1298 AA+AC genotype (P<0.05). Conclusion: MTHFR 1298 AC and AA genotypes might help maintain a normal methylation status of the Dact1 gene, aberrant CpG island methylation of which is closely related to the genesis and progression of transitional cell carcinoma.

사이버초등학교 모형에 관한 연구 (A Study on the Model of Cyber Elementary School)

  • 인터넷을 중심으로 하는 정보사회로의 진행은 우리에게 사이버 공간이라는 또 하나의 삶의 공간을 제공해 주었고, 우리 사회에 사이버 공간이 막대한 영향력을 행사하고 있는 현실에서 초등학교에도 사이버 대학의 연착륙을 경험 삼아 사이버 학교에 대한 논의가 이루어져야 할 때라 생각하여 본 연구에서는 초등학교에 적용 가능한 사이버 초등학교의 모형을 제시하고, 모형별 운영방안에 대해 논의하였으며, 모형 분류 및 운영에 대한 현장 교사들의 의견을 설문을 통해 알아보고 결과를 분석하였다. 학교지원형 사이버초등학교, 학교연합형 사이버초등학교, 독립형 사이버초등학교, 과도기형 사이버초등학교로 나누어 각 모형별로 개념, 주요 교육 대상 및 목적, 운영 체제, 교육과정 및 내용을 연구한 후, 설문을 통해 현장 교사들의 의견을 수렴한 결과, 초등학교에 사이버초등학교가 도입될 경우 과도기형과 학교지원형의 형태로 운영되는 것이 바람직하다고 나타났다.

양성 뇌수막종의 조직학적 아형 및 이형성 뇌수막종에서 PCNA와 Ki-67 표지지수의 비교 (Comparison of Proliferative Activity in Each Histological Subtypes of Benign and Atypical Intracranial Meningiomas by PCNA and Ki-67 Immunolabeling)

  • 목 적 : 양성 뇌수막종에 비하여 이형성 및 악성 뇌수막종이 나쁜 임상적인 예후 및 양상을 보이는 것은 잘 알려져 있으나, 양성 뇌수막종에 있어서 각각의 병리조직학적 아형에 따른 생물학적 양상의 차이에 대해서는 잘 알려지지 않거나 일부 논란이 되고있다. 본 연구에서는 이형성 뇌수막종 및 양성 뇌수막종의 각각의 병리조직학적 아형에 따른 증식능의 차이여부를 알아보고자 PCNA와 Ki-67표지지수를 분석하였다. 방 법 : 본원에서 뇌수막종으로 수술을 시행하여 얻은, 재발을 보여 재수술을 시행한 2례를 포함하여, 파라핀에 포매시킨 27개의 조직을 대상으로 병리학적인 증식능을 분석하기 위해, PCNA에 대한 단일항체 및 MIB-1 단일항체를 이용한 면역조직화학적 염색을 시행하였다. 조직학적 분류상 meningothelial type이 8례, transitional type이 9례, fibroblastic type이 5례였으며, 이형성 수막종이 5례였다. 결 과 : PCNA표지지수의 평균값은 양성 수막종에서 meningothelial type이 $4.82{\pm}5.10%$, transitional type이 $9.01{\pm}4.25%$, fibroblastic type이 $5.66{\pm}5.32%$를 보였으나 이형성 수막종에서는 $27.62{\pm}19.67%$의 높은 지수를 나타냈고, Ki-67 표지지수의 평균값은 양성 수막종의 아형에서 각각 $0.43{\pm}0.85%$, $0.44{\pm}1.08%$, $0.24{\pm}0.18%$를 보이고, 역시 이형성 수막종에서는 $0.84{\pm}0.59%$의 높은 지수를 보였다. 즉, 양성 수막종에서 각각의 아형에 따른 PCNA 및 Ki-67 표지지수는 통계학적으로 의미있는 차이는 없었으나(p>0.05), 이형성 수막종에서는 의미있는 높은 표지지수를 보여(p<0.05) 양성 수막종에서 보다 높은 증식능을 보임을 알 수 있었다. 결 론 : PCNA 및 Ki-67 표지지수를 이용한 증식능의 비교결과, 양성 뇌수막종에서는 각각의 아형에 따른 생물학적 양상이나 예후는 차이가 없을것으로 생각되나, 이형성 수막종에서는 높은 증식능을 보여 이에 대한 예후를 예상할 수 있을것으로 생각되며, 또한 이러한 표지지수가 병리조직학적으로 양성과 이형성의 감별에 많은 도움이 될것으로 사료된다.

평판 근접 후류에서 경계층의 유동조건에 따른 난류유동장 (Turbulent Flow Field on Boundary Layer Flow Conditions in the Near-Wake of a Flat Plate)

  • An experimental study was quantitatively carried out in order to investigate the influence of flow conditions on a boundary layer in the near-wake of a flat plate. Tripping wires attached at various positions were selected to change flow conditions of a boundary layer in the vicinity of trailing edge. The flows such as laminar, transitional, and turbulent boundary layer at 0.98C from the leading edge are imposed to investigate the evolution of symmetric and asymmetric wake. Measurements were made at freestream velocity of 6.0m/s, and the corresponding Reynolds number is $2.8{\times}10^5$. An x-type hot-wire probe(55P61) was employed to measure at 8 stations in the near-wake region. Test results show that the near-wake of the flat plate for the case of a laminar and transitional boundary layer is sensitive to mean flow shear generated after separation but for the case of turbulent boundary layer is insensitive.

둥근형 무전극 램프의 페라이트 코어와 냉점의 온도 의존성 (Dependence of Round Type Electrodeless Lamp according to Ferrite Core and Cold Spot Temperature)

  • Generally Lighting system consists of lamp and luminaire. When a fluorescent lamp is installed in luminaire, power and light output is changed by ambient temperature. Particularly electrodeless lamp depends on the changes that are mercury pressure with amalgam temperature and magnetic properties with ferrite temperature. It has finally influence on optical efficiency. In this study, the temperature change of ferrite and cold spot, vessel are measured at transitional state and then same characteristics are measured with increase of ambient temperature. At transitional state, luminous flux is related to temperature change of cold spot that compare with behavior of mercury pressure and light output. At increase of ambient temperature, we analyzed change that efficiency and electrical, optical characteristics of elecrodeless lamp are related to ferrite core and cold spot temperature.

Growth Pattern of Pandalus gracilis (Decapoda: Pandalidae) in the Southern Coastal Waters of Korea

  • The population dynamics of Pandalus gracilis was investigated in the southeastern coastal area of Korea between May 1998 and April 2000. Of the 4,127 specimens, 57% were identified as females, 39% as males, and 4% as transitional hermaphrodites. The number of females was greater than that of males and transexuals. A significant correlation was observed between the number of transitional hermaphrodites and ambient seawater temperature. Growth parameters were estimated using the modified von Bertalanffy growth function model incorporating seasonal variation in growth using the program ELEFAN. Females grew faster and reached a larger size-at-age than males (K=0.65/y and $L_{\infty}$=17.86 mm carapace length [CL] for females; K=0.51/y and $L_{\infty}$=14.70 mm CL for males). Mean size and age (95% confidence limits) at sex transition, calculated from growth parameters, were 7.07 mm carapace length and 1.05 years, respectively. The reproductive strategies of pandalid shrimps are discussed in terms of the type of sex transition.

재즈 음악내의 일시적 전조에 관한 연구 (A Study about Transitional Modulations in Jazz Music)

  • 재즈 스탠다드 곡들에 있어서 많은 곡들이 1개의 조성 안에 있는 것처럼 보이지만 적어도 한 개 이상의 새로운 조가 비록 짧은 순간이라 할지라도 성립되는 경우가 많다. 게다가 어떤 곡들은 여러 차례에 걸쳐 일시적인 조성의 변화를 내포하기도 하는 데, 이 경우 코드 진행을 '동형진행'하여 조성을 일시적으로 변화시키는 경우가 많으며 이를 '동형진행을 통한 일시적 전조'라고 지칭한다. 동형진행하게 되는 코드진행으로는 장조 및 단조의 II-V-I 진행이 많이 쓰이며, 동형진행적 일시적 전조기법에 있어서 가장 많이 쓰이는 패턴은 장2도 혹은 단2도 간격으로 하행하는 패턴이다. 장2도 간격으로 하행하는 전조 패턴은 가장 많이 사용되는 기법으로, 일례로 두 번을 사용하게 되면 인접하는 상황에서 3개의 조성이 나타나게 된다. 우리는 이 기법을 통하여 감정이 고조되었다가 급격히 가라앉는 현상을 효과적으로 표현할 수 있다. 반음 아래로의 전조는 더욱 극적이라고 표현할 수 있다. 반 음 아래로의 전조 기법은 인접하는 두 개의 조성 간의 대비적인 측면을 더욱 높일 수 있어서 한층 더 드라마틱한 효과를 낼 수 있다.

Cyclooxygenase-2 Expression in Urinary Bladder Transitional Cell Carcinoma and its Association with Clinicopathological Characteristics

  • Background: Transitional cell carcinoma (TCC) is the most predominant type of urinary bladder tumor. As cyclooxygenase (COX)-2 is recently introduced as an attractive target molecule in bladder TCC, we evaluated the immunohistochemical expression of this marker and its association with several clinicopathological characteristics. Materials and Methods: This cross-sectional study was performed in the Pathology department of Sina Hospital in Tehran, Iran during 2006-2011. Ninety-two paraffin embedded blocks were selected from patients with urinary bladder TCC who underwent cystectomy or transurethral resection (TUR). Then, we assessed COX-2 expression by immunohistochemical staining using antibody against COX-2. Staining in more than 5% of tumor cells was considered as positive expression. Results: COX-2 was expressed in 50 % of our patients. This marker was markedly expressed in high grade bladder TCC (62.1%) versus other grades and there was statistically a significant difference in COX-2 expression between various grades (p=0.008). In addition, patients' age, lymphatic and perineurial invasion were associated with the expression of COX-2 (p=0.001, 0.015 and 0.039, respectively). However, other parameters such as stage, tumor size, venous invasion and lymph node metastasis did not show any significant relationship with this marker (all, p>0.05). Conclusions: COX-2 was expressed in urinary bladder TCC especially in high grade forms, advocating its probable role in the differentiation of this tumor. Accordingly, COX-2 could be a valuable biological target molecule in the evaluation and treatment of patients with bladder TCC.

  • The purpose of this study was to examine how the interface between "place of work" and "place of living" in the housing sphere has changed, and to understand its social background. During the korean modernization period, changes in economic structure toward industry has an influence on many aspects of modem life in addition to occupations. The traditional mixing of heterogeneous activities in the home-such as between reproduction and household affairs, first changed into a coexistence of two spaces with different functions within the boundaries of home, and finally into a spatial separation between functions in an urban dimension. As a result of this process, the primary role of the modem home is as a place for relaxation without work. One important kind of home, in which a retail shop is integrated with it, was researched as a transitional form. Its spatial layout showed a various combined usage of both spaces. In conclusion, changes in relevant macro-social aspects are very concretely reflected m the function and spatial organization of the home.
