Systematic Review on the Study of Sasang Typology Published in Korea from 2000 to 2009 (최근 10년간 사상의학 연구동향에 대한 체계적 고찰연구)

  • The purpose of this study was to review the current situation of Sasang typology studies and get a recommendation for ameliorating it by systematically reviewing articles published in Korea from 2000 to 2009. Six hundred seventy-seven articles were found by searching the representative Korean databases such as KISS, RISS, KISTI, DBPIA, NDSL and also by hand search. Yearly analysis was performed by academic field, journal, author and affiliation, type of article and content. The analysis showed that the number of published articles has increased from 61 in 2000 to 86 in 2009. 607(89.6%) were related to medical field, 627(92.6%) were published in Korean Citation Index-listed journals, and 413(68.8%) were published in the Journal of Sasang Constitutional Medicine. The authors listed on one article were 3.98 persons and affiliated in 1.66 departments in average. Sixty six percents are for the clinical studies or case reports, and twenty-five for Sasang type diagnosis. This study showed that more active collaborations with diverse research fields and various research methods are needed for the improvement of Sasang typology studies and the scope expansion of Korean traditional medicine.

Fermentation Increases Antidiabetic Effects of Acanthopanax Senticosusbhpark@chonbuk.ac.kr (발효에 의한 오가피의 항당뇨 활성 촉진)

  • Extract of Acanthopanax senticosus has recently been demonstrated to possess significant antidiabetic potential, in accordance with the traditional use of this plant as an antidiabetic natural health product. The present study evaluated the effects of fermented extract (FE) of this plant on glucose-stimulated insulin secretion, glucose uptake, and streptozotocin-induced type 1 diabetes model. A 3 h pretreatment with FE prevented $IL-1{\beta}$ and $IFN-{\gamma}$ toxicity in isolated rat islets. However, it did not affect insulin-stimulated glucose uptake in C2C12 myotubes. In addition, pretreatment of mice with FE blocked the destruction of streptozotocin-induced islets and the development of type 1 diabetes. FE reduced blood glucose level, increased insulin secretion, and improved glucose tolerance in streptozotocin-treated mice, whereas nonfermented extract (NFE) had moderate effects. Immunohistochemical staining for insulin clearly showed that pretreatment with FE blocked the STZ-induced islets destruction and restored the number of islet cells that secreted insulin to the level of the control. Although the active principles and their mechanisms of action remain to be identified, FE may nevertheless represent a novel complementary therapy and a source of novel therapeutic agents against type 1 diabetes mellitus.

A Feasibility Study of Pulse Rate Per Respiration as an Indicator for the Reaction to Cold Stress (냉자극에 대한 맥율 변화 예비 연구)

  • Cold and Heat pattern identification(CHPI) in traditional East Asian medicine(TEAM) is one of the major indicator to distinguish characteristics of disease and to determine treatment method. Basic parameters to determine CHPI include the pulse rate, respiration rate, and pulse power. Studies to associate physiological responses of human body by cold stress(CS) with CHPI in TEAM were rarely done so far. This study aims to explore the effects of cold stress on pulse signal via a feasibility study for three subjects and investigate some indices which can reflect autonomic nerve reaction(ANR). We measured radial pulse signals and respiration signal of the investigated subjects before the CS, during the CS which continues for 5 minutes, and immediately after the CS, respectively. Finally, we analyzed the pulse rate (P), respiration rate (R), pulse power, pulse depth, and pulse rate per respiration (P/R ratio). As a result, the P/R ratio showed a consistently decreasing tendency through the CS stimulation process, while other parameters behaved more complex and in subject-specific ways. It implies that, among candidate parameters, the P/R ratio is a simple but the most probable parameter that can be used as the ANR indicator. This result is also consistent with the theory in TEAM scripts, in which the P/R ratio is predicted to be a direct indicator for the CHPI. This pilot study shows that P/R ratio can be more appropriately associated with the ANR than heart rate or respiration rate alone. Extensive studies will be necessary to verify or confirm the P/R ratio as an appropriate and well defined parameter for ANR.

Genotoxicity Study of GST Extract (GST 추출물의 유전독성평가)

  • This study aimed to evaluate the genotoxicity of GST (Gamisasangja-tang). For examining genotoxicity, we carried out bacterial reverse mutation assay, chromosome aberration assay, micronucleus induction test according to OECD guidelines. Bacterial reverse mutation assay: In GST treating group, regardless of existence S9 mix, revertant colonies counts appeared to be less than twice of negative control group and dose dependent increase. In positive control group, revertant colonies counts were shown to be more than twice of negative control croup. Chromosome aberration assay: All cell line showed repetition rate of abnormal chromosome aberration less than 5%, regardless of treating time, existence of S9 mix, and no significant change ($p{\succeq}0.05$) compared with negative control group. Micronucleus induction test: Micronucleated polychromatic erythrocytes (MNPCE) repetition rate of Polychromatic erythrocytes (PCE) showed no significant changes compared with negative control group ($p{\succeq}0.05$). PCE portion of total erythrocytes also showed no significant changes ($p{\succeq}0.05$). Our results showed that GST didn't induce any genotoxicity.

Preliminary Study to Develop the Instrument on Pattern Identification for Lung Cancer (폐암 변증도구 개발을 위한 기초연구)

  • This study is to develop a standard tool for pattern identifications in Korean Medicine for Lung Cancer. The advisor committee for this study was organized by 12 Korean Medicine professors from the Korean Association of Traditional Oncology. The item and structure of instrument were based on review of published literature. We took the consultation twice from the advisor committee and additional advices by e-mail correspondences. We divided the symptoms and signs of lung cancer into 6 pattern identifications. - lung spleen both deficiency (肺脾兩虛), lung kidney both deficiency (肺腎兩虛), dampness phlegm obstructing the lung (濕痰蘊肺), qi stagnation blood stasis (氣滯血瘀), yin deficiency toxin heat (陰虛毒熱), and heat toxin accumulation (熱毒蘊結). We obtained the mean weights which reflected the standard deviations from each symptoms of the 6 pattern identifications which were scored on a 5-point scale by 12 experts. We designed the Korean medicine pattern identification tool for lung cancer. It was composed of 57 questions in the question-and-answer format. Though there are some limits that this study is not proved about validity and reliability, the instrument is meaningful and expected to be applied to the subsequent research.

Effect of CST on atopic dermatitis related inflammatory cytokines (청기소독탕(淸肌消毒湯)이 아토피피부염 염증 관련 인자에 미치는 영향)

  • In vitro tests were performed using CST to investigate its role on oxidative damages and inflammatory cytokines. 90% or higher cell viability was observed in CST treated groups from 25 to 200 ${\mu}g/m{\ell}$ using Raw 264.7 cells. CST showed dose-dependent DPPH scavenging activity, with 91.3% and 92.2% scavenging activities at 400 and 800 ${\mu}g/m{\ell}$ concentrations, respectively. CST showed dose-dependent suppression activity of ROS production, especially at 200 ${\mu}g/m{\ell}$ of 41.3%. CST decreased NO production activity, with significant decrease of 16.2% and 33.5% at 100 and 200 ${\mu}g/m{\ell}$ concentrations, respectively. IL-$1{\beta}$, IL-6, MCP-1 production rate were significantly decreased by 30.0%, 27.2%, 22.1% when Raw 264.7 cells were treated with LPS and with CST of 200 ${\mu}g/m{\ell}$. Also, TNF-${\alpha}$ production rate was decreased by 28.6%. The results above indicated therapeutic effect of CST on the AD through anti-oxidative and immune modulatory effect. Various blending of drug substances with CST should be clinically tested.

Combined Treatment with Coptidis Rhizoma Extract and Arsenic Trioxide Enhanced Apoptosis through Diverse Pathways in H157 Cells

  • Coptidis rhizoma (huanglian) is an herb that is widely used in traditional Chinese medicine that has recently been shown to possess anticancer activity. However, the molecular mechanism underlying the anticancer effects of this herb is poorly understood. In this study, we investigated the anticancer activity of a combination of CR extract and arsenic trioxide, as well as the apoptotic pathway associated with its mechanism of action in human lung cancer H157 cells. Combined treatment of H157 cells with CR extract and arsenic trioxide resulted in significant apoptotic death. In addition, combined treatment with CR extract and arsenic trioxide acted in concert to induce a loss of mitochondrial membrane potential (${\Delta}{\Psi}$), the release of cytochrome c from mitochondria, and an increase in the expression of pro-apoptotic p53 and Bax protein, which resulted in activation of caspases and apoptosis. CR extract combined with arsenic trioxide also increased the lipid peroxidation, mRNA expression of DR4 and DR5 and caspase-8 activity. These data indicate that combined treatment with CR extract and arsenic trioxide enhanced apoptotic cell death in H157 cells through diverse pathways, including mitochondrial dysfunction and death receptors, particularly DR4 and DR5. Thus, this treatment may be an effective from of chemotherapy.

Herbal Cocktail Sagunja Protects $H_2O_2$-induced H9c2 Cardiomyoblast Cell Death through the Induction of Heme Oxygenase-1

  • Sagunjatang (Sagunja), containing Radix Astragali, Radix Ginseng, Fructus Schizandrae, Radix Ophiopogonis and Radix Glycyrrhizae, has been used as a prescription for ischemic heart and brain diseases in Korean traditional medicine. This study was designed to investigate the protective mechanisms of Sagunja on $H_2O_2$-induced cytotoxicity of H9c2 cardiomyocytes. Treatment with $H_2O_2$ resulted in death of H9c2 cells, characterized by apparent apoptotic features, including the fragmentation of nucleus and increase in sub-GO/G1fraction of cell cycle. However, Sagunja markedly suppressed the apoptotic characteristics of H9c2 cells induced by $H_2O_2$ with decrease of intracellular peroxide level. In addition, Sagunja suppressed the features of mitochondrial dysfunction, including change of mitochondrial membrane potential, in $H_2O_2$- treated cells. Additionally, Sagunja induced the expression of HO-1 protein in both time-and dose-dependent manner. The role of HO-1 in ROS-scavenging activity of Sagunja is proposed.

Studies on the Extraction Method and Polysaccharide of Tricholoma matsutake using the Supersonic wave and Microwave (초음파와 극초단파를 이용한 송이버섯의 추출법과 다당체에 관한 연구)

  • In order to optimize the extract condition and improve physiological activity of the extract form Tricholoma matsutake, experiments related to extraction methods, totale yield, content of total soluble polysaccharide, SOD-like activity, total polyphenol amount, and volatile flavor compound and the others were carried out, results were obtained as following: Compare with traditional hydrothermal extraction method (Hot water extraction : HWEW), it illustrates that the low temperature extraction method which combines a supersonic waves and microwave (Supersonic microwave extraction : SMEW) causes of increasing the total yield, total soluble polysaccharide. As to the anti-oxident effect, SMEW method leds to increasing of the SOD-like activity, total polyphenol amount as well. Also, cytotoxic effect and growth inhibitory effect against cancer cell line are much higher in SMEW method than HWEW method, especially SMEW5 extracts treated by supersonic 15 min. and microwave 120W, 3 min. and 2 times. The main volatile flavor compound and infinitesimal volatile flavor compound both increase significantly by SMEW method. It is concluded the main components of the volatile flavor compounds extracted from Tricholoma matsutake are 1-octen-3-0l, Methyl cinnamate, 2-octeno1 et al. alcohol typies. Consequently, SMEW5 method is considered as the most effective one for anti-oxidant and is prior to any other methods. And the optimun conditions of this method are : supersonic waves (supersonic, 25KHz, 50W) 15 minutes, microwave spectroscopy (microwave, 2,450MHz, 120W) 3 minutes, and every treatment is performed once followed twice repeats.

Characterization of Hibiscus Latent Fort Pierce Virus-Derived siRNAs in Infected Hibiscus rosa-sinensis in China

  • Although limited progress have been made about pathogen system of Hibiscus rosa-sinensis and Hibiscus latent Fort Pierce virus (HLFPV), interaction between plant host and pathogen remain largely unknown, which led to deficiency of effective measures to control disease of hibiscus plants caused by HLFPV. In this study, infection of HLFPV in Hibiscus rosa-sinensis was firstly confirmed for the first time by traditional electron microscopy, modern reverse transcription polymerase chain reaction and RNA-seq methods in China (HLFPV-Ch). Sequence properties analyzing suggested that the full-length sequences (6,465 nt) of HLFPV-Ch had a high sequence identity and a similar genomic structure with other tobamoviruses. It includes a 5'-terminal untranslated region (UTR), followed by four open reading frames encoding for a 128.5-kDa replicase, a 186.5-kDa polymerase, a 31-kDa movement protein, 17.6-kDa coat protein, and the last a 3'-terminal UTR. Furthermore, HLFPV-Ch-derived virus-derived siRNAs (vsiRNAs) ant its putative target genes, reported also for the first time, were identified and characterized from disease Hibiscus rosa-sinensis through sRNA-seq and Patmatch server to investigate the interaction in this pathogen systems. HLFPV-Ch-derived vsiRNAs demonstrated several general and specific characteristics. Gene Ontology classification revealed predicted target genes by vsiRNAs are involved in abroad range of cellular component, molecular function and biological processes. Taken together, for first time, our results certified the HLFPV infection in China and provide an insight into interaction between HLFPV and Hibiscus rosa-sinensis.
