  • This study was devised to observe an inhibitory reaction toward an lipolytic action of toxohormone-L from KEUMSUNYEON powder. Toxohormone-L is known to be a lipolytic factor that was purified from the ascites fluid of sarcoma 180-bearing mice and of patients with hepatoma. KEUMSUNYEON powder was found to inhibit toxohormone-L induced lipolysis at its concentration of 0.5 mg/ml.

  • Toxohormone-L is a lipolytlc factor, found in ascites fluid of sarcoma 180-bearing mice and of patients with hepatoma. A substance that inhibited the lipolytic action of Toxohormone-L was isolated from Korean red ginseng powder. This substance had a pectin-like a-1,4-polygalacturonan backbone with some acetoxyl groups, and so was an acidic polysaccharide. Acidic polysaccharide was found to inhibit significantly toxohormone-L-induced lipolysis at its concentration of 10$\mu\textrm{g}$/ml.

  • This study was devised to observe the inhibitory effect of 7 kinds of the acidic polysaccharide fraction(PGI, PGa, PG3, PGa, PGs, PG6 and PG7) from Korean white ginseng on a lipolytic action of Toxohormone-L. Toxohormone-L is a lipolytic factor, found in ascites fluid of .sarcoma -180 bearing mice and of patients with hepatoma. A substace that inhibited the lipolytic action of Toxohormone-L was isolated from white ginseng powder. This substance was an acidic polysaccharides. In vitro test showed that the inhibitory effect of PGs fraction of the lipolysis by Toxohormone-L was highest percent among other treatments at concentration of 50, 10, 200, 500 and 1,000 Ug/ml of reaction mixture. And total inhibitory activity(units) of PGI and PG4 was highest among other treatments at the same concentration and that of 10 Ug/ml.

  • This study was devised to observe the inhibitory effect of 7 kinds PG(PG1, PG2, PG3, PG4, PG5, PG6 and PG7) and of 5 kinds PG4(PG41, PG42, PG43, PG44 and PG45) of the acidic polysaccharide fraction from Korean red ginseng on a lipolytic action of Toxohormone-L. A substance that inhibited the lipolytic action of toxohormone-L was isolated from red ginseng powder. This substance was an acidic polysaccharides In vitro test showed that the inhibitory effect of PG4 and PG43 fraction of the lipolysis by Toxohormone-L was highest percent among other treatments at concentration of 50, 100, 200, 500 and 1,000ug/ml of reaction mixture. And total inhibitory activity(units) of PG1 and PG4, and PG4 s was highest among other treatments at the same concentration.

  • Toxohormone-L is a lipolytic factor, found in ascites fluid of sarcoma 180-bearing mice and of patients with hepatoma A substance that inhibited the lipolytic action of Toxohormoue-L was isolated and purified from Korean red ginseng powder. This substance had a pectin-like $\alpha$-1,4-polygalacturonan backbone with some acetoxyl groups, and so was an acidic polysaccharide It inhibited Toxohormone-L Induced liploysis in a dose dependent manner at concentrations higher than 10 Ug/ml. Purified acidic Polysaccharide yield(PG4-3 and PG4-4 fraction) was about 0.03%. And also pectic acid that inhibited the lipolytic action of Toxohormone-L.

  • This study was divised to observe an Inhibitory effect toward a lipolytic action of toxohormone-L from large root and small root Nepal pseudo ginseng (NPG ; Nepal products) components by water extract and ethanol precipitate in vitro. Toxohormone-L is known to be a lipolytic factor that was partially purified from the ascites fluid of Sarcoma 180-bearing mice and of patients with hepatoma. The inhibitory effect that inhibited the lipolytic action of toxohormone-L by ethanol Precipitate component of large root NPG (mean 55.5%) was higher (mean 1.37 times) than that of water extract component in final reaction concentration of 500 and 1, 000ug/ml, on the other side inhibitory effect of water extract component in small root NPG (mean 55.5%) was higer (mean 1.14 times) than that of ethanol precipitate component. In a way inhibitory effect of precipitate component In large root NPG(47.6%), when final reaction concentration of sample were 1, 000ug/ml, was about 40% lower than that of Korean red ginseng.

  • This study was divised to observe an inhibitory toward a lipolytic action of toxohormone-L from large root and small root Nepal pseudo ginseng(NPG ; Nepal products) components by water extract and ethanol precipitate in vitro. Toxohormone-L Is known to be a lipolytic factor that was partially purified from the ascites fluid of sarcoma 180-hearing mice and of patients with hepatoma. The inhibitory effect that inhibited the lipolytic action of toxohormone-L by ethanol precipitate component of large root NPG(mean 46.8%) was higher (mean 1.8 times) than that of water extract component in final reaction concentration ,5001g1m1, on the other side inhibitory effect of water extract component in small root NPG(mean 43.9%) was higher(mean 1. 2 times) than that of ethano1 precipitate component, respectively. In a way inhibitory effect of ethanol precipitate component in large root NPG(47.6%), when final reaction concentration of sample were 1,000 U g/ml, was about 4095 lower than that of Korean red ginseng, respectively.

  • 여러 종류의 종양 환자에서 점차적인 체중 감소가 일어나는 과정에서, 체내에 저장된 지방이 고갈됨이 관찰되었다. 종양이 커가는 동안의 체내 지방의 고갈은 혈장의 유리지방산의 증가와 연관이 있다. 최근에, 우리는 간암, 또는 난소암 환자의 복수액과 악성 임파종 환자의 늑막액은(in vitro에서) 흰쥐의 지방조직에 유리지방산의 방출을 촉진시킴을 발견하였다. Toxoh-ormone-L(독성 호르몬-L)이라고 명명한 이 지방분해 인자를 간암 환자의 복수액에서 분리, 정제하였다. 이 분리된 물질은 disc gel electrophoresis와 ${\btea}$-m-ercaptoethanol 존재하의 sodium dodecyl sulfate(SD5)-acrylamide gel electrophoresis상에서 단일띠를 나타내었다. 이것의 분자량은 각각 SDS-acrylamide gel electrophoresis와 analytical 초원심분리에 의하여 $70,000{\~}75,000$과 65,200의 분량을 가짐이 확인되었다. 흰쥐에 Toxohormone-L의 주사는 사료와 물의 섭취를 현저히 억제하였으며, 이 Toxohormone-L의 주사 5시간후에 이러한 억제효과가 나타났다. Toxohormone-L의 지방분해의 작용을 억제하는 억제물질이 고려인삼 뿌리의 물 추출물에 존재함이 입증되었다. 즉, 인삼 분말을 5배량의 증류수에 현탁시켜 추출하고 감압 농축한 다음 증류수에 투석시켰다. 투석막안의 지방 성분을 석유 에테르로 제거하고, 사포닌은 수포화된 부탄올로 제거하였다. 이렇게 제거하고 남은 여액을 활성탄으로 처리하고 DEAE-cellulose column에 통과시켰다. 이러한 과정으로 유효한 분획 (인삼분획 3)을 얻었으며, 보다 더 순수한 정제가 지금 진행중에 있다. 이러한 실험 결과를 바탕으로 볼 때, 암환자에 고려인삼 분말의 투여는 암조직에서 분비되는 Toxohormone-L(독성 호르몬-L)의 작용을 억제함으로서 암환자의 체중 감소를 방지하고, 식욕감퇴를 개선할것으로 생각된다.

  • This study was devised to observe the inhibitory effects of 3 kinds of petroleum ether extracts (percolation by petroleum ether) from Korean red ginseng, Chinese red ginseng and American white ginseng on a lipolytic action of Toxohormone-L which has been known as lipolytic and anorexigenic factors. Toxohormone-L was obtained by partial purification of the ascites fluid from mice which had been Inoculated with sarcoma-180. The yields of petroleum ether extract from Korean red ginseng, Chinese red ginseng and American white ginseng were 0.64, 0.47 and 0.58 and respectively, indicating that the yield of Korean red ginseng was the highest. In vitro, at the concentration of 2 mg /ml, the inhibition rate of lipolysis by the petroleum ether extract of Korean red ginseng, Chinese red ginseng and American white ginseng were 55.1, 50.0 and 44.9% respectively, and the total inhibitory activity per gram of ginseng material were 18, 12 and 13 unit respectively, indicating that the Korean red ginseng was the most effective in the inhibition of the lipolysis.
