Higher Extraction of Phytochemical Compounds from Tartary Buckwheat Seeds by the Application of Surfactant Formulation

  • The aim of this study was to determine the suitability of surfactant to extract higher phenolic compound, flavonoid and antioxidant activity from Tartary buckwheat and evaluate the potentiality of surfactant as a screening agent for breeding purpose. Primarily, we employed two types of surfactant (Hydrophilic: Tween 20 and Lipophilic: Span 80) to select the suitable surfactant agent for the extraction of optimum bioactive compounds. Between two surfactants, Tween 20 showed highest efficiency at 4 mM concentration to extract total phenolic content (TP), total flavonoid (TF) and antioxidant activity (AA). Tween 20 at 4 mM concentration was fixed for further analysis along with hot water ($90^{\circ}C$) treatment as a control. In our findings, highest TP (118 mg/g), TF (38 mg/g) and AA (76%) was achieved in KW21 and KW22 among the fifteen accessions of Tartary buckwheat. In other way, TP, TF and AA was 200%, 120% and 110% higher in surfactant formulation compared with control treatment, respectively.

Antioxidant Activity of Major Cultivars Prunus mume in Korea (국내 주요 재배 매실 품종의 항산화 활성)

  • This study examined the total phenolic and flavonoid compounds, and the antioxi dant activity of major cultivars of Prunus mume in Korea. The total phenolic content ranged from 64.13 to $93.43mg{\cdot}100g^{-1}$, with the content in 'Osuku' being higher than that in the other cultivars; a difference in the content among cultivars was observed. The total flavonoid content was $6.16{\sim}18.57mg{\cdot}100g^{-1}$, showing a significant difference, with the highest content being noted in 'Gojirou'. The DPPH radical scavenging activity was measured and was relatively high for Nanko, Kamakami, Natsuka, Dana, and Osuku cultivars. When ABTS radical scavenging activity was measured, all the cultivars showed a scavenging activity over 80%, with the scavenging activity of 'Osuku' being the highest. With respect to nitrite scavenging ability, all cultivars exhibited a high scavenging activity of over 50%, with the highest activity observed for 'Nanko' (76.03%), 'Gojirou' (70.56%), and 'Yourou' (70.32%). Based on the study results, it is considered that Prunus mume can be a useful resource because of various substances with functional physiological activities.

Measurement of Antioxidant Activities and Phenolic and Flavonoid Contents of the Brown Seaweed Sargassum horneri: Comparison of Supercritical CO2 and Various Solvent Extractions

  • Seaweed Sargassum horneri extracts were obtained using supercritical carbon dioxide ($SC-CO_2$) and different solvents. $SC-CO_2$ was kept at a temperature of $45^{\circ}C$ and pressure of 250 bar. The flow rate of $CO_2$ (27 g/min) was constant during the entire 2-h extraction period, and ethanol was used as a cosolvent. Six different solvents [acetone, hexane, methanol, ethanol, acetone mix methanol (7:3), and hexane mix ethanol (9:1)] were used for extraction and agitated by magnetic stirring (250 rpm) in the dark at $25^{\circ}C$ for 20 h; the ratio of material to solvent was 1:10 (w/v). Antioxidant properties of S. horneri extracted using $SC-CO_2$ with ethanol and different solvents have shown good activity. The highest activity belongs to $SC-CO_2$ with ethanol extracted oil, showing DPPH, ABTS, total phenolic content, and total flavonoid levels of $68.38{\pm}1.21%$, $83.51{\pm}1.25%$, $0.64{\pm}0.02mg/g$, and $5.57{\pm}0.05mg/g$, respectively. The S. horneri extracts showed a significant correlation between the antioxidant activity and phenolic content. Based on these results, the $SC-CO_2$ extract (ethanol) of the seaweed extract from brown seaweed may be a valuable antioxidant source.

Fabrication of Electrospun PVA Nanofibers Loaded with Artemisia capillaris Thunberg Extracts (인진쑥 추출물 함유 PVA 나노섬유 제조)

  • This study extracted Artemisia capillaris Thunberg with distilled water and ethanol to investigate its antioxidant effect. We then investigated the possibility of producing nanofibers by an electrospinning process by adding the extracts to polyvinyl alcohol (PVA). The electrospinning method used a PVA concentration of 12wt.%, an applied voltage of 10kV, and a tip-to-collector distance of 15cm. Total polyphenol and total flavonoid contents were measured to verify the antioxidant activity of Artemisia capillaris Thunberg extracts (ACEs). The total polyphenol content of the distilled water extract and the ethanol extract were measured as 218.47 and 271.26mg/g, respectively, and the total flavonoid content of the distilled water extract and the ethanol extract were measured as 141.68 and 34.98mg/g, respectively. As the content of the ACEs in the PVA nanofibers increased, the Diameters of the nanofibers and the uniformity of the diameters decreased. The electrospinning process was fabricated in a relatively uniform form without beads, and the diameters of the nanofibers that were produced ranged from 340 to 390nm. The results of FT-IR, XRD and DSC analyses confirmed that the ACEs were well mixed with the PVA molecules and were electrospun.

A Study on the Antioxidant Effect of Doenjang Prepared with Vegetable Water (채소수로 제조한 된장의 항산화 효과 연구)

  • This study compared and analyzed the antioxidant effect of Doenjang prepared from vegetable water, and explored the optimal addition ratio of vegetables of 5 kinds and the possibility of application to Doenjang. The sample is three kinds of vegetable water (VW1, VW2, VW3) prepared by adding different ratios of radish, carrot, green onion, onion and shiitake mushroom and Denjang prepared using it. Doenjang was aged and fermented at about 40℃ for 40 days, and then separated and used only solids. The content of their antioxidant compounds was measured the content of total phenolic acid contents and total flavonoid contents. In addition, the antioxidant effect was measured by electron donating activity, SOD-like activity, ABTs radical scavenging activity and reducing power. The total phenolic acid contents and total flavonoid contents were high at VW3 and that Doenjang made with VW3. Electron donating activity and SOD-liked activity were high at VW2 and Doenjang made with VW2. ABTs radical scavenging activity was high in Doenjang made of VW3, and Reducing power was high in VW3. Therefore, if Doenjang is prepared with vegetable water prepared by properly mixing 5 types of vegetables, the possibility of developing Doenjang with high antioxidant effect was suggested.

Radical Scavenging Activity of Kemenyan Resin Produced by an Indonesian Native Plant, Styrax sumatrana

  • Kemenyan resin from Styrax sumatrana is a unique non-timber forest product (NTFP) native from Sumatera Island, Indonesia. It possesses a wide range of applications in the pharmaceutical, perfume, and cosmetics industries. In this paper, six kemenyan resin samples were investigated to evaluate their free radical scavenging activity using 1,1-diphenyl-2-picrylhydrazyl (DPPH) reagent. The kemenyan resin samples, which originated from North Tapanuli, Pakpak Bharat, and Humbang Hasundutan, showed high antioxidant activity with $IC_{50}$ < 16 mg/L. The antioxidant activity of common kemenyan resin constituents, i.e., cinnamic acid, ethyl cinnamate, gallic acid, and vanillin was also investigated as positive control, although they exhibited lower antioxidant activity ($IC_{50}$ < 1000 mg/L), except for gallic acid ($IC_{50}$ = 5,23 mg/L). The total phenolic and flavonoid contents (TPC and TFC) for all samples were 44-66 mg gallic acid equivalents (GAE)/g sample and 143-160 mg quarcetin equivalents (QE)/g sample. The results revealed that kemenyan resin has high potency as an antioxidant and could be used as a natural antioxidant resource.

Radical Scavenging Activities of Fruits of Crataegus pinnatifida BUNGE Major. from Korea

  • Screenings of potential antioxidant activities of Crataegus pinnatifida BUNGE Major. fruits extracted 80% methanol were performed using four antioxidant assays. Significant differences were observed both in total phenolic contents (TPC) and total flavonoid contents (TFC), DPPH radical scavenging activity, nitric oxide scavenging activity, ABTS radical scavenging assay, and reducing power assay. The total polyphenol content and total flavonoid content in the extract were measured to be $224.4{\pm}0.52$ mg GAE/100 g and $12{\pm}0.25$ mg QE/100 g, respectively. When the tested concentration was $500{\mu}g/mL$, DPPH and ABTS radical-scavenging activities of methanolic extracts were 84.15% and 88.8%, respectively. The reducing power and nitric oxide scavenging activity were increased at the manner of dose-dependently. These results suggest that methanolic extracts of Crataegus pinnatifida Bge. fruits possess excellent radical scavenging activities and may serve as a potential source of natural antioxidant.

Effects of Hot Air Drying on the Antioxidant Capacity of Actinidia arguta × A. deliciosa cv. Mansoo, a Hardy Kiwifruit (열풍 건조 가공 공정이 다래 과실(품종명 만수)의 산화방지능에 미치는 영향)

  • A hardy kiwifruit is a good source of phenolics and antioxidants. This study was aimed at investigating the effects of hot air drying at three different temperatures (35, 50, and $70^{\circ}C$) for 24 h on the total phenolic and flavonoid contents and the antioxidant capacity of hardy kiwifruits of Actinidia arguta ${\times}$ A. deliciosa cv. Mansoo. Dried kiwifruit extracts were produced using 80% (v/v) aqueous ethanol under homogenization. Hot air drying lowered the total phenolic and flavonoid contents and decreased the antioxidant capacity in the dried kiwifruits compared with their fresh counterparts. The results also revealed high positive linear correlations of antioxidant capacity with total phenolic and flavonoid contents, indicating that phenolics are the major contributors to antioxidant capacity. Our results suggested that an optimal drying process must be determined for industrial application in order to obtain dried kiwifruits with increased antioxidant capacity for consumers.

Antioxidative Activity and Tyrosinase Inhibitory Activity of the Extract and Fractions from Arctium lappa Roots and Analysis of Phenolic Compounds (우엉 뿌리 추출물의 항산화 및 Tyrosinase 저해 활성과 Phenolic Compound 분석)

  • In this study, we investigated on antioxidative activities and tyrosinase inhibitory activities of methanol extract and its fractions from roots of Arctium lappa. The total phenolic compound and flavonoid content of the ethylacetate fraction was found to be 818.29 mg/g and 360.59 mg/g as the highest content. In the measurement of DPPH radical scavenging ability and tyrosinase inhibitory activity, the ethylacetate fraction was higher than the other fractions and the extract. In addition, comparative analysis of phenolic compounds by liquid chromatography-mass spectrometry (MS)/MS system under the multiple-reaction monitoring (MRM) with negative-ion electrospray ionization mode. The main phenolic compounds in the extract and fractions of roots from Arctium lappa were cynarin and chlorogenic acid. The main phenolic compound of the ethylacetate fraction was cynarin. n-Butanol fraction had a significantly higher chlorogenic acid content than other samples. In conclusion, DPPH radical scavenging ability and tyrosinase inhibitory activity of the cynarin-riched ethylacetate fraction showed the highest activity.

Antioxidant activities of Erythrina stricta Roxb.using various in vitro and ex vivo models

  • Erythrina stricta, a deciduous tree widely used traditionally in indigenous system of medicine for various ailments such as rheumatism, fever, leprosy, epilepsy etc. The leaves of Erythrina stricta was extracted with ethanol (70%) and used for the evaluation of various in vitro antioxidant assays which includes H - donor activity, nitric oxide scavenging, superoxide anion scavenging, reducing ability, hydroxyl radical, hydrogen peroxide scavenging, total phenolic content, total flavonoid content, total antioxidant activity by thiocyanate and phosphomolybdenum method, metal chelating, $\beta$-carotene bleaching, total peroxy radical assays. The pro-oxidant activity was measured using bleomycin-dependent DNA damage. Ex vivo models like lipid peroxidation and erythrocyte haemolysis were also used to study the antioxidant property of the extract. The various antioxidant activities were compared with suitable standard antioxidants such as ascorbic acid, butylated hydroxyl toluene, $\alpha$-tocopherol, curcumin, quercetin and Trolox. The generation of free radicals viz. $O_2^{{\cdot}-}$, $OH^{\cdot}$, $H_2O_2$, $NO^{\cdot}$ and peroxyl radicals were effectively scavenged by the ethanolic extract of Erythrina stricta. In all the methods, the extract offered strong antioxidant activity in a concentration dependent manner. The total phenolic content, flavonoid content and total antioxidant activity in Erythrina stricta were determined as microgram (g) pyrocatechol, quercetin and $\alpha$-tocopherol equivalent/mg respectively. The extract did not exhibit any prooxidant activity when compared with ascorbic acid. The results obtained in the present study clearly indicates that Erythrina stricta scavenges free radicals and reduces lipid peroxidation, ameliorating the damage imposed by oxidative stress in different disease conditions and serve as a potential source of natural antioxidant.
