Isoflavone Composition within Each Structural Part of Soybean Seeds and Sprouts

  • Isoflavone content in various parts of six soybean cultivars and soybean sprout during germination was analyzed by high performance liquid chromatography. The parts analyzed were seed coat, cotyledon, and axis for seeds and whole sprout, root, hypocotyl, and cotyledon for sprout. Two cultivars, Aga3 which is known to have the smallest seed size and the highest isoflavone content among the Korean soybean cultivars and Pungsannamulkong which is the most widely being used as soy-sprout, were selected for sampling from 1 to 10 days after germination. At the same weight, the order of isoflavone content increased from seed coat to cotyledon to axis. The highest total isoflavone(isoflavone$\times$dry weight) content was observed in the cotyledon and the lowest in the seed coat. The cotyledon of the Aga3 variety had the highest total isoflavone content and the lowest was measured in the Pungsannamulkong variety. The highest total isoflavone content, $10,788{\mu}g/g$, was observed in whole sprouts(cotyledon+hypocotyl+root) on day 7 for Aga3. After day 7, there was a decreasing trend in isoflavone content as the germination period increased. Total isoflavone content in the cotyledon of Aga3 significantly increased after seed germination, whereas the isoflavone content in the cotyledon of Pungsannamulkong decreased. However, total isoflavone content in the root of both varieties increased while isoflavone content in the hypocotyls decreased after seed germination.

66품종 나물콩과 콩나물의 isoflavone 분석 (Analysis of Isoflavones for 66 Varieties of Sprout Beans and Bean Sprouts)

  • Isoflavone함량이 높은 우량나물콩 품종과 우량 콩나물을 선별하기 위해 우리나라의 나물콩 66품종과 이들로 재배한 30종 콩나물의 isoflavone함량을 분석하여 다음과 같은 결과를 얻었다. 66종 나물콩의 총 isoflavone의 평균 함량은 $1209.0{\pm}470\;mg/kg$, 함량범위는 $247{\sim}2,256\;mg/kg$이었고 daidzein과 genistein이 약 1:1의 비율로 함유되어 있었다. 66개 나물콩 품종 중 총 isoflavone 함량이 높은 상위 10개 품종은 KLG10618(2256 mg/kg), KLG11118(2233 mg/kg), KLG10600(1984 mg/kg), KLG10022(1982 mg/kg), KLG10855(1903 mg/kg), 소호콩(1843 mg/kg), KLG11078(1809 mg/kg), KLG11051(1792 mg/kg), KLG10783(1781 mg/kg) 그리고 KLG10873(1762 mg/kg)이었다. 시료 나물콩의 종피색별 총 isoflavone평균 함량은 녹색종에서 가장 높았고 종자크기별 층 isoflavone 평균 함량은 중립종($16{\sim}24\;g$/100립)에서 높은 편이었다. 30종 콩나물의 총 isoflavone 평균 함량은 1898.3 mg/kg이었고, 함량범위는 $768{\sim}3,343\;mg/kg$이었으며, 소호콩(3343 mg/kg), 명주나물콩(2943 mg/kg), KLG11051(2676 mg/kg), KLG11118(2650 mg/kg), KLG10650(2562 mg/kg) 순으로 높았다. 콩에서 콩나물로 재배시 총 isoflavone함량은 평균 1.4배 증가하였다. 콩나물의 총 isoflavone의 함량은 재배일수에 따라 증가하여 5일째에 가장 높은 함량 나타내었다. 콩나물의 부위별 총 isoflavone 함량은 뿌리, 자엽, 배축 순으로 높게 나타났고 daidzein의 함량은 뿌리에서 가장 높았고 genistein의 함량은 자엽에서 가장 높았다.

  • 콩 발아기간 중 몇 가지 면역활성제(자스몬산, 키토산, 베타글루칸 및 게르마늄)와 녹차 및 인삼 추출물이 이소플라본 함량 증대에 미치는 영향에 대하여 알아보았다. 1. 대부분의 처리에서 발아시간이 경과함에 따라 이소플라본 함량이 높아졌는데 면역활성제 중에서는 키토산 처리가 이소플라본 함량 증대에 가장 큰 효과가 있었으며 약초추출물 처리는 효과가 미미하였다. 2. 품종별 이소플라본 함량은 모든 처리에서 대풍2호>우람>풍산나물콩의 순서로 높았으며 이소플라본 함량은 품종과 biotic 물질 처리간 상호작용도 영향을 미치는 것으로 판단된다. 3. 개별 이소플라본 종류별 함량을 분석한 결과 malonyl-glucoside>glucoside>aglycone>acetyl-glucoside 순서로 함량이 높았으며 발아가 진행되면서 aglycone 함량이 증가하는 경향이었다. 따라서 콩 발아기간 중 biotic 물질 처리는 어느 정도 이소플라본 함량을 증대시키며 품종과 물질처리간 상호작용 효과도 있음을 확인할 수 있었다.

다낭성 난소 증후군 환자에서 Isoflavone 섭취에 따른 혈중 생화학적 지표의 변화 (The Effect of Isoflavone Supplementation on Plasma Biochemical Parameters of Women with Polycystic Ovary Syndrome)

  • Objective: This study was performed to evaluate the effect of isoflavone supplementation on hormone levels, lipid profiles and total antioxidant status in patients with polycystic ovary syndrome. Methods: Total 11 women with polycystic ovary syndrome were supplemented daily with 150mg of isoflavone for 6 months. Blood samples were collected 0, 3, and 6 months after supplement of isoflavone for analysis of LH, FSH, E2, testosterone, free testosterone, SHBG levels, serum lipid profiles, and total antioxidant status (TAS). Results: After 6 months isoflavone supplementation, the hormone levels did not change significantly. Serum lipid profiles did not show any significant change in total cholesterol, LDL-cholesterol, triglyceride, lipoprotein(a), and free fatty acid levels. However, there was significant increase in HDL-cholesterol (p<0.05) for 3 months. Total antioxidant status was increased significantly after isoflavone supplementation for 6 months (p<0.05). Conclusion: Isoflavone supplementation showed positive effects on the HDL-cholesterol and total antioxidant status. it is implicated that isoflavone supplementation will may have a effect on cardiovascular disease in patients with polycystic ovary syndrome.

이소플라본 섭취 수준이 주폐경기와 폐경 후 여성의 혈청 내 지질 패턴과 Total Antioxidant Status에 미치는 영향 (Dose Respone Relationship of Isoflavone Supplementation on Plasma Lipid Profiles and Total Antioxidant Status in Perimenopausal and postmenopausal Women)

  • There is growing evidence that soy isoflavones play beneficial role in the prevention of cardiovascular disease. However, current data are not sufficient to determine the effective dose of isoflavone on improving plasma lipid profiles and total antioxidant capacity in Korean menopausal women. In the present study, 24 perimenopausal and 23 postmenopausal women were divided into 3 groups and were given soy isoflavone supplements of 100, 150, 200mg/day for 12weeks. Blood samples were taken before and after 6, 12weeks of soy isoflavone supplementation for analysis of total cholesterol, LDL cholesterol, HDL cholesterol, triglyceride and total antioxidant status(TAS). In the perimenopausal women, serum total cholesterol level decreased significantly in all three groups after 12weeks of isoflavone supplementation(p< 0.05). However, serum LDL cholesterol level significantly lowered only in 150 and 200mg groups, and serum triglyceride level decreased significantly in 200mg group(p<0.05). In the postmenopausal women, the 150mg isoflavone supplementation group demonstrated significant effects on lowering serum total cholesterol, LDL cholesterol levels and raising HDL cholesterol level after 12weeks of isoflavone supplementation(p<0.05). Two hundred mg isoflavone supplementation group had a significantly higher TAS than other groups in both perimenopausal and postmenopausal women. (Korean J Nutrition 34(3) : 322∼329, 2001)

이소플라본과 포도종실유의 보충이 난소절제 흰쥐의 혈중지질 패턴 및 골격강도에 미치는 영향 (The Effect of Isoflavone and/or Grape Seed Oil Supplementation on Blood Lipid Profiles and Bone Strength in Ovariectomized Female Rats)

  • The study was performed to investigate the effects of isoflavone and/or grape seed oil supplementation on serum and liver lipid profiles and bone strength in ovariectomized female rats. Female Spraque-Dawley rats were assigned into 4 groups of ovariectomized (O), isoflavone (0.085 mg/100 g b.w/day) in ovariectomized rats, grape seed oil in ovariectomized rats and isoflavone and/or grape seed oil in ovariectomized rats. After 8 weeks, biochemical profiles of serum, liver and bone were analyzed. Total food intakes, body weight gains and FER (food efficiency ratio) were not statistically significantly different among groups. Total cholesterol, triglyceride and LDL-cholesterol levels in serum were decreased by fed of isoflavone and/or grape seed oils. However, crude lipid and total cholesterol contents in liver were not affect of isoflavone and/or grape seed oil. The hepatic glutathione contents were increased by isoflavone and/or grape seed oil fed. The hepatic glutathione-S-transferase activity in isoflavone and/or grape seed oil supplemented groups were higher than that O group. Bone (scapular and femur bone) dry weight, femur of max weight and bending strength were no significant difference among groups. Our finding suggest that isoflavone/grape seed oils might have potential role for serum lipid profiles improvement and bone strength in vivo.

국내 콩 유전자원의 Isoflavone 함량 변이 (Variation of Isoflavone Contents in Korean Soybean Germplasms)

  • 고 isoflavone 함유 콩 유전자원 선발 및 품종육성의 기초자료로 활용하기 위해 국내 콩 주요품종 43계통 및 보존 유전자원 648계통의 isoflavone 함량을 분석한 결과를 요약하면 다음과 같다. 1. 한국산 콩 주요 43품종의 평균 isoflavone 함량은 daidzein이 $484.4{\pm}223.9{\mu}g/g$, genisteinol $448.0{\pm}156.2{\mu}g/g$, glyciteinol $84.9{\pm}48.7{\mu}g/g$, total isoflavoneol $1,017.3{\pm}390.0{\mu}g/g$을 나타내었다. 2. 공시된 주요 품종의 isoflavone 함량은 석양풋콩이 $2,002{\mu}g/g$으로 가장 높은 함량을 나타내었고, 황금콩이 $277{\mu}g/g$으로 가장 낮은 함량을 나타내었다. 또한 콩 주요 43품종을 용도별로 분류하여 isoflavone 함량을 비교한 결과 풋콩 및 올콩이 $1,438{\pm}493.7{\mu}g/g$을 나타내어 조사된 용도별 콩 분류 중 가장 높은 양상을 나타내었다. 3. 국내 보존 콩 유전자원 648계통을 대상으로 isoflavone 함량을 조사한 결과 IT 180001계통이 $3,479{\mu}g/g$의 함량을 나타내어 가장 높은 양상을 나타내었고, IT 910878 계통은 $82.61{\mu}g/g$의 total isoflavone을 함유하여 가장 낮은 계통으로 조사되어 보존 유전자원간에 큰 함량변이를 나타내었다.

Quantitative Variation of Total Seed Isoflavone and its Compositions in Korean Soybean Cultivars (Glycine max (L.) Merr.)

  • The variation of content of 12 soybean seed isoflavone components was determined in the aglycone, glucoside, malonylglucoside and acetylglucoside groups of 44 Korean soybean cultivars grown in 2016 as well as in 2017. The total isoflavone content of the 44 cultivars averaged at $2935.4{\mu}g/g$ and was in the range of 950.6 to $5226.3{\mu}g/g$ for two years. Malonylglucoside group averaged at $2437.2{\mu}g/g$ with the highest proportion of isoflavone composition (83.0%). Significant differences were observed between cultivars, years and their interactions for both the total isoflavone and each composition group contents (P < 0.0001); however, no year-wise differences were observed for daidzein and genistin. The broad-sense heritability ($h^2$) within the set of 44 Korean soybean cultivars was as high as 0.93 for the total isoflavone content and was in the range of 0.8-0.92 for each composition group of isoflavone except for acetylglucoside. The total isoflavone content in cultivar group for soy-sprout was higher ($3850.4{\mu}g/g$) than that for the other cultivar groups of soy-paste and tofu ($3082.8{\mu}g/g$), black or green soybean cooked with rice ($2345.8{\mu}g/g$), and early maturity group ($1298.6{\mu}g/g$). The total isoflavone content of 'Sowonkong', a soybean cultivar for soy-sprout, was the highest ($5226.3{\mu}g/g$). In the cultivar group for soy-paste and tofu, the average isoflavone contents of 'Daepung', 'Daepung2ho', 'Saegeum', 'Uram', and 'Jinpung' were higher than $4000{\mu}g/g$. With the exception of small seeded cultivars with low isoflavone contents such as 'Sohwang' and 'Socheongja', the seed size and total isoflavone content were significantly negatively correlated in 2016 and 2017, respectively ($r=-0.47^{**}$ and $-0.49^{**}$). The number of days of growth from flowering to maturity did not affect the variations observed in isoflavone content.

  • 본실험은 조생종인 하대두형 및 중국수집종 콩과 중숙${\sim}$만숙종의 국내 육성 종과의 isoflavone 함량변이를 비교하며, 파종기 이동 및 콩의 생육단계별 생육기간에 따른 isoflavone함량의 차이를 분석하고자 수행하였으며, 결과를 요약하면 다음과 같다. 1. 전체 isoflavone함량은 파종기 이동에 관련 없이 국내 수집종이 하대두형 및 중국수집 종에 비하여 놓았으며, 하대 두형콩과 중국수집 종과 비슷한 함량 분포를 나타냈다. 2. 파종기이동에 따른 총 isoflavone 함량은 파종기를 늦게 할수록 하대두형, 중국수집종 및 국내육성종 모두에서 증가하는 경향 이었다. 3. 콩의 개화기${\sim}$성숙기의 기간 및 생육일수가 길어질수록 isoflavone 함량도 높아지는 경향을 나타냈으며, 통계적으로도 부합되었다. 4. 콩의 isoflavone 함량은 입중이 무거울수록 함량이 낮아 졌으며, 상관계수는 4월${\sim}$5월 파종구에서 각각 -0.57, -0.51 및 -0.38로 분석되었다.

Isoflavone 섭취가 흰쥐의 지방대사 및 면역능력에 미치는 영향 (The Effects of Isoflavone on Lipid Metabolism and Immune Responses in SD Rats)

  • This study was performed to investigate the effects of isoflavone consumption on plasma and liver lipid profiles and immune responses in Sprague-Dawley male rats. Experimental animals fed isoflavone at various doses for 4 weeks (0, 1095, 2190, 4380 isoflavone mg/kg diet). Exposure to isoflavone decreased the food consumption and final body weights of rats without decreasing the relative weights of organs, hemoglobin and hematocrit. And the plasma cholesterol and LDL-cholesterol, liver total lipid, cholesterol and triglyceride concentrations were significantly decreased by isoflavone intakes. The absolute and relative weights of thymus were significantly decreased in groups fed isoflavone than in control. Also splenocyte proliferations with Con A or PHA were decreased according to isoflavone consumption in rats, although there was not significant. These results demonstrate that isoflavone intakes significantly improve lipid profiles in plasma and liver. But the effects of isoflavone intakes on immune responses are needed further experiments.
