  • The object of this study was to observe the effects of Hwanggeum-tang (HGT) aqueous extracts on Streptozotocin-induced rat's diabetes and related complications. Three different dosages of HGT extracts were orally administered oncea day for 28 days from 3 weeks after Streptozotocin treatment (60 mg/kg, single intraperitoneally administered). All the rats were checked at 3 weeks after Streptozotocin treatment as follows. Changes on the body weight, blood glucose level, kidney weight, serum BUN and creatinine level, liver weight, serum AST and ALT level, serum LDL, HDL, triglyceride and total cholesterol level were observed with changes on the pancreatic MDA content and GSH content. The results were compared with a potent antioxidant silymarin 100 mg/kg in which the effects on Streptozotocin-induced diabetes and related complications were already confirmed. As results of Streptozotocin-injected diabetes and related complications, dramatical decreases on the body weight, increase of the kidney and liver weight, increase of serum BUN, creatinine, AST, ALT, LDL, triglyceride, total cholesterol level and decreases of serum HDL level were detected in streptozotocin control as compared with intact control. In addition, marked increases of pancreatic MDA content and decreases of GSH content were also detected in streptozotocin control as compared with intact control. However, these diabetes and related complications, and inhibition of antioxidant effects induced by Streptozotocin were inhibited by 28 days continuous treatment of 50, 100 and 200 mg/kg of HGT extracts in the present study. HGT have favorable effects on the diabetes and various diabetic complications. Therefore, more detail mechanism studies should be conducedin future with the efficacy tests of individual herbal composition of HGT and the screening of the biological active compounds in herbs.

  • Extraction of Sedum sarmentosum Bunge by super critical carbon dioxide was operated under $40-50^{\circ}C$and 200-250 atm, thus, yield of extraction was very low as 4 wt%. Rats were administrated with the extract orally once a day for succesive 6 days, followed by treatment with carbon tetrachloride ($CCl_4$) on the sixth day. The activities of aminotransferase, alkaline phosphatase, ${\gamma}$-glutamyl transpeptidase, lactate dehydrogenase and contents of triglyceride, total cholesterol in the extract-pretreated rats were decreased compared to the $CCl_4$controled rats, whereas content of HDL-cholesterol was increased. Especially content of hepatic malondialdehyde (MDA) and atherogenic index (AI) were decreased and HTR was increased in the extract-pretreated rats, and reduced peroxidative liver damage in the $CCl_4$-induced hepatotoxicity rats. In addition, activities of hepatic superoxide dismutase, catalase, glutathione peroxidase in the extract-pretreated rats were significantly decreased compared to the $CCl_4$ controled rats, but the content of glutathione was significantly increased. These results suggest that extract of Sedum sarmentosum Bunge has hepatoprotective effect in the $CCl_4$-intoxicated rats.

  • This study was conducted to investigate the supplementary effects of Suwon 464 developed by Rural Development Administration, which has over two times of dietary fiber content compared with normal rice (Ilpum), on lipid metabolism in diabetic mice. We supplied 5 kinds of experimental diets (com starch diet as a control (CO), Ilpum polished rice diet (IP), Ilpum brown rice diet (IB), polished rice diet (SP) and brown rice diet (SB) of Suwon 464) to diabetic mice for 8 weeks, after analyzing dietary fiber contents of 5 experimental diets. Diet intake, body weight, organ weights, and lipids levels of serum, liver and feces were measured. The dietary fiber contents in CO, IP, IB, SP, and SB diets were 1.0, 1.2, l.4, 1.4, and 2.0% respectively. Body weight and liver and epididymal fat pad weights were lower in SB group than the other groups though there was no significant difference in diet intake among experimental groups. The concentrations of serum triglyceride was lower in SP and SB groups than CO and IP groups. The levels of hepatic total lipid and total cholesterol were significantly lower in SP and SB groups than CO group, and the level of hepatic triglyceride was lower in IB, SP and SB groups than CO group. The levels of total lipid and triglyceride excreted in feces were higher in IB, SP and SB, and the level of total cholesterol in feces was higher in SP and SB groups than CO group. These results suggested that the high dietary fiber rice (Suwon 464) decrease the triglyceride or total cholesterol concentrations of serum and liver by increasing of fecal lipid excretion in diabetic mice. • In order to investigate experimental effects of Yangkyuksanwha-Tang, which is the typical prescription for diseased SOYANGIN, on diabetes rats, the observation and measurement was made on the content of volume of blood glucose, total cholesterol, triglyceride, phospholipid, total protein, electrolyte($K^+$, $Na^+$) in serum, by dosing the cooked Y.S.T. to diabetic rats, induced by streptozotocin and following result was obtained. 1. The volume of blood glucose, in the diabetes rats, was decreased significantly, and remarkable decrease of blood glucose was shown in the Y.S.T. (12.5ml/kg) dosed group, after 4weeks of experiment. 2. The volume of total cholesterol of blood serum was decreased notably, especially remarkable reduce of total cholesterol was shown in the Y.S.T. (12.5ml/kg) dosed group after a week. 3. The volume of triglyceride in the blood serum of diabetic rats, induced by the streptozotocin was decreased after 4 weeks, and remarkable decrease was shown in the Y.S.T. (12.5ml/kg) dosed group after 2 weeks of experiment. 4. The volume of the phospholipid in the diabetes rats, was decreased significantly in the Y.S.T. (12.5ml/kg) dosed group after 2 weeks of experiment. 5. The volume of total protein of diabetes rats, was increased in all group, and the remarkable increase was shown in the Y.S.T. (12.5ml/kg) dosed group after 4 weeks. 6. The volume of the sodium in the Y.S.T. (12.5ml/kg) dosed group, was notable in each Y.S.T. dosed group (1.25ml/kg, 12.5ml/kg) after a week experiment. According to above results, It is expected that Y.S.T. applied to SOYANGIN, can be used for medical treatment of diabetes with effect. But because the effect can be somewhat different depending upon dosage, appropriate operation on clinic is needed, for the best curing effect.

  • Objective: An experiment was conducted to evaluate the effect of different levels of crude protein (CP) and two rearing systems (cage and floor), on blood parameters and digestive and reproductive organ development of brown laying hens. Methods: A total of 400 Hisex Brown laying hens between 30 and 45 weeks of age were distributed in a completely randomized design and a 2×4 factorial arrangement, with main effects including two rearing systems (cage and floor) and levels of CP (140, 150, 160, and 180 g/kg), in a total of eight treatments and five replicates of 10 birds each with initial body weight of 1,877 g (laying hen in cage) and 1,866 g (laying hens in floor). The parameters evaluated were plasma total protein, albumin, uric acid, total cholesterol, relative weights of oviduct, abdominal fat, liver, gizzard, crest and dewlap, length of small intestine and oviduct. Results: The blood parameters were similar in birds reared in cage and floor systems. The birds reared on the floor showed greater small intestine and oviduct weight (%) and lower liver and pancreas weight (%). A significant interaction was observed between factors for the relative gizzard, crest and dewlap weight, serum protein, uric acid, and total cholesterol (p<0.05). The diets with 140 g/kg CP resulted in lower serum protein and lower cholesterol in birds reared in floor system, while birds reared in cage system showed no effect of CP on both parameters. Birds reared in cage and fed with 140 and 150 g/kg CP presented lower uric acid. The group of birds reared in floor system fed 180 g/kg had greater uric acid. Conclusion: The dietary protein level can be reduced up to 140 g/kg for Hisex Brown hens (30 to 45 weeks of age) without an important effect on metabolic profile and organ development in both rearing systems.

  • The ethanol extraction yield of Liriope platyphylla(Lp) was about 30.7% by extract apparatus. This study was done to investigate the carbohydrate metabolism related enzyme activities effects of Lp in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) were significantly decreaed in Lp treated group compared to the those of STZ-control group, also content of Total cholesterol was decreased. High density lipoprotein (HDL)-cholesterol was increased in Lp treated group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in Lp treated group. Also the activity of glucokinase(Gk) was increaed in Lp treated group. The content of hepatic glycogen was significantly increaed in Lp treated group. These results indicated that ethanol extract of Lp would have antidiabetic effect in STZ-induced diabetic rats.

  • This study explored the effects of dietary calcium level and Hijikia fusiforme supplementation on bone indices and serum lipid levels using 36 female Sprague-Dawley rats as a model. Rats received low Ca diet for 3 weeks after ovariectomy. The rats were then divided into six dietary groups and fed low (0.1% Ca), normal (0.5% Ca) and high (1.5% Ca) Ca diets (CaL, CaN, CaH) and low, normal, high Ca diets with Hijikia fusiforme supplementation (CaLH, CaNH, CaHH) for 3 weeks. After each experimental periods, 24 hour urine and/or blood samples, left and right femurs were collected for analysis. Serum Ca concentration showed no significant difference by dietary Ca levels and Hijikia fusiforme supplementation. Alkaline phosphatase activity was significantly higher in normal and high Ca group compared to low Ca group. Serum total cholesterol, triglyceride and total lipid were not significantly different among groups. HDL-cholesterol showed no significant difference by Hijikia fusiforme supplementation. However, the normal and high Ca groups showed significantly higher HDL-cholesterol compared to the low Ca group. Urinary hydroxyproline and hydroxyproline/creatinine ratio were not significantly different among groups. The wet weight of the femur was significantly higher in low Ca group compared to normal or high Ca group. The dry weight, wet weight/body weight, length and breaking force of the femur were not significantly different among groups. Ash contents/wet weight of the femur was significantly increased as dietary Ca levels up and significantly higher in Hijikia fusiforme supplementation groups. The Ca content of the femur were significantly higher in the normal and high Ca groups than the low Ca group. However, there was no significant difference in Ca content by Hijikia fusiforme supplementation.

  • The effects of stearic(18 : 0, SA), oleic(18 : 1 cis, OA) and elaidic acid(18 : 1 trans, EA) on the cell growth, contents of cellular lipids, and the fatty acid composition of cellular and medium lipids in Hep-G$_2$cells were evaluated. The cells were incubated in serum-free medium containing 25, 50, 100 and 200$\mu$M of a fatty acid combined with albumin for 2 days. The fatty acid concentration up to 100$\mu$M showed the normal growth, but the cell growth decreased in the presence of 200$\mu$M fatty acid. The treatment of cells with 100$\mu$M of a fatty acid for two days significantly(p<0.05) increased the cellular triglyceride(TG) content in all fatty acid groups compared to control, but TG contents was not significantly different among all treatment group, but total cholesterol(TC) was the highest level in EA group. The level of free cholesterol(FC) and cholesteryl ester(CE) was similar to those of TC in all fatty acid treated group. The cellular phospholipid(PL) contents were similar between the control and all fatty acid groups. The treatment of cells with SA has no notable effects on the fatty acid composition of TG, CE and PL. The OA treatment caused significant increases in CE(51.2%) and PL(29.8%), but not in TG. The EA treatment resulted in 10.1, 10.7 and 7.8% of $C_{18:1\;trans}$ content in cellular TG, CE, and PL. The TG, CE and PL of medium were relatively similar between SA and OA groups. In EA treated group, TG, CE and PL of medium contained 17.0%, 0.7% and 5.6% of $C_{18:1\;trans}$, respectively.
