Production of a Fermented Korean Pear Puree using a New Strain Leuconostoc mesenteroides KACC 91495P Isolated from Kimchi (김치에서 신규 Leuconostoc mesenteroides KACC 91495P 균주의 분리 및 이를 이용한 배 발효물의 제조)

  • A lactic acid bacterial strain showing fast growth and high acid production in Korean pear puree was isolated from Kimchi. This strain was analyzed by API 50 CHL kit and 16S rRNA sequencing analysis and identified as Leuconostoc (Ln.) mesenteroides KACC 91495P. Korean pear puree was fermented using Ln. mesenteroides KACC 91495P strain at $30^{\circ}C$ for 18 h. The changes of pH, titratable acidity and viable cell number during fermentation were investigated. The pH and titratable acidity were reached to pH 3.86 and 1.09% after 18 h fermentation, respectively. The viable cell population of Ln. mesenteroides KACC 91495P was rapidly increased to $2.0{\times}10^9\;CFU/g$ during the 9 h of cultivation. The contents of lactic acid, acetic acid and malic acid were determined to be 0.213, 0.259, and 0.217% after 18 h fermentation, respectively. The content of polyphenolic compounds, known as antioxidants, in pear puree were enhanced by Ln. mesenteroides KACC 91495P cultivation. The level of total polyphenolic compounds was increased to around 140% of initial concentration. When the fermented pear puree was kept at $4^{\circ}C$, pH, titratable acidity and number of viable cells population were nearly maintained for 13 days.

Effects of Organic fertilizer Application on Growth and Medicinal ingredients of Platycodon grandiflorum Radix (유기질 비료시용에 따른 도라지의 생육 및 약용성분에 미치는 효과)

  • This study was conducted to elucidate the effect of organic fertilizer on growth characteristics, saponin contents and antioxident activity of Platycodin grandiflorum Ridix roots for organic farming. As basal fertilizers, chemical fertilizer, mixed organic fertilizer, bacterial cultures and fermented oil cake and decomposed manure were treated based on 3 kg/10a level before transplanting Platycodin grandiflorum Ridix. In root length, when chemical fertilizer plot and mixed organic fertilizer plot were treated, root length was recorded the highest scores (25.3 and 24.0 cm) Root width was recorded the highest score (26.6 cm) in chemical fertilizer plot. The number of fine-roots was the highest in mixed organic fertilizer plot and chemical fertilizer plot (20.0 and 17.0), respectively. Fresh weight, which affects directly yield, was the highest in organic fertilizer plot (55.7 g/plant). The content of Platycodin D was shown to 327.4~373.8 mg/100 g, the highest values were observed in organic fertilizer plot. The total polyphenol and flavonoid contents were recorded the highest scores (15.5 and 15.3 mg/g, respectively) in organic fertilizer plot. In this study we confirmed that application of organic fertilizer was effective to increase yield and pharmacological effect through increase of the number of fine-roots with high saponin contents.

  • Population changes of serveral bacterial groups and chemical changes of nutrients applied were compared between soils suspensed with pentachlorophenol(PCP) and amended with different nutrients. The number of total bacteria and Gram-negative bacteria were dramatically increased for 1st week, after remained at its maximum level for a short time, decreased gradually in all treatments for 5th week, the rate of decrease were faster at addition of only PCP than at addition of nutrients and PCP. While, the addition of nutrients in the soil suspension containing PCP suppressed the number of PCP-degrading bacteria. The dissipation of PCP were faster at single addition of PCP than at the combined addition of nutrients with PCP. The nutrients added with PCP were rapidly degraded for 1st week, among the nutrients the rate of degration of glucose was faster than that of glutamic acid or glycine. pH values in the suspension showed which addition of nutrients with PCP caused its changes in comparsion with addition of only PCP where changes was slightly.

Ultrafiltration for Quality Improvement of Wine (한외여과공정을 이용한 포도주의 품질개선)

  • Physicochemical and microbiological changes of grape wine fermented and aged at 25 and $15^{\circ}C$ for 2 and 14 weeks, respectively, were investigated. Viable bacterial cell number, $3.3{\times}10^2\;CFU/mL$ at the beginning of fermentation, increased to $2.3{\times}10^6\;CFU/mL$ after 2 weeks, then decreased to $1.9{\times}10^3\;CFU/mL$ after 14 weeks. Viable yeast cell number increased from $2.8{\times}10^2\;to\;2.2{\times}10^7\;CFU/mL$ during fermentation, then decreased to $1.6{\times}10^4\;CFU/mL$ after aging. Turbidity, pH, total sugar content, reducing sugar content, and solid content of grape wine decreased during fermentation, whereas acidity and alcohol content increased to 0.64 and 8.4%, respectively. Most physicochemical properties did not change significantly during aging. When grape wine was filtered through $0.45-{\mu}m$ nitrocellulose membrane, followed by various ultrafiltration membranes with different molecular weight cut-off values, Biomax 100K membrane with $100\;L/m^2/hr$ (LMH) of initial flux was chosen for ultrafiltration process. These membrane filtration treatments resulted in complete removal of microorganisms and decreases in turbidity, reducing sugar, and solid content. Physicochemical properties of wine did not change, and no microorganisms were found during storage at $30^{\circ}C$ for 12 weeks.

Major Microbial Composition and Its Correlation to the Taste of Jeonju Kimchi (전주 김치의 적숙기 주요 미생물 조성과 맛의 상관성)

  • The 21 Kimchi samples collected by a contest for the best Kimchi homemaker in Jeonju were analysed to find major microbial composition and the correlation between sensory scores and the microbial properties, which include total microbial numbers, dominant strain number, subdominant strain number, yeast strain numbers, and floral diversity. The most common microbial composition of the Kimchi samples was Leuconostoc mesenteroides as dominant strain, and Lactobacillus sakei as subdominant strain, the pattern of which was also observed in all three Kimchi samples with the highest sensory scores, but not in all three Kimchi samples with the lowest sensory scores. However none of the other examined properties showed any significant correlation to sensory scores. This result means that the taste of fermented Kimchi depends mainly on the composition of bacterial species and that of Leuconostoc mesenteroides as dominant strain and Lactobacillus sakei as subdominant strain is the best for good taste. Yeast strains were found in the range of $3.8{\sim}4.6log\;CFU/g$ from 8 Kimchi samples and all the strains were identified to be Saccharomyces servazzii. However, fungi were not detected from any of the Kimchi. samples.

  • A series of field and laboratory experimentes were conducted to find out the effects of kinds of crops residues and the different amount of fertilizer to the microbial interaction, chemical properties, plant growth and their interaction under continous cultivation of hot pepper, soybean, sesame and upland rice. The results obtained were summerized follows : 1. Total number of bacteria and actinomycetes were enhanced by cultivation of upland rice and soybean while no appreciable effects were obtained by the cultivation of hot pepper and sesame. 2. The number of bacteria, actinomycetes and fungi were increased by return of crop residues when the cultivation of hot pepper, soybean, sesame and upland rice. Specially, actinomycetes was remarkably increased by upland rice cultivation. 3. Increased amount of fertilizer were remarkably affected to decrease of number of soil microorganisms. Specially, actinomycetes succession was appearently affects while plant growing time. 4. The number of identified soil bacterial species were obtained high in order of Bacillus sp.>Rhizobium sp.>Agrobacterium sp.>Pseudomonas sp. The number of Gram positive bacteria were superior than that of Gram negative bateria. 5. Interaction between microbial succession and crops cultivation, the number of Bacillus sp. was increased in hot pepper, Rhizobium sp. was in soybean, and sesame, and Agrobacterium sp. were increased in soybean, respectively. 6. Survival and habitate of soil microorganisms were differ with kinds of crop, application of residue and fertilizers. Most high number of Bacillus sp. Rhizobium sp. and Pseudomonas sp. were obtained on the rhizoplane and rhizosphere while Agrobacterium sp. and Fusarium sp. were high in rhizosphere. 7. Factors in relation to change of soil microbial population was obtained high in order of climates>crops>organic>matter>fertilizer.

  • Extensive studies on the commercial fermented milks, which had been distributed over four months from July to October in 1978, were carried out for their microbial characterization including an investigation on the variations of bacterial populations under various storage conditions. 1. Total number of viable cells in the products were in between $5{\times}10^7$ cells/ml and $73{\times}10^7$ cells/ml, and $62{\times}10^7$ cells/ml, however no coliform bacteria were detected in the products. 2. Acidities of the products were not very high but fit in the standard as appeared to be in between 0.4869% and 0.6689%. 3. The acidities of the products showed little changes when stored at $5^{\circ}C$, but rises in 22.55% at $20^{\circ}C$ and in 117.66% at $30^{\circ}C$. 4. Total viable counts weren't varied much upto 96 hours when stored at $5^{\circ}C$, but increased during first 24 hours then decreased gradually at the higher temperatures. 5. Viable counts of lactic acid bacteria were decreased markedly with ascending the storage temperature, showing minimal variations when stored at $5^{\circ}C$.

Quantitative and Qualitative Studies of Commensal Bacterial Flora of Clam, Ruditapes philippinarum in Hadong Area (하동 지역에 서식하는 바지락의 미생물총 분포에 관한 정량 및 정성적 분석)

  • Characteristics and distribution of the natural commensal flora in the surrounding environment and tissues of clam in Hadong area were studied under varying conditions of growth media and incubation temperatures. Total numbers of bacteria present in intestinal tract, gill, body fluid and surrounding mud were found to be not influenced by the used BHIA, STA and SNA media. Although the growth rate of bacteria at the condition of $15^{\circ}C$ incubation temperature was slower than that of $25^{\circ}C$ and $35^{\circ}C$, it showed the highest number of total bacteria compared with other two different conditions of incubation temperature. Interestingly, the proportion of bacteria able to form colony on several selective media was higher in replica analysis from nutrient media to selective media than that in direct smearing from samples. The generic diversity of bacteria isolated from the tissues and analyzed by API 20E and API 20NE kit showed similar pattern with each other and distinct from that of environment. The distribution of bacteria in the surrounding mud or mantle fluid of clam indicated a high diversity comparable to that found for the gill or intestinal tract microflora, with Pseudomonas being the prevalent group. It implies that the tissues of clam may probide a selective habitat for a commensal microflora.

Antioxidant Properties of Various Microorganisms Isolated from Arctic Lichen Stereocaulon spp. (북극 지의류 Stereocaulon spp로부터 분리한 여러 미생물의 항산화 성질)

  • Lichens are symbiotic organisms composed of fungi, algae, or cyanobacteria which are able to survive in extreme environmental conditions ranging from deserts to polar areas. Some lichen symbionts produce a wide range of secondary metabolites that have many biological activities such as antibacterial, antifungal, antiviral, antitumor, antioxidant and anti-inflammatory etc. Among the symbionts of lichens, of the bacterial communities of lichen symbionts little is known. In this study, we isolated 4 microbial species from the Arctic lichen Stereocaulon spp. and evaluated their antioxidant properties using 1,1-diphenyl-2-picryl-hydrazyl assay as well as 2,2'-azino-bis(3-ethyl benzothiazoline-6-sulphonic acid) assay. Total phenolic contents and total flavonoid contents were also measured. A potent radical scavenging activity was detected in a number of the lichen extracts. Among the 4 species tested in this study, the ethyl acetate extract of Bosea vestrisii 36546(T) exhibited the strongest free radical scavenging activity, with an inhibition rate of 86.8% in DPPH and 75.2% in ABTS assays. Overall, these results suggest that lichen-bacteria could be a potential source of natural antioxidants.

  • For efficient and economical production of Btrc,illus tlz~rr.ingi~r1~sstirsa in NT0423 as amicrobial-control agents, a new culture medium and culture condition were developed. Total cell number in all SW media showed similar values, hut SW32 lnediilm wasthe most efficient in the development of spore, which amo~~ntetod 3.7 x 10XC FUImI. Also. at the pHranging frorn 6.2 to 7.3 in the mediiun~ no ad\:erse effect was not made in the culture of B. thur-ingicnsisstrain NT0423.
