INFLUENCE OF AMINO ACID SUPPLEMENTS TO A STRAW-MAIZE-BASED UREA DIET ON DUODENAL DIGESTA FLOW AND DIGESTION IN SHEEP

  • Amino acid (AA) substituted diets had no influence on rumen levels of total volatile fatty acids (VFA), ammonia and ${\alpha}$-amino-N, but tended to increase molar proportions of isovalerate and counts of total viable AA utilizing and celluloytic bacteria in the rumen as compared with the control urea diet. The AA diets did not affect daily flow to the duodenum of dry matter (DM), organic mater (OM) and acid detergent fibre (ADF), and rumen digestibility of these nutrients. However, the AA diets, in particular the 10 essential AA (EAA) diet improved total digestibility of DM, OM and ADF by decreasing faecal output of these fractions. Although N flow to the duodenum and N retention were not affected with the dietary treatments, duodenal bacterial flow appeared to increase by the AA diets when it was estimated by means of 2,6-diaminopimelic acid (DAP) and nucleic acid-purine bases (PB) as markers. The results suggest that AA supplements to a urea diet could improve feed utilization by stimulating microbial activity and proliferation in the rumen but and increased microbial activity per se is not necessarily associated with improvement of feed conversion.

국내 보존음료수(생수)의 성분에 관한 연구 (A Study on the Water Quality of Domestic Mineral Water (Saeng Su))

  • This study was performed to analyze for the mineral water (Saeng Su) from 12 pure beverage Co. products, which was obtained during March to May in 1988. The results were as follows: 1. The analytical results of contaminant materials in all the samples were suitable for drinking water standard. The mean value of NH$_{3}$-N, NO$_{3}$-N, Cl-and KMnO$_{4}$ consumption were ND, 49mg/l, 1.08mg/l and 8.14mg/l, respectively. 2. Total bacterial counts were 3/cc in average and Coliforms was negative in all samples. 3. The analytical results of Heavy metals were not detected or detected in a small amount, and the contents were different from each company. The CN, Hg, P were not detected in all the samples and the mean value of Cu, Fe, Mn were 0.0087mg/l, 0.0303mg/l, 0.018mg/l, respectively and it's contents were low. 4. The mean value of Pb, Zn, Cr$^{+6}$, As and Cd were 0.016mg/l, 0.015mg/l, 0.0025mg/l. 0.0059mg/l and 0.0011mg/l respectively.

Microbial Dynamics of Commercial Makgeolli Depending on the Storage Temperature

  • Market fresh makgeolli was stored at different temperatures of $4^{\circ}C$ and $25^{\circ}C$ to assess the change of the microbial diversity according to the storage temperature and period. Yeast counts increased until day 3 of storage and decreased thereafter. General and lactic acid bacterial counts continuously increased during storage. The data indicated that the control of growth of microorganisms, particularly general bacteria and lactic acid bacteria (LAB), is essential. Total acid levels started to decrease in the makgeolli stored at $4^{\circ}C$, and increased from day 6 of storage in the makgeolli stored at $25^{\circ}C$. The increase of total acid in the non-refrigerated condition greatly affected the quality of makgeolli. In both the fresh makgeolli samples stored at $4^{\circ}C$ and $25^{\circ}C$, yeast (Saccharomyces cerevisiae) and molds (Aspergillus tubingensis, Candida glaebosa, and Aspergillus niger) were noted. Denaturing gradient gel electrophoresis (DGGE) band patterns were almost constant regardless of the storage period. As for bacteria, Lactobacillus crustorum, L. brevis, and Microlaena stipoides were found in the makgeolli stored at $4^{\circ}C$, and L. crustorum, Lactobacillus sp., L. plantarum, L. brevis, L. rhamnosus, and L. similis were found in the makgeolli stored at $25^{\circ}C$. In particular, in the makgeolli stored at $25^{\circ}C$, L. crustorum and L. plantarum presented dark bands and were identified as the primary microorganisms that affected spoilage of fresh makgeolli.

Assessment of the potential of algae phycobiliprotein nanoliposome for extending the shelf life of common carp burgers during refrigerated storage

  • This study is focused on the effect of phycobiliprotein extraction of Gracilaria on the quality of common carp burgers, and the application of nanoliposomes containing pigment in the improvement of its antimicrobial and antioxidant activity of burgers during refrigerated storage in 18 days. Burgers were incorporated with phycobiliprotein and liposomal phycobiliprotein (2.5% and 5% w/w), and their chemical and microbial changes in terms of pH, peroxide value (PV), thiobarbituric acid (TBA), total volatile basic nitrogen (TVB-N), total viable counts (TVC), psychrotrophic bacterial counts (PTC), and sensory characteristics were evaluated. Results presented a nanoliposome size of about 515.5 nm with capable encapsulation efficiency (83.98%). Our results showed non-encapsulated phycobiliprotein could delay the deterioration of common carp burgers, as a reduction in PV, TBA, and TVB-N, TVC, and PTC values in burgers treated with free and nano encapsulated phycobiliprotein. Moreover, the potential of phycobiliprotein was improved when it was encapsulated into chitosan coated liposomes. Burgers treated with 5% nanoliposomes displayed the lowest amount of lipid oxidation and microbial deterioration in comparison to others during storage. According to chemical, microbial and sensory evaluation, the shelf life of common carp burgers was increased in samples treated with encapsulated phycobiliprotein at 2.5% and 5%, as compared to the control (p ≤ 0.05).

Pediocin 처리가 생면의 저장 중 pH와 미생물에 미치는 영향 (Effects of Pediocin Treatment on the Microbial Quality of Wet Noodles during Storage)

  • Pediocin 처리에 따른 생면의 저장성 연장효과를 얻기 위하여 pediocin을 처리한 후 생면의 미생물의 변화를 알아 보았다. pH의 변화는 pediocin 처리 농도가 높을수록 낮은 경향을 보였으며, 저장 후 2일에서 약간 증가 하다가 이후부터 점차 감소함을 보였다. 저장기간이 경과함에 따른 일반 세균수의 변화는 점차 균수가 증가하는 경향이었으며, 1,000 ppm 처리구의 경우 저장 1일 후 일시적으로 균수의 감소되어 살균효과를 관찰할 수 있었으나 이후 균수는 저장 4일까지 서서히 증가하는 양상을 보였다. 대장균군의 변화는 1,000 ppm 처리구의 경우 저장 4일까지 대장균군의 검출이 없었으나, 대조구의 경우 저장 후 2일에서 대장균군이 검출되었다. 효모와 곰팡이군의 변화는 pediocin의 처리농도가 높을수록 균의 성장을 억제하였으며, 대조구의 경우 균수가 빠르게 증가하여 저장 후 4일에서는 5 log CFU/mL 이상의 균수를 보였다. 따라서 pediocin 처리는 생면의 저장성 연장에 효과가 있음을 확인하였다.

Long-Term Starvation Induces the Viable-but-Nonculturable Condition in Lactobacillus crispatus KLB46

  • In a previous study, we have isolated a number of lactobacilli from Korean women, and one of them (KLB46) was identified as Lactobacillus crispatus by 16S rRNA gene sequencing. For the ecological treatment of bacterial vaginosis (BV) cell suspension of L. crispatus KLB46 was instillated into BV patients. L. crispatus KLB46 was found to persist for several days in cell suspension with no nutrients. In this study, in order to assess the influence of starvation on physiological activity, we compared the viability and culturability of KLB46 following suspension in various buffer solutions. A pair of in situ fluorescent dye was used to assess viability (i.e. membrane integrity) and the culturability was examined by plate count assay. A rapid epifluorescence staining method using the LIVE/DEAD Bacterial Viability Kit $(BacLight^{TM})$ was applied to estimate both viable and total counts of bacteria in cell suspension. $BacLight^{TM}$ is composed of two nucleic acid-binding stains ($SYTO\;9^{TM}$ and propidium iodide). $SYTO\;9^{TM}$ penetrates all bacterial membranes and stains the cells green while propidium iodide only penetrates cells with damaged membranes, therefore the combination of the two stains produces red fluorescing cells. Optimal staining conditions for $BacLight^{TM}$ were found to be with 0.0835M $SYTO\;9^{TM}$ and 0.05M propidium iodide for 15 min incubation at room temperature in dark. When cells were microscopically examined during 140 hours of starvation, the culturability decreased markedly while the viability remained relatively constant, which suggests that large fraction of KLB46 cells became viable but non-culturable (VBNC) upon starvation.

서울에서 시판중인 굴의 세균학상 오염과 분리된 비브리오속 균의 항생제 내성 양상 (Antibiotic Resistance of Vibrio spp. and Bacterial Contamination of Commercial Oysters in Seoul, Korea)

  • The purpose of this investigation was to measure the level of bacterial contamination of oysters, and observe antibiotic resistance pattern of Vibro spp. found in oysters. For this experiment, 100 oysters were collected from 100 markets around seoul area from October, 2004 to January, 2005. Bacterial contaminations of the oysters were confirmed by performing the method of standard plate count and desoxycholate lactose agar plate. Total plate counts were $2.7\times$$10^{2}$$\sim$$1.2\times$$10^{5}$ cfu/g. Coliform group were $1.0\times$$1.0^{1}$$\sim$$3.2\times$$10^{5}$ cfu/g. Results have shown that Vibrio spp. was present in $64\%$ of the 100 sampls. Antibiotic resistance pattern of Vibrio spp. showed that $MIC_{50}$ of chloramphenicol was 0.2 g/mL, $MIC_{90}$ was 25 $\mu$g/mL, and $MIC_{50}$ of tetracycline was < 0.05 $\mu$g/mL, $MIC_{90}$ was 25 $\mu$g/mL, and $MIC_{50}$ of ciprofloxacin was 0.01 g/mL, $MIC_{90}$ was 10> $\mu$g/mL. Three of the six strains were identified as Vibrio alginolyricus.

자리돔의 식품위생학적 연구 (STUDIES ON SANITARY QUALITY OF JARHI-DOM CHROMIS NOTATUS)

  • 1975년 5월부터 10월까지 자리돔의 식품위생학적인 안전성을 검토하기 위하여 자리돔의 서식해수, 어획직후의 자리돔, 시판자리돔, 자리물회 및 자리무침 회의 일반세포수, coliform group MPN 및 parahaemolyticus의 유무를 조사한 결과를 요약하면 다음과 같다, 1) 서식해수의 일반세균수는 평균 $1.7\times10^2/ml$ 정도였고 coliform group MPN은 4/ml이하였다. 2) 어획직후의 자리돔은 일반세균수가 평균 $3.2\times10^3/g$였고 coliform group MPN은 300g이하였다. 3) 시판되고 있는 자리돔의 일반세균수는 8월 중순경에는 평균 $1.1\times10^6/g$ 9월중순경에는 $2.2\times10^6/g$ 였다. 4) 자리무침회 및 자리물회의 일반세균수는 각각 $1.1\times10^7/g,\;4.9\times10^5/g\~1.7\times10^7/g$였다. 5) 자리무침회와 자리물회에서는 V.purahamolyticus 균이 검출 되지 않았으나 서식해수와 자리돔에서는 V. parahaemolyticus 균이 검출 되었는데 식중독을 일으킬수 있는 균량은 아니였다. 5) 자리돔 및 각종 자리돔회의 일반세균수, coliform group MPN 및 V. parahaemolyticus는 8월과 9월에 오염도가 가장 높았다. 본 연구의 지도와 많은 조언을 주신 부산수산대학 식품공학료 최위경, 이강호, 이응호, 박사들과 실험을 도와 준 제주대거 이창환, 박철우, 이진영 제군에게 감사를 드리는 바이다.

Role of Organic Spices in the Preservation of Traditionally Fermented Kunun-zaki

  • Kunun-zaki, produced by submerged fermentation of a combination of millet and sorghum, is a popular beverage in Northern Nigeria. Owing to the nature of the process involved in its production, kunun-zaki is highly susceptible to contamination by food spoilage microorganisms, leading to inconsistent quality and short shelf-life. In this study, we investigated various food spices, including cinnamon, garlic, and nutmeg, as potential preservatives that could be used to extend kunun-zaki shelf-life. Kunun-zaki varieties were fermented with each of these spices mentioned above and subjected to bacterial, nutritional, sensory, and quality maintenance assessments (using a twelve-member sensory panel to evaluate the organoleptic properties of kunun-zaki). Bacterial counts in the final products ranged between 105-7 CFU/ml. We identified two bacterial genera, Weissella and Enterococcus, based on partial 16S rRNA gene amplicon sequencing. Three amino acids, namely leucine, aspartate, and glutamate, were abundant in all kunun-zaki varieties, while the total essential amino acid content was above 39%, suggesting that kunun-zaki could potentially be considered as a protein-rich food source both for infants and adults. The kunun-zaki products were also rich in carbohydrates, crude proteins, ash, crude fiber, and fat, with contents estimated as 81-84, 8-11, 0.8-4.0, 2.9-3.58, and 5.1-6.3%, respectively. However, this nutritional content depreciated rapidly after 24 h of storage, except for kunun-zaki fermented with garlic, which its crude protein and fat content was maintained for up to 48 h. Our results revealed that organic spices increased the nutritional content of the kunun-zaki varieties and could be potentially be used as natural preservatives for enhancing the kunun-zaki shelf-life. However, garlic might be considered a better alternative based on our preliminary investigation. The presence of the isolated microorganisms in the analyzed kunun-zaki samples should be highlighted to raise awareness on the possible health hazards that could arise from poor handling and processing techniques.

Ensiling Characteristics and the In situ Nutrient Degradability of a By-product Feed-based Silage

  • This study was conducted to evaluate the ensiling characteristics and the in situ degradability of a by-product feed (BF)-based silage. Before ensilation, the BF-based mixture was composed of 50% spent mushroom substrate, 21% recycled poultry bedding, 15% ryegrass straw, 10.8% rice bran, 2% molasses, 0.6% bentonite, and 0.6% microbial inoculant on a wet basis and ensiled for up to 4 weeks. The BF-based silage contained on average 39.3% moisture, 13.4% crude protein (CP), and 52.2% neutral detergent fiber (NDF), 49% total digestible nutrient, and 37.8% physically effective $NDF_{1.18}$ on a dry matter (DM) basis. Ensiling the BF-based silage for up to 4 weeks affected (p<0.01) the chemical composition to a small extent, increased (p<0.05) the lactic acid and $NH_3$-N content, and decreased (p<0.05) both the total bacterial and lactic acid bacterial counts from $10^9$ to $10^8$ cfu/g when compared to that before ensiling. These parameters indicated that the silage was fermented and stored well during the 4-week ensiling period. Compared with rice or ryegrass straws, the BF-based silage had a higher (p<0.05) water-soluble and filterable fraction, a lower insoluble degradable DM and CP fraction (p<0.05), a lower digestible NDF (p<0.05) fraction, a higher (p<0.05) DM and CP disappearance and degradability rate, and a lower (p<0.05) NDF disappearance and degradability rate. These results indicated that cheap, good-quality BF-based roughage could be produced by ensiling SMS, RPB, rice bran, and a minimal amount of straw.
