포장된 전통 된장 및 고추장의 저장 중 $CO_2$ 발생과 특성변화 (Carbon Dioxide Production and Quality Changes in Korean Fermented Soybean Paste and Hot Pepper-Soybean Paste)

  • One hundred fifty grams of Korean fermented soybean paste and hot pepper-soybean paste were packaged in glass jar of 232 mL and Sotred at 5, 13, 22 and 30℃. During the storage, the changes in their microbial flora and quality attributes were monitored. Carbon dioxide production rate from the stored pastes were also determined from initial change of CO₂concentration in headspace of the pack. Hot pepper-soybean pate showed much higher CO₂ production rate higher dependence of CO₂ production on temperature compared to soybean paste. Total aerobic bacteria count and lactic acid bacteria count did not change significantly through the storage. Yeast count in soybean paste decreased slowly after initial uprise while that of hot pepper-soybean paste steadily decreased. Surface color of hot pepper paste changed to dark red with slight decrease in 'L' value and slight increase in 'a' and 'b' values, whereas any significant color change was not observed in soy paste. Titratable acidity increased with time with higher increase in soybean paste, but pH stayed at constant level for both pastes. All the rates of quality change were higher with higher temperature. Pressure buildup due to CO₂ production needs to be considered first in designing the packages of the fermented pastes before their color changes and other chemical quality changes.

서부 경남지역 토마토 농장에서의 위생 미생물의 분포 (Sanitary Microbial Distribution at the Tomato Farms in Western Gyeongnam)

  • This study were conducted to investigate the microbial contamination level in 5 tomato farms in Western Gyeongnam. A total of 130 samples was examined for sanitary indicator bacteria, such as aerobic plate count (APC), coliforms, and Escherchia coli, and pathogenic bacteria such as E. coli O157:H7, Salmonella spp., Staphylococcus aureus, and Listeria monocytogenes. APC and coliform count ranged $0\~6.62\;and\;0\~4.52 log_{10}\;CFU/(ml,\;g,\;100\;cm^2,\;hand)$, respectively, and $32.5\%$ were contaminated with E. coli. Especially, most of the samples from employees are high as above $4.0\;log_{10}\;CFU/(ml,\;g,\;100\;cm^2,\;hand)$ in APC. S. aureus, detected at $10.7\%$, was found in employees' hands, irrigation water, and hydroponic solution. whereas E. coli $O157:H7$, Salmonella spp, and L. monocytogenes were not detected. These results will provide fundamental microbiological information for introduction of good agricultural practice (GAP)system in tomato farms.

어린이 급식소의 배식용기 표면에 대한 ATP(Adenosine Triphosphate) 위생 모니터링 가이드라인 설정 (Setting Guidelines for ATP Bioluminescence Hygiene Monitoring of Distribution Trays in Used Children's Foodservices)

  • The purpose of this study was to set new guidelines for adenosine triphosphate (ATP) bioluminescence hygiene monitoring of distribution trays at children's foodservices. Five dietitians visited 223 foodservices (95 institutional, 128 small) to examine whether they adhered to the norms of 'Keeping distribution tray sanitary by washing/sanitizing' and 'Performing food distribution in a clean and appropriate way'. In this visit, dietitians swabbed 100 ㎠ area of the distribution trays twice, once for obtaining ATP measurements and the second time for Aerobic Plate Counts (APC) using 3M Petrifilm Plates. Chi-square test and ANOVA were applied using SPSS 23.0 software. SPSS 23.0 was used to conduct graphical and statistical analysis of the raw data of ATP measurements, which were further transformed by a Box-Cox transformation. The mean of APC from all the subjects inspected was 3.8×102±2,102.0 CFU/100 ㎠. A total of 208 (93.3%) trays were observed within the acceptable limits of APC (Pass<5.0×102 CFU/100 ㎠). APCs taken at institutional foodservices showed significantly lower levels (1.4×102±600.0 CFU/100 ㎠, P<0.01) compared to the small foodservices (5.5×102±2,718.7 CFU/100 ㎠). No significant differences were observed between the two groups in ATP measurements and in the performance rate of 2 checklist items. As against the 93.3% APC adequacy from the total subjected inspection, total ATP adequacy (Pass≤300 RLU/100 ㎠) was only 71.7%. Therefore, more practical guidelines should be prepared for the assessment of the hygiene of distribution trays. In the graphical and statistical analysis, levels below 250 RLU/100 ㎠ was considered 'Pass', while equal to or greater than 350 RLU/100 ㎠ was considered 'Fail' for distribution trays.

퀴노아를 첨가한 청국장의 발효기간에 따른 품질 변화 (Changes in Quality Characteristics of Cheonggukjang added with Quinoa during Fermentation Period)

  • In this study, the quality characteristics of cheonggukjang with addition of different quinoa were investigated. We evaluated the quality and sensory characteristics of cheonggukjang, including the pH, amino nitrogen, slime contents, color value, and total aerobic bacteria. Moreover, The anti-oxidant activities were measured as total polyphenol content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and superoxide dismutase (SOD) activity. The pH and amino nitrogen content were noted to significantly increase during the fermentation period in all the samples. The slime content of cheonggukjang added with quinoa increased with increased fermentation time, but cheonggukjang with addition of 20% of quinoa was decreased. The L value and b value decreased significantly with increased fermentation time, but the a value increased significantly. The microbial tests of cheonggukjang with addition of quinoa showed that the aerobic micro-organisms count was 7.45~9.10 Log cfu/g. Total polyphenol contents increased in all groups during the fermentation period, and activity increased with an increased percentage of added quinoa. The DPPH radical scavenging activity and SOD-like activity of cheonggukjang (with addition of quinoa) were also significantly higher than those of the control. The sensory quality of quinoa 10% cheonggukjang was stronger in flavor, and taste, and demonstrated a higher level of overall acceptability, when compared to the other groups.

Characteristics of proteolytic microorganisms and their effects on proteolysis in total mixed ration silages of soybean curd residue

  • Objective: The objective of this study was to isolate proteolytic microorganisms and evaluate their effects on proteolysis in total mixed ration (TMR) silages of soybean curd residue. Methods: TMRs were formulated with soybean curd residue, alfalfa or Leymus chinensis hay, corn meal, soybean meal, a vitamin-mineral supplement, and salt in a ratio of 25.0: 40.0:30.0:4.0:0.5:0.5, respectively, on a basis of dry matter. The microbial proteinases during ensiling were characterized, the dominate strains associated with proteolysis were identified, and their enzymatic characterization were evaluated in alfalfa (A-TMR) and Leymus chinensis (L-TMR) TMR silages containing soybean curd residue. Results: Both A-TMR and L-TMR silages were well preserved, with low pH and high lactic acid concentrations. The aerobic bacteria and yeast counts in both TMR silages decreased to about 105 cfu/g fresh matter (FM) and below the detection limit, respectively. The lactic acid bacteria count increased to 109 cfu/g FM. The total microbial proteinases activities reached their maximums during the early ensiling stage and then reduced in both TMR silages with fermentation prolonged. Metalloproteinase was the main proteinase when the total proteinases activities reached their maximums, and when ensiling terminated, metallo and serine proteinases played equally important parts in proteolysis in both TMR silages. Strains in the genera Curtobacterium and Paenibacillus were identified as the most dominant proteolytic bacteria in A-TMR and L-TMR, respectively, and both their proteinases were mainly with metalloproteinase characteristics. In the latter ensiling phase, Enterococcus faecium strains became the major sources of proteolytic enzymes in both TMR silages. Their proteinases were mainly of metallo and serine proteinases classes in this experiment. Conclusion: Proteolytic aerobic bacteria were substituted by proteolytic lactic acid bacteria during ensiling, and the microbial serine and metallo proteinases in these strains played leading roles in proteolysis in TMR silages.

한국형 편의 육가공 제품의 가공 및 포장 개발 연구 (Development of Processing and Packaging Method for A Korean Seasoned Meat Product)

  • A Korean seasoned meat product, seasoned bulgogi was developed in processing and packaging for improved convenience and extended shelf life. The optimal process conditions were found to consist of aging for $20{\sim}30$ hours at $5^{\circ}C$ and air heating for 20 minutes at $210^{\circ}C$ Aging of 24 hours could attain soft texture with little change in pH, color and total aerobic bacterial count. Appropriate drip level could be achieved at air heating of 20 minutes at $210^{\circ}C$, while the air heating at $180^{\circ}C$ could not give suitable level of texture. Modified atmosphere packaging of 60% $CO_2$ and 40% $N_2$ could preserve the product significantly better compared to control of air package and vacuumed package.

레토르트 및 감마선 조사에 의한 화닭 덮밥 소스의 미생물 제어 효과 비교 (Antimicrobial Effects of Retort and Gamma Irradiation on Bacterial Populations in Spicy Chicken Sauce)

  • 극한 환경에서 취식이 가능하도록 안전성 및 기호성이 확보된 덮밥 소스류 개발을 위한 기초연구로 많이 소비되고 있는 화닭 덮밥소스의 위생화를 위해 감마선 조사 기술을 적용하고 레토르트 살균과의 비교연구를 진행하였다. 화닭 덮밥 소스의 미생물 오염도 평가 결과 레토르트 비처리군은 총세균수가 4.5 log CFU/g 검출되었고 레토르트 처리군은 2.1 log CFU/g의 총 세균이 검출되었다. 반면 3 kGy의 감마선 처리군 에서는 검출한계 이하의 세균이 검출되었다. 검출된 미생물을 동정한 결과 Bacillus spp.로 확인되었으며, B. cereus 및 B. subtilis에 대한 방사선 감수성 측정 결과 각각 0.39 및 0.28 kGy로 확인되었다. SOS chromotest를 이용한 유전독성학적 안전성 평가 결과 감마선 조사에 의한 돌연변이원성은 관찰되지 않았다. 이상의 결과를 종합하여 볼 때, 화닭 덮밥 소스의 효과적인 위생화를 위하여 3 kGy의 감마선 조사처리가 레토르트 처리보다 효과적으로 미생물을 사멸하는 것으로 확인되었다.

Identification of Microorganisms in Duck Meat Products Available in Korea and the Effect of High Hydrostatic Pressure

  • The objective of this study was to investigate the microbial count of duck meat and duck meat products commercially available in Korea. High hydrostatic pressure (HHP) treatment was applied at 0.1, 300, 400, and 500 MPa for 5 min to enhance the microbiological safety of duck meats. The levels of total aerobic bacteria were in the ranges of 3.53-6.19 and 3.62-6.85 Log CFU/g in raw and smoked duck products, respectively. By DNA sequence analysis, we identified microorganisms responsible for spoilage, with the most common species in the raw and smoked duck products being Aeromonas spp. or Pseudomonas spp. and Leuconostoc mesenteroides, respectively. HHP treatment significantly reduced the levels of total aerobic bacteria in raw and smoked duck products. This study demonstrates that HHP treatment may be used to effectively improve the safety of raw and smoked duck meat products.

Effects of vacuum and high-oxygen modified atmosphere packaging on physico-chemical and microbiological properties of minced water buffalo meat

  • Objective: In this study, the effects of vacuum (VP) and high-oxygen modified atmosphere ($80%\;O_2+20%\;CO_2$) packaging (HiOx-MAP) on physico-chemical and microbiological properties of minced water buffalo meat were investigated. Methods: After minced meat preparation, samples were packaged under VP and HiOx-MAP and stored at $2^{\circ}C{\pm}0.5^{\circ}C$ for 14 days. Samples taken on certain days were subjected to total aerobic mesophilic bacteria, total aerobic psychrotrophic bacteria, lactic acid bacteria, Pseudomonas, Enterobacteriaceae and yeast-mold counts as well as pH, color ($L^*$, $a^*$, and $b^*$) and thiobarbituric acid reactive substances (TBARS) analyses. Results: In minced water buffalo meat packaged under HiOx-MAP, TBARS value exceeded 1 mg malondialdehyde/kg on the 4th day of the storage. In VP samples, TBARS value remained close to initial TBARS value during storage. According to the findings, $a^*$ value was determined to be high in the HiOx-MAP samples within initial days of the storage. However, no significant changes in $a^*$ value were observed in VP samples during storage. In contrast, the mean value of $L^*$ was detected as higher in HiOx-MAP sample than VP samples. The count of psychrotrophic bacteria increased more than that of mesophilic bacteria during storage. The growth of Enterobacteriaceae and Pseudomonas was delayed in both the packaging methods. However, lactic acid bacteria exhibited more growth in VP samples compared to MAP samples. Conclusion: The lipid oxidation proceeded faster than expected in minced water buffalo meat packed with HiOx-MAP method. This situation adversely affected the $a^*$ value. On the other hand, similar microbiological results were obtained in both packing methods.

떡국 떡의 품질유지에 미치는 변형기체포장(MAP) 효과 (Effect of Modified Atmosphere Packaging on Quality Preservation of Rice Cake (Ddukgukdduk))

  • Packages of different atmospheres (air (control), 100% CO2, vacuum, and vacuum + O2 absorber) were prepared for 0.4 kg rice cake (ddukgukdduk) using gas-barrier plastic film and stored at 10℃ for 11 days. The stored products were evaluated in their packages atmosphere, total aerobic bacteria, yeast and molds, texture and sensory quality during storage period. In the air package, the O2 concentration decreased from initial 21% to 16% on storage 4 days and the CO2 concentration increased to 23% on storage of 11 days, which resulted from the growth of microorganisms. CO2 concentration decreased from initial 98% to 36% on storage 11 days in the 100% CO2 package. It is reasoned that CO2 was dissolved into the product reducing the volume of the package. Vacuum and vacuum +O2 absorber package maintained shrunk vacuum condition until 11 days of storage. Total aerobic bacteria count increased significantly in the control package (6.41 log (cfu/g) after 11 days) compared to the 100% CO2 package (4.96 log (cfu/g) after 11 days). Yeast and molds were 6.66 in control package, 3.43 in 100% CO2 package, 4.66 in vacuum package, and 3.78 in vacuum + O2 absorber package after 11 days. There was no significant difference between control and the other treatments for the texture of the stored products. Sensory quality was the worst in control package on the storage of 8 days. All treatment groups except control improved the quality preservation, but vacuum and vacuum + O2 absorber packages suffered from cracking of the product. Thus 100% CO2 flushing is suggested as a desired packaging condition.
