체액 검체에서 톨루이딘 블루 염색의 유용성 (Availability of Toluidine Blue Stain in Body Fluid)

  • We evaluated the availability of toluidine blue stain in body fluids, such as peritoneal and pleural fluid and urine. Nine hundreds specimens, i.e., 400 pleural and 400 peritoneal fluids and 100 urine samples, respectively, from Jan. 1995 to May 1996 were included. We obtained the result of high sensitivity and high specificity. In toluidine blue stained body fluid in comparison with Papanicolaou stained result. Additionally, we found the diagnostically important crystals in chylothorax and some urine samples, which can not be seen in routine Papanicolaou stain. We thought the toluidine blue stain in body fluid is one of very useful diagnostic methods.

  • 자연계에 풍부한 천연고분자물질인 키틴을 탈아세틸화하여 얻어지는 키토산을 색소의 흡착제로 이용하고자, 색소 Toluidine Blue O(TBO)에 대한 키토산의 흡착 특성을 연구하였다. 키토산과 TBO의 흡착반응에는 키토산 입경, 키토산의 양, TBO의 초기 농도, 반응액의 pH, 반응시간 등이 중요한 인자로 작용하는 것으로 밝혀졌다.

  • 1. 10種染料 (neutral red, toluidine blue, pyronin, methylene blue, alcian blue, trypan blue, carmine, orange G, aniline blue, Janus green B, and India ink)로 處理된 흰쥐의 腹腔內細胞 特히 肥滿細胞의 식作用 및 複屈折性의 有無를 位相差顯微鏡法 및 偏光顯微鏡法으로 分析하였으며 附隨的으로 cytomorphic change 도 觀察하였다. 2. 腹腔內 肥滿細胞는 大部分 染料에 對하여 아무런 식作用을 보이지 않았으며 India ink 群에서 輕微한 식作用을 보였다. 大식細胞와 各種 白血球는 大體로 旺盛한 식作用을 나타내었다. 3. 腹腔內 肥滿細胞는 大部分의 染料를 處理하여도 아무런 複屈折性을 보이지 않았으며 trypan blue 와 aniline blue 處理群에서 매우 輕微한 反應을 보였을 뿐이었으며 白血球는 全然 反應을 보이지 않았다. 4. 各種染料에 對한 肥滿細胞의 細胞形態學的 反應은 一定하지 않았으며 大體로 輕微한 變化를 일으켰다. 白血球의 同染料에 對한 反應은 特히 trypan blue, orange G, India ink群에 서 顯著하였다.

꿩 전위의 비만세포에 관한 형태학적 연구 (Morphological Study on the Mast Cell of Proventriculus in Pheasant (Phasianus colchicus))

  • 꿩 비만세포는 원형, 난원형, 방추형 또는 부정형이었고 세포질돌기를 가지고 있었다. 꿩 비만세포는 Karnovsky 용액으로 고정했을 때 가장 좋은 염색성을 얻었고, Toluidine blue로 염색을 했을 때 비만세포의 과립은 이염색성(meta-chromatic)을 나타내었다. 비만세포의 세포질에는 막으로 둘러싸인 과립과 발달이 미약한 세포소기관으로 채워져 있었다. 비만세포의 과립은 원형과 난원형이었고 일부 과립은 막이 함몰된 오목한 모양과 밖으로 돌출된 볼록한 모양이었다. 과립 속의 구조는 동질성, 입자형 또는 망상형이었다.

Comparison of Different Screening Methods in Estimating the Prevalence of Precancer and Cancer Amongst Male Inmates of a Jail in Maharashtra, India

  • Aim: To compare the effectiveness of self-examination, clinical examination and screening methods using 1% toluidine blue and Lugol's iodine in estimating the prevalence of lesions at risk of malignancy and oral malignant disease amongst the male inmates of Yerwada Central Jail, Pune. Material and Methods: Study was carried out on male inmates in two phases. In the first phase self-examination and clinical examination was carried out on 2,257 male inmates. 164 suspicious cases were subjected to phase II of the study out of whom, 82 participants were screened with 1% toluidine blue and 2% Lugol's iodine followed by biopsy procedure. Results: Sensitivity and specificity for self-examination with clinical examination was 92.2% and 96.6% respectively. Sensitivity, specificity, positive (PPV) and negative predictive values (NPV) and positive (+LR) and negative likelihood ratio (-LR) for Toluidine Blue were 88.1%, 66.6% 97.1%, 30%, 2.63 and 0.17 respectively while for Lugol's Iodine they were 94.7%, 83.8%, 98.6%, 55.5%, 5.67 and 0.06 respectively. Conclusions: Prevalence of lesions at risk of malignancy and oral malignant disease by self-examination was 7.8% and by clinical examination was 6.3%. Self-examination is an effective tool in early detection of oral cancer. Use of Lugol's iodine as a screening tool for oral lesions is highly effective in inmate populations.

Staining Characteristics of Mast Cell Tumor in a Hedgehog

  • A 2-year old female African hedgehog (Atelerix albiventris) weighing 645 grams was presented with a well circumscribed mass in the subcutaneous of the left shoulder. It measured $2{\times}2cm$ in size and had a hard texture. There were no clinical signs. In a fine-needle aspiration (FNA) cytological evaluation, mast cells were identified with Wright-Giemsa stain. Mast cell tumor was confirmed and characterized by surgical sampling and histopathological evaluation. Four stains (hematoxylin-eosin, toluidine blue, periodic acid-Schiff, and Safranin O) were examined in order to find an applicable stain for a mast cell tumor histopathological evaluation in hedgehog. This case showed that periodic acid-Schiff and toluidine blue staining were the most useful for assessment of a mast cell tumor in a hedgehog.

The utility of sperm DNA damage assay using toluidine blue and aniline blue staining in routine semen analysis

  • Objective: The aim of the present study was to examine the relationship among male age, strict morphology, and sperm chromatin structure and condensation. Methods: Sperm samples from a total of 100 men underwent semen analysis, and sperm chromatin structure and condensation were assessed with toluidine blue (TB) and aniline blue (AB) tests. Results: Prevalence of strict morphology of less than 4%, and abnormal sperm chromatin structure and condensation did not show any statistically significant differences according to male age (p=0.605, p=0.235, and p=0.080). No significant correlation was demonstrated among age of male partners, strict morphology, and abnormal sperm chromatin structure using TB and AB tests. However, abnormal sperm chromatin condensation was positively associated with sperm chromatin structure (r=0.594, p=0.000) and showed negative correlation with strict morphology (r=-0.219, p=0.029). Conclusion: The tests for sperm chromatin condensation showed a significant association with strict morphology. Further study is needed to elucidate the relationship between clinical outcome and sperm chromatin tests.

가토 치수에 있어서 KMnO4투여후 형성된 경조직의 성상에 관한 연구 (HISTOCHEMICAL STUDY ON THE CALCIFIED TISSUE INDUCED BY POTASSIUM PERMANGANATE IN THE RABBIT DENTAL PULP)

  • The author observed histochemically the nature of the calcified tissue in the rabbit dental pulp, induced by pulpal injection of potassium permanganate. The pulp of rabbit mandibular incisors were exposed and enlarged by a dental hand reamer. The exposed pulps were injected with 0.05ml of 20mM solution of potassium permanganate dissolved in Ringer's solution in experimental tooth. Also the control tooth received a pulpal injection of 0.05ml of Ringer's solution. After pulpal injection, the tooth was plugged with a gutta-percha root canal point. The staining techniques were hematoxylin-eosin stain, van Gieson stain, PAS reaction, toluidine blue stain, alcian blue-hematoxylin stain and colloidal iron-picric acid stain. The results were as follows: 1. The pulp on experimental tooth showed osteodentin-like calcified tissue. Also, in some areas, false denticle-like substance were observed. 2. The central portion of the calcified matrix showed metachromasia in toluidine blue stain had strong staining capacity in alcian blue stain. 3. The peripheral portion of the calcified tissue revealed marked van Gieson positive reaction for collagen. But their staining ability in alcian blue was slight and metachromasia was not appeared.

폐수처리시스템에서의 그람 양성 세균 모니터링 방법 (A Rapid Method for Monitoring of Gram-positive Bacteria in Wastewater Treatment Systems)

  • 폐수 처리 시스템의 그람 양성 세균을 간편하고 신속하게 모니터링할 수 있는 그람 염색 방법을 개발하였다. 각각 4종류의 그람 양성 세균과 음성 세균의 배양액을 polyethersulfone membrane filter와 Toluidine Blue-O로 여과 및 염색하였다. 염색된 여과막을 건조시키고 스캔하였으며, 영상 분석 프로그램을 이용하여 빨강색, 녹색, 파랑색의 강도를 측정하는 정량적 색상 이미지 분석법을 개발하였다. 그람 양성 세균의 경우 그람 음성 세균보다 빨간색과 초록색 수치가 높게 나타났다. 본 방법을 그람 양성 세균이 혼합된 활성슬러지에 적용한 결과, 활성슬러지의 경우 플록의 불규칙한 크기와 형태로 측정이 어렵다 할지라도 플록 분산 기술을 이용하여 활성슬러지의 그람 양성 세균의 변화를 모니터링 할 수 있었다. 활성슬러지에서도 그람 양성 세균의 비율이 높아질수록 빨간색과 초록색 수치가 증가하였다. 본 연구에서 개발된 방법은 폐수처리 시스템에 존재하는 그람 양성 세균을 신속하고 정량적으로 모니터링 할 수 있다.

Modified Toluidine Blue: an Alternative Stain for Helicobacter pylori Detection in Routine Diagnostic Use and Post-eradication Confirmation for Gastric Cancer Prevention

  • Background: Modified toluidine blue staining (MTBs) is a simple, inexpensive and time saving method to detect H. pylori in gastric biopsy specimens. As a metachromatic stain, it simultaneously highlights intestinal metaplasia, a gastric cancer precancerous lesion. The aim of this study was to assess the reliability of MTBs compared with hematoxylin-eosin (H&E) for H. pylori detection using immunoperoxidase staining as the gold standard. This technique would be beneficial for a routine diagnosis and confirmation of H. pylori eradication in developing countries where endoscopic-based approaches are dominant. Materials and Methods: Esophagogastroduodenoscopy with triple site gastric biopsies was undertaken in 207 dyspeptic patients at Thammasat University Hospital, Thailand between 1997 and 1999. H&E, MTBs and immunoperoxidase staining were applied to each specimen. The presence or absence of H. pylori with each stain was interpreted separately and the sensitivity, specificity, positive and negative predictive values of H&E and MTBs were calculated. Results: A total of 282 specimens from 207 patients were evaluated. Using immunoperoxidase staining, organisms were positive in 117 specimens (41%). MTBs proved almost equally sensitive as immunoperoxidase (99%) and significantly more sensitive than H&E (85%). It has comparable specificity (96% vs 96%), PPV (95% vs 94%), and NPV (99% vs 90%) to H&E, using immunoperoxidase staining as gold standard. MTBs compared with immunoperoxidase staining, is cheaper (2 USD vs 12 USD) and faster (20 min vs 16 hrs) compared to immunoperoxidase staining. Conclusions: MTBs is effective, economical and easy to use in daily practice for the detection of H. pylori in gastric biopsy specimens. In addition to saving time in evaluating H. pylori associated gastritis, with a high sensitivity and ability to demonstrate intestinal metaplasia, the technique may have a role in confirmation of H. pylori eradication for gastric cancer prevention in a developing country setting.
