Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is considered as an antitumor agent owing to its ability to induce apoptosis of cancer cells without imparting toxicity toward most normal cells. TRAIL is produced in poor yield because of its insoluble expression in the cytoplasm of E. coli. In this study, we achieved soluble expression of TRAIL by fusing maltose-binding protein (MBP), b'a' domain of protein disulfide isomerase (PDIb'a'), or protein disulfide isomerase at the N-terminus of TRAIL. The TRAIL was purified using subsequent immobilized metal affinity chromatography and amylose-binding chromatography, with the tag removal using tobacco etch virus protease. Approximately 4.5 mg of pure TRAIL was produced from 125 ml flask culture with a purification yield of 71.6%. The endotoxin level of the final product was $0.4EU/{\mu}g$, as measured by the Limulus amebocyte lysate endotoxin assay. The purified TRAIL was validated and shown to cause apoptosis of HeLa cells with an $EC_{50}$ and Hill coefficient of $0.6{{\pm}}0.03nM$ and $2.41{\pm}0.15$, respectively. The high level of apoptosis in HeLa cells following administration of purified TRAIL indicates the significance and novelty of this method for producing high-grade and high-yield TRAIL.
Lower leaves of aromatic tobacco are also much lower in Quality than upper leaves. So feasibility test of no harvesting and curing of lower leaves was conducted under high planting density and high nitrogen conditions with conventional cultural system. Effect of harvesting time on yield and Quality were investigated under 2 nitrogen levels. Among harvesting methods of conventional harvest with priming under high planting density, no-harvest of first priming, removal of lower leaves which relevant to first prime stalk before maturity, no-harvest of first and second priming. no-harvesting or pruning of first prime stalk before maturity was best in yield, price and in crude income. The shortor the harvest period became, the lower the yield, price and contents of reducing sugar and nicotine became, but reverse in this trends with total nitrogen and protein nitrogen. So 6 or 8 days interval of harvest is most recommendable.
Fundamental data for new type of product development from Lycii fructus was experimentally determined. The chemical composition, and Hunter value changes, optimum extraction, volatile components of extract under different extraction conditions and heat treatment were mainly studied. Results are summarized as follows. The proximate compositions of dried Lycii fructus were water 21.8%, total sugar 27.6%, reducing sugar 15.2%, crude protein 14.29%, crude fat 5.65%, crude fiber 7.48%, and ash 7.98% in percent stale, respectively. Extracted yield on the basic of solid extract was getting increased when more solvent was used for extraction. The most recommendable extraction was 1 to 10 part of sample to solvent ratio. When water was employed as extraction solvent, the highest amount of solid extract was obtained. Extract of Lycii fructus in terms of yields and color was most acceptable when raw sample was treated 8 minute roasting at $180^{\circ}C$ and 60 minute heating at $120^{\circ}C$, respectively. Major volatile components of fresh Lycii fructus were to hexadecanoic acid, methyl linoleate, benzyl alcohol, dimetane benzene by GC/MS. By the roasting of raw sample, the compounds of 2-methyl-2buthenal, 1,4-dimethyl benzene, and benzyl alcohol were reduced. Wheras, methyl thiopropanol, benzene acetaldehyde and ethyl linoleate were slightly increased.
Purpose - During the last two years, convenient stores (CS) are emerging as one of the most fast-growing retail trades in Korea. The goal of this work is to forecast and to analyze sales at CS using ARIMA-Intervention model (IM) and exponential smoothing method (ESM), together with sales at supermarkets in South Korea. Considering that two retail trades above are homogeneous and comparable in size and purchasing items on off-line distribution channel, individual behavior and characteristic can be detected and also relative superiority of future growth can be forecasted. In particular, the rapid growth of sales at CS is regarded as an everlasting external event, or step intervention, so that IM with season variation can be examined. At the same time, Winters ESM can be investigated as an alternative to seasonal ARIMA-IM, on the assumption that the underlying series shows exponentially decreasing weights over time. In case of sales at supermarkets, the marked intervention could not be found over the underlying periods, so that only Winters ESM is considered. Research Design, Data, and Methodology - The dataset of this research is obtained from Korean Statistical Information Service (1/2010~7/2016) and Survey of Service Trend of Korea Statistics Administration. This work is exploited time series analyses such as IM, ESM and model-fitting statistics by using TSPLOT, TSMODEL, EXSMOOTH, ARIMA and MODELFIT procedures in SPSS 23.0. Results - By applying seasonal ARIMA-Intervention model to sales at CS, the steep and persisting increase can be expected over the next one year. On the other hand, we expect the rate of sales growth of supermarkets to be lagging and tied up constantly in the next 2016 year. Conclusions - Based on 2017 one-year sales forecasts for CS and supermarkets, we can yield the useful information for the development of CS and also for all retail trades. Future study is needed to analyze sales of popular items individually such as tobacco, banana milk, soju and so on and to get segmented results. Furthermore, we can expand sales forecasts to other retail trades such as department stores, hypermarkets, non-store retailing, so that comprehensive diagnostics can be delivered in the future.
Hydrolysis properties of ginseng saponins in processing of extraction with vacuum impulse system extraction method were compared with multi-stage extraction methods. Crude saponin content of the extract produced by vacuum impulse system extraction method was $11.5\%,$ compared with multi-stage extraction method (about $8.13\%).$ Also the yield of the extract increased about $6.7\%.$ The flavor and aroma of ginseng extract with vacuum impulse system extraction method are stronger than multi-stage extraction methods and people have a tendency to like more. The color was similar to existing extraction items and the liquidity ratio was high. Vacuum impulse system extraction method could save human resources because of short extraction time and automatic operation of processing. With HPLC pattern, We could ascertain the truth that hydrolysis properties of ginseng saponin was restrained in the extraction processing, vacuum impulse system extraction method.
This experiments were carried out determine planting date for highest Yields and select an covering material for enhancement of environments in Cassia tora L.The highest Yields by regressions estimate produced 352 kg/10a at May 18. of coure, it is possible to plant at June and early-July in spite of slight decreas of Yields, therefore Cassia tora L. is benefit to establish cropping system with barley, wheat or other crops. The covering materials for highest yielding was Black-white nonporous PE(low density) and it out yielded about 53% than conventional cultivations. Cassia tora L. is possible to produce Without agricultural medicines because of little occurence of decrease except occuring of Damping off(Rhizoctonia solanikuhn) at early planting.
This study was conducted to find out the stalk curing methods that can reduce photobleaching, and shorten the curing period in burley to bacco stalk curing. The results obtained summarized as follows. In the ventilation window-plot, the temperature within the curing house was high and the curing period was shortened, too. The rate of photobleached leaves of whole plant stalk curing-plot was higher than those of any other plots. In the ventilation window-plot, the labor of operating the curing house was saved and the yield and visual quality of cured leaves were high, too. For reducing the photobleaching of leaves, following procedures are desirable; to prevent the lower leaves from drying up and overripening at maturing stage, cut the stalk after first or second priming, shorten the curing period as possible when the 80% of leaf bodies changed brown.
The enzymatic transgalactosylation of L-ascorbic acid was investigated to synthesize a chemically stable form of L-ascorbic acid by using commercially available ${\beta}-galactosidases$. Among various enzymes examined, Aspergillus oryzae ${\beta}-galactosidase$ was found to be formed the derivative of ascorbic acid in a high yield from ascorbic acid and lactose. The reaction product was isolated by ion exchange chromatography on a $Dowex\;1\;{\times}\;8$ (Cl - form) resin and Toyopearl HW-40S gel chromatography. The product was identified as $6-O-{\beta}-_D-galactopyranosyl-_L-ascorbic\;acid$ on the basis of various experimental results, viz., UV, IR, $^1H-NMR,\;^{13}C-NMR$ and mass spectral data.
To ascertain the effect of over-expressed maize calreticulin in tomato plant on tobamovirus movement in addition to validating potentiality of the gene (ZmCRT) as a means for the virus-resistance resource, four ZmCRT-expressing homozygous lines were generated from the T0 plants as using an Agrobacterium-mediated transformation, nucleic acid analyses, and a conventional breeding method. Of them, a line was subjected to the bioassay for tolerances to tobacco mosaic virus-U1 (TMV-U1) and tomato mosaic virus (ToMV) followed by RT-PCR and a chlorophyll fluorescence quenching analyses. Both transgenic plants transcribing ZmCRT and wild-type plants showed no symptom by 20 days after viruses inoculation, however the photosystem II quantum yield parameter measured from the upper leaves of ToMV-inoculated plants revealed that ZmCRT transgenic plants have higher photosynthetic ability than wild-type ones at that time, which indirectly implies that over-expressed ZmCRT product acts as a barrier to the cell-to-cell and/or systemic movement of ToMV. Moreover, ZmCRT transgenic plants showed remarkably longer shoot length than wild-type ones in 40 days after TMV-U1 or ToMV inoculation each, which might be resulted from higher photosynthetic ability during the phase not yet showing any external symptoms. Collectively, over-expressed ZmCRT protein in tomato plants is able to interrupt the systemic movement of infected TMV-U1 and ToMV even though not perfect.
Field trials were conducted in Suwon, Andong, Bongwha and Taean to evaluate an anthracnose forecaster in the integrated pest management system (IPM) including anthracnose, Phytophthora blight and tobacco budworm for hot pepper in 2008-2009. Percentage of diseased fruits by pepper anthracnose and yield were compared among three treatments, no-fungicide, forecaster and conventional. The incidence and yield of the forecaster at Bongwha on two varieties were 3.1-3.5% and 30-33 kg/40 plants. These were little less than those of the conventional's 0.3-0.8% and 35-36 kg/40 plants and much higher than those of the no-fungicide's 23-27% and 18-24 kg/40 plants. The number of spray at the forecaster was 5 times, whereas those at the conventional was 11-19 times depending on the farmers. In addition, the results of an anthracnose forecaster at the other three field trials allowed in protecting hot pepper from anthracnose while reducing the amount of pesticides used and the total cost of the pathogen control. However, it is needed to improve for anthracnose forecaster when anthracnose pressure was high such as Taean where 60-80% of the incidence. Because the control values of the forecaster at Taean were between those of no-pesticide and conventional treatment at best. If anthracnose is severe early in the season, additional preventive control should be considered when the fruits were fully grown yet.
