Control Effects of an Antibiotic Produced by Streptomyces sp. B25 on Tobacco Mosaic Virus and Determination of Its Molecular Structure

  • The culture filtrate of Streptomyces sp. B25, which was identified in this experiment, was tested for the control of tobacco mosaic virus (TMV) with the susceptible tobacco cultivar, NC 82, under the field conditions following the preliminary examination of its characters for TMV control. Control efficacy of the culture filtrate against TMV infection continued over 50% up to 6 days after treatment, and its systemic effect was about 30% of the direct effect. In field conditions control efficacy of the culture filtrate against TMV infection was 95.3 % at 2 weeks after TMV inoculation, and decreased to 58.3 % at 3 weeks after inoculation. Five fold-dilution of the culture filtrate showed about half of the control efficacy by the stock culture filtrate. Analysis of the antibiotic material responsible for the inhibition of TMV infection through nuclear magnetic resonance experiments revealed that the antibiotic is antimycin $A_1$, which is firstly reported as an anti-phytoviral antibiotic in this experiment.

STUDIES ON THE FRAGILITY OF TOBACCO RAW MATERIAL (잎담배의 부스러짐 특성에 관한 연구)

  • The fragility, which express the fineness index, increased linearly with the moisture content for various tobacco shreds and showed a high correlation to the moisture content. Then, fineness index (FIs) at reference moisture content(l2.5%) was obtained by following equation. FIs=FIm+A(12.5-Mm) A" denote the gradient of simple linear regression equation between the fineness index and moisture content. FIm; the fineness index measured at the standard moisture content(12.5%). The characteristics of fragility concerning with some kinds of tobacco raw materials, stalk position, moisture content and storaged time were investigated. The following results were obtained. 1. The moisture content range of less fragile of tobacco raw material was from 16% to 20%. whereas the rolled process sheet tobacco was from 12% to 16%. 2. The Bright yellow and the paper process sheet tobacco were less fragile than that of the other tobacco raw materials. 3. The leaves were more distant from middle stalk position, and its become more fragile. 4. Physical properties of the storaged tobacco leaves were determined as a function of each storageunction of each storage time. The long-term storaged leaves(more than 3 years) were slightly lighter weight, more fragility and lower equilbrium moisture content than thos of the mid-term storaged leaves.(less than 3 years). 5. Contents of chemical component of the storaged leaves were determined every storage time. The long-term storaged leaves were slightly lower content of nicotine and chlorine than those of mid-term storaged leaves, but contents of crude fiber, total ash and calcium were increased significantly.

  • A procedure for the simultaneous determination of reducing sugar, nicotine alkaloid, total sugar and ammonia has been developed. The four components of tobacco extracted from ground tobacco sample with 2 % acetic acid were deter mined with a Technicon autoanalyzer system. The relative errors in burley and flue-cured tobacco leaves were 3.6% in comparison with each official methods.

  • Most of studies on tobacco have researched it as economic or cultural factor. These studies, certainly, have the value of their own. But from time to time these studies have brought about romantic view regarding tobacco, and have been an obstacle to approach it as a medical factor. Thus this thesis will introduce documents of tobacco as medical factor. Through it we will be able to furnish researchers with information to treat poison caused by tobacco.

  • This experiment was conducted to investigate the effect of physical and chemical characteristics of lower grade tobacco with treatment of ozone. In ozonization of low grade tobacco, optimum moisture was 20 percentage, and decrease ratio of content of tobacco leaf was 30 percentage in nicotine , 15.9 percentage in total volatile base, 10.5 percentage in petroleum-ether-extract, 29.5 percentage in solanesol, and decrease ratio of cigarette smoke composition was 31 percentage in nicotine, 6.08 percentage in tar.

  • Pot experiment was conducted to find out the effect of decomposing degree of rice straw on growth characteristics of flue-cured tobacco, NC82. Tobacco growth was hindered by fresh straw of rice application. Generally, it was known that if organic matter of high Carbon/Nitrogen ratio had applied in soil, there was temporary nitrogen deficiency in plant caused by soil microorganism utilized nitrogen contained organic matter. In pot experiment, it was supposed that tobacco growth hindered by fresh straw of rice application was not nitrogen deficiency by soil microorganism, but gas toxicity by fresh straw of rice application.
