Bioaerosols generated from wastewater treatment plants may create health risks for plant workers and nearby residents. To determine the levels of culturable airborne bacteria and fungi in bioaerosols, samples were seasonally collected above and near the aeration tanks of one feces-urine and three sewage treatment plants in Ulsan, Korea with an impaction-type sampler. In the feces-urine treatment plant, concentrations of heterotrophic bacteria were between $1.3({\pm}0.2){\times}10^3$ and $2.6({\pm}1.2){\times}10^4$ MPN/$m^3$ above the aeration tank and between $1.7({\pm}1.0){\times}10^2$ and $7.2({\pm}2.2){\times}10^3$ MPN/$m^3$ near the aeration tank. Coliform bacteria were detected both above and near the aeration tank. In cases of sewage treatment plant, the numbers of heterotrophic bacteria ranged from $1.9({\pm}1.2){\times}10^1$ to $1.8({\pm}1.2){\times}10^4$ MPN/$m^3$ above the aeration tank and from $5.0({\pm}2.8){\times}10^0$ to $6.6({\pm}2.0){\times}10^3$ MPN/$m^3$ near the aeration tank. At reference sites, the concentrations of heterotrophs in ambient air were measured between $7.0{\times}10^0$ and $2.7{\times}10^1$ MPN/$m^3$. When we isolated and tentatively identified heterotrophic bacteria, Pseudomonas luteola was the most dominant species in bioaerosols from wastewater treatment plants, whereas the most abundant one in reference samples was Micrococcus sp. When we measured fungal concentrations in bioaerosols, they were rather similar regardless of sampling locations and seasons, and such genera as Cladosporium, Alternaria, and Penicillium were commonly identified.

This study was performed to investigate the antimicrobial effects against food-borne pathogens and antioxidant activity of Rhododendron brachycarpum ethanol-extract. The antimicrobial activity of the extract was determined using a paper disc-diffusion method, and the diameter of the clear zone was measured. The diameter of the clear zone in the presence of 10 mg of extract was maximal against Bacillus cereus among the three tested Gram-positive bacteria and against Escherichia coli O157:H7 among the five tested Gram-negative bacteria. Analysis of the minimum inhibitory concentration (MIC) showed that the extract exhibited a similar efficacy as that of sorbic acid, a well-known chemical preservative. The growth inhibitory effects of the extract at concentrations of 250, 500, 1,000, and 2,000 mg/L on food-borne pathogens were determined against Staphylococcus aureus, Listeria monocytogenes, Salmonella Typhimurium, and Escherichia coli O157:H7. Growth of the microorganisms was not affected by the extract at concentrations up to 250 mg/L, but it was significantly (p<0.05) inhibited by the extract at concentrations higher than 1,000 mg/L. The antioxidant effects of the extract were examined via measurement of DPPH radical scavenging activity, inhibition of reactive oxygen species (ROS) generation using fluorescent dichlorofluorescien (DCF) assay, and prevention of peroxyl radical- and hydroxyl radical-induced supercoiled DNA breakage. The $IC_{50}$ of the extract for DPPH radical scavenging activity was about half that of ${\alpha}$-tocopherol, which was used as a positive control. DCF fluorescence intensity decreased as the concentration of the extract increased, demonstrating that ROS generation was inhibited in a concentration-dependent manner. The ROS inhibitory effect of the extract was higher than that of ascorbic acid. The extract prevented supercoiled DNA strand breakage induced by peroxyl radical and hydroxyl radical. Thus, the results of the present study demonstrate that the extract exhibits antimicrobial effects against food-borne pathogens as well as potent antioxidant capacity, suggesting that R. brachycarpum could be used as a natural antibacterial agent and effective antioxidant in food.

This study evaluated the effects of water velocity, substrates, and phosphorus concentrations on the growth of filamentous periphytic algae (FPA) in the two types of artificial channel systems using treated wastewater. Controlled parameters included 5 ${\sim}$ 15 cm $s^{-1}$ for the water velocity; 10 and 20 mm wire meshes, natural fiber net, gravel and tile for the substrates: and 0.05 ${\sim}$ 1.0 mgP $L^{-1}$ for the P concentration. Algal growth rate of FPA was compared using both chi. a and dry weight change with time. Under the controlled water velocity range, the growth of FPA increased with the velocity, but the maximum growth rate was shown in the velocity of 10 cm $s^{-1}$. The substrate that showed the maximum growth of FPA differed between the artificial channel and indoor channel, due to the influence of suspended matters which caused the clogging of the meshed substrates. Under the controled range of P concentration, the growth rates of all three FPA species (Spirogyra turfosa, Oedogonium fovelatum, Rhizoclonium riparium) increased with the P increase, but they showed the differential growth rates among different P concentrations. The results of this study suggest that under the circumstance having an large amount of nutrients FPA develop the biomass rapidly and that even a little increase over the threshold velocity causes the detachment of filamentous periphytic algae. Thus, FPA dynamics in eutrophic streams, such as those receiving treated wastewater, seem to be sensitive to the water velocity. On the other hand, detached algal filaments could deteriorate water quality and ecosystem function in receiving streams or down-stream, and thus they need to be recognized as an important factor in water quality management in eutrophic streams.

To improve understanding of the sources and chemical properties of particulate pollutants on the Korean Peninsula, An Aerodyne High Resolution Time of Flight Aerosol Mass Spectrometer (HR-ToF-AMS) measured non-refractory fine particle ($NR-PM_1$) from 2013 to 2015 at Baengnyeong Island and Seoul metropolitan area (SMA), Korea. The chemical composition of $NR-PM_1$ in Baengnyeong island was dominated by organics and sulfate in the range of 36~38% for 3 years, and the organics were the dominant species in the range of 44~55% of $NR-PM_1$ in Seoul metropolitan area. The sulfate was found to be more than 85% of the anthropogenic origin in the both areas of Baengnyeong and SMA. Ratio of gas to particle partition of sulfate and nitrate were observed in both areas as more than 0.6 and 0.8, respectively, representing potential for formation of additional particulate sulfate and nitrate. The high-resolution spectra of organic aerosol (OA) were separated by three factors which were Primary OA(POA), Semi-Volatility Oxygenated Organic Aerosol (SV-OOA), and Low-Volatility OOA(LV-OOA) using positive matrix factorization (PMF) analysis. The fraction of oxygenated OA (SOA, ${\fallingdotseq}OOA$=SV-OOA+LV-OOA) was bigger than the fraction of POA in $NR-PM_1$. The POA fraction of OA in Seoul is higher than it of Baengnyeong Island, because Seoul has a relatively large number of primary pollutants, such as gasoline or diesel vehicle, factories, energy facilities. Potential source contribution function (PSCF) analysis revealed that transport from eastern China, an industrial area with high emissions, was associated with high particulate sulfate and organic concentrations at the Baengnyeong and SMA sites. PSCF also presents that the ship emissions on the Yellow Sea was associated with high particulate sulfate concentrations at the measurement sites.

Purpose: The long-term administration of antibiotics interferes with bacterial culture in the middle ear fluids (MEFs) of young children with otitis media with effusion (OME). The purpose of this study is to determine whether molecular diagnostics can be used for rapid and direct detection of the bacterial pathogen in culture-negative MEFs. Methods: The specificity and sensitivity of both polymerase chain reaction (PCR) and loop-mediated isothermal amplification (LAMP) to the lytA gene of Streptococcus pneumoniae were comparatively tested and then applied for pneumococcal detection in the clinical MEFs. Results: The detection limit of the PCR assay was approximately $10^4$ colony forming units (CFU), whereas that of LAMP was less than 10 CFU for the detection of S. pneumoniae. Both PCR and LAMP did not amplify nucleic acid at over $10^6$ CFU of H. influenzae or M. catarrhalis, both of which were irrelevant bacterial species. Of 22 culture-negative MEFs from children with OME, LAMP positivity was found in twelve MEFs (54.5%, 12/22), only three of which were PCR-positive (25%, 3/12). Our results showed that the ability of LAMP to detect pneumococcal DNA is over four times higher than that of PCR (P<0.01). Conclusions: As a high-resolution tool able to detect nucleic acid levels equivalent to <10 CFU of S. pneumoniae in MEFs without any cross-reaction with other pathogens, lytA -specific LAMP may be applied for diagnosing pneumococcus infection in OME as well as evaluating the impact of a pneumococcal conjugate vaccine against OME.

Fusaric acid (FA) is a mycotoxin produced by Fusarium species. Its toxicity is relatively low but often associated with other mycotoxins, thus enhancing total toxicity. To date, biosynthetic genes or enzymes for FA have not been identified in F. oxysporum. In order to explore the genetic element(s) for FA biosynthesis, restriction enzyme mediated integration (REMI) procedure as an insertional mutagenesis was employed using FA producing-F. oxysporum strains. Genetic transformation of two F. oxysporum strains by REMI yielded more than 7,100 transformants with efficiency of average 3.2 transformants/${\mu}g$ DNA. To develop a screening system using phytotoxicity of FA, eleven various grains and vegetable seeds were tested for germination in cultures containing FA: Kimchi cabbage seed was selected as the most sensitive host. Screening for FA non-producer of F. oxysporum was done by growing each fungal REMI transformant in Czapek-Dox broth for 3 weeks at $25^{\circ}C$ then observing if the Kimchi cabbage seeds germinated in the culture filtrate. Of more than 5,000 REMI transformants screened, fifty-three made the seeds germinated, indicating that they produced little or fewer FA. Among them, twenty-six were analyzed for FA production by HPLC and two turned out to produce less than 1% of FA produced by a wild type strain. Sequencing of genomic DNA regions (252 bp) flanking the vector insertion site revealed an uncharacterized genomic region homologous (93%) to the F. fujikuroi genome. Further study is necessary to determine if the vector insertion sites in FA-deficient mutants are associated with FA production.

This study was carried out to investigate polymorphism, gene diversity, and geographical relationships of 81 Korean wild (Glycine soja) and 130 cultivated soybeans (G. max) using seven simple sequence repeat (SSR) markers. A total of 144 alleles were observed in 211 accessions with an average of 20.6. Each SSR loci showed 13 (Satt532) to 41 (Sat_074) multialleles. The range of alleles within the loci was wider in wild soybean than the cultivated soybeans. The average genetic diversity values were 0.88 and 0.69 in wild and cultivated soybeans, respectively. In a scatter diagram of wild and cultivated soybeans based on canonical discriminant analysis, CAN1 accounted for 84.2% while CAN2 did 8.5%. Two species were grouped into three: group I (G. max), group II (G. soja), and group III (complex of G. max and G. soja). The geographical relationships of wild soybean were distinguished into two groups: Gyeonggi for Group I, and Gyeongsang, Jeolla, Gangwon, and Chungcheong for Group II. Those of cultivated soybeans were distinguished into Gyeonggi, Gangwon, and Gyeongsang for Group I, and Jeolla and Chungcheong for Group II. Therefore, the geographical relationships of wild soybeans were well typified based on the ecosystems of the Korean peninsula.
Bile formation is a complex process comprised of three separate physiologic mechanism operating at two anatomical sites. At present time, it was known that at least two processes are responsible for total canalicular secretion at the bile canaliculus. One of the processes is bile salt-dependent secretion (BSDS) hypothesis that the active transport of bile salts from plasma to bile provided a primary stimulus for bile formation: the osmotic effect of actively transported bile acid was responsible for the movement of water and ions into bile. The other process is bile salt-independent secretion (ESIS), which is unrelated to bile salt secretion at the canaliculus and which may involve the active transport of sodium. The third process for bile formation involves the biliary ductal epithelium. Secretin-stimulated bile characteristically contained bicarbonate in high concentration. Therefor, it was suggested that secretin stimulated water and bicarbonate secretion from the biliary ductules. One the other hand, it was found that a large amounts of cAMP was present in canine bile but no apparent relationship between bile salt secretion and cAMP content in dog bile. However, bile flow studies in human have demonstrated that secretin and glucagon increase bile cAMP secretion as does secretin in baboons. Secretin increases baboon bile duct mucosal cAMP levels in addition to bile CAMP levels suggesting that in that species secretin-stimulated bile flow may be cAMP mediated. It has been postulated that glucagon and theophylline which increase the bile salt-independent secretion in dogs might act through an increased in liver cAMP content. In a few studies, the possible role of cAMP on bile formation has teen tested by administration of an exogenous derivative of cAMP, dibutyryl cAMP. In the rat, DB cAMP did not modify bile flow, but injection of DB cAMP in the dog promoted an increase in the bile salt-independent secretion. Because of these contradictory results, this study was carried out to examine the relationship between cyclic nucleotides and bile flow due to various bile salts as well as secretin or theophylline. Experiments were performed in rabbits with anesthesia produced by the injection of seconal(30 mg/kg). Rabbits had the cystic duct ligated and the proximal end of the divided common duct cannulated with an appropriately sized polyethylene catheter. A similar catheter was placed into the inferior vena cava for administration of drugs. Bile was collected for determination of cyclic nucleotides and total cholate in 15 min. intervals for a few hours. The results are summerized as followings. 1) Administrations of taurocholic acid or chenodeoxycholic acid increased significantly the concentrations of cAMP and cGMP in bile of rabbits. 2) Concentration of cAMP in bile during the continuous infusion of ursodeoxycholic acid, was remarkedly increased in accordance with the increase of bile flow, while on the contrary concentration of cGMP in bile was decreased significantly. 3) Dehydrocholic acid and deoxycholic acid significantly increased bile flow, total cholate output and cyclic nucleotides in bile. 4) Only cAMP concentration in bile was significantly increased from control value by secretin, while theophylline increased cAMP as well as cGMP in rabbit bile. 5) In addition, the administration of secretin to taurocholic acid-stimulated bile flow increased cAMP while theophylline produced the increases of cAMP and cGMP in bile. 6) The administration of insulin to taurocholic acid-stimulated bile flow decreased cAMP concentration, while on the contrary cGMP was remarkedly increased in rabbit bile.
When we investigated seed infestation by fungal pathogens from 51 varieties in 9 crops, the contamination rate of rice and sesame seeds was high. Therefore, to control seed-borne diseases, we obtained extracts from commercial products of Kimchi, Gochujang, Doenjang, Ganjang, Makgeolli and Tohajut and their suppressive effects against seed-borne diseases were studied. In addition, bacterial strains were screened to control rice seed-borne diseases in vitro and in vivo. Among forty food extracts, eleven food-extracts suppressed incidence of seedling rots in vitro and five food extracts increased 8-33% of healthy seedling in the greenhouse. Among 218 isolates from 40 fermented foods, 43 isolates showed high antifungal activity against seven fungal pathogens. When we tested 43 isolates for the reduction of rice seed borne disease, 32 isolates were able to reduce the rice seed borne disease. Among 32 isolates, 17 isolates reduced significantly seedling rot and increased healthy seedlings, the other isolates except for Kc4-2 and Mkl 2-2 increased shoot emergence and the percentage of healthy plants. Thirty isolates with high antifungal activity and suppressive effect against rice seedling rots were identified by 16S rRNA sequencing. Twenty one of thirty isolates were identified as Bacillus spp. Three isolates from Makgeolli were identified as Saccharomyces cerevisiae. B. amyloliquifaciens were isolated from six Korean traditional fermented foods except for Ganjang. B. amyloliquifaciens were majority in the effective bacterial population of Gochujang and Jutgal. Relatively diverse Bacillus species including B. subtilis, B. pumilus, and B. amyloliquificiens were isolated from Kimchi. The selected effective microorganisms from Korean fermented foods founded to be effective for controlling seed-borne diseases of rice in vitro and in the greenhouse. We think that Korean fermented foods and their useful microorganisms can be used as biocontrol agents for suppressing rice seed-borne diseases based on above described results.

This experiment was conducted to compare forage production and feed value of winter forage crops at the experimental field of Livestock Division, National Institute of Subtropical Agriculture from 2007 to 2008. The experiment was arranged in a randomized block design with three replications. The five winter forage crops used in this experiment were barely ('Youngyang' and 'Yuyeon'), oats ('Samhan' and 'Swan'), wheat ('Kumkang' and 'Woori'), triticale ('Shinyoung') and italian ryegrass ('Hwasan 101' and 'Florida-80'). The species having the fastest heading date was wheat (5th April) while the heading date of oats ('Samhan') was the latest as 28th April. The dry matter (DM) content of wheat was the highest among the five winter forage crops. Italian ryegrass ('Hwasan 101') showed the highest CP content by 11.2% and oats (Swan) showed the lowest content by 9.8%. Average acid detergent fiber (ADF) and neutral detergent fiber (NDF) content were 28.6 and 50.8% respectively. The five winter forage crops have narrow range of total nutrient digestibility (TDN) from 64.0% to 69.1%. Italian ryegrass ('Hwasan 101') showed the highest DM yield by 25,951 kg/ha, but the DM yield of barely and wheat low by 13,892 kg/ha ('Yuyeon') and 14,815 kg/ha ('Woori'), respectively. Italian ryegrass and oats have higher forage yield and feed value in Jeju. Also, oats has high grain and forage yield. The results demonstrated oats can cultivate as alternative winter forage crop for whole crop silage barely in Jeju.
