  • Effect of PCBs on the skin and gill development of olive flounder, Paralichthys olivaceus were investigated by histological methodology and morphometric data. The olive flounder were exposed to limit concentration of effulent of PCBs (3.0 ${\mu}g/L$) for 60 days. Skin development can be classified into four stages: SSEL (simple squamous epithelial layer), MCA (mucous cell appearance) & CCA (club cell appearance) DLA (dermal layer appearance), and SEL (startification of epidermal layer) stages. Gill development had five stages: GFA (gill filament appearance), IGFE (identification of early gill filament epithelial cell) MCA (mucous cell appearance), PCA (pillar cell appearance), and FGL (formation of gill lamella) stages. The periods of structural completion of skin and gill were 22-30 days and 23-30 days after hatching in the exposure group respectively. The process of development of skin and gill of the exposure group was very similar to that of the control group. Therefore, PCBs (3 ${\mu}g/L$) have no influence on the development of skin and gill in the larval stage of olive flounder.

  • Objectives The purpose of this study is to confirm the effect of alleviating inflammation through creating skin fat barrier from Scutellaria baicalensis extract. Methods Four-week-old Balb/C mice were divided into four groups: control group (Ctrl), lipid barrier eliminated group (LBEG), dexamethasone (DM) treated group after lipid barrier elimination (DMTG), and Scutellaria baicalensis (SB) treated group after lipid barrier elimination (SBTG). Scutellaria baicalensis extract were administered for 5 days after removal of the fat barrier. Changes in skin condition, improvement of the fat barrier, and relief of inflammation were observed in each group. Results Compared to LBEG and DMTG, pathological skin damage and tissue changes were less in SBTG, and transepidermal water loss (TEWL) and pH were also significantly reduced. Filaggrin was also significantly increased in SBTG. KLK7, PAR-2, and TSLP in SBTG also showed significant reduction compared to the LBEG and DMTG. Conclusions Scutellaria baicalensis extract restores skin barrier and relieves inflammation through the creation of skin fat barrier. This means that the Scutellaria baicalensis extract can regulate Th2 differentiation through the creation of the epithelial fat barrier.

  • The transdermal therapeutic system (TTS) of scopolamine has various advantages over its oral dosage forms. The ideal scopolamine TTS requires high skin permeation rate in short time after it is applied on the skin. In order to increase the initial skin permeation rate of scopolamine from TTS, various permeation enhancers were employed. Enhancers employed were fatty acids (oleic and linolenic acids), cyclic monoterpenes (menthol, camphor, cineole and limonene) and others (isopropyl myristate, sodium lauryl sulfate and glyceryl monostearate). The concentration of enhancers in the base were fixed to 5% (w/w). While fatty acids had little enhancing effect on the skin permeation of scopolamine, cyclic monoterpenes, isopropyl myristate and sodium lauryl sulfate resulted in $1.5{\sim}2.6-fold$ higher skin permeation rate of the drug compared to the control. However, lag time was not affected by enhancers studied.

  • Polyphenolic compounds have been reported to exhibit a wide range of pharmacological properties. In this study. we investigated the hepatoprotective effect of skin and seed of grape which contain abundant polyphenol compounds on dimethylnitrosamine(DMN)-induced liver damage in rats. Ingestion of skin and seed of grape (10% diet, daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase, and bilirubin levels. • To evaluate the effect of antioxidant on the cytotoxicity induced by oxidative stress of reactive oxygen species (ROS) in cultured human skin melanocytes, colorimeric assay of XTT and tyrosinase activity assay were adopted after human skin melanocytes were preincubated for 2 hours in the media containing various concentrations of superoxide dismutase (SOD) before the treatment of hydrogen peroxide. Light microscopic study was carried out in same cultures. The results of this study were as follows 1. Cell viability of human skin melanocytes was significantly decreased by 30 and $40{\mu}M$ of hydrogen peroxide($H_2O_2$), respectively. 2. XTT50 was determined at $30{\mu}M$ after human skin melanocytes were treated with $10{\sim}40{\mu}M$ of hydrogen peroxide for 6 hours. 3. The cell viability of cultured human skin melanocytes pretreated with SOD was increased than that of cultured human skin melanocytes treated with $H_2O_2$ dose-dependently. 4. In tyrosinase activity of human skin melanocytes, the cell treated with SOD showed brown stain compared with $H_2O_2$ treated cells, dark stain. 5. In light microscopy, cultured human skin melanocytes exposed to $H_2O_2$ showed morphological changes such as the decreased cell number and cytoplasmic processes, compared with control. 6. In light microscopy, cultured human skin melanocytes pretreated with SOD showed the increase of cell number and cytoplasmic processes compared with $H_2O_2-treated$ group. From these results, it is suggested that oxidative stress of ROS such as $H_2O_2$ has cytotoxicity by showing the decreased cell viability, the increased tyrosinase activity and mophological changes of the decreased cell number and cytoplasmic processes. While, antioxidant like SOD was effective in the prevention of oxidative stress-mediated cytotoxicity by the increased cell viability, decreased tyrosinase activity and the protection of degenerative morphological changes in cultured human skin melanocytes.
