Analysis of Residual Solvents of [F-18]FDG Using Gas Chromatography (기체크로마토그래프법을 이용한 [F-18]FDG의 잔류용매 분석)

  • Purpose: The general test method of the Korean Pharmacopeia specifies the test method on the clauses of quality control after manufacturing. According to KFDA Guidance for Medicines, standards of residual solvents regulates the maximum permissible dose of acetonitrile as 400 ppm, ethanol as 5,000 ppm, and acetic acid as 5,000 ppm. This study aims at identifying the type of resiual solvents in the final [F-18]FDG vial of an automatic synthesizer and measure its residual quantity. Materials and Methods: The center carried out residual solvents test of [F-18]FDG injection using Agilent Technologies 7890A with a Flame Ionization Detector. The column of Agilent Technologies 7890A used in measuring of residual solvents was CP WAX column ($30m{\times}0.53mm{\times}1.0{\mu}m$) and analysis condition was split mode 1:1 at the initial temperature $70^{\circ}C$ which was increased $20^{\circ}C/minute$ after two minutes and maintained at the final $140^{\circ}C$ for two minutes. The analysis method was as following: Firstly, ethanol-acetonitrile-acetic acid mixture was classified into four types of concentration (250-25-250 ppm, 1,000-100-1,000 ppm, 3,000-300-3,000 ppm, and 6,000-600-6,000 ppm), and $1.0{\mu}L$ of each type of concentration was injected into gas chromatography followed by an analysis of its peak domain. Then, a calibration-curve by the external standard method was drawn based on the analysis result. Results: While ethanol and acetonitrile were detected in TRACERlab MX, FASTlab had additional acetic acid. The residual quantity of the ethanol-acetonitrile-acetic acid mixture evaluated using the calibration-curve was average 72 ppm ethanol, 54 ppm acetonitrile, and 1030 ppm acetic acid for FASTlab, whereas average 439 ppm ethanol and 79 ppm acetonitrile for TRACERlab MX. This indicated that both of them were within the maximum permissible dose. Conclusion: Solvent residues in the [F-18]FDG injection were all within maximum permissible doses and proper to be used to examine a patient. The result indicated that types and quantities of solvent resides of radioactive pharmaceuticals vary depending on the automatic synthesizer.

  • Massive Multiplayer Online Role Playing Games(MMORPGs) headed by some Korean game companies such as NC Soft, NHN, and Nexon have exploded in recent years. However, it becomes one of the major challenges for the MMORPG developers to design their games to appeal to gamers since only a few MMORPGs succeed whereas they require a huge amount of initial investment. Under this background, our study derives the major elements for designing MMORPG from the literature, and identifies the ones critical to the users' satisfaction and their willingness to pay among the derived elements. Though most previous studies on the design elements of MMORPG have used analytic hierarchy process(AHP), our study adopts artificial neural network(ANN) as the tool for identifying key success factors in designing MMORPG. The results of our study show that the elements of the game contents quality have a bigger effect on the user's satisfaction, whereas the ones of the value-added systems have a bigger effect on the user's willingness to pay. They also show that user interface affects both the user's satisfaction and willingness to pay most. These results imply that the strategies for the development of MMORPG should be aligned with its goal and market penetration strategy. They also imply that the satisfaction and revenue generation from MMORPG cannot be achieved without convenient and easy control environment. It is expected that the new findings of our study would be useful forthe developers or publishers of MMORPGs to build their own business strategies.

Development of Analytical Reference Material for Proficiency Test of Pesticide Multi-residue Analysis in Green-pepper (풋고추 농약다성분분석 정도관리용 분석표준물질 개발)

  • This study was to develop the analytical reference material of green-pepper for multi-residue analysis of pesticides. According to the ISO Guide 35, ISO Guide 13528 and EURL-PT protocol, the homogeneity, stability, assigned value and uncertainty were calculated to assess if it was suitable to be used as the proficiency test or quality control. The values of the within-bottle standard variation ($s_{wb}$) and the between-bottle standard variation ($s_{bb}$) were 1.7~3.7% of assigned value according to the requirement of the ISO guide 35. And, the uncertainty ($u^*{_{bb}}$) due to inhomogeneity was 0.8~1.1% for all pesticides. The storage stabilities of ten-pesticides at various conditions were assessed. For all target pesticides, the slop ($b_1$) values were smaller than the corresponding values of $[t_{0.95,n-2}{\times}s(b_1)]$ specified by the ISO guide 35, indicating that there were no statistically significant decreases in the concentration of the target pesticides when the analytical reference material was stored at room temperature ($20{\sim}30^{\circ}C$) for 7 days, freezing ($-20^{\circ}C$) for 30 days and deep freezer ($-80^{\circ}C$. except for bifenthrin, fenpropathrin) for 245 days. For proficiency test by using it developed by Korea Institute of Toxicology, inter-lab test was performed with eight organization performing the residual pesticide analysis. We found that there were some different results among them. Some were assessed as questionable or unacceptable for two pesticides and one organization didn't analyze the six pesticides. From these results, this green-pepper analytical reference material containing ten-pesticides could be used as a tool for the proficiency test to improve the reliability or consistency for pesticide residue's results.

Fertility Status in Northeastern Alpine Soils of South Korea with Cultivation of Vegetable Crops (강원도 고랭지 채소 재배지의 토양 비옥도관리 현황과 전망)

  • Total upland area for cultivating the vegetable crops in the Alpine soils of Northeastern South Korea has been extending its limit to meet the increasing demand of vegetable food in recent decades. About 70% of these alpine soils are located in over 7% of the slope and most of vegetable crops have been cultivated intensively without practicing the best management systems. Thus, soil erosion and continuous cropping system have degenerated the soil fertility and shown detrimental effects on water quality. We initiated an intensive and extensive investigation to characterize the fertility problems encountered in these uplands. Objectives of this paper were to characterize the fertility status in the Alpine soils cultivated with vegetable crops for many years and to provide the recommendations for adequate soil management measures including fertilization and erosion control. Soils in general have good drainage with textural classes of loam or sandy loam. Their topographical characteristics tended to lead them to shallow plow layers, and the steepness of the terrain created erosion hazard. Of the soils examined, about 11% of uplands over 30% gradient was found in need of an urgent reforestation. A high content of gravel and firm hardness of soil attributed to inhibit the utilization of farm machinery and plant-root development. The average soil pH 5.6 was slightly low relative to pH 5.70 of the national average. Organic matter content was high compared with 2.0% of national average, but decreased with the prolonged cultivation periods. Available $P_2O_5$ concentration was unusually high due to the consequence of over dose application with chemical and organic fertilizers. Exchangeable cations as Ca, Mg, and K were appeared to be decreased in these regions with prolonging the cultivation periods. There were no significant differences in cation exchange capacity (CEC) and electrical conductivity (EC) among locations. Heavy metal contents were mostly lower than the threshold of danger level designated by Soil Environment Conservation Law of South Korea. Results indicated that a proper countermeasure and the best management practice should be immediately implemented to conserve the top soil and fertility in the Alpine regions.

Preservation of Strawberries and Cucumbers Packaged by Low density polyethylene film impregnated with antimicmbial agent, Scutellariae baicalensis extract (황금추출물을 함유한 항균성 포장필름을 이용한 딸기와 오이의 저장효과)

  • To develop a wrapping film, which suppresses the microbial decay through the storage and prolongs the selflife of fruits and vegetables, the antimicrobial packaging films were prepared and applied to the preservation of strtwberries and cucumbers. Low density polyethylene(LDPE) film of 50㎛ thickness was faricated with 1% of Scutellariae baicalensis extract. The LDPE film impregnated with Scutellariae baicalensis extract showed antimicrobial activity on the disk test against Bacillus cereus, Escherchia coli and Fusarium sp.. The antimicrobial film changed the color and light transmittance, but did not affect heat shrinkage, mechanical tensile strength and wattability. Strawberries and cucumbers were separately wrapped with packaging films in the state of closely-adhered packaging as well as modified atmosphere packaging(MAP). The wrapped strawberries and cucumbers were stored for 21 days at 5$\^{C}$ and for 40 days at l0$\^{C}$, respectively. For the packaged strawberries and cucumbers at 5$\^{C}$ and 10$\^{C}$, the LDPE film impregnated with Scutellariae baicalensis extract showed the reduced growth of total aerobic bacteria, molds and yeasts and did not give any negative effect on other quality attributes during storage in comparison with conttrol film without any additive.

Analysing the effect of impervious cover management techniques on the reduction of runoff and pollutant loads (불투수면 저감기법의 유출량 및 오염부하량 저감 효과 분석)

  • Impervious covers(IC) are artificial structures, such as driveways, sidewalks, building's roofs, and parking lots, through which water cannot infiltrate into the soil. IC is an environmental concern because the pavement materials seal the soil surface, decreasing rainwater infiltration and natural groundwater recharge, and consequently disturb the hydrological cycle in a watershed. Increase of IC in a watershed can cause more frequent flooding, higher flood peaks, groundwater drawdown, dry river, and decline of water quality and ecosystem health. There has been an increased public interest in the institutional adoption of LID(Low Impact Development) and GI(Green Infrastructure) techniques to address the adverse impact of IC. The objectives of this study were to construct the modeling site for a samll urban watershed with the Storm Water Management Model(SWMM), and to evaluate the effect of various LID techniques on the control of rainfall runoff processes and non-point pollutant load. The model was calibrated and validated using the field data collected during two flood events on July 17 and August 11, 2009, respectively, and applied to a complex area, where is consist of apartments, school, roads, park, etc. The LID techniques applied to the impervious area were decentralized rainwater management measures such as pervious cover and green roof. The results showed that the increase of perviousness land cover through LID applications decreases the runoff volume and pollutants loading during flood events. In particular, applications of pervious pavement for parking lots and sidewalk, green roof, and their combinations reduced the total volume of runoff by 15~61 % and non-point pollutant loads by TSS 22~72 %, BOD 23~71 %, COD 22~71 %, TN 15~79 %, TP 9~64 % in the study site.

Physicochemical Characteristics of Yanggaeng with Pear Juice and Dried Pear Powder Added (배즙과 배 건조분말을 첨가한 양갱의 물리화학적 특성)

  • This study was conducted to investigate the physicochemical characteristics of yanggaeng prepared with pear juice and dried pear powder. The proximate composition and total polyphenol content, antioxidant activities, Hunter's color values, texture, sensory properties of pear yanggaeng variants [pear juice (PJ), pear juice and hot-air-dried pear powder (PJH), pear juice and freeze-dried pear powder (PJF) yanggaeng] were examined. The moisture content was highest (27.9%) in the control (C) yanggaeng, but the latter's energy and carbohydrate content were lower than in the pear yanggaeng variants. The differences between the pear yanggaeng variants were not significant.The total polyphenol content was highest in the 20.7 mg/100g PJF yanggaeng. The antioxidant activities of PJF yanggaeng were higher than those of the other pear yanggaeng varients. The Hunter color value results showed that the lightness (L) values were highest in C yanggaeng whereas the redness(a) and yellowness(b) values were highest in PJH yanggaeng. The springiness in texture was lowest in C yanggaeng and not significant in the others. The hardness and chewiness were highest in PJH and PJF yanggaeng, but adhesiveness was lowest therein. The results of the sensory test showed that PJF yanggaeng was the highest in flavor, color, taste, hardness, chewiness, and overal quality. Based on these results, it can be concluded that pear juice and freeze-dried pear powder yanggaeng has excellent physicochemical and antioxidant activities.

Effects of Bambusae Caulis in liquamen on inhibition of angiotensin-converting enzyme in vitro and blood flow in mouse model (죽력의 in vitro에서의 angiotensin-converting enzyme 저해효과와 마우스모델에서 혈행개선효과)

  • The purpose of this study was to investigate the effects of the Bambusae Caulis in liquamen (BCL) on blood circulation in animal models. Previous studies on BCL have shown effects on thrombolytic activity and angiotensin-converting enzyme (ACE) inhibitory activity. In the mouse model, the triglyceride content were 301.5 mg/dL in the high fat diet+BCL II 0.01% group, 289.2 mg/dL in the high fat diet+BCL II 0.05% group, which was significantly lower than the high fat diet group. The total cholesterol content was 311.9 mg/dL in high fat diet+BCL II 0.01% and 293.7 mg/dL in high fat diet+BCL II 0.01% 0.05%, respectively, which was significantly lower than the high fat diet group. The HDL-cholesterol level was 206.0 mg/dL for the high fat diet, 196.6 mg/dL for the high fat diet+BCL II, and 189.2 mg/dL for the high fat diet+BCL II. There was no significant difference between the 0.01% and 0.05% groups. The high-fat diet+0.05% group was significantly improved in the blood flow compare to the high fat diet and the high fat diet+0.01% group. Platelet aggregation inhibition ability was inhibited in the high fat diet+0.01% and 0.05% groups compared to the high fat diet group.

A Study on the Selection of Evaluation Indicators of Amenities (국토 어메니티 평가지표 개발)

  • This study aims to, first, classify the approach system of utilizing national and local amenities into three groups: evaluation of the level of the amenity, evaluation of the value of amenity resources, and utilization of amenity resources. Secondly, the study aims to derive an evaluation index which is required according to the approach system. As for a strategy of utilizing amenities, the self-diagnosis based on the evaluation of the level of local amenities is required. In this study, three sectors, ten items and 43 detailed items have been derived though specialists, FGIs(Focus Group Interview), and the cases within and outside of the country. In the survey with specialists, a unit-space key index which can be used in accordance with the unit-space characteristics(such as urban and rural area, fishing villages, mountain villages, and littoral areas), environmental area, important index and optional index have been derived. As for the evaluation index which is needed for the development of typical local amenities resources, 14 detailed evaluation indexes from three sectors have been derived. These have been classified again into six key indexes, six important indexes and two optional indexes. In the evaluation index of amenity resources, natural and environmental value-such as ecological value of preservation, uniqueness of aesthetic landscape and ecological value of restoration-historic and cultural value of preservation and individuality of the resources, and aesthetic and cultural values are derived as being important. As for utilization of amenity resources, 15 items from three sectors-such as reservation, restoration, intentional use, and industrial use-have been derived. Also, through a survey with specialists, key indexes and important indexes were derived by unit-space. As a result, in urban areas, seven items-including creative development of space, aesthetic landscape control and development of cultural contents-have been derived as key indexes; in rural areas, mountain villages, fishing villages and littoral areas, eight items-including brand of region, brand of the products, and brand of tourism resources-have been derived. In environmental areas, six items-including reservation of natural resources, historic and cultural heritage, and restoration of ecosystem-have been derived. Indexes derived from this study can be utilized in order to establish a strategy for amenity plans and an implementation strategy, but can be readjusted, allowing for various regional characteristics. It is, thus, advisable that local governments selectively utilize indexes according to regional characteristics or newly develop them for the maximum utilization of regional amenities.

A Study of Working Environments and Workers Exposed to Chromium (크롬 취급 사업장의 작업환경 및 근로자 폭로 실태에 관한 연구)

  • For the purpose of providing the basic data for health management of workers who are exposed to chromium and for improving the quality of working environment, the authors evaluated blood and urinary level of chromium, the occupational history. AST, ALT, Hb, Hct, nasal specular examinaton on 287 workers who have been dealed chromium compounds in 56 manufacturing Industries of five types, that is, 38 metal plating services(plating),4 manufacture of other fabricated metal products (fabricated metal product), 5 manufacturing of dyestuff(dyestuff), 6 dressing and dyeing of leather(leather), 3 others(manufacture of pottery and ceramic househod wares, motor vehicles, electronic valves and tubes and other electronic components) and also measured the level of chromium in air from February to October 1993. The results were as follows ; 1. The utilized type of chromium compounds was the hexavalent state in plating fabricated metal product dyestuff, leather and the trivalent state in .other, and atmosperic chromium concentration as geometric mean was $0.0138mg/m^3(0.001{\sim}0.068mg/m^3)$ in plating, $0.0115mg/m^3(0.006{\sim}0.015mg/m^3)$ in fabricated matal product, $0.068mg/m^3(0.002{\sim}0.019mg/m^3)$ in dyestuff, $0.0083mg/m^3(0.002{\sim}0.028mg/m^3)$ in leather $0.0039mg/m^3(0.003{\sim}0.005mg/m^3)$ in other by the type of industry and it exceeded TLV-TWA ($0.05mg/m^3$) in five (13.6%) of plating services. 2. The geometric mean of chromium in blood was $1.54{\mu}g/dl(0.10{\sim}3.62{\mu}g/dl)$ in Plantng, $0.94{\mu}g/dl(0.27{\sim}2.82{\mu}g/dl)$ in fabricated metal product, $0.51{\mu}g/dl(0.10{\sim}3.25{\mu}g/dl)$ in dyestuff, $0.87{\mu}g/dl(0.15{\sim}8.00{\mu}g/dl)$ in leather, $0.55{\mu}g/dl(0.20{\sim}2.28{\mu}g/dl)$ in other by the type of industry(p<0.001). 3. The geometric mean of chromium in urine was $14.47{\mu}g/l(6.90{\sim}28.00{\mu}g/l)$ in planting, $4.63{\mu}g/l(0.24{\sim}43.00{\mu}g/l)$ in fabricated metal product, $5.93{\mu}g/l(1.00{\sim}33.00{\mu}g/l)$ in dyestuff, $11.09{\mu}g/l(0.80{\sim}48.00{\mu}g/l)$ in leather, $12.41{\mu}g/l(10.10{\sim}41.00{\mu}g/l)$ in other by the type of industry(p<0.001). 4. As the result of nasal specular examination, twenty four cases (8.4%) of nasal septal perforation among 287 total subjects was observed, and there were 17 (9.7%) cases in plating, 4 csaes (14.3%) in dressing and dyeing of leather. In the comparison of chromium concentration in blood and urine between the perforated group and non-perforated group, the perforated group showed a significantly higher value as $1.883{\pm}3.055{\mu}g/dl\;and\;0.793{\pm}0.815{\mu}g/dl$(P<0.001), $21.31{\pm}34.610{\mu}g/L\;and\;9.304{\pm}11.079{\mu}g/L$ (P<0.001). 5. The mean concentration of chromium in blood, urine and the mean level of AST, ALT, Hb and Hct in exposure group were higher than those of control group(p<0.001).
