Experimental Analysis of a Supersonic Plasma Wind Tunnel Using a Segmented Arc Heater with the Power Level of 0.4 MW (0.4 MW 급 분절형 아크 히터를 이용한 초음속 플라즈마 풍동 특성 실험)

  • Experimental analyses on a supersonic plasma wind tunnel of CBNU (Chonbuk National University) were carried out. In these experiments, a segmented arc heater was employed as a plasma source and operated at the gas flow rates of 16.3 g/s and the total currents of 300 A. The input power reached ~350 kW with the torch efficiency of 51.4 %, which is defined as the ratio of total exit enthalpy to the input power. The pressure of plasma gas in the arc heater was measured up to 4 bar while it was down to ~45 mbar in a vacuum chamber through a Laval nozzle. During this conversion process, the generated supersonic plasma was expected to have a total enthalpy of ~11 MJ/kg from the measured input power and torch efficiency. In addition to the measurement of total enthalpy, a cone type probe was inserted into the supersonic plasma flow in order to estimate the angle between shock layer and surface of the probe. From these measurements, the temperature and the Mach number of the supersonic plasma were predicted as ~2,950 K and ~3.7, respectively.

  • In Bongseon-ri site, the central type and the local type of the Baekje tombs co-exist together. Many swords with ring pommel which to regional leaders in Seocheon the Beekje government granted are excavated from these tombs. These represent that the regional leaders were controlled by the Baekje government gradually. Four swords with ring pommel and one simple sword are investigated metallurgically and we seek what swords have a function of weapon or what sword produce for grave goods. The simple sword is made by solid carburizing technology and quenching of heat treatment so it's possibly used as a weapon. In contrast, four swords with ring pommel has little chance of weapon because the blade of swords consists of microstructure that have low hardness and the back of swords don't have microstructure can absorb the shock. The most identical characteristic of the simple sword and swords with ring pommel is that they are possible to classify clearly into practical/non-practical tools. Additionally, according to non-metallic inclusion analysis result of these swords, w$\ddot{u}$stite and glass phase are found together. Thus, it is able to say that reduction method in low temperature is applied during smelting process.

  • The development of industry and the increase in the use of fossil fuels have accelerated the process of global warming and climate change, resulting in more frequent and intense natural disasters than ever before. Since electricity facilities are often installed outdoors, they are heavily influenced by natural disasters and the number of related accidents is increasing. In this paper, we analyzed the statistical status of domestic electrical fires, electric shock accidents, and electrical equipment accidents and hence analyzed the risk associated with climate change. Through the analysis of the electrical accidental data in connection with the various regional (metropolitan) climatic conditions (temperature, humidity), the risk rating and charts for each region and each equipment were produced. Based on this analysis, a basic electric risk prediction model is presented and a method of displaying an electric hazard prediction map for each region and each type of electric facilities through a website or smart phone app was developed using the proposed analysis data. In addition, efforts should be made to increase the durability of the electrical equipment and improve the resistance standards to prevent future disasters.

Artificial Spawning, Larval and Spat Developments of the Bay Scallop, Argopecten irradians (해만가리비, Argopecten irradians의 인공산란과 유생 및 치패의 발생)

  • Artificial spawning, larval and spat developments of the bay scallop, Argopecten irradians, which was transplanted from China on 16 August 1996, were investigated monthly until August 1997 in the Deukyang Bay, Jangheung-gun, Jeollanam-do, Korea. Sufficient amount of cultured microalgae supplemented seawater were supplied as food (6 kinds of phytoplankton) for fully grown adult individuals at 17.1-23.2$^{\circ}C$ for 44 days. A total of 45,320,000 eggs were spawned by way of 2 times of artificial spawning inductions such as exposure stimulus to the air and thermal shock (with water temperature) on 29 January and 31 January in 1997. Artificially fertilized eggs were developed to D-shaped larvae (77.5 ${\times}\;63.8\;{\mu}m)$ and metamorphosed to larvae (191.8 ${\times}\;181.2\;{\mu}m)$ 181.2 m) in the attached larval stage on the collectors. A total of 110,000 spats (average 3.04 mm in shell length) were produced at 22.8-26.3$^{\circ}C$ and 31.0-34.4 psu in the indoor rearing tank from 14 February through 7 May in 1997. In case of Argopecten irradians, if the attached larvae in the attachment stage are detached from the collector, they could not live. Accordingly, it is assumed that survival (%) of the attached larvae of A. irridians showed very low because of weak power attached to the collector due to the small number of the byssuses of the attached larva, not the short attachment period by the byssus as seen in other scallops such as Argopecten balloti.

Gene Structure and Altered mRNA Expression of Metallothionein in Response to Metal Exposure and Thermal Stress in Miho Spine Loach Cobitis choii (Cobitidae; Cypriniformes) (미호종개 metallothionein 유전자의 구조 및 중금속 노출과 고온 자극에 대한 MT mRNA의 발현 특징 분석)

  • Gene and promoter structures of metallothionein(MT) from Miho spine loach (Cobitis choii; Cypriniformes) were characterized, and the transcriptional responses to experimental exposures to heavy metals and heat stress were examined. The C. choii metallothionein displayed well-conserved features of teleostean metallothioneins at gDNA, mRNA and amino acid levels. Bioinformatic analysis predicted that the C. choii MT regulatory region potentially possessed various motifs or elements targeted by various transcription factors associated with metal-coordinating regulation (e.g., metal transcription factor-1), immune responses (e.g., nuclear factor kappa B), and thermal modulations (e.g., heat shock factor). Acute heavy-metal exposures to 0.5 or $1.0\;{\mu}M$ of cadmium (Cd), copper (Cu), manganese (Mn), nickel (Ni) or zinc (Zn) showed that MT transcription was significantly stimulated by Cd (9.6-fold relative to non-exposed control) and Cu (10.4-fold), only moderately by Mn (2.4-fold), but hardly by Ni and Zn. Elevation of water temperature from $25^{\circ}C$ to $31^{\circ}C$ caused a rapid modulation of MT mRNAs toward upregulation to 9.5-fold; however, afterward the elevated mRNA level slightly decreased during further incubation at $31^{\circ}C$ for 6 h. Results from this study suggest that MT-based expression assay could be a useful basis for better understanding the metal- and/or heat-caused stresses in this endangered fish species.

  • The application of high pressure on cellular morphology, proliferation and protein expression of Jurkat cells (human T lymphocyte cell line) has been extensively investigated. In the present study, we manufactured a novel pressure chamber that modulates 5% $CO_{2}$, temperature and pressure (up to 3 ATA). Jurkat cells was incubated 2 ATA pressure and analyzed cellular morphology and growth using an electron microscopy and MTT assay. The cells showed the morphological changes in the cell surface, which appeared to cause a severe damage in cell membrane. The growth rate of the cells under 2 ATA pressure decreased as cultured time got increased. Furthermore, a long term exposure of high pressure on Jurkat cells may act as one of the important cellular stresses that leads to inducing cell death. Cellular proteomes were separated by 2-dimensional electrophoresis with pH 3-10 ranges of IPG Dry strips. And many proteins showed significant up-and-down expressions with hyperbaric pressure. Out of all, 10 spots were identified significantly using matrix-assisted laser desorption/ionization-time of fight (MALDI-TOF) mass spectrometry. We and found that 9 protein expressions were decreased and one protein, heat shock protein HSP 60, was increased in Jurkat cells under 2 ATA. Identified proteins were related to lipid metabolism and signal transduction.

Quasi-monochromatic Parallel Radiography Achieved with a Polycapillary Plate

  • Fundamental study on quasi-monochromatic parallel radiography using a polycapillary plate and a plane-focus x-ray tube is described. The x-ray generator consists of a negative high-voltage power supply, a filament (hot cathode) power supply, and an x-ray tube. The negative high-voltage is applied to the cathode electrode, and the transmission type target (anode) is connected to the ground potential. The maximum voltage and current of the power supply were -100 kV (peak value) and 3.0 mA, respectively. In this experiment, the tube voltage was regulated from 20 to 25 kV, and the tube current was regulated by the filament temperature and ranged from 1.0 to 3.0 mA. The exposure time is controlled in order to obtain optimum film density, and the focal spot diameter was about 10 mm. The polycapillary plate is J5022-21 made by Hamamatsu Photonics Inc., and the outside and effective diameters are 87 and 77 mm, respectively. The thickness and the hole diameter of the polycapillary are 1.0 mm and 25 ${\mu}$m, respectively. The x-rays from the tube are formed into parallel beam by the polycapillary, and the radiogram is taken using an industrial x-ray film of Fuji IX 100 without using a screen. In the measurement of image resolution, we employed three brass spacers of 2, 30, and 60 mm in height. By the test chart, the resolution fell according to increases in the spacer height without using a polycapillary. In contrast, the resolution slightly fell with corresponding increases in the height by the polycapillary. In angiography, fine blood vessels of about 100 ${\mu}$m are clearly visible.

Semen Treatment to Maintain Good Quality of Post-thaw Motility and Viability of Canine Spermatozoa Frozen Using Methanol (Methanol을 이용한 개 정액 동결시 융해후 양호한 활력 및 생존율을 나타내는 정액 처리 조건)

  • This experiment was carried out to investigate the renditions to maintain good post-thaw motility and viability of canine spermatozoa when the semen was frozen using methanol. The semen from two male dogs which had been proven to be fertile in the previous one year was treated with different compositions of semen diluent and was frozen at different freezing temperatures, When canine semen was frozen at-2$0^{\circ}C$, -6$0^{\circ}C$, or -8$0^{\circ}C$, the spermatozoa frozen and stored at -2$0^{\circ}C$ showed very low post-thaw motility and viability from day 2 to 7 and showed no viability since day 15 after freezing. The spermatozoa frozen and stored at -6$0^{\circ}C$ or -8$0^{\circ}C$ showed higher post-thaw motility and viability on day 2, 1, 15 and 30 after freezing than that frozen and stored at-2$0^{\circ}C$(p<0.01), with no difference between two groups. Among different composition groups of the semen diluents of control(tris + egg yolk + glycerol), egg yolk-free, 히ycerol-free, and tris-free, Prior to freezing, the egg yolk-free diluent showed significantly love. motility and viability than the other diluents(p<0.05). On each thawing day (from day 2 to 15 after freezing), control group showed considerably higher motility and viability than the other groups(p<0.01). The canine spermatozoa frozen and stored at -6$0^{\circ}C$ and -8$0^{\circ}C$ showed gradual decrease of motility from day 2 to 30 after freezing and the spermatozoa of these two groups thawed on day 30 showed considerably love. motility than those thawed on day 2 after freezing, respectively(p<0.01). These results indicate that the freezing temperature of either -6$0^{\circ}C$ or -8$0^{\circ}C$ can be applicable to the freezing method using methanol and also all of the components of the semen diluent including cryoprotectant, buffer and cold-shock buffer are very important to maintain motility and viability of canine spermatozoa in the freezing and thawing procedure.

  • Rice (Oryza sativa) cultivars show an impairment of growth and development in response to abiotic stresses such as drought, salinity, heat and cold at the early seedling stage. The tolerance to heat stress in plants has been genetically modulated by the overexpression of heat shock transcription factor genes or proteins. In addition to a high temperature-tolerance that has also been altered by elevating levels of osmolytes, increasing levels of cell detoxification enzymes and through altering membrane fluidity. To examine the heat tolerance in transgenic rice plants, three OsOPT10 overexpressing lines were characterized through a physiological analysis, which examined factors such as the electrolyte leakage (EL), soluble sugar and proline contents. We further functionally characterized the OsOPT10 gene and found that heat induced the expression of OsOPT10 and P5CS gene related proline biosynthesis. It has been suggested that the expression of OsOPT10 led to elevated heat tolerance in transgenic lines.

Overexpression of NtHSP70-1 Protects Chlorophyll from High Temperature in Plants (NtHSP70-1에 의한 클로로필의 고온 내성 효과)

  • Heat shock protein 70 (HSP70) is known as molecular chaperone, the fundamental protein participating in various processes, from nascent protein synthesis to protection of proteins during abiotic stresses and developmental programs. However, their biological functions in plants are not yet well known. Here, NtHSP70-1 (AY372069), HSP70 of Nicotiana tabacum induced by heat stress was investigated. To analyze the protective role of NtHSP70-1, transgenic tobacco plants, which constitutively overexpressed NtHSP70-1 as well as contained either the vector alone or having NtHSP70-1 in the antisense orientation, were constructed. The altered NtHSP70-1 levels in plants were confirmed by western blotting and transgenic sense lines exhibited tolerance to heat stress. Seedlings with the constitutively expressed NtHSP70-1 grew as green or healthy plants after heat stress. In contrast, transgenic vector or antisense lines exhibited yellowing of leaves or some delay in growth, which finally led to death. Evaluation of chlorophyll contents of heat-shocked transgenic tobacco seedlings indicated that NtHSP70-1 contributes to thermotolerance by preventing chlorophyll synthesis in plants.
