Meat Quality, Digestibility and Deposition of Fatty Acids in Growing-finishing Pigs Fed Restricted, Iso-energetic Amounts of Diets Containing either Beef Tallow or Sunflower Oil

  • The influence of dietary beef tallow (BT) versus sunflower oil (SO) on meat quality and apparent digestibility and deposition of individual fatty acids in the whole carcass was investigated in pigs fed diets containing either BT or SO. The diets contained equal amounts of energy in the form of the variable fats and were fed on an iso-energetic, restricted basis. Crude fat in the SO diet was better digested (p<0.001) than in the BT diet. The dietary fat type had no effect on growth performance, physical properties of the carcass and meat quality. The pigs fed the BT diet showed lower (p<0.001) apparent digestibilities for palmitic and linoleic acid, but those of oleic and ${\alpha}$-linolenic acid were not affected. The ratio of deposition in the carcass to intake of digestible fatty acids for the whole feeding period was decreased (p<0.01) for oleic and linoleic acid in pigs fed the SO diet. The pigs fed the SO diet instead of the BT diet had a lower (p<0.05) deposition:intake ratio for mono-unsaturated fatty acids. The calculated minimum de novo synthesis of saturated fatty acids was increased for the SO diet, but that of mono-unsaturated fatty acids was not different. In conclusion, the iso-energetic replacement of BT by SO had a marked impact on the fatty acid composition of tissues, but did not affect carcass and meat quality traits in spite of the marked difference in the deposition of linoleic acid in adipose tissues, loin muscle and the whole body. In addition, it became clear that the type of dietary fat had marked, specific effects on the synthesis and oxidation of fatty acids.

  • 본 연구는 햄버거 패티 제조시 우지를 대체한 canola oil, olive oil, corn oil 및 sunflower oil 등의 식물성 유지와 결착 보조제로 egg white powder와 wheat fiber 를 첨가하여 품질 특성을 조사하였다. 식물성유를 첨가한 햄버거 패티는 일반적인 우지를 첨가한 패티보다 낮은 칼로리와 가열감량 및 직경감소율을 나타내었고, 물리적 측면 및 관능적 특성에서는 우지를 사용한 패티와 큰 차이를 나타내지 않았다. 지방산 조성 분석 결과, 우지를 사용한 패티에 비해 낮은 트랜스 지방산 함량과 낮은 포화지방산 함량을 나타내었다. 따라서 식물성유를 대체할 때 대두단백, egg white powder 및 wheat fiber를 일정량 혼합하여 첨가한다면, 트랜스 지방산의 함량을 낮추고, 많은 불포화 지방산의 함량이 높은 웰빙형 햄버거 패티의 개발이 가능할 것으로 사료된다.

  • 어유식이(魚油食餌)가 간장조직내 lipogenic enzyme의 활성도에 미치는 영향을 조사하고자 고등어유를 식이(食餌) 10%(w/w) 되게 먹이를 조제하여 13일동안 사육한 흰쥐의 간장조직의 glucose-6-phosphate dehydrogenase와 malic enzyme의 활성도는 lard를 섭취한 군(群)보다 $3{\sim}6$일의 섭취로 훨씬 저하되었으며 콩기름에 비하여는 같거나 또는 약간 낮은 값을 보였다. 고등어 또는 장어유를 어유(魚油)로 사용하여 먹이의 $0{\sim}10%$로 함량을 변화시키며 총지방량을 10%로 일정히 하고자 콩기름 또는 쇠기름을 배합유로 사용하였을 때 glucose-6-phosphate dehydrogenase의 활성은 어유(魚油)가 2 % 포함하였을 때부터 malic enzyme의 활성은 5% 포함하였을 때, 어유(魚油)를 포함하지 않은 군(群)에 비하여 감소를 보였다.

고지방식이 및 고탄수화물식이가 흰쥐의 혈청 Leptin 농도 및 지질함량에 미치는 영향 (Effect of High Fat and High Carbohydrate Diet on Serum Leptin and Lipids Concentration in Rat)

  • The purpose of this study was to investigate the effects of diet composition on serum leptin and lipids profiles in rats. At the baseline, seven 4-week-old Sprague-Dawley rats, male, were sacrificed and the remaining rats were divided into five groups and each group was fed one of the following five dietary regiments for 2 weeks and 6 weeks;the control diet AIN76(CAL, n=2l), high-carbonhydrate(rice)(HCR, n=2l), high-carbohydrate(flour)(HCF, n=2l), high-fat(corn oil)(HFO, n=2l), high-fat(beef tallow)(HFB, n=2l). Serum leptin was determined by a double antibody ELISA assay at the baseline(n=7), 6 week(n=35) and 10 week of age(n=70). At 6 weeks of age, the increase in the Food Efficiency Ratio(FER) was related to adipocyte hyperplasia in rats on HFB diets. The serum triglyceride, total cholesterol, and LDL-cholesterol increased significantly in HFB group, and decreased in HFO group compared to control group. The HFC group showed significant increase in serum triglyceride level compared with control group. After 2 weeks and 6 weeks, noticeably high increases in epididymal adipose tissue fat cell mass and numbers were observed with the HFB fed group. Serum leptin levels increased as body weight increased over the period of time(4weeks; 1.50$\pm$0.13ng/ml versus 10weeks; 2.08$\pm$-.13ng/ml). And this result shows that there are 193% higher in rats fed high fat-beef tallow diet than the control diet. Serum leptin levels of the HFB group(4.01$\pm$0.39mg/ml) were significantly higher than that of the HFO(2.06$\pm$0.5613ng/ml), CAL(2.08$\pm$0.1313ng/ml), HCR(2.41$\pm$0.2113ng/ml) and HCF(2.80$\pm$0.4713ng/ml) at p<0.05. The serum leptin concentration was positively correlated with the amount of epididymal fat pads(r=0.47 p<0.01), serum triglyceride(r=0.49, p<0.001), tatal cholesterol(r=0.48, p<0.001), LDL-cholesterol(r=0.58, p<0.001), atherogenic index(r=0.67, p<0.001), and inversely correlated with HDL-cholesterol(r=-0.65, p<0.001). In conclusion, the changes in composition of dietary fat and carbohydrate intake could affect changes in concentration of serum lipids and leptin. Especially, the high-fat diet with animal fat source could increase circulating leptin level. (Korean J Nutrition 34(2) : 123-131, 2001)

Butylated Hydroxytoluene첨가 식이 및 2-Acetylaminofluorene 투여가 식이지방을 달리한 쥐간의 Microsomal Mixed Function Oxidase계에 미치는 영향 (Effect of Butylated Hydroxytoluene and 2-Acetylaminofluorene Administration and Microsomal Mixed Function Oxidase System in Young Rats fed different Fats)

  • Sprague-dawley 숫쥐를 식이지방을 달리하여(I:Soybean oil p/s 4.0, II:beef tallow p/s 0.08) 이유후 8주동안 사육하였다. 이때 I,II군은 각각 기초식이군, 기초식이군에 0.3% butylated hydroxytoluene(BHT)를 첨가신킨 군, 생후 5~7주 사이에 4번의 2-Acetylaminofluorene(2-AAF)를 투여한 군 2-AAF를 투여하고 BHT도 먹인 군으로 나누었다. BHT는 이유 후부터 식이에 섞어 먹였으며 2-AAF를 투여하지 않은 군읜 2-AAF 주사에 의해 얻는 stress와 같은 효과를 주기위해 placebo로 polyethylene glycol 300을 투여하였다. Mixed function oxidase(MFO)계의 효소인 cytochrome p-450, cytochrome b$_5$및 cytochrome p-450 reductase와 과산화지질 등을 측정하였다. 성장기에 2-AAF의 투여는 성장지연을 초래하였으며 지질과산화물은 지방의종류, 2-AAF,BHT 등에 의해 큰 차는 없었다. Cytochrome p-450은 2-AAF에 의해 I-BHT-AAF와 II-AAF군에서 증가되었고 BHT에 의해서는 차이가 나지 않았다. 불포화지방을 먹인경우 cytochrome p-450과 cytochrome p-450 reductase가 2-AAF에 의해 증가되기보다는 오히려 감소하거나 I,II군에 비해 별 차이가 없었는데 2-AAF의 농도와 식이의 불포화도가 높아 세포막이 손상되었기 때문이라 사료된다. Cytochrome $b_5$는 각군 사이에 별 차가 없었다. Cytochrome p-450과 과산화지질(r=0.2475, p<0.05), cytochrome p-450 reductase와 cytochrome $b_5$ (r=0.2475, P<0.05)가 각각 양의 상관관계를 나타내었다. 따라서 2-AAF를 대사시키는 MFO계는 식이지방의 종류 및 BHT의 존재에 따라 영향을 받고, 특히 불포화지방식이인 경우 2-AAF를 대사시킬 수 있는 cytochrome p-450 유도 및 합성능력이 매우 저조함을 알 수 있으며 2-AAF는 어린 쥐의 성장을 지연시켰다.

식이칼슘 수준이 콜레스테롤 첨가 또는 무첨가 고지방식 섭취 흰쥐의 지질대사에 미치는 영향 (Effects of Dietary Calcium Levels on Lipid Metabolism in Rats Fed High Fat Diet with or without Supplemental Cholesterol)

  • 본 연구에서는 성숙한 흰쥐에게 동물성 포화지방(식이중 beef tallow로서 18%)과 식이에 콜레스테롤을 첨가하지 않은 조건과 첨가한 조건(1%) 하에서 식이칼슘을 수준별로(식이에 0.1%, 0.5%, 1.5% 첨가) 4주간 동안 급여한 후 혈액과 조직의 지질함량, 소장내용물 중 지질의 분포 및 분중 지질배설량 등이 측정되었다. 그 결과, 콜레스테롤 무첨가의 고지방식을 4주간 급여한 경우에는 고콜레스테롤혈증은 유발되지 않았으며, 식이칼슘의 투여효과도 인정되지 않았다. 한편 콜레스테롤 첨가 고지방식을 급여한 경우에는 고콜레스테롤혈증의 유발이 가능하였다. 이때 식이칼슘의 투여효과는 식이칼슘의 수준 증가에 따라 혈청 콜레스테롤 농도가 저하하는 경향을 보였다. 즉 식이칼슘 섭취 부족에 따른 혈중 콜레스테롤 상승효과(hypercholesterolemic effect)와 칼슘섭취 증가에 따른 혈중 콜레스테롤 저하효과(hypocholesterolemic effect)가 인정되었다. 또 칼슘 섭취가 체내 지질대사에 미치는 효과에 대한 하나의 작용메카니즘으로서 콜레스테롤의 소화율과 장관내 흡수양상이 검토되었는데, 이때 분중 지질성분의 배설량은 고칼슘식이군에서 유의적으로 높았고 콜레스테롤의 소화율은 저칼슘식이군에서 유의적으로 높았다. 이것은 칼슘이 지질 성분의 배설을 촉진하므로서 지질 성분의 흡수를 저하시킨다고 하는 장관내 지질 흡수 메카니즘에 대한 “지질-칼슘 soap 형성”으로 칼슘의 역할이 설명될 수 있다. 본 과제에서 처음으로 시도된 소장내용물의 지질과 칼슘성분의 존재양상은 지질 흡수와 칼슘의 상호작용을 보다 직접적인 방법으로 측정할 수 있었으나, 얻어진 결과에 대한 해석에는 아직 어려움이 있었다.

난황 콜레스테롤과 지방산 조성에 관한 서로 다른 지방의 첨가 효과 (Effects of Dietary Fats on the Cholesterol Content and Fatty Acid Composition of Egg Yolk)

  • 계란 품질, 난황 콜레스테롤 함량 및 지방산 조성의 변화에 관한 산란계 사료 내 서로 다른지방의 첨가효과를 조사하였다. 36주령의 Hyline brown 산란계 총 200수(4처리구${\times}$50수/반복)를 공시하여 우지, 옥수수유, 아마인유 및 어유를 각각 5.0% 함유하는 실험사료를 10주간 급여하였다. Haugh unit, 파란 강도, 난각 두께 및 난황색에 대한 처리구 간 유의적 차이는 나타나지 않았다. 첨가지방은 계란 콜레스테롤 함량과 고도 불포화지방산 조성을 현저하게 변화시켰다. 난황의 n-6지방산 함량은 옥수수유 첨가구가 31.61%로써 가장 높았고 우지, 아마인유 및 어유 첨가구순으로 낮았으며, n-3 지방산 함량은 아마인유와 어유 첨가구가 각각 9.74%, 5.16%로써 유의적 증가 경향을 나타냈다(p<0.05). 난황지방 산 조성의 n-3지방산 증가는 계란 콜레스테롤 함량 감소를 초래하였다. 난황 콜레스테롤(mg/g yolk) 및 계란 콜레스테롤(mg/60g egg) 범위는 각각 $15.98{\sim}18.37mg$$227{\sim}261mg$으로써 우지 첨가구가 가장 높았고 어유, 아마인유 및 옥수수유 첨가구 순으로 낮아지는 경향을 보였다(p<0.05). 난황 콜레스테롤의 감소율은 우지 첨가구와 비교하였을 때 어유 13.01%, 아마인유 11.49% 및 옥수수유 6.91%로써 유의적인 차이를 나타냈다(p<0.05). 이상의 결과는 산란계 사료 내 첨가되는 지방급원을 조절해 줌으로써 계란 콜레스테롤을 낮출 수 있으며 n-6와 n-3 지방산을 난황으로 축적하는 것이 가능함을 시사해준다.

Effects of n-3 Polyunsaturated Fatty Acids-enriched Diet Supplemented with Different Levels of α-Tocopherol on Lipid Metabolism in Laying Tsaiya Ducks

  • The objective of this experiment was to determine the effects of n-3 polyunsaturated fatty acids (n-3 PUFAs)-enriched diet supplemented with different levels of $\alpha$-tocopherol on the activities of hepatic lipogenic-related enzymes and the contents of liver and plasma lipid fractions in laying Tsaiya ducks. A total of 180 30-wk-old laying Tsaiya ducks, at the beginning of peak production, were allotted into 6 treatments with 3 replicates each. Ducks were fed one of the 6 experimental diets, containing 4% tallow (control), and 4% fish oil supplemented with graded levels of $\alpha$-tocopheryl acetate ($\alpha$-tocopherol) at 0, 100, 200, 300 and 400 mg/kg, respectively, for 6 wks. Feed and water were supplied ad libitum throughout the experimental period. The results indicated that the n-3 PUFAsenriched diet supplemented with different levels of $\alpha$-tocopherol did not affect (p>0.05) egg weight, feed intake, body weight change or liver and abdominal fat weights. Egg production, egg mass and feed efficiency significantly (p<0.05) improved as dietary $\alpha$-tocopherol levels increased. The activities of hepatic lipogenic-related enzymes including acetyl-CoA carboxylase (EC 6. 2. 1. 3; ACC), glucose-6-phosphate dehydrogenase (EC 1. 1. 1. 49; G-6-PDH), ATP-citrate cleavage enzyme (EC 4. 1. 3. 8; CCE), NADP-malate dehydrogenase (EC 1.1.1.40; NADP-MDH) and fatty acid synthetase (FAS) were higher (p<0.05) in birds fed with the tallow diet than in those fed with fish oil diets and increased with increasing dietary $\alpha$-tocopherol levels. None of the dietary treatments significantly affected the contents of triglyceride and total cholesterol in the liver, or total cholesterol, phospholipid and total lipid in the plasma. However, the contents of phospholipid and total lipid in the liver, and triglyceride in the plasma increased as dietary $\alpha$-tocopherol levels increased. Increasing dietary $\alpha$-tocopherol levels decreased the non-esterified fatty acid (NEFA) content in the plasma and trended to decrease the cholesterol contents in the egg yolk. The lipid metabolism of laying Tsaiya ducks was influenced not only by the dietary fat but also by the supplementation levels of $\alpha$-tocopherol.

식이의 n-3 지방산과 지방의 불포화도가 혈장지질 조성에 미치는 영향 (Effect of Dietary n-3 Fatty Acids and Fat Unsaturation on Plasma Lipids and Lipoproteins in Rats)

  • This study was to compare the effects of dietary n-6 and n-3 fatty acids and fat unsaturation on plasma lipids and chemical composition of VLDL and LDL fraction and lipogenic enzymes activity in rat liver under the conditions providing 1) a similar amount of n-6, n-3 fatty acids(LA, ALA, EPA+DHA) in diets and 2) the various degree of fat unsaturation. Male Sprague-Dawley rats weighing 420g were treated for 6-n with six experimental diets providing 25% of energy as fat and which were different only in fatty acid composition. The fats used for a source of each fatty acid were beet tallow for saturated fatty acid corn oil for n-6 linoleic acid(LA) perilla oil for n-3 $\alpha$-linolenic acid(ALA) and fish oil n-3 eicosapentaenoic acid (EPA) and n-3 docosahexaenoic acid(DHA). Plasma cholesterol level was increased by corn oil to compare with beef tallow but was decreased by perilla oil or fish oil. Plasma TG level was significantly decreased by perilla oil or fish oil. Fish oil significantly reduced the level of HDL-Chol and the proportion of Chol in LDL fraction and that of TG in vVLDL fraction. Overall there was a singificant negative correlation between the level of each plasma lipid(Chol TG, VLDL-TG, LDL-C) and the degree of fat unsaturation. However this rerlationship is not always true when compared the hypolipidemic effect of each fatty acid at a similar level of fat unsaturation. There was a trend such taht glucose 6-P dehydrogenase 6-phosphogluconate dehydrogenase and malic enzyme activites were reduced by n-3 fatty acids. Perilla oil significantly increased the incorporation of c20:5 and c22:5 into liver tissue and fish oil suignificantly increased the incorporation of c20:5, c22:6 into liver tissue and the effect of long chain n-3 fatty acid incorporation was greater by fish oil. therefore the hypotriglyceridemic effect of n-3 fatty acid could be resulted from the interference of hepatic lipogenesis by long-chain n-3 fatty acids and the reduced proportion of TG in VLDL fraction and its effect was greater by n-3 EPA+DHA than n-3 ALA even though plasma Chol and TG levels were also influenced by the degree of dietary fat unsaturation.

The Effects of Vitamin D Supplementation to Peak-producing Hens Fed Diets Differing in Fat Source and Level on Laying Performance, Metabolic Profile, and Egg Quality

  • This experiment was designed to examine the effects of supplemental vitamin D on laying performance, metabolic profile and egg quality of hens fed diets containing different fat sources and levels. Lohman strains (n = 480) were assigned to one of 10 diets: basal diet (BD), BD plus 2.5 and 5.0% sunflower oil (SO) or tallow (T) at vitamin D provided $1{\times}$ and $3{\times}$ of the current recommendation. The experiment lasted from week 30 to 44 of age. Each diet was tested in 12 replicate cages of 4 hens. Production, metabolism, and egg quality data were subjected to three-way ANOVA. Both fats decreased feed intake (FI) as compared to BD. Increasing SO and T levels linearly decreased and quadratically increased FI, respectively. The dietary factors did not affect egg production (EP) and egg weight. Vitamin D supplementation increased and decreased EP when diets contained SO and T, respectively. Feed conversion efficiency (FCE) for hens fed SO was lower than for hens fed T. However, increasing T level improved FCE, whereas increasing SO level worsened FCR. Vitamin D supplementation increased serum vitamin D and glucose concentrations. Vitamin D supplementation also caused a decrease and an increase in serum vitamin D concentration when diets contained SO and T, respectively. Serum glucose concentration for hens fed SO was lower than hens fed T. Increasing fat level linearly increased serum triglyceride and VLDL concentrations, regardless of the fat type. Increasing SO level linearly decreased serum cholesterol concentration. Vitamin D supplementation did not alter lipid metabolites. The dietary factors did not affect serum total protein, Ca, and P concentrations. As compared with BD, feeding SO decreased dry tibia and ash weights more than feeding T. Vitamin D supplementation tended to increase dry tibia weight and decrease tibia ash weight. Eggshell strength and thickness, yolk and albumen indexes, and Haugh unit were not responsive to the dietary factors. Eggshell strength quadratically increased with increasing T level. Yolk color for hens fed SO was lower than for hens fed T. The dietary factors did not affect most of yolk fatty acids. Increasing SO level quadratically decreased yolk $C_{18:2}$ concentration. Vitamin D supplementation increased and decreased yolk $C_{18:2}$ concentration when diets contained SO and T, respectively. In conclusion, increasing fat level improved laying performance without altering metabolic profile and egg quality. Vitamin D supplementation had minor alteration effects on laying performance, metabolic profile, and egg quality in response to fat feeding.
