Whitening Effect of Black Tea Water Extract on Brown Guinea Pig Skin

  • To evaluate the whitening effect of black tea water extract (BT), BT was topically applied to artificially hyperpigmented spots on the back skins of brown guinea-pigs (weight: 450~500 g) induced by 1,500 mJ/$cm^2$ of ultraviolet B (UVB) irradiation. The test compounds of 30 ${\mu}l$ were applied twice a day, six days a week, for four weeks. The artificially hyperpigmented spots were divided into 5 groups: control (UVB + saline, C), vehicle control [UVB + propylene glycol: ethanol: water (5 : 3 : 2), VC], positive control (UVB + 2% hydroquinone, PC), experimental 1 (UVB + 1% BT), experimental 2 (UVB + 2% BT). After 4-week application, the spots were removed by biopsy punch under anesthetic condition and used as specimens for the histological examination. The total polyphenol and flavonoid contents of BT were 104 and 91 mg/g, respectively. The electron-donating ability of BT revealed a dose-dependent response, showing the excellent capacities of 86% at 800 ${\mu}g$/ml. The artificially hyperpigmented spots treated with the PC and BT were obviously lightened compared to the C and VC groups. At the fourth week, the melanin indices for the PC and BT groups were significantly lower (p < 0.001) than those of the C and VC groups. In histological examination, PC and BT groups were significantly reduced in the melanin pigmentation, the proliferation of melanocytes and the synthesis of melanosomes compared to the C and VC groups. It is found that BT inhibits the proliferation of melanocytes and synthesis of melanosomes in vivo using brown guinea pigs, thereby showing a definite skin whitening effect.

  • The crystallization behaviour and the machinability of mica glass-ceramics with the content of F1 were studied. The material was made from the $K_2O-MgO-Al_2O_3-B_2O_3-SiO_2-F$ glasses by the heattreatment at 80$0^{\circ}C$-110$0^{\circ}C$ where the content of F-1 was changed in the range from 1, 3wt% to 6.1wt%. X-ray diffraction phase analysis and optical observation were adopted to study the crystallization behaviour. The machinability was measured by a manual sawing test and MOR. The crystal phases of these glass-ceramics identified by XRD were chondrodite fluoborite and norbergite at low temperature but fluorophlogopite at high temperature. The crystallization of glasses containing 1.3wt% -2.5wt% F-1 were predominately controlled by surface crystallization while the crystallization of glasses containing 3.8 wt% -6.1wt% F-1 were controlled by volume crystallization. Among the test the best machinability and strength value were obtained from those specimens contained fluoride 4.2wt% -4.4wt% and when the heattreatment was performed at 95$0^{\circ}C$-110$0^{\circ}C$ for 2 hours.

Improved Production of Long-Chain Fatty Acid in Escherichia coli by an Engineering Elongation Cycle During Fatty Acid Synthesis (FAS) Through Genetic Manipulation

  • The microbial biosynthesis of fatty acid of lipid metabolism, which can be used as precursors for the production of fuels of chemicals from renewable carbon sources, has attracted significant attention in recent years. The regulation of fatty acid biosynthesis pathways has been mainly studied in a model prokaryote, Escherichia coli. During the recent period, global regulation of fatty acid metabolic pathways has been demonstrated in another model prokaryote, Bacillus subtilis, as well as in Streptococcus pneumonia. The goal of this study was to increase the production of long-chain fatty acids by developing recombinant E. coli strains that were improved by an elongation cycle of fatty acid synthesis (FAS). The fabB, fabG, fabZ, and fabI genes, all homologous of E. coli, were induced to improve the enzymatic activities for the purpose of overexpressing components of the elongation cycle in the FAS pathway through metabolic engineering. The ${\beta}$-oxoacyl-ACP synthase enzyme catalyzed the addition of acyl-ACP to malonyl-ACP to generate ${\beta}$-oxoacyl-ACP. The enzyme encoded by the fabG gene converted ${\beta}$-oxoacyl-ACP to ${\beta}$-hydroxyacyl-ACP, the fabZ catalyzed the dehydration of ${\beta}$-3-hydroxyacyl-ACP to trans-2-acyl-ACP, and the fabI gene converted trans-2-acyl-ACP to acyl-ACP for long-chain fatty acids. In vivo productivity of total lipids and fatty acids was analyzed to confirm the changes and effects of the inserted genes in E. coli. As a result, lipid was increased 2.16-fold higher and hexadecanoic acid was produced 2.77-fold higher in E. coli JES1030, one of the developed recombinants through this study, than those from the wild-type E. coli.

  • This study was designed to investigate the effect of silkworm (Bombyx mori L.) powder on lipofuscin, acetylcholine (ACh) and its related enzyme activities in brain of rats. Sprague-Dawley (SD) male rats ($160{\pm}10g$) were fed basic diet (control group), and experimental diets (SWP-200 and SWP-400 groups) added 200 and 400 mg/kg BW/day for 6 weeks. In case of liver membranes, lipofuscin (LF) levels resulted in a slight decreases (4.6% and 11.5%, respectively) in SWP-200 and SWP-400 groups compared with control group. But in case of barin as the most sensitive organ, LF levels were remarkably inhibited about 16.7% and 20.0% in SWP-200 and SWP-400 groups compared with control group. There were no significant differences in acetylcholine (ACh) syntheses as a very important neurotransmitter, and choline acetyltransferase (ChAT) activities as a synthesis enzyme of ACh, and acetylcholinesterase (AChE) activities as a hydrolysis enzyme, which were concerned in transmission of neuron through synapses in brain of SWP-200 and SWP-400 groups compared with control group. Monoamine oxidase-B (MAO-B) activities were significantly inhibited (about 10.2%) in brain of SWP-400 groups compared with control group. These results suggest that inhibiting effects of LF accumulation and MAO-B activity of silkworm powder (SWP) may play a pivotal role in attenuating a various age-related changes for improvement of brain function.

Isolation, Cloning and Co-Expression of Lipase and Foldase Genes of Burkholderia territorii GP3 from Mount Papandayan Soil

  • Lipases are industrial enzymes that catalyze both triglyceride hydrolysis and ester synthesis. The overexpression of lipase genes is considered one of the best approaches to increase the enzymatic production for industrial applications. Subfamily I.2. lipases require a chaperone or foldase in order to become a fully-activated enzyme. The goal of this research was to isolate, clone, and co-express genes that encode lipase and foldase from Burkholderia territorii GP3, a lipolytic bacterial isolate obtained from Mount Papandayan soil via growth on Soil Extract Rhodamine Agar. Genes that encode for lipase (lipBT) and foldase (lifBT) were successfully cloned from this isolate and co-expressed in the E. coli BL21 background. The highest expression was shown in E. coli BL21 (DE3) pLysS, using pET15b expression vector. LipBT was particulary unique as it showed highest activity with optimum temperature of $80^{\circ}C$ at pH 11.0. The optimum substrate for enzyme activity was $C_{10}$, which is highly stable in methanol solvent. The enzyme was strongly activated by $Ca^{2+}$, $Mg^{2+}$, and strongly inhibited by $Fe^{2+}$ and $Zn^{2+}$. In addition, the enzyme was stable and compatible in non-ionic surfactant, and was strongly incompatible in ionic surfactant.

Combined Effects of Container Volume and Fertilizer Level on Plant Growth, Physiological Characteristics, and Nutrient Uptake of Vinca Plant (Catharanthus roseus)

  • The aim of our study was to investigate the interactive effects of container size and nutrient supply on plant growth, chlorophyll synthesis, transpiration, $CO_2$ assimilation, water use efficiency (WUE), and nutrient uptake of vinca plant (Catharanthus roseus). A complete experiment utilizing four concentrations of fertilizer and three volumes of containers was conducted. As the container size was increased, the plant height, leaf area, and dry weight of vinca significantly increased regardless of fertilizer level. The leaf area and dry weight of vinca were highly sensitive to the container size. However, the chlorophyll contents of vinca 20 days after the transplant significantly increased with decreasing container sizes and increasing fertilizer concentrations. Significant differences in transpiration and $CO_2$ assimilation occurred with the use of differentfertilizer solutions, but the highest values for transpiration and $CO_2$ assimilation were in plants grown in the 15 cm-diameter containers. The highest water use efficiency was observed in the plants grown in 10 cm-containers with 4 dS/m of fertilizer, and there were no significant differences in WUE values among container sizes with fertilizer concentrations of 0, 1, or 2 dS/m. No significant difference in nutrient uptake was observed among the fertilizer levels or among the container sizes. However, at a fertilizer concentration of 4 dS/m, the uptake of several nutrients, including N, P, K, Ca, Mg, B and Fe, was higher in small containers than in larger ones.

스테로이드제가 Surfactant Protein A의 유전자 발현과 총단백량에 미치는 영향에 관한 실험적 연구 (The Effect of Dexamethasone on Gene Expression and Total Amount of Surfactant Protein A)

  • 연구배경: SP-A는 surfactant의 분비, 합성 및 재순환에 관여하는 중요한 역할을 담당한다. Glucocorticoid는 폐의 형태학적 발생을 촉진시키며, surfactant인지질의 생산을 증가시키고, surfactant B와 C의 축적 및 폐탄성을 향상시킨다. 시험관 내에서의 실험을 통하여 관찰 한 바에 의하면 glucocorticoid의 투여량에 따라 SP-A mRNA와 SP-A 단백의 총량이 증가하기도하고 감소하기도 한다. 그러나 실험동물 내에서의 SP-A mRNA와 SP-A 단백량에 대한 스테로이드의 효과에 관한 보고는 드물다. 방 법: 저자들은 실험동물 내에서의 현황을 파악하고자 dexamethasone을 백서에 투여한 후 SP-A의 유전자 발현양상은 filter hybridization방법으로, SP-A 단백량은 double sandwich ELISA 방법으로 각각 검색하여 dexamethasone의 투여양과 투여기간에 따라 실험동물 내의 SP-A 유전자 발현과 총 SP-A 단백량에 대한 dexamethasone의 효과를 관찰하였다. 결 과: 1) Dexamethasone 투여량에 따른 SP-A mRNA량은 dexamethasone을 일일 0.2 mg/kg 투여한 지 24시간 경과후에 38.8%가 증가하였다. 2) Dexamethasone투여기간에 따른 SP-A mRNA량은 dexamethasone을 일일 2 mg/kg씩 일주일간 투여 후에 대조군에 비하여 49.7%가 유의하게 증가하였다(P<0.01). 3) 폐의 총 SP-A 단백량은 dexamethasone을 일일 2 mg/kg씩 일주일간 투여 후에 대조군에 비하여 373.7%가 유의하게 증가하였다(P<0.005). 결 론: 이상의 결과는 실험동물 내 dexamethasone양에 따른 SP-A mRNA량과 총 SP-A 단백량은 소량을 투여하였을때보다는 대량을 투여 하였을 때, 단기간보다는 장기간 사용하였을 때 유의한 증가가 있음을 알 수 있었다.

PLANT BIOCHEMISTRY OF GINSENG SAPONINS(III) Radioactive Studies (2). Sodium Acetate-U-$C^{14}$ Experiment

  • The radioactive compound sodium $acetate-U-C^{14}$ (C-14 acetate) was administered to two- and four-year-old July and September American ginseng (Panax quinquefolium L.) plants and cuttings. The C-14 acetate uptake was approximately $99\%.$ The autoradiochromatograms suggest that the saponins(panaquilins) isolated by preparative thin-layer chromatography contained impurities, especially those isolated from the leaf and stem extracts. The root and fruit methanol extracts yielded relatively pure saponins. The large amounts of panaquilin B and its proximity to panaquilin C on preparative thin-layer plates resulted in some admixing. The average concentration $(\%$ plant dry weight) of semipurified saponins were high in the leaves $(13.8\%),$ compared to fruits $(9.8\%),\;stems\;(7.9\%)\;and\;roots\;(6.3\%).$ The average percentage of C-14 acetate incorporation into panaquilins was $4.8\%.$ The average percentage of C-14 acetate incorporation into panaquilins B and C was higher $(1.40\%\;and\;1.13\%,$ respectively) than that into panaquilin C, (d), G-1 and G-2 $(0.75\%,\;0.65\%,\;0.13\%\;and\;0.53\%,$ respectively). Panaquilin synthesis may be depending upon the part collection period and age of the plant. The average percentage of C-14 acetate incorporation into panaquilin B is high in roots $(0.58\%)\;and\;stems\;(0.48\%);$ that into panaquilins C and (d) high in leaves $(0.40\%\;and\;0.45\%,$ respectively); and that into panaquilin E high in roots and leaves $(0.55\%\and\;0.50\%,$ respectively). Panaquilin G-2 was synthesized in all parts of plants. The panaquilins appear to be biosynthesized more actively in July than September (exception-panaquilin G-l). Panaquilins B, C and G-1 may be biosynthesized more actively in four-year-old plants and panaquilins (d) and E more actively in two-year-old plants. The results from expectance with cuttings suggest that the panaquilins are synthesized de novo in the above-ground parts of ginseng plants, and that panaquilin G-l may be synthesized de novo in the leaf. It is known from the tissue culture studies that panaquilins are produced by leaf, stem and root callus tissues and callus-root cultures of American and Korean ginseng plants. Panaquilins may actively be synthesized de novo in most any cell or organ of the ginseng plants. It was verified that C-14 acetate was incorporated into the panaxadiol portions of the panaquilins of two-year-old plants (sp. act., 0.56 $m{\mu}Ci/mg$) and four-year-old plants (sp. act., 0.54 $m{\mu}Ci/mg$).

Glucose Kinetics for Milk Synthesis in Etawah Crossbred Goats Fed King Grass Silage Prepared with Manure

  • A study was carried out to determine glucose kinetics, nutrient balance and milk production of lactating Etawah crossbred goats. The animals (27.2 to 29.1 kg BW) were randomly divided into four levels of dietary treatment groups: the first group R1 received 100% (3 kg) fresh king grass (Penisetum purpuroides), the second group R2 received 75% king grass and 25% king grass silage prepared with chicken manure, the third group R3 received 50% king grass and 50% silage, and the fourth group R4 received 100% silage. In addition to the roughage, each group received 800 g of concentrate (CP 14.77% of DM; 17.26 MJ/kg). Animals fed king grass silage made with chicken manure were found to be superior to the group fed king grass alone. Glucose kinetics and retained energy were significantly affected. Calculations showed that glucose requirements for maintenance and milk production can be met for the groups with high levels of silage (R3 and R4). The values of glucose flux were in the range of 2.52 to 4.50 mg/min.kg $BW^{0.807}$ which are lower, but close to, the values for the temperate lactating dairy cow. The present glucose flux value for the lactating Etawah crossbred goat is higher than the previous value published from this laboratory.

한약재 12종의 열수추출물이 ${\alpha}$-melanocyte stimulating hormone에 의해 유도된 B16F10 흑색종 세포의 멜라닌형성에 미치는 영향 (Effects of Aqueous Extracts from Twelve Herbs on ${\alpha}$-melanocyte Stimulating Hormone-induced Melanogenesis in B16F10 Mouse Melanoma Cell)

  • We investigated the effects of aqueous extracts from twelve medical herbs on ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis in B16F10 mouse melanoma cell. The cells were incubated with ${\alpha}$-MSH and aqueous extracts 5 days and 2 days and then analysed melanin amount and tyrosinase activities, respectively. Nine aqueous extract of herbs examined at 1 mg/m${\ell}$ level decreased melanin synthesis in B16F10 cell, especially in Agastache rugosa, Leonurus siviricus and Murus bombycis. The significant decrease of released extracellular melanin were also observed in treated cells with aqueous extract from Leonurus siviricus, Murus bombycis and Ledebouriella seseioides. The ${\alpha}$-MSH-induced activation of tyrosinase was inhibited in cells treated with aqueous extract from Cuscutae semen, Angelica tenuissima and Agastache rugosa. These results suggest that herbs inhibiting melanogenesis through tyrosinase activity may apply to develop whitening drugs and cosmetics.
