Municipal Solid Waste-derived Syngas Fermentation Process by Pressurization (가압을 통한 도시형 생활 폐기물 기반 합성가스발효 공정 개발)

  • Global efforts are focused on achieving carbon neutrality due to the increases in the levels of greenhouse gases. Moreover, the greenhouse gases generated from the disposal of municipal solid waste (MSW) are the primary sources of emissions in South Korea. In this study, we conducted the biological conversion of syngas (CO, H2, and CO2) generated from MSW gasification. The MSW-derived syngas was used as a feed source for cultivating Eubacterium limosum KIST612, and pressurization was employed to enhance gas solubility in culture broth. However, the pH of the medium decreased owing to the pressurization because of the CO2 in the syngas and the cultivation-associated organic acid production. The replacement of conventional HEPES buffer with a phosphate buffer led to an approximately 2.5-fold increase in acetic acid concentration. Furthermore, compared with the control group, the pressurized reactor exhibited a maximum 8.28-fold increase in the CO consumption rate and a 3.8-fold increase in the H2 consumption rate.

  • 바이오에탄올 생산공정은 당 (Sugar)을 기반으로 하는 공정과 합성가스를 이용하는 공정으로 분류할 수 있다. 이 가운데 합성가스를 이용하는 공정은 촉매를 이용한 화학적 공정과 혐기성 발효에 의한 생물학적 공정의 두 가지로 나뉜다. Clostridium ljungdahlii는 일산화탄소와 수소가 주요 성분으로 구성되는 합성가스를 이용하여 에탄올과 아세트산을 생산할 수 있는 균주 중의 하나로 알려져 있다. 합성가스 발효공정에서 pH는 미생물의 증식과 에탄올 등의 생산에 아주 중요한 요인 중의 하나이다. 본 연구에서는 pH 조건이 미생물의 생장과 에탄올 생산성에 미치는 영향을 조사하였다. C. ljungdahlii 배양은 엄격한 혐기성 조건에서 100 ml의 serum bottle과 pH 제어가 가능한 반응기를 이용한 실험결과, 회분식 배양 조건에서는 미생물의 생장과 에탄올 생산을 위한 최적 초기 pH는 7.0로 나타났다. 미생물 농도는 0.57 g/L, 에탄올 농도 0.91 g/L로 나타났다. pH 4.5 이하에서는 미생물의 생장이 멈추는 것으로 나타났다. pH 제어가 가능한 생물반응기에서는 pH 6.0 일때 에탄올 생산량이 pH 7.0 일때 보다 높게 나타났다. 일정 수준의 미생물 농도를 유지한 조건에서 합성가스를 기포식으로 주입하고 pH 5.9에서 5.4까지 제어하였을 때 미생물량과 에탄올 농도가 증가하였다. 60 시간이 지난 후에 미생물의 농도는 0.498 g/L, 에탄올은 1.056 g/L까지 이르렀다.

Effect of Culture Medium Vitamin Concentration of Culture Medium on Ethanol Production in Syngas Fermentation (합성가스 발효에서 배지 내 Vitamin 농도의 에탄올 생산에 대한 영향)

  • In this study, we assessed the effect of vitamin components (such as biotin, thiamine-HCl, and folic acid) on microorganism microbial growth and ethanol production was examined to enhance increase the ethanol concentration in the Clostridium autoethanogenum culture process using syngas as a sole carbon source. Biotin and folic acid concentrations of 0.2, 2, 20, and 100 ㎍/L were used in the culture experiments at 0.2, 2, 20, and 100 ㎍/L concentrations. The maximum ethanol concentrations of 2.81 g/L and 3.12 g/L were obtained by adding at 0.2 ㎍/L biotin and folic acid, respectively. Moreover, Thiaminethiamine--HCl at concentrations of 0.5, 5, 50, and 250 ㎍/L were was examined evaluated to in the culture experiments. The maximum ethanol concentration of 2.84 g/L was observed at 0.5 ㎍/L of thiamine--HCl. As a resultThus, the optimized concentrations of biotin, thiamine--HCl, and folic acid were determined at 0.2, 0.5, and 0.2 ㎍/L, respectively, for enhancing increasing the ethanol production. In conclusion, the maximum ethanol production was obtained by adding the minimal concentration of vitamins in C. autoethanogenum culture.

  • Cellulosic alcohol fermentation has recently gained more attention in the production of ethanol, butanol, and 2,3-butanediol. However, it was revealed that the process had several hurdles, such as, an expensive cost for biomass decomposition to yield fermentable sugars and a production of byproduct lignin. As an alternative for the process through biomass saccharification, the alcohol production through syngas from biomass has been studied. In this study, we reviewed acetogen and its central metabolic pathway, Wood-Ljungdahl route, capable of utilizing syngas. Furthermore, the metabolic engineering strategies of acetogen for bio-alcohol production from syngas was also reviewed with a brief perspective.

Effect of Tryptone and Tungsten in Medium on Syngas Fermentation Using Clostridium ljundahlii (Clostridium ljundahlii를 이용한 합성가스 발효에서 배지 내 tryptone 및 tungsten의 영향)

  • In this study, an experiment was conducted to investigate the effect of the concentrations of tryptone, an organic nitrogen supplement, and sodium tungstate on the growth of microbial and the production of acetic acid and ethanol in the culture of Clostridium ljungdahlii. Microbial growth increased by 144.6%, and ethanol and acetic acid production improved by 8.6% and 36.7%, respectively, when 2.5 g/L of tryptone was added to the medium of the control experiment (0 g/L tryptone). In the experiment with 1 µM Na2WO4·2H2O, which is 100 times higher than the condition of the medium used in the control experiment (0.01 µM Na2WO4·2H2O), there was no significant difference in microbial growth or total production of C2 metabolites, but ethanol production increased and acetic acid production decreased. As a result, the ethanol/acetic acid production ratio increased significantly from 0.24 in the control experiment to 0.56.

Effect of Heavy Metal on Syngas Fermentation Using Clostridium autoethanogenum (Clostridium autoethanogenum을 이용한 합성가스 발효에 대한 중금속의 영향)

  • In this work, we investigated the effect of the concentration of medium components on microbial growth and ethanol production in order to improve ethanol productivity in the Clostridium autoethanogenum culture process using syngas as a sole carbon source. Molybenum, nickel and cobalt (as heavy metal ions) were selected as examined components, and the effects of components concentration on the cell growth and ethanol production was examined. Among molybdenum concentrations of 0, 0.001, 0.01 and 0.1 g/L. a slight increase in ethanol production was observed at 0.001 g/L, but significant differences in the microbial growth and ethanol production were not observed in the examined concentration range. In the case of nickel concentration of 0, 0.001, 0.01 and 0.1 g/L, the change in the microbial growth and ethanol production was investigated, and it was found that the ethanol production using 0.001 g/L increased by 26% compared to that of using the basal medium concentration (0.01g/L). The effect of cobalt concentrations (0, 0.018, 0.18 and 1.8 g/L) on the microbial growth and ethanol production was also investigated, and the inhibition of microbial growth was observed when the cobalt usage was over 0.18 g/L. In conclusion, cobalt did not show any further improvement of ethanol production by changing concentration, however, molybdenum and nickel showed increases in the produced ethanol concentration compared to that of using 1/10 times of the basal medium concentration.

Effect of Vitamin and Sulfur Sources on Syngas Fermentation Using Clostridium autoethanogenum (Clostridium autoethanogenum을 이용한 합성가스 발효에 대한 비타민과 황 공급원의 영향)

  • In this work, the effect of the culture medium composition on microbial growth and ethanol production in Clostridium autoethanogenum culture was investigated to enhance the ethanol productivity. D-Ca-pantothenate, vitamin B12 (as vitamins), and sodium sulfide (as sulfur source) were selected as examined components, and the effects of components' concentration on cell growth and ethanol production was investigated. For D-Ca-pantothenate concentrations varing from 0.5, 5, 50 and 500 mg/L, a slight increase in the ethanol production was observed at the 0.5 mg/L, but negligible differences in microbial growth and ethanol production were measured for the concentration ranges examined. The effect of vitamin B12 concentrations from 0.1, 1.0, 10, and 100 mg/L on the microbial growth and ethanol production was investigated, and it was found that the ethanol production using a 0.1 mg/L of vitamin B12 concentration increased by 245% compared to that of using the basic medium concentration (10 mg/L). The effect of sodium sulfide concentrations (0.5, 5, and 10 g/L) on the microbial growth and ethanol production was also studied, and the inhibition of microbial growth was observed when the sodium sulfide usage was over 0.5 g/L. In conclusion, changes in D-Ca-pantothenate and sodium sulfide concentrations did not affect the ethanol production, whereas even a 100 times lower concentration of vitamin B12 than that of the basic medium improved the production.
