Gelation Properties and Industrial Application of Functional Protein from Fish Muscle-1. Effect of pH on Chemical Bonds during Thermal Denaturation (기능성 어육단백질의 젤화 특성과 산업적 응용-1. 가열변성 중 화학결합에 미치는 pH의 영향)

  • The effect of pH on surface hydrophobicity, sulfhydryl group, infrared spectrum, SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis) pattern and enthalpy was investigated in recovered protein from mackerel and frozen blackspotted croaker by alkaline processing. Hydrophobic residue in myofibrillar protein exposed to the surface of protein, and hydrophobic interaction were the highest around 6$0^{\circ}C$. The surface hydrophobicity was different between myofibrillar protein and myofibrillar protein including sarcoplasmic protein (recovered protein). The peak at 1636 c $m^{-l}$ was increased with pH, and the recovered protein was unfolded in alkali pH. Difference of surface and total sulfhydryl group at pH 7.0 and 10 was comparative high, and decrease of surface sulfhydryl group indicated formation of S-S bonds. Mackerel and frozen blackspotted croaker in alkaline pH showed bands of polymerized myosin heavy chain on SDS-PAGE pattern. The transition temperatures of recovered protein were 33.1, 44.3 and 65.5$^{\circ}C$. Gelation of recovered protein from alkali processing was estimated by increase of $\beta$-sheet structure by pH treatment, S-S bonds by oxidation of surface sulfhydryl group in heating, polymerization of myosin heavy chain in order.r.

Surface and Corrosion Protection Properties of Fluorine Doped PVDF by Plasma Fluorination (플라즈마 불소화에 의해 제조된 불소 도핑 PVDF의 표면 및 부식방지 특성)

  • Polyvinylidene fluoride (PVDF) is a promising coating material because of its outstanding processability. The PVDF coating, however, has limitations in anti-corrosion application due to its weak hydrophobicity compared to that of other fluoropolymers. In this study, plasma fluorination was performed using carbon tetrafluoride (CF4) gas to improve anti-corrosion properties of PVDF. The fluorine content and hydrophobicity of PVDF were investigated in different CF4 flow rates, followed by the determination of anti-corrosion properties. The fluorine content on the surface of the PVDF film increased by up to 46.70%, but the surface free energy was independent of CF4 flow rate. Meanwhile, the surface roughness of the PDVF film tended to increase by up to 150% and then decrease with increasing CF4 flow rate. It is considered that the plasma fluorination affects the surface free energy due to the introduction of fluorine functional groups and surface etching. In addition, the degree of corrosion of the PVDF-coated Fe plate was significantly reduced from 49.2% to 19.0% compared to that of the uncoated Fe plate. In particular, the degree of corrosion of the fluorinated PVDF-coated Fe plate was 13.6%, which was 28.4% lower than that of the PVDF-coated Fe plate, showing improved anti-corrosion protection.

Monitoring of Structural Changes during in vitro Unfolding and Refolding of Recombinant Human Growth Hormone (재조함 인성장호르몬의 in vitro 풀림과 재접힘 과정의 구조변화 모니터링)

  • Using recombinant human growth hormone as a model protein, we carried out unfolding by adding a denaturant such as urea, guanidine HCl, or SDS followed by refolding by dilution and dialysis. The objectives were to monitor the structural changes during in vitro refolding process and, based on the results, to develop a quantitative method of refolding progress assessment. The changes in surface hydrophobicity were measured by fluorescence tagging of 1-anilinonaphthalene-8-sulfonate(1,8-ANS) to the hydrophobic portions, and those in the secondary structure were monitored by using far UV-CD(circular dichroism) spectroscopy. Also, we used RP-HPLC to separate and quantify the folded and unfolded proteins to correlate the result with the structure analysis. Our results indicate the surface hydrophobicity are well correlated with the formations of the secondary structure, primarily ${\alpha}$-helices, as well as the disulfide bridges. We expect this monitoring technique can be applied in industrial fields as a means to quantitatively assess the progress of in-vitro refolding of recombinant proteins.

Improving Microalgal Biomass Productivity and Preventing Biofouling in Floating Marine Photobioreactors via Sulfonation of Selectively Permeable Membranes (부유형 해양 광생물반응기의 선택적 투과막의 술폰화 반응을 통한 Biofouling 억제 및 미세조류 생산성 향상)

  • The purpose of this study was to inhibit biofouling on a selectively permeable membrane (SPM) and increase biomass productivity in marine photobioreactors (PBRs) for microalgal cultivation by chemical treatment. Surfaces of a SPM, composed of polyethylene terephthalate (PET), was sulfonated to decrease hydrophobicity through attaching negatively charged sulfonic groups. Reaction time of sulfonation was varied from 0 min to 60 min. As the reaction time increased, the water contact angle value of SPM surface was decreased from $75.5^{\circ}$ to $44.5^{\circ}$, indicating decrease of surface hydrophobicity. Furthermore, the water permeability of sulfonated SPM was increased from $5.42mL/m^2/s$ to $10.58mL/m^2/s$, which reflects higher nutrients transfer rates through the membranes, due to decreased hydrophobicity. When cultivating Tetraselmis sp. using 100-mL floating PBRs with sulfonated SPMs, biomass productivity was improved by 34% compared with the control group (non-reacted SPMs). In addition, scanning electron microscopic observation of SPMs used for cultivation clearly revealed lower degree of cell attachment on the sulfonated SPMs. These results suggest that sulfornation of a PET SPM could improve microalgal biomass productivity by increasing nutrients transfer rates and inhibiting biofouling by algal cells.

Probiotic Potential of Pediococcus pentosaceus BCNU 9070 (프로바이오틱 Pediococcus pentosaceus BCNU 9070 균주)

  • Lactic acid bacteria are generally recognized as beneficial probiotic organisms. Recent studies revealed that the potential of probiotic strains was essentially dependent on the bacterial-binding and adhesion capabilities to gut epithelial cells and the hydrophobicity of the cell surface. In this study, we screened some indigenous lactic acid bacteria from Kimchi and selected one lactic acid bacterium as a potential probiotic based on its cell surface hydrophobicity. Analysis of the 16S rRNA gene sequences of probiotic isolates indicated that the selected isolate (BCNU 9070 strain) was a member of Pediococcus pentosaceus. P. pentosaceus BCNU 9070 showed resistance to bile acids and acidic pH. The P. pentosaceus BCNU 9070 strain also inhibited the cell growth of six food-borne pathogens including Listeria monocytogenes and Shigella sonnei. In addition, the P. pentosaceus BCNU 9070 strain expressed bile salt hydrolase activity and showed an ability to assimilate cholesterol in vitro. On the basis of these results, P. pentosaceus BCNU 9070 is considered to have probiotic potential for applications in functional foodstuffs.

Epicuticular Waxes and Stomata of Adult Scale Leaves of the Chinese Juniper Juniperus chinensis

  • Leaf surface structures were investigated in the Chinese juniper Juniperus chinensis by scanning electron microscopy. Adult scale leaves were collected from the tree, air-dried at room temperature, and sputter-coated with gold without further specimen preparation. Approximately fi ve stomata were locally distributed and arranged in clusters on the leaf surface. Stomata were ovoid and ca. 40 ${\mu}m$ long. The epicuticular wax structures of J. chinensis leaves were tubules and platelets. Numerous tubules were evident on the leaf regions where stomata were found. The tubules were cylindrical, straight, and ca. 1 ${\mu}m$ in length. They almost clothed the stomatal guard cells, and occluded the slit-shaped stomatal apertures. Moreover, the wax ridges were flat crystalloids that were connected to the surface by their narrow side. They did not have distinct edges, and their width/height ratio varied. In particular, the wax ridges could be discerned on the leaf regions where stomata were not present nearby. Since the wax ridges did not have distinct edges on their margin, they were identified as platelets. Instances were noted where platelets were oriented either parallel to each other or perpendicular to the cuticle surface. These results can be used in biomimetics to design the hierarchical structures for mimicking the plant innate properties such as hydrophobicity and self-cleaning effects of the leaf surface.

PDMS Surface Modification for hydrophilicit Using 2-Hydroxyethyle Methacrylate without Monomethyl Ether Hydroquinone (친수성 유지를 위해 Monomethyl Ether Hydroquinone이 제거된 2-Hydroxyethyle methacrylate로 처리하는 PDMS 표면개질)

  • In this paper, the surface modificatioin of PDMS (polydimethyl-siloxane) which is a useful material of microfluidic devices is presented. PDMS-based devices can be fabricated by casting the polymer in a mold, but the porosity and the hydrophobicity of PDMS make difficult to use as bio-medical devices. To overcome these disadvantages, the PDMS surface is grafted with HEMA (2-Hydroxyethyle methacrylate) treatments and $O_2$ plasma process. The $O_2$ plasma process is performed for 20 sec after curing PDMS, and PDMS is put in the prepared HEMA without Monomethyle Ether Hydroquinone. Residual monomers and homopolymers of HEMA-treated PDMS surface are removed using soxhlet extractor. The PDMS surface modification using HEHA without Monomethyle Ether Hydroquinone is experimented, and compare to when additing $FeCl_2{\cdot}2H_2O$. A method with a soxhlet extractor compare to the existing rinse method. The hydrophilicity is confirmed by the measurement of a contact angle, and we observe whether the hydrophilicity is retained.

  • Surface modification of silicone rubber by low temperature plasma process was investigated to improve quality of silicone EVD tube by reducing tackiness and hydrophobicity. Treatment with nonpolymer-forming plasmas and thin film deposition with polymer-forming plasmas were tried. Tackiness could significantly be reduced, especially by thin film deposition. As a result, the tube became slippery and less vulnerable to contamination in laboratory environment. Inner as well as outer surface of the tube could be changed to be hydrophilic if the plasma contained oxygen. As a result, initial hydrodynamic resistance was reduced. The surface modification did not give any bad influence on mechanical properties of the silicone tube in most cases. Rather, some properties such as Young's modulus, ultimate tensile strength and elongation at break were improved.

Surface Characterization of Silicone Rubbers by Corona Discharge (코로나 방전처리에 따른 실리콘 고무의 표면특성 변화)

  • In this paper we investigated the changes of surface hydrophobic characteristics on silicone rubbers by corona discharge treatment and also investigated the distribution and the behavior of low molecular weight(LMW) silicone fluid which was extracted by solvent-extraction with gel permeation chromatography(GPC). It was shown that contact angle was $110.5^{\circ}$ on initial sample but contact angle was approximately decreased to $10^{\circ}$ after 45 minutes. However the surface hydrophobic characteristic on silicone rubbers which were removed from corona discharge was recovered within 5 hours. It was shown that corona discharge insured the increase of diffusible LMW chains, which could lead to recover the surface hydrophobicity. The surface hydrophobic characteristics on silicone rubbers and the recovery mechanism based on our results were discussed.

  • Magnesium and its alloys are prone to be corroded, thus surface treatments improving corrosion resistance are always required for practical applications. As a surface treatment of magnesium alloys, plasma electrolytic oxidation (PEO), creating porous stable oxide layer by a high voltage discharge in electrolyte, enhances the corrosion resistance. However, due to superhydrophilicity of the porous oxide layer, which easily allow the penetration of corrosive media toward magnesium alloys substrate, post-treatments inhibiting the transfer of corrosive media in porous oxide layer are required. In this work, we employed a hydrophobizing method to enhance the corrosion resistance of PEO treated Mg alloy. Three types of hydrophobizing techniques were used for PEO layer. Thin Teflon coating with solvent evaporation, self-assembled monolayer (SAM) coating of octadecyltrichlorosilane (OTS) based on solution method and SAM coating of perfluorodecyltrichlorosilane (FDTS) based on vacuum method significantly enhances corrosion resistance of PEO treated Mg alloy with reducing the contact of water on the surface. In particular, the vacuum based FDTS coating on PEO layer shows the most effective hydrophobicity with the highest corrosion resistance.
