Effect of Phenazine Dioxide Derivatives on the Hepatic Xanthine Oxidase Activity (Phenazine dioxide 유도체가 간 Xanthine Oxidase 활성에 미치는 영향)

  • 8-Acyl-2-hydroxyphenzaine -5, 10-dioxides and 8-acyl-2-aminophenazine-5, 10-dioxides bearing n-butanoyl, n-hexanoyl and n-octanoyl groups were synthesized. It was attempted to observe the effect of phenazine dioxide derivatives on the hepatic xanthine oxidase activity in this study. As the activity of xanthine oxidase, the key enzyme in the generation of superoxide anion radical ($O_2$), was measured in the presence of phenazine dioxide derivatives, the action of 8-acyl-2-hydroxyphenazine-5,10-dioxide derivatives inhibited with increase of numbers of carbon atom bearing 8-acyl group. Moreover, when plotted on double reciprocal form, the Vmax value decrease as increase of numbers of carbon atom bearing acyl groups without affecting the Km value. However, the hepatic xanthine oxidase activity was not changed by 8-acyl-2-aminophenazine-5,10-dioxide derivatives.

The biological activities of Zelkova leaves and the quality characteristics of Zelkova leaf rice cake according to leaf levels (느티잎의 생리활성 및 느티잎 첨가량에 따른 느티떡의 품질특성)

  • The objectives of this study were to determine the antioxidant and cytotoxic activities of Zelkova leaf extract and the sensory and textural characteristics of Zelkova leaf rice cake. The ethyl acetate layer of Zelkova leaf extract showed the most effective DPPH radical and superoxide anion scavenging activities. The residual layer of Zelkova leaf extract exhibited potent cytotoxicity against the HeLa cell line. In the sensory evaluation, the color and taste of Zelkova leaf rice cakes were not significantly different. Zelkova leaf rice cakes with 20%(5.18), 25%(4.94) and 30%(4.88) added Zelkova leaves showed higher overall acceptability than 0%(4.06) added Zelkova leaves. In the textural analysis, the hardness of the Zelkova leaf rice cake increased slowly with increasing leaf content during 3 day storage. The cohesiveness of 25% and 30% Zelkova leaf rice cakes showed no variation with storage time but that of 0% Zelkova leaf rice cakes decreased with increasing storage period. The 'L' value of Zelkova leaf rice cake decreased with increasing leaf content while the 'a' value of 0% Zelkova leaf rice cake was lower than that of 20%, 25% and 30% Zelkova leaf rice cakes.

  • This study was undertaken to evaluate the gastrotherapeutic effects of the extract of Rosa rugosa. It can be regarded that the antigastric and anti-ulcerative effect of Rosa rugosa is originated from the reduction of total acid output identified by gastric secretion test. To clarify the protective mechanism of the Rosa rugosa extract, the gastropathy was induced in rats with alcohol-salicylate and the activities of the free radical scavenging enzymes were examined. The acitivity of superoxide dismutase and glutathione peroxide were significantly increased and the total content of glutathione was recovered. We concluded that the protective effect of the extract of Rosa rugosa on gastropathy in rats is its ability increased the activities of the free radical scavenging enzymes.

Augmentation of antioxidant system: Contribution to antimalarial activity of Clerodendrum violaceum leaf extract

  • Reactive oxygen species are known to mediate various pathological conditions associated with malaria. In this study, the antioxidant potential of Clerodendrum violaceum leaf extracts, an indigenous antimalarial remedy, was evaluated. Total phenol, flavonoid, selenium, vitamins C and E contents of Clerodendrum violaceum leaf extracts were determined. The free radical scavenging activities of the extracts against DPPH, superoxide anion and hydrogen peroxide coupled with their reducing power were also evaluated in vitro. Moreover, responses of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR) in a rodent malaria model to a 4-day administration of Clerodendrum violaceum leaf extracts were also evaluated. The methanolic extract was found to contain the highest amounts of antioxidant compounds/element and also demonstrated the highest free radical scavenging activity in vitro. The results showed a significant decrease (p < 0.05) in SOD and CAT activities with a concurrent significant (p < 0.05) increase in GPx and GR activities in both erythrocytes and liver of untreated Plasmodium berghei NK65-infected animals compared to the uninfected animals. The extracts were able to significantly increase (p < 0.05) SOD and CAT activities and significantly reduce (p < 0.05) GPx and GR activities in both the liver and erythrocytes compared to those observed in the untreated infected animals. The results suggest the augmentation of the antioxidant system as one of the possible mechanisms by which Clerodendrum violaceum extract ameliorates secondary effects of malaria infection, alongside its antiplasmodial effect in subjects.

A Prolyl Endopeptidase-lnhibiting Antioxidant from Phyllanthus ussurensis

  • A prolyl endopeptidase inhibitor was isolated from the ethyl acetate soluble fraction of Phyllanthus ussurensis. The active compound was identified as an ellagitannin, corilagin. It was shown to non-competitively inhibit prolyl endopeptidase (PEP) with the $IC_{50}$ value of $1.17 \times $10^{-6}\mu$M. The Ki value was $6.70 \times 10^{-7}$ M. Corilagin was less inhibitory to other serine proteases such as chymotrypsin, trypsin, and elastase, indicating that it was relatively a specific inhibitor of PEP. Corilagin also effectively inhibited reactive oxygen species such as hydroxide and superoxide anion radical, hydrogen peroxide, and DPPH. Especially, corilagin showed potent scavel1ging activity on the superoxide anion radical in the ESR method ($IC_{50} =3.79 \times 10^{-6}$M) as well as xanthine oxidase system.

  • This study was undertaken to examine the effect of Jwagyuyeum and Woogyuycum, being known to reinforce Kidney-yin and yang, on the activities of endogenous antioxidant enzymes and the production of oxygen free radicals in the liver and kidney tissues, Alterations in enzyme activities were observed after in vivo treatment in rats, Jwagyuyeum and Woogyuyeum caused a significant increase in the activities of superoxide dismutase(SOD) and catajase Jwagyuyeum significantly increased the activity of glutathione peroxidase in both liver and kidney, but the enzyme activity was not significantly altered by Woogyuyeum. Treatment in vitro of Jwagyuyeum and Woogyuyeum decreased the production of oxygen free radicals in a dose-dependent fashion. These results suggest that Jwagyuyeum and Woogyuyeum stimulate the activities of antioxidant enzymes and inhibit directly the production of oxygen free radicals. These effects of both herbs may contribute to prevent the oxygen free radical-induced impairment of cell function.

Some Physiological Activities Observed from Ethanol Extracts of Germinated Specialty Rough Rice

  • This study analyzed the some of the physiological activities of 70% ethanol extracts of specialty rough rice ('Goami2', 'Keunnunbyeo', and 'Heugkwangbyeo' rough rice cultivars) before and after germination. The physiological activities of the extracts were analyzed for their inhibition of linoleic acid peroxidation, hydrogen peroxide radical scavenging activity, superoxide dismutase-like activity, xanthine oxidase inhibitory activity, and nitrite scavenging effects. The most active rough rice cultivar was 'Heugkwangbyeo' and germinated 'Heugkwangbyeo' rough rice was more effective than ungerminated rough rice, having higher inhibition activity of linoleic acid peroxidation (1.2-fold), hydrogen peroxide radical scavenging activity (1.4-fold), superoxide dismutase-like activity (1.2-fold), and xanthine oxidase inhibitory activity (1.1-fold). The nitrite scavenging effect was increased after germination (2.1-fold) and decreased as pH increased. According to the results obtained in this study, 70% ethanol extracts from germinated 'Heugkwangbyeo' rough rice can be used for industrial applications such as the functional food materials.

Anti-Oxidant Effects of Highly Bioavailable Curcumin Powder in High-Fat Diet Fed- and Streptozotocin-Induced Type 2 Diabetic Rats

  • Curcumin is a hydrophobic polyphenol extracted from turmeric that exhibits a variety of biological functions has albeit with limited efficacy as a functional food material owing to its low absorption when administered orally. The newly developed curcumin powder formulation exhibits improved absorption rate in vivo. This study evaluates the anti-oxidant effects of $Theracurmin^{(R)}$ (TC), which is highly bio-available in curcumin powder. The antioxidant activity of TC was investigated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity, ferrous reducing antioxidant power (FRAP) assays, NO radical, superoxide radical, $H_2O_2$ scavenging activity, and total antioxidant capacity (TAC). Additionally, we evaluated the antioxidant activity of TC in high-fat diet (HFD)-fed streptozotocin (STZ)-induced Type 2 diabetic rats. As a result of oral administration of TC for 13 weeks in type 2 diabetic rats, the group administration of 2,000 mg/kg significantly increased FRAP, superoxide dismutase (SOD), and reduced the level of glutathione (GSH) in liver tissue 1.9, 1.2, and 1.2-times, respectively. Furthermore, serum TAC levels increased by 1.3-fold after the rats were administered with a dose of 500 mg/kg. These results were consistent with the in vitro assay results. In conclusion, TC exhibited its potential as a functional food material through its antioxidant properties.

Induction of Iron Superoxide Dismutase by Paraquat and Iron in Vitreoscilla $C_1$ (Vitreoscilla $C_1$에서 paraquat와 Iron에 의한 Iron Superoxide Dismutase의 유도)

  • Superoxide dismutase which is metalloenzyme that decomposes superoxide radicals into hydrogen peroxide and molecular oxygen. Vitreoscilla has FeSOD. Expression of FeSOD to paraquat was largely constitutive. This suggests that the basal level of FeSOD is sufficient to provide protection against superoxide generated during normal aerobic metabolism. Induction of SOD by iron supports that insertion of the active site metal into the corresponding apoprotein. The effect of paraquat on induction by iron seemed that iron brought the synergism effect in SOD activity with paraquat. It suggests that the relief of growth inhibition is due to protection against the lethality of O$_2$afforded by the elevated SOD. There may be control of FeSOD activity posttranslationally. Posttranslation control of enzyme function is particularly feasible for a metalloenzyme, for which conversion of apo- to holoenzyme may be the rate-limiting or regulatory step.

The antioxidative effects of Ampelopsis brevipedunculata extracts (개머루덩굴 추출물의 항산화 효과)

  • This study was carried out to investigate the antioxidative capacity of Ampelopsis brevipedunculata 95% ethanol extracts. The concentration of A. brevipedunculata extract at which DPPH radical scavenging activity was inhibited by 50% was 0.42 mg/mL as compared to 100% by pyrogallol as a reference. Total antioxidant status was examined by total antioxidant capacity against ABTS radical reactions. Total antioxidant capacities of A. brevipedunculata extract at the concentrations of 0.1 and 1 mg/mL were 0.65 and 3.71 mM Trolox equivalents, respectively. Oxygen radical absorbance capacities of A. brevipedunculata extract at the concentrations of 5 and $100\;{\mu}g/mL$ were 22.75 and $131.25\;{\mu}M$ gallic acid equivalents, respectively. Superoxide scavenging activities of A. brevipedunculata extract at the concentrations of 0.1 and 1 mg/mL were 27.7 and 56.0%, respectively. Total phenolic contents of A. brevipedunculata extract at the concentrations of 0.5 and 2.0 mg/mL were 0.55 and 2.06 mM gallic acid equivalents, respectively. A. brevipedunculata extract at the concentration of 0.1 mg/mL inhibited 0.2 mM and 0.5 mM tert-butyl hydroperoxide induced cyototoxicity by 36.2 and 23.3%, respectively, in HepG2 cell culture system. Thus strong antioxidant and cytotoxicity-inhibiting effects of A. brevipedunculata extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation as well as high levels in total phenolic contents.
