Screening of antioxidant and antimicrobial activities of Caesalpinia bonducella Flem., leaves (Caesalpiniaceae)

  • The study was aimed at evaluating the antioxidant and antimicrobial activities of methanol extract of Caesalpinia bonducella leaves (MECB) (Family: Caesalpiniaceae). The effect of MECB on antioxidant activity, reducing power, free radical scavenging (DPPH radical, nitric oxide radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide radical scavenging), total phenolic content and antimicrobial activities were studied. The antioxidant activity of MECB increased in a dose dependent manner. About 50, 100, 250 and 500 g of MECB showed 53.4, 61.2, 69.1 and 76.2 % inhibition respectively on peroxidation of linoleic acid emulsion. Like antioxidant activity, the effect of MECB on reducing power increased in a dose dependent manner. The free radical scavenging activity of MECB was determined by DPPH radical scavenging method. The potency of this activity was increased with increased amount of extract. MECB was found to inhibit the nitric oxide radicals generated from sodium nitroprusside $(IC_{50}\;=\;102.8\;g/ml)$ whereas the $IC_{50}$ value of curcumin was 20.4 g/ml. Moreover, the MECB was found to scavenge the superoxide generated by photoreduction of Riboflavin. MECB was also found to inhibit the hydroxyl radical generated by Fenton reaction, where the $IC_{50}$ value is 104.17 g/ml compared with catechin 5 g/ml, which indicates the antioxidant activity of MECB. The MECB capable of scavenging hydrogen peroxide in a concentration-dependent manner. The amounts of total phenolic compounds were also determined. Antimicrobial activities of MECB were carried out using disc diffusion methods with five Gram positive, four Gram negative and four fungal species. The results obtained in the present study indicate that MECB leaves are potential source of natural antioxidant and antimicrobial agents.

Free Radicals Scavenging Activity of Bulro Kugi (Lycium chinense Mill) Fruit, Leaf and Root (불로 구기 부위별 자유라디칼 소거효과에 관한 연구)

  • Free radical-scavenging activities of Korean Bulro Kugi (Lycium chinense Mill) fruit, leaf, and root were evaluated. Total phenolic contents of fruit, leaf, and root were $1,078.4{\pm}61.0$, $939.9{\pm}19.8$, and $3,792.2{\pm}106.6mg/L$, and their flavonoids were $396.7{\pm}15.2$, $1,952.9{\pm}21.3$, and $425.3{\pm}13.5mg/L$, respectively (p<0.001). HPLC revealed main polyphenolic compounds in fruit were p-coumaric and syringic acids in fruit, p-coumaric, syringic, and procatechuic acids in leaf, and p-coumaric, syringic, caffeic, and procatechuic acids in root. Highest radical-scavenging activities of superoxide anion and hydroxyl were found in leaf and root, respectively (p<0.001).

Antioxidative Effect of Rhus javanica Linne Extract Against Hydrogen Peroxide or Menadione Induced Oxidative Stress and DNA Damage in HepG2 Cells

  • The free radical scavenging activities and the protective effects of Rhus javanica extracts against oxidative damage induced by reactive oxygen species (ROS) were investigated. n-Hexane, ethyl acetate and water fractions were prepared from a methanol extract. DPPH radical, superoxide anion and hydroxyl radical scavenging activities were estimated. Intracellular ROS formation was quantified using fluorescent probes, 2', 7'-dichlorofluorescin diacetate (DCFH-DA) for hydroxyl radical and dihydroethidium (DHE) for superoxide anion. The oxidative DNA damage was investigated by the comet assay in HepG$_2$ cells exposed either to $H_2O$$_2$ or to menadione. The highest $IC_{50}$/ values for DPPH radical scavenging activity was found in the ethyl acetate fraction with a value of 5.38 $\mu\textrm{g}$/mL. Cells pretreated with $\geq$ 1 $\mu\textrm{g}$/mL of the ethyl acetate extract had significantly increased cell viability compared to control cells, which were not pretreated with the extract. Intracellular ROS formation and DNA damage in HepG$_2$ cells, which were pretreated with the various concentrations of Rhus javanica ethyl acetate extract and then incubated either with $H_2O$$_2$ or with menadione, reduced in a dose-dependent manner. These findings suggest that Rhus javanica might have biologically active components which have strong protective effects against ROS induced oxidative damages to the biomolecules, such as cell membranes and DNA.

Antioxidant Activities in Enzymatic Hydrolysates of Pumpkin Powder (Cucurbita spp.) (호박분말 효소가수분해물의 항산화활성)

  • This study was conducted to investigate total polyphenol contents and antioxidative effects of the enzymatic hydrolysates digested by several kinds of carbohydrases from the powder of ripened pumpkin (Cucurbita moschata), sweet pumpkin (C. maxima) and green pumpkin (C. moschata). The total polyphenol contents of all enzymatic hydrolysates from green pumpkin powder were higher than those of ripened and sweet pumpkins. DPPH radical scavenging activities of the enzymatic hydrolysates digested by AMG and Termamyl from green pumpkin were also very strong compared to ripened and sweet pumpkins. However, the most enzymatic hydrolysates of ripened and sweet pumpkin powders, except Viscozyme digestion, were higher superoxide anion scavenging activities than green pumpkin powder. Hydrogen peroxide scavenging activities in the enzymatic digests (not Ultroflo) of green pumpkin were potent compared to other pumpkin powders whereas hydroxyl radical scavenging activities were low at less than 14% in hydrolysates of all pumpkin species. Nitric oxide scavenging activities were very effective in Viscozyme digests of sweet and green pumpkin, and other enzymatic hydrolysates also showed higher activity than $\alpha$-tocopherol control (not BHT).

Antioxidant Properties of Flavone-6(4')-Carboxaldehyde Oxime Ether Derivatives

  • The in vitro antioxidant properties of some flavone-6(4)-carboxaldehyde oxime ether deriva-tives (Ia-f, lIa-f) were determined by their effects on the rat liver microsomal NADPH-dependent lipid peroxidation (LP) levels by measuring the formation of 2-thiobarbituric acid reactive substances. The free radical scavenging properties of the compounds were also examined in vitro by determining their capacity to scavenge superoxide anions and interact with the stable free radical 2, 2-diphenyl-1-picrylhydrazyl (DPPH). The most active compounds, lib (Flavone-4'-carboxaldehyde-O-ethyl oxime) and Id (Flavone-6-carboxaldehyde-O-[2-(1-pyrolidino) ethyl] oxime), caused 98 and 79% inhibition of superoxide anion production and DPPH stable free radical at $10^{-3}{\;}M$, respectively.

A Study of Effects of Crataegus pinnatifida var. psilosa Extracts (좁은잎산사나무 열매 추출물의 효능에 관한 연구)

  • The purpose of this study was to investigate effects of Crataegus pinnatifida var. psilosa extracts obtained from 95% methanol and water. Methylene chloride, ethyl acetate, and methanol were used to fractionate the 95% methanol extract and each fraction was testified total polyphenol contents, electron donating abilities, the scavenging abilities of superoxide anion radical, as well as hydrogen peroxide. Extraction yields of 95% methanol and water from Crataegus pinnatifida var. psilosa were 25.40%, 23.12% respectively. Total polyphenol contents were $28,708.0{\pm}1,755.05{\mu}g\;GAE/mL$ in 95% methanol, revealing the highest among them, $12,726.67{\pm}479.33{\mu}g\;GAE/mL$ in water extract, $15,854.67{\pm}498.38{\mu}g\;GAE/mL$ in methanol fraction, $11,810.67{\pm}584.48{\mu}g\;GAE/mL$ in ethyl acetate fraction, and $5294.67{\pm}190.36{\mu}g\;GAE/mL$ in methylene chloride fraction. Total polyphenol contents revealed significant differences (p<0.05) between the solvents. In the experiment of the electron donating ability, water extract revealed $84.33{\pm}0.1%$ scavenging ability, the highest. Other extracts and fractions were $81.8{\pm}1.11%$ for water, $79.73{\pm}1.32%$ for ethyl acetate fraction, $75.73{\pm}2.17%$ for methylene chloride fraction, and $42.1{\pm}5.01%$ for methanol fraction, the lowest electron donating ability. Electron donating abilities revealed significant difference (p<0.05) between the solvents. In the experiment of superoxide anion radical scavenging ability, ethyl acetate fraction($0.0026{\pm}0.0002$) had the highest scavenging ability, and the others revealed slight increase rather than decrease in scavenging ability. Hydrogen peroxide scavenging ability revealed the highest in methanol fraction ($-0.00206{\pm}0.00165$) and the others were as follow; water extract ($0.00157{\pm}0.00249$), 95.0% methanol extract ($0.005{\pm}0.0036$), methylene chloride fraction ($0.0039{\pm}0.00364$), and ethyl acetate fraction ($0.0002{\pm}0.00059$).

Radical Scavenging and Antioxidant Effects of Juglandis Semen Extract(JSE) (호도약침액(胡桃藥鍼液)의 유리기(遊離基) 소거(消去)와 항산화(抗酸化) 효과(效果)에 대한 실험적(實驗的) 연구(硏究))

  • This study was performed to determine if Juglandis semen extract(JSE) has free radical scavenging and antioxidant activities. Superoxide anion generation by xanthine oxidase/xanthine and in neutrophils activated by phorbol-12, 13-dibutyrate was inhibited by JSE and its effect was dose-dependent. JSE also inhibited generation of $H_2O_2$ induced by glucose oxidase/glucose and in opossum kidney cells treated with antimycin A. JSE exerted a direct $H_2O_2$ scavenging effect. Exposure of opossum kidney cells to 1mM tBHP caused a significant increase in lipid peroxidation, which was prevented by JSE. JSE also prevented tBHP-induced LDH release. These data suggest that JSE has free radical scavenging and antioxidant activities. However, further studies should be carried out to find the active ingredient(s) of JSE that exerts radical scavenging action.

Antioxidative and Antiallergic Effects of Aronia (Aronia melanocarpa) Extract (아로니아(Aronia melanocarpa)로부터 유래한 추출물의 항산화 및 항알레르기 효능)

  • The purpose of this study was to investigate the antioxidative and antiallergic effects of aronia extract. The aronia extract was tested for various antioxidative potentials (scavenging activity of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and superoxide anion radical) and inhibitory effect of 5-lipoxygenase (5-LO) and cyclooxygenase (COX). In aronia extract, polyphenol content was $81.6{\pm}3.3\;mg/g$ and flavonoid content was $5.15{\pm}0.16\;mg/g$. DPPH radical-scavenging activity ($SC_{50}$) of aronia extract was $6.15{\pm}0.343\;ppm$. $SC_{50}$ value of aronia extract against superoxide anion radical was $6.99{\pm}1.28\;ppm$. In addition, $IC\_{50}$ values of aronia extract against 5-LO, COX-1, and COX-2 were $42.07{\pm}0.15\;ppm$, $24{\pm}4.29\;ppm$, and $24.3{\pm}3.5\;ppm$, respectively. These results suggest that aronia extract may be useful for the prevention or treatment of allergic disease.

Screening on Radical Scavenging Activity of Salt Marsh Plants (염생식물로부터 천연항산화 활성소재 탐색)

  • The antioxidant activities of methanol and dichloromethane extracts of eighteen salt marsh plants were tested by using 1,1-Diphenyl-2-picryl-hydrazyl(DPPH) at a $100{\mu}g/ml$ concentrations. The methanol extracts of four salt marsh plants(Rosa rugosa Thunberg, Erigeron annuus, Ixeris tamagawaensis kitamura, Artemisia capillaris Thunberg) were found to be most effective in DPPH radical scavenging activity. The DPPH radical scavenging effect of these plants was Artemisia capillaris Thunberg(88.67%), Rosa rugosa Thunberg(87.51%), Erigeron annuus(78.49%), Ixeris tamagawaensis kitamura(69.99%) in order and was comparable with that of natural antioxidant, L-ascorbic acid(96.64%). The next effective ones were the methanol extracts of Teragonia tetragonoides(58.66%) and the dichloromethane extracts of Artemisia capillaris Thunberg(54.67%) and the others did not show a considerable activity. These salt marsh plants were evaluated using the pyrogallol UV-VIS spectrophotometeric method to generate superoxide anion. Among them, the dichloromethane extracts of four salt marsh plants and the methanol extracts of fifteen ones were weak superoxide dismutase-like activities.

The Inhibition of UVA-induced Matrix Metalloproteinase-1 in Human Dermal Fibroblasts and the Improvement of Skin Elasticity by Cirsium setidens Extract (고려엉겅퀴 추출물의 사람 섬유아세포에 있어서 자외선으로 유도된 MMP-1발현 저해와 피부 탄력 개선 효과)

  • In this study, we measured the anti-oxidative activity of Cirsium setidens and investigated its effect on UVA-induced MMP-1 expression in human dermal fibroblats. And then we examined possible improvement in skin elasticity by topical treatment with fomular including Cirsium setidens extract. The ethanol extract of C. setidens showed free anion radical scavenging effect(87.47 % at 1 mg/mL) and superoxide anion radical scavenging effect(61.71 % at 1 mg/mL) in the xanthine/xanthine oxidase system, respectively. At the concentration of 100 ${\mu}g/mL$, C. setidens extract showed 95.54% inhibition on lipid peroxidation of linoleic acid. UVA-induced MMP-1 expression in human dermal fibroblasts was reduced to 54.69 % by treatment with 100 ${\mu}g/mL$ of C. setidens extract. A human clinical study, in which oil-in-water emulsion with C. setidens extract was topically applied, showed significant increase in skin elasticity. These results suggest that the C. setidens extract can be effective anti-aging ingredient for cosmetics applications.
