참다래 '홍양' 품종의 차등발현유전자 분석 (Analysis of Differentially Expressed Genes in Kiwifruit Actinidia chinensis var. 'Hongyang')

  • 적색 과육 '홍양' 품종에서 차등발현하는 유전자를 찾기 위하여 mirror orientation selection (MOS)과 결합된 suppression subtractive hybridization (SSH) 실험을 수행하였다. 그 결과, 288개의 cDNA clone을 확보하였으며, colony PCR을 통해 192개의 positive clone을 선발하였고, 이들을 sequencing하였다. NCBI/Genbank 데이터베이스의 BLAST 검색를 이용하여 염기서열을 분석한 결과, 30개의 clone에서 기존에 알려진 유전자기능과의 유사성을 확인할 수 있었으며, 10개의 clone이 특이유전자였다. 그 중 3개의 clone(AcF21, AcF42, AcF106)은 과실 후숙과 관련된 ACC-oxidase와 상동성이 있었다. SSH의 결과를 통해 얻어진 이 유전자들의 차등발현양상을 확인하기 위하여 reverse transcription PCR(RT-PCR)과 quantitative real-time PCR(qReal-time PCR) 분석을 실시하였다. qReal-time PCR분석과 RT-PCR분석에서 모두 동일한 결과를 확인할 수 있었으며, 3개 clone 모두 '홍양'에서의 유전자발현수준이 '헤이워드'보다 더 높았다. AcF21은 다른 유전자들보다 가장 높은 발현수준을 나타내었는데, 만개 후 120일과 160일 모두 '홍양'에서의 발현수준이 높았다.

  • 음악은 수많은 악기가 내는 사운드로 구성되어 있다. 그 중에서도 사람의 목소리인 '보컬' 사운드는 그 어떤 악기보다도 가장 즉각적으로 잘 인지되는 파트이다. 곡에서 중요한 부분을 차지하는 보컬을 여러 사운드 속에서 완벽하게 믹싱하는 데에는 많은 요소들이 고려되어야 하고 또 다양한 단계들이 존재한다. 그 단계 중 EQ에 집중하여 연구하고자 한다. 따라서 본 논문에서는 EQ작업과 관련된 사항들에 대해 EQ의 개념 및 종류로 부터 시작해 보컬 레코딩 시 EQing방법, Subtractive EQ, Additive EQ 등에 관해 알아보도록 할 것이다. EQ는 사운드 믹싱 과정에서 특히나 보컬 사운드를 다루는데 있어서 가장 중요한 도구 중 하나로 손꼽힌다. 보컬 이큐잉은 보컬이 완벽히 트랙 믹스 안에 어우러지게 하기 위해 각 주파수 대역을 부스트 하고 또 컷팅하여 음색을 다듬는 과정이다. 프로페셔널한 보컬사운드를 얻기 위해서는 그 무엇보다도 자신이 스타일적으로 의도하는 사운드가 무엇인지 확실한 방향성을 가지고 가야하고 그를 위해 레퍼런스 트랙을 이용하는 것도 아주 효과적이다. EQing이외에도 컴프레션, 리버브, 코러스, 딜레이 등의 이펙터 작업과 백킹 보컬과 하모니의 조정 등의 다양한 복잡한 단계의 작업이 있고 그 또한 매우 중요한 작업들이다. 그러나 믹싱의 시작에 해당하는 작업인 EQing은 무엇보다도 그 중요도가 큰 관계로 위 사항들을 두루 고려하여 세밀한 작업을 하여야 할 것이다.

최적화된 pRBF 뉴럴 네트워크에 이용한 삼상 부분방전 패턴분류에 관한 연구 (A Study on Three Phase Partial Discharge Pattern Classification with the Aid of Optimized Polynomial Radial Basis Function Neural Networks)

  • In this paper, we propose the pattern classifier of Radial Basis Function Neural Networks(RBFNNs) for diagnosis of 3-phase partial discharge. Conventional methods map the partial discharge/noise data on 3-PARD map, and decide whether the partial discharge occurs or not from 3-phase or neutral point. However, it is decided based on his own subjective knowledge of skilled experter. In order to solve these problems, the mapping of data as well as the classification of phases are considered by using the general 3-PARD map and PA method, and the identification of phases occurring partial discharge/noise discharge is done. In the sequel, the type of partial discharge occurring on arbitrary random phase is classified and identified by fuzzy clustering-based polynomial Radial Basis Function Neural Networks(RBFNN) classifier. And by identifying the learning rate, momentum coefficient, and fuzzification coefficient of FCM fuzzy clustering with the aid of PSO algorithm, the RBFNN classifier is optimized. The virtual simulated data and the experimental data acquired from practical field are used for performance estimation of 3-phase partial discharge pattern classifier.

변압기 권선을 이용한 자속구속형 초전도 전류제한기의 전류제한 및 전압강하 보상 특성 (Current Limiting and Voltage Sag Compensation Characteristics of Flux-Lock Type SFCL Using a Transformer Winding)

  • The superconducting fault current limiter (SFCL) can quickly limit the fault current shortly after the short circuit occurs and recover the superconducting state after the fault removes and plays a role in compensating the voltage sag of the sound feeder adjacent to the fault feeder as well as the fault current limiting operation of the fault feeder. Especially, the flux-lock type SFCL with an isolated transformer, which consists of two parallel connected coils on an iron core and the isolated transformer connected in series with one of two coils, has different voltage sag compensating and current limiting characteristics due to the winding direction and the inductance ratio of two coils. The current limiting and the voltage sag compensating characteristics of a SFCL using a transformer winding were analyzed. Through the analysis on the short-circuit tests results considering the winding direction of two coils, the SFCL designed with the additive polarity winding has shown the higher limited fault current than the SFCL designed with the subtractive polarity winding. It could be confirmed that the higher fault current limitation of the SFCL could be contributed to the higher load voltage sag compensation.

현대패션 활용을 위한 천연염색에 의한 검정색 구현(1): 견직물을 중심으로 (Developing Black Color by Natural Dyeing for Contemporary Fashion: Dyeing of Silk Fabrics)

  • The aim of this study is to develop an effective dyeing process for black color on the silk fabrics, using natural Indigo, Madder, Amur cork tree, Alder, Logwood, and Gallnut. All natural dye materials were water-extracted, concentrated, and freeze-dried. Seven different processes were carried out and the color differences(${\Delta}E$) of black color with each process from the chemical black dyed fabric were compared. The light, washing, and rubbing fastness were evaluated. The black dyeing of the silk fabrics was well carried and excellent black color was obtained in every methods. The profound black color could be obtained by the subtractive mixture of the three primary colors of red, yellow, and blue in the order of indigo(blue) - amur cork tree(yellow) - madder or lac(red). Black color was efficiently obtained by iron mordant when logwood or alder fruit was used. The color difference(${\Delta}E$) from the black color with chemical dyestuff was the lowest in the indigo - amur cork tree - lac - iron method. Light fastness and washing fastness were excellent in all dyeing processes, showing grade 5. The fastness to rubbing was excellent as shown grade 5 in the dry samples of logwood - iron process and alder fruit - iron process.

Elevated Levels of PDGF Receptor and MDM2 as Potential Biomarkers for Formaldehyde Intoxication

  • Formaldehyde has been identified as the most prevalent cause of sick building syndrome (SBS), which has become a major social problem, especially in developing urban areas. However, studies on the molecular mechanisms associated with formaldehyde toxicity have been limited, probably because it is difficult to relate the experimental results obtained from in vitro studies to human exposure in vivo. Using polymerase chain reaction-based suppression subtractive hybridization, we recently identified 27 different formaldehyde-inducible genes including platelet-derived growth factor receptor alpha gene (PDGFRA) and mouse double minute 2 (MDM2) gene which were increased significantly in both formaldehyde-exposed human trachea cells, 680.Tr, and rat tracheas. To establish a possible relationship between induction of these formaldehyde-inducible genes and symptoms of SBS, we examined expression levels of these genes in peripheral lymphocytes of residents of new apartments. Here, we report that the expression of PDGFRA and MDM2 transcripts was significantly higher in peripheral blood lymphocytes obtained from 15 residents in new buildings than in seven control individuals. Our results suggest that the elevated levels of PDGFRA and MDM2 may be associated with the formaldehyde-induced pathophysiology that is closely related with SBS, and that they deserve evaluation as potential biomarkers for formaldehyde intoxication.

  • The improved flux-lock type superconducting fault current limiter (SFCL) is composed of a series transformer and superconducting unit of the yttrium-barium-copper-oxide (YBCO) coated conductor. In this paper, we investigated current limiting characteristics through winding direction of coil 2 and variable number of coil 1 and coil 2 in improved flux-lock type SFCL. The better fault current characteristics and the burden of YBCO coated conductor can be confirmed from the experimental result in the higher turn ratio of coil 1 and coil 2 in the additive conditions. In case of subtractive condition, we can confirm a similar result in the same case of experimental conditions. but the burden of YBCO coated conductor has been increased from an increase in winding numbers of coil 2.

  • When the current of the superconducting element exceeds its critical current by the fault occurrence, the quench of the high-$T_C$ superconducting fault current limiter (HTSC) comprising the flux-lock type superconducting fault current limiter (SFCL) occurs. Simultaneously, the magnetic flux in the iron core induces the voltage in each coil, which contributes to limit the fault current. In this paper, the fault current limiting characteristics of the flux-lock type SFCL as well as the load voltage sag suppressing characteristics according to the flux-lock type SFCL's winding direction were investigated. To confirm the fault current limiting and the voltage sag suppressing characteristics of the this SFCL, the short-circuit tests for the simulated power system with the flux-lock type SFCL were carried out. The flux-lock type SFCL designed with the additive polarity winding was shown to perform more effective fault current limiting and load voltage sag suppressing operations through the fast quench occurrence right after the fault occurs and the fast recovery operation after the fault removes than the flux-lock type SFCL designed with the subtractive polarity winding.

Isolation of Two cDNAs Encoding a Putative Peptidohlycan Recognition Protein Gene from the Domestic Silkworm, Bombyx mori

  • Peptidohlycan recognition protein (PGRP) is one of the pattern recognition proteins in innate immunity of insect. We isolated differentially expressed two cDNAa, BTL-LPI and BTL-LP2, in the fat body of Bombyx mori larvae injected with bacteria by subtractive hybridization method. These two clones showed amino acid sequence divergence of 30.4%. In the comparison with other insect PGRP genes, BTL-LP2 showed 48.8% and 45.2% of sequence homology to the known PGRP genes of Bombyx mori and Tricoplusia ni, respectively, and BTL-LP2 was 31.8% and 30.9% , respectively. Phylogenetic analysis showed relatively close relationship of the BTL-LP2 to the known insect PGRP, unlike BTL-LPI, which was equidistant both to insect and mammals, suggesting a divergent relationships of the two newly cloned B. mori PGRP genes. Northern blot analyses confirmed an induction of the expression of BTL-LP2 by the bacterial infection in the Int body of B. mori, suggesting the involvement of the gene in the insect immunity.

Selection and Target-Site Mapping of Peptides Inhibiting HCV NS5B Polymerase Using Phage Display

  • A series of pep tides binding to the HCV NS5B polymerase was selected from phage display peptide libraries. A conserved motif of Ser-Arg-X-Arg/Leu was identified among the selected peptides, and Pep2 (Trp-Ser-Arg-Pro-Arg-Ser-Leu) was chosen for further characterization. The binding of Pep2 to HCV NS5B in vivo was shown by a yeast two-hybrid assay and by subcellular colocalization analysis using immunofluorescence confocal microscopy. The in vitro interaction was also confirmed by GST pulldown assay. The replication of the HCV 1b subgenomic replicon was efficiently inhibited by the presence of the peptide. By using a subtractive biopanning against Pep2, the binding site of the peptide was mapped at the pocket of Pro388 to Pro391 in the thumb subdomain of the polymerase. A yeast two-hybrid analysis using Pro388Ala and Pro391Ala mutants of NS5B confirmed the binding.
