Isolation of Coat Protein Gene from Cucumber Mosaic Virus and Its Introduction into Tobacco (오이 모자이크 바이러스 외피 단백질 유전자 분리 및 담배로의 형질전환)

  • The coat protein (CP) gene was cloned from RNA genome of the Cucumber Mosaic Virus strain ABI (CMV-ABI) isolated in Korea. The comparisons of the nucleotide sequence of the cloned CP gene and its deduced amino acid sequences with other CP genes revealed that the CMV-ABI belongs to subgroup I (type I), CMV-ABI developed the typical mosaic symptom in infected plants. Tobacco plants (Samsun and NC82) were transformed by leaf-disc transformation via Agrobacterium, temefaciens LB4404 harboring pVCP, witch CMV-ABI CP gene was inserted into the pBI121, and a number of mature transgenic tobacco plants were developed. Southern and PCR analysis of genomic DNA from the transgenic plants showed that the CP gene was integrated into the genomes of the most of the transgenic plant. Result of the segregation patterns of resistance in T1 seedlings of the plants to kanamycin showed that the transgenic plants containing l,2 and 3 copies of CP gene were50%, 39% and 11% of the total transgenic plants, respectively.

Morphological characteristics and RAPD analysis of Epimedium spp. (국내외 수집 삼지구엽초의 형태적 특성 및 유연관계 분석)

  • Morphological characteristics of nine Epimedium spp was analyzed on the basis of six morphological characters. To analyse the genetic relationship among Epimedium spp., polymerase chain reaction (PCR) was performed with total genomic DNA of 17 Epimedium spp. by using random 8 primers. The genetic diversity and genetic distance among nine Korean collections, seven Japanese and one China collection were used to generate a dendrogram showing phylogenie relationship. Seventeen Epimedium spp were classified into two groups of group I and II, since they were divided into two major groups at the similarity coefficients value of 0.65. In addition, one of the two group, group I was divided into three sub-groups including Epimedium koreanum, Chul-won collections 1, 2, and 3, Yanggu, Hongchon, Hwachun, Chunchon, China, Maehwa (Japanese), E. diphyllum and E. violaceum (Japanese), while group II included Chulwon collection 4 and 5 and Japanese collection. The samples collected at Chulwon district showed close similarity with Japanese collection. Similarity indexes between collection and genetic relationship were related at the levels ranging from 0.6 to 0.9.

Diversity and Phylogenetic Analysis of Fluorescent Pseudomonads Isolated from Soil-Root System of Red Pepper in Greenhouse (비닐하우스 고추재배지의 토양과 근계로부터 분리된 형광성 Pseudomonads의 계통 분류 및 다양성)

  • Among the fluorescent pseudomonad isolates from soil- root system of red pepper in Chinju, Kyunsangnam-Do, the phylogenetic analysis for 35 isolates were conducted. The partial 16S ribosomal DNA sequences were used as taxonomic key for phylogenetic analyses, and these sequences were enabled to identification of the fluorescent pseudomonad isolates on the species level. The 17 isolates among them were classified into Pseudomonas putida group, and consisted of the strains isolated mainly from soil. This group were subdivided into 4 subgroups (I, II, III, and IV). The subgroup I and IV were unique ones which were relatively remotely related with subgroup II and III including the type strain of P. putida. The 15 isolates among 35 isolates were grouped along with the type strain of Pseudomonas fluorescens, and 3 isolate were characterized as intermediates of P. fluorescens and Pseudomonas chlororaphis. Most of strain isolateds from the rhizosphere soil and rhizoplane of red pepper were identified as P. fluorescens and closely related with each other. In this study, root of red pepper was supposed to be colonized by a specific strain or strains of P. fluorescens.

Facial Nerve Decompression for Facial Nerve Palsy with Temporal Bone Fracture: Analysis of 25 Cases (측두골 골절후 발생한 안면마비 환자의 안면신경감압술: 25명 환자들의 증례분석)

  • Purpose: The aim of this study is to present a retrospective review of patients who had a sudden onset of facial palsy after trauma and who underwent facial nerve decompression. Methods: The cases of 25 patients who had traumatic facial palsy were reviewed. Facial nerve function was graded according to the House-Brackmann grading scale. According to facial nerve decompression, patients were categorized into the surgical (decompression) group, with 7 patients in the early decompression subgroup and 2 patients in the late decompression subgroup, and the conservative group(16 patients). Results: The facial nerve decompression group included 8 males and 1 female, aged 2 to 86 years old, with a mean age of 40.8. In early facial nerve decompression subgroup, facial palsy was H-B grade I to III in 6 cases (66.7%); H-B grade IV was observed in 1 case(11.1%). In late facial nerve decompression subgroup, 1 patient (11.1%) had no improvement, and the other patient(11.1%) improved to H-B grade III from H-B grade V. A comparison of patients who underwent surgery within 2 weeks to those who underwent surgery 2 weeks later did not show any significant difference in improvement of H-B grades (p>0.05). The conservative management group included 15 males and 1 female, aged 6 to 66 years old, with a mean age of 36. At the last follow up, 15 patients showed H-B grades of I to III(93.7%), and only 1 patient had an H-B grade of IV(6.3%). Conclusion: Generally, we assume that early facial nerve decompression can lead to some recovery from traumatic facial palsy, but a prospective controlled study should and will be prepared to compare of conservative treatment to late decompression.

An in Vitro Study of the Effects of Different Dentin Bonding Agents on the Prevention of Tooth Discoloration and the Sealing Ability of Calcium Silicate-Based Cement in Regenerative Endodontic Procedures (치수재혈관화 술식에서 다양한 상아질 접착제가 치아 변색과 칼슘 실리케이트 시멘트의 밀폐 효과에 미치는 영향에 대한 실험실적 연구)

  • This study aimed to compare the effects of applying different types of dentin bonding agents (DBAs) on tooth and on the sealing ability of CSC in regenerative endodontic procedures (REPs). Specimens were made from 120 human first premolars and categorized into the following groups based on the presence and the type of DBA: Group I, no bonding; Group II, One-Step; Group III, Clearfil SE Bond; and Group IV, Single Bond Universal. Subsequently, the groups were categorized into Subgroup a and Subgroup b, in which Endocem MTA® Premixed Regular and Biodentine were used, respectively. The CIE L*a*b* on the photos taken prior to TAP placement (S0), 1 week after TAP placement (S1), 2 weeks after TAP placement (S2), 4 weeks after TAP placement (S3) and 2 weeks after CSC placement (S4) was used to assess discoloration. The samples were submerged in a 2% methylene blue solution and magnified by 30 times under a stereomicroscope to assess microleakage. The application of DBA during REP reduced crown discoloration, while Biodentine showed minimal discoloration with or without DBA and regardless of the DBA type. Additionally, the application of DBA did not increase microleakage.

  • The purpose of this study was to evaluate the effect of the canal filling sealer to resin cement When posts were cemented in the endodontically treated teeth, 86 incisors were used. The coronal portion of the teeth were removed at the cemento-enamel junction, every tooth was done treatment of canal. And the teeth was divided was into 12 groups. After 5 days at cementation of post, teeth with cemented posts were mounted on a retention jig and the failure loads of the specimens were measured by an Instron Universal Testing Machine. The results were as follows. 1. The results of failure loads were $15.5{\pm}7.1kg$ in group I b, $21.6{\pm}5.4kg$ in group II b and $20.1{\pm}18.1kg$ in group III b, and there was no statistically significant , difference between each group(p>0.05). 2. The results of failure loads were $19.0{\pm}6.7kg$ in group IV b, $17.3{\pm}6.5kg$ in group V b.and $18.9{\pm}7.9kg$ in group VI b and there was no significant difference between each other(p>0.05). 3. In same condition, the failure load of subgroup a was largely higher the subgroup b. But there was no significant difference between each other(p>0.05).

Isolation and Characterization of Human scFv Molecules Specific for Recombinant Human Heat Shock Protein (HSP) 70.1

  • Background: The heat shock proteins (HSPs) play an important role in cellular protection mechanisms against physical or chemical stresses. In this study scFv antibodies specific for human HSP70.1 were isolated from a semi-synthetic human scFv library with the ultimate goal of developing anti-HSP70.1 intracellular antibody (intrabody) that may offer an attractive alternative to gene targeting to study the function of the protein in cells. Methods: A semi-synthetic human scFv display library ($5{\times}10^{8}$ size) was constructed using pCANTAB-5E vector and the selection of the library against bacterially expressed recombinant human HSP70.1 was attempted by panning. Results: Three positive clones specific for recombinant HSP70.1 were identified. All three clones used $V_{H}$ subgroup III. On the other hand, $V_{L}$ of two clones belonged to the kappa light chain subgroup I, but the other utilized $V_{k}$ subgroup IV Interestingly, these scFv molecules specifically reacted to the recombinant HSP70.1, yet failed to recognize native HSP70 induced in U937 human monocytic cells by heat treatment. Conclusion: Our results indicated that affinity selection of an scFv phage display library using recombinant antigens produced in E. coli might not guarantee the isolation of scFv antibody molecules specific for a native form of the antigen. Therefore, the source of target antigens needs to be chosen carefully in order to isolate biofunctional antibody molecules.

Isolation and Molecular Analysis of Methanol Oxidation Genes in an Obligate Methylotrophic Bacterium, Metheylobacillus sp. Strain SK-5

  • Methanol dehydrogenase (MDH) is a key enzyme in the process of methanol oxidation in methylotrophic bacteria. However, information on MDH genes from genus Methylobacillus is limited. In this study, a 6.5-kb HindIII DNA fragment of Methylobacillus sp. SK-5 chromosomal DNA was isolated from the genomic library of the strain by using a degenerate oligonucleotide probe that was designed based on JV-terminal amino acid sequence of the MDH $\alpha$ subunit purified from the strain. Molecular analysis of the fragment revealed four tightly clustered genes (mxaFJGI) involved in the methanol oxidation. The first and fourth genes were very similar to mxaF (77% identity for nucleotides an 78% identity for amino acids) and mxaF (67% Identity for nucleotides and 68% Identity for amino acids) genes, respectively, from Methylovorus sp. SSI. Genes mxaF and mxaI encode $\alpha$ and $\beta$ subunits of MDH, respectively. The two subunits were identified from purified MDH from Methylobacillus sp. SK-5. A dendrogram constructed by comparison of amino acid sequences of MDH u subunits suggests that MxaF from Methylobacillus sp. SK-5 belongs to a subfamily cluster of MDH u subunits from $\beta$-subgroup Proteobacteria. The subfamily cluster is separated from the other subfamily that consists of $\beta$- and $\gamma$-subgroup Proteobacteria. This study provided information on mn genes from a methylotrophic bacterium in $\beta$-subgroup Proteobacteria, which would aid to better develop a gene probe to detect one-carbon metabolizing bacteria.

Effect of the amount of thickness reduction on color and translucency of dental monolithic zirconia ceramics

  • PURPOSE. This study investigated the effect of amount of thickness reduction on color and translucency of dental monolithic zirconia ceramics. MATERIALS AND METHODS. One-hundred sixty-five monolithic zirconia specimens ($16.3mm{\times}16.3mm{\times}2.0mm$) were divided into 5 groups (Group I to V) according to the number of A2-coloring liquid applications. Each group was then divided into 11 subgroups by reducing the thickness up to 1.0 mm in 0.1-mm increments (Subgroup 0 to 10, n=3). Colors and spectral distributions were measured according to CIELAB on a reflection spectrophotometer. All measurements were performed on five different areas of each specimen. Color difference (${\Delta}E^*{^_{ab}}$) and translucency parameter (TP) were calculated. Data were analyzed using one-way ANOVA and multiple comparison $Scheff{\acute{e}}$ test (${\alpha}=.05$). RESULTS. There were significant differences in CIE $L^*$ between Subgroup 0 and other subgroups in all groups. CIE $a^*$ increased (0.52<$R^2$<0.73), while CIE $b^*$ decreased (0.00<$R^2$<0.74) in all groups with increasing thickness reduction. Perceptible color differences (${\Delta}E^*{^_{ab}}$>3.7) were obtained between Subgroup 0 and other subgroups. TP values generally increased as the thickness reduction increased in all groups ($R^2$>0.89, P<.001). CONCLUSION. Increasing thickness reduction reduces lightness and increases a reddish, bluish appearance, and translucency of monolithic zirconia ceramics.
