Assessment of Salinity-Induced Antioxidative Defense System of Diazotrophic Cyanobacterium Nostoc muscorum

  • The present study examined the salinity-induced oxidative damage and differential response of enzymatic and nonenzymatic antioxidants of Nostoc muscorum. As compared with carotenoid content that showed induction, the chlorophyll and phycocyanin contents were inhibited after salt stress. Acceleration of lipid peroxidation and peroxide production suggested the onset of oxidative damage. The activities of all studied enzymatic antioxidants were significantly increased by salt stress, with maximum induction occuring with superoxide dismutase (154.8% at 200 mM NaCl treatment). Interestingly, under severe stress condition (250 mM NaCl), ascorbate peroxidase seemed to be more crucial than catalase for peroxide scavenging. Among the studied nonenzymatic antioxidants, ${\alpha}$-tocopherol was induced maximally (56.0%); however, ascorbate and reduced glutathione were increased by only 8.9% after 250 mM NaCl treatment as compared with control cells. Therefore, salinity was found to induce the antioxidative defense system of N. muscorum.

반하후박탕(半夏厚朴湯)의 사회·심리적 스트레스에 대한 실험적 연구(實驗的 硏究) (Effects of Banhahubak-Tang Extract (BHTe) on Sociopsychological Stress)

  • Objectives: This study is designed to investigate the effects of Banhahubak-Tang Extract (BHTe) on stress. Methos: BHTe was fed to ICR male mice ($20{\pm}2g$) orally with the dose of 100 mg/kg/day for five days. Mice were then exposed to sociopsychological stress by observing foot shock stressed mice for one hour for five days while restrained. Results: 1) The BHTe-administered group showed a tendency of decreasing of serum corticosterone secretion compared with the control group. 2) The BHTe-administered group showed an increase in noradrenalin secretions in the dorsal cortex of the brain, but it was not significant. 3) BHTe administration had no effect on the brain level of lipid peroxidation. 4) BHTe administration decreased the serum level of lipid peroxidation. 5) BHTe administration decreased the Cu,Zn-SOD in the brain. 6) BHTe administration had no effect on catalase activity in the brain. 7) BHTe administration had no effect on the brain level of GSH. 8) BHTe administration increased the serum level of DPPH free radical scavenging activity, but not significantly enough to make a comparison with BHT. 9) The elevated plus-maze test is designed to detect the effect of anxiolytic drugs. The BHTe group showed a significant increase in latency time. Conclusions: These results suggest that BHTe can effectively rid the subject of the effects of sociopsychological stress.

Aerobic and Graduated Treadmill Exercise Decreases Blood Glucose Levels, Lipid Levels and Oxidative Stress in an Animal Model of Type 1 Diabetes Mellitus

  • Purpose: Exercise has been shown to be a simple and economical therapeutic modality that may be considered as an effective aid for diabetic mellitus. For example, exercise training increases insulin sensitivity in type 2 diabetes. But we found no reported of how exercise affect type 1 diabetes. This study investigated the impact of aerobic and graduated treadmill exercise regimens on body weight, glucose and insulin concentrations, lipid profiles, and oxidative stress indicators in rats with streptozotocin (STZ) induced diabetes. Glycosylated hemoglobin ($HbA_{1c}$) was determined as an indicator of glucose control during exercise. Methods: In our study, a total of 40 rats were used. Three groups of 10 rats each were given STZ to induce diabetes. The remaining 10 rats became the normal group. After 28 days we determined biochemical parameters such as glucose, glycosylated hemoglobin ($HbA_{1c}$), insulin concentration, serum total cholesterol (TC), triglycerides (TG), and high-density lipoprotein (HDL). Superoxide dismutase (SOD) and catalase activities were also measured. Results: Concentrations of blood glucose and $HbA_{1c}$ in the moderated exercise groups were significantly decreased after 28 days compared with the control group (p<0.05). There was a significant reduction in serum TC and TG in the experimental groups. The activity of SOD increased significantly by 17.70% and 48.25% respectively. Conclusion: These results indicate that physical training and exercise training affects body weight, fasting blood glucose, $HbA_{1c}$, insulin, lipid profiles, and antioxidant status in rats with streptozotocin-induced diabetes. We suggest that graduated treadmill exercise may have therapeutic, preventative, and protective effects against diabetes mellitusby improving glycemic control, oxidant defenses, and lipid metabolism.

Treadmill에서 운동에 따른 지질의 과산화와 항산화제 수준의 변화 (Antioxidant Levels and Lipid Peroxidation in Plasma and Erythrocyte Following Treadmill Running)

  • The relationship between exercise intensity, oxidative stress and antioxidant status has been studied in sixteen trained male athletes aged 20-25years. Subjects performed 30-40minutes of treadmill running at 65% of VO2 max(high intensity exercise). Blood samples were taken before and immediately after two exercise bouts for measurement of blood antioxidants, indices of lipid peroxidation and susceptibility of crythrocyte to peroxidation. Plasma concentrations of cholesterol(7.3%), vitamin C(7.5%) and uric acid(2.1%) were elevated a little after exercise at 65% of VO2 max(13.7%)were significantly high than before exercise(p<0.05). However, these exercise-induced changes could be partly due to significant decreases in plasma volume which occurred after both exercise bouts(p<0.05). Plasma volume decreased 5.85$\pm$2.06% and 11.25$\pm$2.87% with exercise at 65% and 85% of VO2 max, respectively. The erythrocyte susceptibility to peroxidation after exercise at 65% of VO2 max was unchanged compared with the value before exercise, whereas after exercise at 85% of VO2 max, it was significantly higher than after exercise at 65% and 85% of VO2 max as well as before exercise(p<0.05). A significant increase at 85% of VO2 max, it was significantly high than after exercise at 85% of VO2 max (29.10$\pm$4.76ug/g Hb)when compared with the level before exercise (24.61$\pm$3.45ug/g Hb)(p<0.05). The results suggest that exercise-induced changes in plasma levels of lipid peroxide and antioxidant need to be evaluated, taking the shift in plasma volume into consideration. Also, exercise at high intensity corresponding to 85% of VO2 max alters the erythrocyte antioxidant status in relation to exercise-induced of oxidative stress.

복분자 미숙과 물 추출물이 마우스의 지질대사 및 산화적 스트레스에 미치는 영향 (Effects of Unripe Black Raspberry Water Extract on Lipid Metabolism and Oxidative Stress in Mice)

  • 본 연구는 복분자 미숙과 물 추출물이 in vivo에서 지질대사 및 산화적 스트레스에 미치는 영향을 살펴보고, 기능성 식품으로서의 가능성을 알아보기 위하여 수행되었다. 실험동물은 C57BL/6J mouse를 사용하였으며, 12주 동안 고지질 식이를 급이하면서, 복분자 미숙과 물 추출물을 경구 투여하여 다음과 같은 결과를 얻었다. 고지질대조군과 비교하였을 때, 복분자 미숙과 첨가군에서 체중증가량, 식이효율이 모두 감소되었고, 혈중 콜레스테롤 및 LDL-콜레스테롤의 농도를 유의성 있게 감소시켰을 뿐만 아니라 간 조직에서 총콜레스테롤의 함량이 유의적으로 감소됨을 확인하였다. 콜레스테롤 합성의 가장 중요한 기작인 LDL receptor와 SREBP2의 활성을 증가시킴으로써, 세포 안으로의 LDL 흡수를 촉진하였고, HMG-CoA reductase의 유전자 발현이 증가되면서 체내 콜레스테롤을 조절하며, ABCA1의 활성화로 인해, 콜레스테롤의 배출을 증가시키는 결과를 도출하였다. 또한, 간조직의 지질산화수준(TBARS)을 유의적으로 감소시켰으며, ox-LDL관련 유전자인 CD36을 억제함으로써, 동맥경화를 예방할 것으로 기대한다. 간의 형태학적 변화를 관찰해본 결과, 고지질대조군에 비하여 지방형성이 현저히 줄어들었음을 확인하였다. 결과적으로 복분자 미숙과 물 추출물이 콜레스테롤의 생합성과 배출 및 산화적 스트레스를 조절하여 고지질 및 동맥경화 질환 예방에 긍정적인 효과를 미치는 것으로 사료된다.

라지 팻치에 감염된 잔디의 산화적 스트레스 발현과 항산화효소의 활력의 변화 I. 산화적 스트레스 발현 (The Oxidative Stress Induction and Response of Antioxidative Enzymes in the Large Patch-Infected Zoysiagrass I. Oxidative stress induction)

  • 한국형 잔디에 있어 병원성 (라지 팻치) 감염이 산화적 스트레스의 발현에 미치는 영향을 규명하기 위해 라지 팻치에 감염된 잔디의 생육, 뿌리 건사율, 과산화수소 농도 및 지질과산화 정도를 감염이되지 않은 대조구와 비교하였다. 처리 후 6일 동안 이틀간격으로 잎과 뿌리 시료에 대해 각각 분석하였다. 라지 팻치에 감염된 잔디 뿌리의 건사율은 대조구에 비해 약 30% 증가하였다. 건물 함량은 4일째까지는 유의적인 차이가 없었으며 6일차에서만 라지 팻치 감염에 따라 잎에서 14% 뿌리에서 20% 각각 감소하였다. 과산화수소의 농도는 라지 팻치에 감염된 잎에서 초기 2일 동안 약 28%의 높은 증가를 하였다가 이후 서서히 감소하여 6일차에는 대조구에 비교하여 11% 낮은 수준이었다. 뿌리에서는 감염에 따른 과산화수소의 증가가 뚜렷하여 6일차에는 대조구에 비해 약 1.7배 높았다. 지질과산화 정도는 잎의 경우, 라지 팻치 감염에 따라 초기 4일간 지속적으로 증가하다가 이후 정체하였으며 뿌리에서는 초기 2일간의 증가가 뚜렷하였다. 이러한 결과들은 잔디에 있어 라지 팻치 감염은 산화적 스트레스를 유도하며, 잎과 뿌리 조직간에 산화적 스트레스 발현정도 및 역동성은 차이가 있음을 보여준다.

Effects of disturbed liver growth and oxidative stress of high-fat diet-fed dams on cholesterol metabolism in offspring mice

  • BACKGROUND/OBJECTIVES: Changes in nutritional status during gestation and lactation have detrimental effects on offspring metabolism. Several animal studies have shown that maternal high-fat diet (HFD) can predispose the offspring to development of obesity and metabolic diseases, however the mechanisms underlying these transgenerational effects are poorly understood. Therefore, we examined the effect of maternal HFD consumption on metabolic phenotype and hepatic expression of involved genes in dams to determine whether any of these parameters were associated with the metabolic outcomes in the offspring. MATERIALS/METHODS: Female C57BL/6 mice were fed a low-fat diet (LFD: 10% calories from fat) or a high-fat diet (HFD: 45% calories from fat) for three weeks before mating, and during pregnancy and lactation. Dams and their male offspring were studied at weaning. RESULTS: Dams fed an HFD had significantly higher body and adipose tissue weights and higher serum triglyceride and cholesterol levels than dams fed an LFD. Hepatic lipid levels and mRNA levels of genes involved in lipid metabolism, including $LXR{\alpha}$, SREBP-2, FXR, LDLR, and ABCG8 were significantly changed by maternal HFD intake. Significantly lower total liver DNA and protein contents were observed in dams fed an HFD, implicating the disturbed liver adaptation in the pregnancy-related metabolic demand. HFD feeding also induced significant oxidative stress in serum and liver of dams. Offspring of dams fed an HFD had significantly higher serum cholesterol levels, which were negatively correlated with liver weights of dams and positively correlated with hepatic lipid peroxide levels in dams. CONCLUSIONS: Maternal HFD consumption induced metabolic dysfunction, including altered liver growth and oxidative stress in dams, which may contribute to the disturbed cholesterol homeostasis in the early life of male mice offspring.

Red Wine Prevents Brain Oxidative Stress and Nephropathy in Streptozotocin-induced Diabetic Rats

  • We have studied the effects of red wine on brain oxidative stress and nephropathy in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in Wistar rats with a single intraperitonally injection of STZ (50 mg/kg). Two weeks before and four weeks after injection, red wine was given orally in both normal and diabetic rats. Blood samples were taken from the neck vascular trunk in order to determine the glucose, triglycerides, total cholesterol, HDL-cholesterol (HDL-c), atherogenic index (AI), total protein, blood urea nitrogen (BUN), creatinine, insulin, lipid peroxidation products, reduced glutathione (GSH) and superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities. As well, we estimated the lipid peroxidtion, GSH and SOD, GSH-Px and catalase activities in brain and renal homogenates, and the excretion of albumin, proteins and glucose in urine over 24 h period. The administration of STZ caused significant increases in levels of glycosuria, proteinuria, albuminuria, glycemia, total cholesterol and AI, as well as in lipid peroxidation products in the brain, plasma and kidney, whereas it decreased the GSH content and SOD, GSH-Px and catalase activities. Treatment with red wine significantly prevented the changes induced by STZ. These data suggested that red wine has a protective effect against brain oxidative stress, diabetic nephropathy and diabetes induced by STZ, as well as it protects against hypercholesterolemia and atherogenic risk.

The effects of physical training on antioxidative status under exercise-induced oxidative stress

  • This study investigated the effect of physical training and oxidative stress on the anti oxidative activity and on plasma lipid profile. Forty eight rats were given either a physical training or no training for 4 weeks and were then subdivided into 3 groups: before-exercise (BE); during-exercise (DE); after-exercise (AE). The antioxidative activity was evaluated with the activities of catalase in plasma and superoxide dismutase (SOD), the ratio of reduced glutathione/ oxidized glutathione (GSH/GSSG) and the level of malondialdehyde (MDA) in liver. The plasma concentrations of triglyceride (TG), total cholesterol (TC), high-density lipoprotein-cholesterol (HDL-C)) were also compared. Compared to those of non-training group. catalase activities of training group were lower before exercise but higher during and after exercise. SOD activities were higher regardless of exercise. GSH/GSSG ratio was higher before exercise but was not significantly different during exercise and even lower after exercise. There were no differences between non-training group and training group in MDA levels regardless of exercise. Compared to those of non-training group, atherosclerotic index of training group was lower after exercise and there were no significant differences before and during exercise. There were no differences between non-training group and training group in HDL-C regardless of exercise. These results suggest that moderate physical training can activate antioxidant defenses and decrease the atherosclerotic index and this beneficial effect is evident under exercise-induced oxidative stress.

Hepatoprotective Effect of Stamen Extracts of Mesua ferrea L. against Oxidative Stress induced by $CCl_4$ in Liver Slice Culture Model

  • Stamens of Mesua ferrea L. are a well-known herbal drug used in Indian System of Traditional Medicine to treat various diseases. The claimed activity of this plant part is necessitated to investigate antioxidant and hepatoprotective activity. Authenticated plant sample was extracted with hexane, ethanol (EtOH) and water (aq.) using ASE 100 accelerated solvent extractor. Antioxidant activity was evaluated by means of different in vitro assays. Hepatoprotective effect was investigated on carbon tetrachloride induced oxidative stress in liver slice culture model. Cytotoxic marker lactate dehydrogenase (LDH) released in culture medium and the activity of lipid peroxidation along with antioxidant enzymes (AOEs) namely superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) were estimated. Hexane and EtOH extracts were significantly inhibited DPPH, NO, SOD and $ABTS^+$ radical in dose dependent manner. The trade of phenol content was: aq. extract < hexane extract < EtOH extract. A significant correlation was shown by total phenol content and free radical scavenging activity of extracts. The culture system treated with hexane extract, EtOH extract or ascorbic acid exhibited significant depletion in LDH, lipid peroxidation, antioxidative enzymes SOD, CAT and GR. Hexane extract and EtOH extracts of stamen of M. ferrea protected liver slice culture cells by alleviating oxidative stress induced damage to liver cells.
