Using the Caenorhabditis elegans model system, the antioxidant activity of methanol extract of the guzeunggupoprocessed Liriope platyphylla F. T. Wang (Liliaceae) tuber was calculated. Between the methanol extracts of guzeunggupo-processed and non-processed L. platyphylla tuber, the processed L. platyphylla tuber showed higher DPPH radical scavenging effect than the non-processed one. The ethyl acetate soluble fraction of the methanol extract of the guzeunggupo-processed L. platyphylla tuber showed the best DPPH radical scavenging activity. The ethyl acetate fraction of the processed sample was measured for the activities of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species level. In addition, to verify the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction of the processed sample, SOD-3 expression was measured using a transgenic strain (CF1553). Consequently, the ethyl acetate fraction of the processed sample, increased SOD and catalase activities, and decreased ROS accumulation in a dose-dependent manner. Furthermore, the ethyl acetate fraction of the processed sample-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

최근 수중폭발로 인한 구조물의 충격응답에 대한 연구는 매우 높은 비용과 소요시간, 민감한 환경문제 등으로 인하여 실제 시험보다는 컴퓨터를 통한 수치해석적 연구가 활발히 진행되어 왔다. 또한 시뮬레이션의 기술 향상과 더욱 정교해진 기능들로 수치 시뮬레이션의 효율성이 증가되었을 뿐 아니라 그 신뢰성까지 증가하였다. 본 연구에서는 유체 표면의 Acoustic Pressure와 구조물 표면 변위의 적절한 관계를 다루는 구조-유체 상호작용(FSI : Fluid-Structure Interaction), 수중폭파 형태를 결정하는 유체의 깊이와 폭발물과 구조물 사이의 거리에 대한 파라미터를 상용 유한요소 프로그램인 ABAQUS에 적용한 시뮬레이션 값과 실험적 이론 값 비교에 중점을 두었다. 수중폭발로 인한 파이프의 충격테스트 응답 분석은 ABAQUS/Explicit을 사용하여 수행되었고, 시간이력에 따른 충격하중, Acoustic Pressure, 타격지점의 응력, 속도, 변형에너지 등 ABAQUS CAE에서 결과를 나타내었다.
Reinforced concrete (RC) structures consist of two different materials: concrete and steel bar. The stress transfer behaviour between the two materials through bond plays an important role in the load-carrying capacity of RC structures, especially when they subject to lateral load such as blast and seismic load. Therefore, bond and slip between concrete and reinforcement bar will affect the response of RC structures under such loads. However, in most numerical analyses of blast-induced structural responses, the perfect bond between concrete and steel bar is often assumed. The main reason is that it is very difficult to model bond slip in the commercial finite element software, especially in hydrodynamic codes. In the present study, a one-dimensional slide line contact model in LS-DYNA for modeling sliding of rebar along a string of concrete nodes is creatively used to model the bond slip between concrete and steel bars in RC structures. In order to model the bond slip accurately, a new approach to define the parameters of the one-dimensional slide line model from common pullout test data is proposed. Reliability and accuracy of the proposed approach and the one-dimensional slide line in modelling the bond slip between concrete and steel bar are demonstrated through comparison of numerical results and experimental data. A case study is then carried out to investigate the bond slip effect on numerical analysis of blast-induced responses of a RC column. Parametric studies are also conducted to investigate the effect of bond shear modulus, maximum elastic slip strain, and damage curve exponential coefficient on blast-induced response of RC columns. Finally, recommendations are given for modelling the bond slip in numerical analysis of blast-induced responses of RC columns.
이전 연구에서 우리는 RDX (hexahydro-1,3,5-trinitro-1,3,5-triazine) 분해세균 Pseudomonas sp. HK-6에서 xenobiotic reductase B를 암호화하는 xenB 유전자의 돌연변이 균주를 이용하여 RDX 스트레스에 대한 xenB 유전자의 역할에 관하여 연구를 보고하였다[Lee et al. (2015) Curr. Microbiol. 70(1): 119-127]. 본 연구에서는 Pseudomonas sp. HK-6 xenA 돌연변이 균주로 연구 범위를 확대하여 RDX 스트레스 조건에서 세포반응과 프로테옴 프로필의 변화를 분석하였다. RDX 첨가 배지에서 xenA 돌연변이 균주는 야생균주와 비교하여 RDX를 약 2배 정도 느리게 분해하였으며, RDX 스트레스 하에서 xenA 돌연변이 균주의 생장률과 생존율은 야생균주와 비교하여 낮았다. RDX 스트레스에 의한 심한 형태적 손상이 xenA 돌연변이 균주의 세포 표면에 발생하는 것이 주사전자현미경을 통해서 확인되었다. RDX 스트레스 하에서 야생균주에서 발현된 충격단백질인 DnaK 및 GroEL의 양은 배양 초기 혹은 상대적으로 낮은 RDX 농도에서는 증가하였으나, 배양시간이 길어지거나 높은 RDX 농도에서는 다소 감소하였다. 그러나 xenA 돌연변이 균주에서는 DnaK와 GroEL의 발현양은 RDX 농도가 증가함에 따라 점차 감소되었다. RT-qPCR에 의해 측정된 야생균주에서 dnaA와 groEL의 전사 수준은 RDX 스트레스가 증가된 상태에서 잘 유지되었으나, xenA 돌연변이 균주에서는 점차 감소되어 결국에는 소멸되었다. RDX 스트레스에서 xenA의 돌연변이에 의한 프로테옴 프로필의 변화를 2-DE PAGE를 통해서 관찰한 결과에 따르면 27개 단백질이 감소하고 3개가 증가한 것으로 나타났다. 이들 결과로 보아, 정상적인 xenA 유전자는 RDX 스트레스 하에서 세포의 온전한 형태 유지와 효율적인 RDX 분해 과정을 수행하기 위해서 필요하다는 것을 의미하였다.

Metabolic engineering is now a well established discipline, used extensively to determine and execute rational strategies of strain development to improve the performance of micro-organisms employed in industrial fermentations. The basic principle of this approach is that performance of the microbial catalyst should be adequately characterised metabolically so as to clearlyidentify the metabolic network constraints, thereby identifying the most probable targets for genetic engineering and the extent to which improvements can be realistically achieved. In order to harness correctly this potential, it is clear that the physiological analysis of each strain studied needs to be undertaken under conditions as close as possible to the physico-chemical environment in which the strain evolves within the full-scale process. Furthermore, this analysis needs to be undertaken throughoutthe entire fermentation so as to take into account the changing environment in an essentially dynamic situation in which metabolic stress is accentuated by the microbial activity itself, leading to increasingly important stress response at a metabolic level. All too often these industrial fermentation constraints are overlooked, leading to identification of targets whose validity within the industrial context is at best limited. Thus the conceptual error is linked to experimental design rather than inadequate methodology. New tools are becoming available which open up new possibilities in metabolic engineering and the characterisation of complex metabolic networks. Traditionally metabolic analysis was targeted towards pre-identified genes and their corresponding enzymatic activities within pre-selected metabolic pathways. Those pathways not included at the onset were intrinsically removed from the network giving a fundamentally localised vision of pathway functionality. New tools from genome research extend this reductive approach so as to include the global characteristics of a given biological model which can now be seen as an integrated functional unit rather than a specific sub-group of biochemical reactions, thereby facilitating the resolution of complexnetworks whose exact composition cannot be estimated at the onset. This global overview of whole cell physiology enables new targets to be identified which would classically not have been suspected previously. Of course, as with all powerful analytical tools, post-genomic technology must be used carefully so as to avoid expensive errors. This is not always the case and the data obtained need to be examined carefully to avoid embarking on the study of artefacts due to poor understanding of cell biology. These basic developments and the underlying concepts will be illustrated with examples from the author's laboratory concerning the industrial production of commodity chemicals using a number of industrially important bacteria. The different levels of possibleinvestigation and the extent to which the data can be extrapolated will be highlighted together with the extent to which realistic yield targets can be attained. Genetic engineering strategies and the performance of the resulting strains will be examined within the context of the prevailing experimental conditions encountered in the industrial fermentor. Examples used will include the production of amino acids, vitamins and polysaccharides. In each case metabolic constraints can be identified and the extent to which performance can be enhanced predicted
본 연구에서는 국내 철도 토공노반 재료로 가장 흔히 사용되는 입도조정쇄석, 화강풍화토, 암버럭-토사 혼합 재료에 대해 평균유효주응력과 축변형률의 함수로 표현되는 회복탄성계수 예측모델을 결정하였다. 회복탄성계수 예측모델은 대표적인 동적물성치인 변형률에 따른 전단탄성계수 감소곡선의 표현과 같이 최대영탄성계수와 정규화 영탄성계수 감소곡선으로 구성된다. 평균유효주응력의 함수로 표현되는 최대영탄성계수의 모델인자는 $A_E$와 $n_E$이고, 비선형 영역의 정규화 영탄성계수 감소곡선은 기준변형률(${\varepsilon}_r$)과 곡률계수(a)를 모델인자로 하는 수정 쌍곡선 모델로 표현된다. 제안된 회복탄성계수 예측모델을 검증하기 위해 3차원 다층탄성해석 프로그램(GEOTRACK)을 이용하여 평택 시험 철도노반의 탄성거동을 평가하였고, 화물열차 및 여객열차가 시험구간을 통과할 때 계측한 노반의 수직 탄성변위와 비교하였다. 현장계측은 자갈도상 아래의 재료가 각각 입도조정쇄석과 양질의 화강풍화토인 두 개소에서 수행되었다. 자갈도상 아래에서 계산된 수직 탄성변위는 대략 0.6mm 이내였고 계측 결과와 잘 일치하였다. 본 연구를 통해 제안된 회복탄성계수 예측모델이 열차하중에 의한 노반의 탄성거동을 적절히 표현하고 있음을 확인하였다.
Ni기 초내열합금인 GTD111 DS는 가스터빈 블레이드에 사용된다. 본 논문에서는 실제 운전조건과 유사한 조건을 설정하여 GTD111 DS의 저주기 피로시험을 실시하였다 상온, $760^{\circ}C$, $870^{\circ}C$의 온도범위와 다양한 변형률에서 저주기 피로시험을 수행하였다. 실험결과 총 변형률이 증가함에 따라 피로수명은 감소하였다. 상온 및 $760^{\circ}C$에서는 주기적 경화반응이 나타났으며 $870^{\circ}C$에서는 주기적 연화반응이 나타났다. $870^{\circ}C$에서 응력완화 현상은 유지시간에 따른 크리프의 영향으로 나타났다. 피로수명과 총 변형률의 관계는 Coffin-Manson 식을 통해 얻었다. 파단면은 SEM을 통해 초기균열 및 피로진전지역을 관찰하였다.
광섬유 브래그 격자(fiber Bragg grating: FBG)를 이용한 스트레인 센서는 출력 값으로 브래그 반사 파장의 변화를 제공하게 된다. 본 논문에서는 브래그 격자 스트레인 센서에서 출력되는 빛을 도파로 어레이 격자(arrayed waveguide grating: AWG)에 입력시켜서 파장의 변화를 측정하는 방법을 제안한다. FBG에 스트레인이 가해지게 되면 브래그 반사 파장이 이동하게 되어 AWG를 지나서 출력되는 채널별 광 파워가 변하게 되며, 이 값들의 조합으로 구해지는 centroid 값을 계산하면 브래그 반사 파장과 인가된 스트레인 값을 얻을 수 있다. 브래그 격자의 반사 스펙트럼의 대역폭이 centroid 값에 미치는 영향을 고려하여 3-dB 대역폭이 5.4 nm인 chirped FBG를 사용하였으며, 저항 측정방식의 기존 스트레인 센서와 비교하여 오차범위가 2% 이내인 결과를 확인 하였다. FBG 센서를 외팔보에 부착하여 진동에 따른 스트레인 값을 실시간으로 측정한 결과 진동 주파수 17.8 Hz, damping 시상수 0.96 초를 얻을 수 있었다.
제안된 모델은 FRP 구속 콘크리트에 대한 압축거동 예측을 위한 것이다. FRP로 구속된 콘크리트의 모델링을 위하여, 3축 응력상태의 콘크리트 아탄성 구성관계를 제시하였다. FRP 구속에 따른 콘크리트 강도 증진은 3축 응력공간의 파괴기준에 따라 결정되며, 이에 대응하는 최대 압축변형률은 본 연구에서 제안된 변형률 증진계수로부터 결정된다. 따라서, 기존의 모델들이 하중단계에 관계없이 구속조건이 초기부터 파괴까지 일정하게 고려되는 반면에, 제안된 모델은 FRP로 구속된 콘크리트의 구속현상을 하중단계에 의존적인 비선형 관계로 제시하였다. FRP 층은 2차원의 적층된 복합재료의 해석에 기초하여 모델링되었다. 개발된 해석모델은 증분법에 의한 압축거동실험에 대한 해석을 수행할 수 있도록 하였다. FRP로 구속된 콘크리트 실린더의 대한 여러 연구자들의 실험 결과와 본 예측모델을 비교한 결과, 제안된 모델은 축방향 변형 뿐만 아니라 횡방향 변형을 포함하여 FRP 층으로 인한 콘크리트의 구속효과의 증진에 관한 거동 특성들을 잘 예측해 주었다.
