When various strains of Streptomyces griseus and S. galbus were examined for the ability on the production of streptomycin in tryptic soy(TS) broth, S. griseus ATCC 27001 was found to be the best. S. griseus ATCC 12475 and ATCC 23345 showed also good growth and favorable production of streptomycin. Examination of various complex media reported in fermentation literatures for the industrial production of streptomycin indicated that glucose-soybean media-sodium chloride (GSS) broth and K (Chucken) broth gave higher yields of streptomycin than others studied by us. Examination of the ingredients of media producing streptomycin in high yield indicated that some components in soybean activated the production of streptomycin. Addition of meat extract enhanced the yield of streptomycin but it could be substituted with distillers solubles without much effect on the yield. Addition of corn steep liquor decreased the production of streptomycin.
Erwinia carotovora subsp. Carotovora (Ecc), a pathogen of tobacco hollow stalk disease, was isolated for testing susceptibility to streptomycin from diseased plants in burley tobacco growing area. Of 157 isolates tested, 17 isolates (108%) were resistant to the antibiotic at the antibiotic from field soils, which streptomycin had been used continuously for three years for control of the disease was three times higher than those of non-used. There was no difference in virulence and generation time between streptomycin-sensitive and resistant strains.
Chemicals including antibiotics and bactericides were screened for suppression of streptomycin-resistant Erwinia cmutouom subsp. cmutovom (Ecc) strains in laboratory and field conditions. Oxytetracycline, ethoquinolac and dichlorophen suppressed the growth of streptomycin-resistant Ecc strains in vitro. Fractional inhibitory concentration (FIC) indices of oxytetracycline and ethoquinolac mixed with streptomycin against the Ecc strains were equal to and less than one, respectively. Consequently the efficacy of those chemicals in mixture with sorptomycin were non-antagonistic But that of dichlorophen mixed with streptomycin was more than one, therefore the efficacy of the mixture was antagonistic. Spray of oxyteoucycline, ethoquinolac and agrimycin-100 on the topped burley tobacco plants was efficacious in reducing tobacco hollow stalk at the same level of sorptomycin treatment in three-year field trials, which suggests that those are promising chemicals to be alternative to streptomycin for control of tobacco hollow stalk.
Recent researches demonstrated well promising anticancer activities for antibiotics. Such effects would be significantly increased while nanoparticle based delivery systems were applied. In this study, the goal was aim to improve anticancer and antitoxic effects of Streptomycin by loading on special kind of dendrimer (anionic-linear-globular second generation). In the current study, Size and zeta potential as well as AFM techniques have been used to prove the fact that the loading was performed correctly. The Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of the drug loaded on dendrimer nanoparticle were determined and compared with both of dendrimer alone and free drug with respect to staphylococcus aureus as the test microorganism. The anticancer activity among three groups including Streptomycin, Streptomycin -G2 dendrimer, and control was measured in vitro. In vitro studies showed that G2 anionic linear-globular polyethylene-glycol-based dendrimer, which loaded on Streptomycin was able to significantly improve the treatment efficacy over clinical Streptomycin alone with respect to proliferation assay. Maximal inhibitory concentration (IC50) was calculated to be $257{\mu}g/mL$ for streptomycin alone and $55{\mu}g/mL$ for Streptomycin -G2 dendrimer. In addition, Streptomycin -G2 dendrimer conjugate prevented the growth of MCF-7 cancerous cells in addition to enhance the number of apoptotic and necrotic cells as demonstrated by an annexin V-fluorescein isothiocyanate assay. Streptomycin -G2 dendrimer conjugate was able to increase Bcl-2/Bax ratio in a large scale compared with the control group and Streptomycin alone. Based on results a new drug formulation based nano-particulate was improved against S. aureus with sustained release and enhanced antibacterial activity as well as anticancer activity shown for functional cancer treatment with low side effects.
It has been known that streptomycin resistance in bacteria can occur as a results of chromosomal mutation or through gene acquisition or both. Chromosomal mutations for resistances are point mutations in the rpsL gene, which alter ribosomal protein S12. Acquired resistance has occurred when an $Sm^R$ plasmid carrying transposon Tn5393 with tandem strA-strB gene is transferred by conjugation. A total of 686 isolates of Xanthomonas smithii subsp. citri causal agent of citrus canker disease were collected from 26 citrus orchards in Jeju Island in 2003 and 2004 seasons. Forty-nine of 111 isolates from streptomycin non-sprayed orchards in 2003 season were resistant to streptomycin. Of 107 isolates from orchards sprayed one time with streptomycin, 58 isolates were resistant, and 166 of 221 isolates from orchards sprayed two times with streptomycin were resistant. In 12 orchards sprayed three or more times with streptomycin, 219 of 247 isolates were resistant to streptomycin. Twenty-five isolates of X. smithii subsp. citri were surveyed to identify the mechanisms of streptomycin resistance in this study. Twenty-one of these 25 isolates were resistant to streptomycin, and it was proven by PCR assay that 18 of the 21 streptomycin resistant isolates have the strB gene. In sixteen of the 21 streptomycin resistant isolates, it was occurred a point mutation altered codon lysine (AAG)-41 of rpsL gene to arginine (AGG). The streptomycin-sensitive isolates easily acquired the resistance by mixed culture with resistant isolates. The strB gene was amplified from the isolates that acquired the resistance by mixed culture, and one isolate of them was also point-mutated in codon 41 of rpsL gene to be resistant. In this study, most of the streptomycin-resistant isolates of X. smithii sub sp. citri in Jeju island expressed the resistance by both chromosomal point mutation and gene acquisition, and the resistance was easily acquired through conjugation by culture mixed with streptomycin resistant and sensitive strains.

The residue depletion of streptomycin was investigated in the olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major) after consecutive three days treatment with dipping water at a dose of 20 g/ton water. Fishes were sampled for muscle on 1st, 2nd, 3rd, 4th, and 5th day after treatment. Streptomycin concentrations were determined by high performance liquid chromatography with tandem mass spectrometry. The recovery rates of streptomycin in muscle samples ranged from 87.2 to 102.3% and from 80.4 to 94.1% for the concentration of 0.05 mg/kg and 0.1 mg/kg, respectively. Streptomycin concentrations detected on the 1st day after treatment were 0.066, 0.058, and 0.073 mg/kg in muscles of olive flounder, rockfish, and red sea bream, respectively. At day 2, residue concentrations of all samples were believed to decrease to lower than 0.05 mg/kg, the detection limit. From results of the present study, a withdrawal period of streptomycin is proposed on 3 days after consecutive three days treatment with dipping administration at a dose of 20 g/ton water to avoid the presence of excessive residues of the edible muscles of olive flounder, rockfish, and red sea bream. The present study showed that residue concentrations of streptomycin decreased to below 0.05 mg/kg after treatment 2nd day.
Chemical control of bacterial canker of kiwifruit caused by Pseudomonas syringae pv. actinidiae was attempted by spraying of streptomycin sulfate ·oxytetracycline WP streptomycin WP streptomycin ·copper hydroxide WP kasugamycin SL kasugamycin·copper oxychloride WP and copper hydroxide WP. The control efficacies of the bactericides were variable depending upon the spraying schedule,. Application of streptomycin WP and streptomycin sulfate·oxytetracycline WP from middle April to early May was found to be the most effective in controlling the bacterial canker. For copper hydroxide WP the spraying from middle January to early February showed the highest control efficacy. Kasugamycin SL was the most effective in controlling the disease by spraying from middle April to early May but it was still relatibvely effective during other spray periods. Foliar application of copper hydroxide WP and copper-antibiotic formulaions after middle April caused severe phytotoxicity. Kasgamycil SL streptomycin WP streptomycin·copper hydroxide WP and copper hydroxide WP were potential bactericides which could substitute streptomycin sulfate·oxytetracycline WP. Selective applications of the bactericides according to their optimum spray time can enhance the control efficacies against bacterial canker of kiwifruit and retard the emergency of resistant strains of P. syringae pv. actinidiae to the bactericides. The optimum spray number of streptomycin sulfate·oxytetracycline WP was 3 times with 15-day-intervals or 4 times with 10-day-intervals. The result suggested that the potential bactericides to bacterial canker of kiwifruit should be also used according to their optimum spray schedules in order to get their highest control efficacies.

Bacterial canker is a devastating disease of kiwifruit caused by the bacterium Pseudomonas syringe pv. actinidiae. Canker disease of kiwifruit in Korea has been controlled using streptomycin for more than two decades. Four streptomycin-resistant strains, belonging to biovar 2, which are found only in Korea, were collected between 2013 and 2014 from different orchards located in Jeju, Korea. The genetic background for streptomycin resistance among P. syringe pv. actinidiae strains were determined by examining the presence of strA-strB or aadA, which are genes frequently found in streptomycin-resistant bacteria, and a point mutation at codon 43 in the rpsL gene. All four streptomycin-resistant strains of P. syringe pv. actinidiae investigated in this study contained strA-strB as a resistant determinant. The presence of the aadA gene and a mutation in codon 43 of the rpsL gene was not identified.
Potato blackleg disease caused by Erwinia carotovora subsp. atroseptica has been a serious problem in Korea. It was previously reported that four mixtures [streptomycin (9.3 ppm) + copper oxide (171.6 ppm)/copper hydroxide (146.3 ppm), streptomycin sulfate (7.0 ppm)+copper oxide (171.6ppm)/copper hydroxide (146.3 ppm)] were effective for the control of E. carotovora subsp. atroseptica. in in vitro test. Using those four mixtures and two antibiotics [streptomycin (81.4 ppm) and streptomycin sulfate (61.3 ppm)], the effectiveness of control for E. carotovora subsp. atroseptica. was conducted in the field. Two antibiotics showed over 60% of control efficacy under different soil conditions, while mixtures of two antibiotics with copper compounds did not show any control effects on the infected seed potato. Two mixtures [streptomycin (27.9 ppm)+copper hydroxide (438.9 ppm), streptomycin sulfate (21.0 ppm) + copper oxide (514.8 ppm)] were effective in the control of potato blackleg disease on the infected potato plants under different climate conditions.

Commercial products with antibiotics like streptomycin as active ingredients have been used to control soft rot disease caused by Pectobacterium species for a long time. In this study, antibiotic resistance of twenty-seven Korean strains of Pectobacterium species including P. carotovorum, P. odoriferum, P. brasiliense, and P. parmenteri, which were previously shown to be susceptible to the bacteriophage phiPccP-1 was surveyed using a disk diffusion assay. While all strains were highly susceptible to ampicillin, kanamycin, chloramphenicol, tetracycline, and rifampicin, some strains showed weak susceptibility to 300 ㎍/ml of streptomycin. Furthermore, some of them are partially or completely resistant to commercial pesticides-Buramycinand streptomycin at the concentration of 250 ㎍/ml that is recommended by the manufacturer for streptomycin-based pesticides. These results indicate the presence of streptomycin-resistant Pectobacterium strains in South Korea, and the development of antibiotic alternatives to control soft rot is needed.
