  • The purpose of this study was to compare the growth-inhibiting effect of sodium fluoride and stannous fluoride on a cariogenic strain of Streptococcus mutans exposed to the different concentration of $SnF_2$ and NaF The result were as follows: 1. The growth rate of the Streptococcus mutans was unaffected by 75 ppmF, slowed by 150 ppmF, and immediately arrested by 300 or 600 ppmF where increments of NaF were added to actively growing 12-hour broth culture. 2. $SnF_2$ slowed the growth rate at 75 ppmF, was to bactericidal initially at 150 and 300 ppmF, and was to totally bactercidal at 600 ppmF. 3. $SnF_2$ has been shown to be more effective than NaF in inhibiting the growth activity of Streptococcus mutans. 4. The inhibitory effect of fluoride compounds on the growth of Streptococcus mutans was increased in the decreased pH at concentration of 600 ppmF.

Streptococcus mutans에 대한 티트리의 항균 효과 (Antibacterial effect of tea tree on Streptococcus mutans)

  • Objectives: The purpose of this study is to investigate the antibacterial effect of Streptococcus mutans of tea tree ingredient. Methods: The experimental groups were each given with different concentrations (30 or 50 vol%) of tea tree prepared in saline solution. The control group applied only saline solution. The tea tree coating of the specimen were examined under a scanning electron microscope. For the antibacterial activity test of the tea tree, the contact angle of the tea tree- coated specimen's surface was analyzed. The antibacterial effect against Streptococcus mutans was determined by counting the colony forming units (CFU). The statical statics were evaluated by using one-way ANOVA and paired t-test. Results: The tea tree treated group of hydrophilic more than non treated group. Antibacterial experiments demonstrated that tee tree solution was effective against Streptococcus mutans. However there was no significant difference in depending solution concentration groups. Conclusions: The antimicrobial activity of the tea tree containing solution showed its potential for use as coating for denture and medical materials.

Inhibitory effects of Coptis chinensis extract on the growth and biofilm formation of Streptococcus mutans and Streptococcus sobrinus

  • Streptococcus mutans and Streptococcus sobrinus play important roles in dental caries. Coptis chinensis is a natural product with antimicrobial activity against enterobacteria; however, its effects on oral streptococci are still unknown. Therefore, the effects of C. chinensis on the growth and biofilm formation of the representative cariogenic bacteria S. mutans and S. sobrinus were investigated for the possible use of C. chinensis as an anticaries agent. The C. chinensis extract was diluted with sterile distilled water, and 0.1-2.5% of the extract was used in the experiment. The effects of the C. chinensis extract on the growth and glucan formation of S. mutans and S. sobrinus were measured by viable cell counting and spectrophotometry at 650 nm absorbance, respectively. Crystal violet staining was also carried out to confirm the C. chinensis extract's inhibitory effect on biofilm formation. The C. chinensis extract significantly inhibited the growth of S. mutans and S. sobrinus at concentrations of ≥ 0.3% as compared with the control group. The viable cell count of colonies decreased by 1.7-fold and 1.2-fold at 2.5% and 1.25%, respectively, compared with the control group. The biofilm formation of S. mutans and S. sobrinus was inhibited by > 20-fold at C. chinensis extract concentrations of ≥ 1.25% as compared with the control group. In summary, the C. chinensis extract inhibited the growth and biofilm and glucan formation of S. mutans and S. sobrinus. Therefore, C. chinensis might be a potential candidate for controlling dental caries.

상백피의 Sanggenon C에 의한 Streptococcus mutans의 생육 및 균부착 저해효과 (The inhibitory Effect of Sanggenon C from the Root-bark of Morus alba L. on the Growth and the Cellular Adherence of Streptococcus mutans)

  • The methanolic extract of the root-bark of Morus alba L.(Mulberry tree) has the potent antibacterial activity against Streptococcus mutans. Its active component was identified to be sanggenon C. The active component had stronger anti-bacterial activity than berberine, having minimum inhibitory concentration(MIC) of $25\;{\mu}g/ml$. Moreover, the inhibitory effect of this component on the cellular adherence of Streptococcus mutans to glass surfaces also was more remarkable than that of berberine in the presence of glucosyltransferase(GTase) and sucrose in vitro. These results indicate that sanggenon C may play an important role in inhibiting plaque formation and caries incidence.

치태형성 억제세균의 분리 (ISOLATION OF THE BACTERIA INHIBITING THE FORMATION OF PLAQUE)

  • 치태의 주요성분인 비수용성 글루캔의 형성을 억제시키는 세균을 분리하기 위하여 유치원 원아 만여명으로부터 타액을 채취하였다. 일회용 큐벧을 사용하여 비수용성 글루캔의 형성을 억제시킨 세균을 분리하였다. 분리된 세균을 그람 염색과 API 20S kit와 API 50 CHL kit를 사용하여 당발효 및 생화학적 특성을 검사한 결과, Streptococcus oralis, Streptococcus mitis, Streptococcus mitior, Streptococcus sanguis, Enterococcus durans, Lactococcus lactis로 동정되었다. 비수용성 글루캔 형성 억제 정도를 판정하기 위하여 일회용 큐벧에서 S. mutans 단독 배양시에는 550nm에서의 흡광도가 1.503이었으나, S. mutans와 Streptococcus oralis, Streptococcus mitis, Streptococcus mitior, Streptococcus sanguis, Enterococcus durans, Lactococcus lactis, Lactobacillus acidophilus 혼합 배양시에는 각각의 흡광도가 0.823, 0.912, 0.894, 0.878, 0.753, 0.845, 1.021로 감소되었다.

  • The aim of this study was to investigate the susceptibility of mutans streptococci (S. mutans and S. sobrinus) and Streptococcus anginosus, for seven antibiotics, penicillin G, amoxicillin, ciprofloxacin, cefuroxime, erythromycin, bacitracin, and vancomycin. The minimum inhibitory concentration (MIC) of seven antibiotics against 3 species (type strains) of mutans streptococci and S. anginosus, 10 strains (wild type) of S. mutans, 7 strains (wild type) of S. sobrinus, and 11 strains (wild type) of S. anginosus, were measured by broth dilution method. All of the type strains of mutans streptococci and S. anginosus had the same susceptibility for penicillin G, amoxicillin, cefuroxime and bacitracin. Type strain of S. anginosus was sensitive in ciprofloxacin, but those of mutans streptococci were not. All of the clinical isolates of mutans streptococci and S. anginosus had the same susceptibility for the seven antibiotics. Our data reveal that mutans streptococci and S. anginosus have similar antibiotic-resistant character. In addition. these results may offer the basic data to verify the antibiotic-resistant mechanism of mutans streptococci and S. anginosus.

Effect of Bacteriocin-Like Inhibitory Substance (BLIS) from Enterococcus faecium DB1 on Cariogenic Streptococcus mutans Biofilm Formation

  • The aim of the study was to investigate the effect of bacteriocin-like inhibitory substance (BLIS) from Enterococcus faecium DB1 on cariogenic Streptococcus mutans biofilm. Crystal violet staining, fluorescence, and scanning electron microscopy analyses demonstrated that the BLIS from Enterococcus faecium DB1 (DB1 BLIS) inhibited S. mutans biofilm. When DB1 BLIS was co-incubated with S. mutans, biofilm formation by S. mutans was significantly reduced (p<0.05). DB1 BLIS also destroyed the preformed biofilm of S. mutans. In addition, DB1 BLIS decreased the viability of S. mutans biofilm cells during the development of biofilm formation and in the preformed biofilm. DB1 BLIS significantly decreased the growth of S. mutans planktonic cells. Furthermore, S. mutans biofilm on the surface of saliva-coated hydroxyapatite discs was reduced by DB1 BLIS. Taken together, DB1 BLIS might be useful as a preventive and therapeutic agent against dental caries caused by S. mutans.

자일리톨 섭취에 따른 Streptococcus mutans의 글루칸 생성관련 유전자 발현 억제효과 (INHIBITION OF GLUCAN SYNTHESIS RELATED GENE EXPRESSION OF STREPTOCOCCUS MUTANS BY XYLITOL TREATMENT)

  • 본 연구에서는 장기적인 자일리톨의 섭취가 Streptococcus mutans의 대표적인 독성인자 중 하나인 글루칸 생성에 미치는 영향을 알아보기 위하여 글루칸 합성효소인 glucosyltansferase의 mRNA 발현을 실시간 역전사 중합효소 연쇄반응을 통해 평가하여 다음과 같은 결과를 얻었다. 1. 24개월 동안 자일리톨껌을 섭취한 군에서 타액 내 Streptococcus mutans의 colony 수는 통계적으로 유의하게 감소하였다(p<0.05). 2. 비수용성 글루칸 합성에 관여하는 유전자인 gtfB, gtfC의 발현은 자일리톨껌을 섭취한 군에서 시간이 지남에 따라 유의 하게 감소하였다(p<0.05). 특히 gtfB의 발현은 12개월과 24개월째 대조군에 비하여 통계적으로 유의하게 낮았고, gtfC의 발현은 24개월째 대조군에 비하여 통계적으로 유의하게 낮았다(p<0.05). 3. 수용성 글루칸 합성에 관여하는 유전자인 gtfD의 발현 역시 자일리톨껌을 섭취한 군에서 시간이 지남에 따라 유의하게 감소하였다(p<0.05). 또한 gtfD의 발현은 12개월과 24개월째 대조군에 비하여 통계적으로 유의하게 낮았다(p<0.05). 이상의 결과들을 종합해 보았을 때, 자일리톨의 섭취는 구강 내 Streptococcus mutans의 글루칸 합성 관련 유전자들의 발현을 억제시킴으로써 Streptococcus mutans의 수적인 감소를 가져오는 것으로 생각된다.

Inhibitory Effect of Continentalic Acid from Aralia continentalis on Streptococcus mutans Biofilm

  • In our present study, we investigated the effects of continentalic acid on Streptococcus mutans (S. mutans) biofilm. Methanol extract of Aralia continentalis (A. continentalis) was suspended in water and sequentially partitioned with CHCl3, ethyl acetate (EtOAc), and n-butanol (n-BuOH). The CHCl3 fraction showed the highest activity and an antibacterial compound against S. mutans was isolated from this preparation through various chromatography methods by bioassay guided fractionation. MS, $^1H-NMR$ and $^{13}C-NMR$ analysis showed that the active principle was continentalic acid which was confirmed to show significant inhibitory effects against S. mutans biofilm. These results may provide some scientific rationale for the traditional use these extracts for the treatment of dental diseases.

  • Dental caries has been reported to be infectious disease by Streptococcus mutans(S. mutans). The ability of S. mutans to produce dental plaque has been shown to be related to glucan synthesis from sucrose by glucosyltransferase (GTase). Glucan is known to play an important role in the initiation of smooth surface caries. For preventing dental caries by traditional medicines, two hundred kinds of natural products were assayed for inhibiting activity against S. mutans and GTase. During the assay course, we chose some active plants against S. mutans and GTase and then applied these plant extracts to toothpaste.
