Copper possesses antimicrobial properties against certain bacteria and fungi. In this study, changes in microbial population, color and the antioxidant properties of dried red pepper powder (RPP) by contact with the surface of a copper plate were investigated. Lightness of RPP increased after treatment on the copper plate for 2 h at 55℃. The numbers of total aerobic mesophilic bacteria in RPP increased by 0.61 and 0.50 log on the surface of stainless steel and copper plates during 24 h at 37℃, respectively. The population of yeast and mold was reduced by 90% in the RPP treated on both copper and stainless steel plates under the same condition. Yeast and mold were significantly reduced on the copper plate during 2 h at 55℃. Scavenging activities of RPP treated on the copper plate for 24 h at 37℃ against DPPH radical were decreased; those for 2 h at 55℃ against ABTS radical were rather enhanced. The results suggest that copper might be applied in the processing and storage equipment in relation to RPP products for antimicrobial purposes.
This study was performed to investigate the microbiological contamination on toothbrushes, toothbrush caps, and tooth cleaning cups in the child care centers and to evaluate the toxin genes, toxin production ability and antibiotic resistance of food-borne pathogens. The average number of total aerobic bacteria and fungi were 5.3 log CFU and 3.2 log CFU. Coliform bacteria were detected in 41 (54.7%) of 75 toothbrushes, 13 (44.8%) of 29 toothbrush caps, and 29 (44.6%) of 65 tooth cleaning cups. Salmonella spp. was not detected in all of samples but Bacillus cereus was isolated from 1 (1.3%) of 75 toothbrushes and 2 (3.1%) of 65 tooth cleaning cups. Staphylococcus aureus was detected in 1 (1.5%) of 65 tooth cleaning cups. The nheA, nheB, nheC, hblC, hblD, hblA and entFM toxin genes were possessed in B. cereus isolated from toothbrush which also produce NHE and HBL enterotoxins. S. aureus was resistant to ampicillin and penicillin, while B. cereus was resistant to ${\beta}-lactam$ antibiotics. These results indicated that the sanitary conditions of toothbrushes and tooth cleaning cups in the child care centers should be improved promptly. The UV sterilization after drying and then storage in dried condition is required to improve the sanitary condition of toothbrushes and tooth cleaning cups in the child care center.
The physiological and sensory properties of breads, prepared with wheat flour substituted with various herbs, were evaluated. The specific gravities and maximum heights of the breads increased when the wheat flour was supplemented with 1% rosemary, lemon balm or lavender, and with 3% rosemary, but not with 3% lemon balm or lavender. The fermentation and cooling losses of the breads with added herbs were smaller than those of the control, but there was no significant difference in the baking loss. The lightness of the breads decreased with increases in the herb contents. In the texture measurements for the breads, the hardness decreased slightly on the addition of 1% of the herb powders, but increased on the addition of 3% of the herb powders. As for the results of the sensory evaluation, the overall acceptances of the breads with 3% added herbs were higher than those of the control and 1% added herb breads. The addition of herbs to the breads inhibited the growth on fungi, and the more herbs substituted, the higher the degrees of this inhibition. The moisture contents were slightly increased, and the staling rates during storage at 25 decreased, in breads with added herbs. These results suggest that the shelf-lives of the breads were extended by the addition of herbs.
To explain the change of the pungent principles of red pepper, capsaicin, during storage, the relation between infecting mold and capsaicin was studied. Results obtained were summarized as follows; 1. Superior strains which showed good growth and high decomposition activity in their culture broths, Aspergillus oryzae and Aspergillus sp. KF-7 were obtained from the contaminated red pepper fruits. 2. The weight of dry matter was increased at low concentration of capsaicin $(2{\sim}3{\mu}g/ml)$ but it was decreased at high concentration. 3. The residual capsaicin in the culture medium was reduced as half as such after 1 week, and almost disappeared after 2 weeks. At $30^{\circ}C$ and $pH\;7{\sim}9$ of culturing condition, the amounts of reducing capsaicin reached to the maximum. 4. At ground red pepper fruits as culture medium, Aspergillus sp. KF-7 could not glow at all, but Aspergillus oryzae showed good growth at 13% of moisture contents and the residual of capsaicin of red pepper fruits after 40days of cultivation time was 45%. 5. At ground red pepper fruits seed as culture medium, Aspergillus oryzae and Aspergillus sp. KF-7 showed good growth, and the residual of capsaicin after 4 weeks of cultivation time was 55% and 38% respectively.
Peanuts in storage were estimated for mycotoxigenic fungi and mycotoxins. Peanut samples collected from storages in Gochang were mainly contaminated with Fusarium (17.2±28.0%), Penicillium (12.4±28.0%), and Aspergillus (8.0±7.6%). Other genera, including Talaromyces, Rhizopus, Rhizoctonia, Trichocladium, Clonostachys, Mucor, Chaetomium, Trametes, Epicoccum, and Humicola, were also found. Although aflatoxins were not detected in the peanut samples, 29 strains of Aspergillus flavus were identified using molecular marker genes. Among them, 17 selected isolates produced aflatoxins in solid culture media ranging from 0.61-187.82 ㎍/kg. All of them could produce both aflatoxin B1 and B2 and some (n=5) produced additional G1, G2, or both. This study is the first report that A. flavus stains obtained from Korean stored peanut are aflatoxigenic.
To control Phytophthora blight of red pepper biologically caused by Phytophthora capsici, we developed Trichoderma harzianum DYMC for commercial product. DYMC was storage at room temperature and was investigated their population every 3 months for 1 year. For investigating the dynamic population of T. harzianum in the pot soils, we applied powder and suspension applications with DYMC, and then investigated for 95 days. The efficacy of powder and suspension applications of DYMC for control of Phytophthora blight of red pepper and plant growth were investigated for 50 days in greenhouse experiment. The population of T. harzianum was decreased at the room temperature for 1 year but there was not statistically significance. After soil treated in the pot with DYMC, the population of Trichoderma spp. was the highest when DYMC powder at 5 g was applied to mix with pot soil, and the population was deceased significantly among treatment means as time goes by ($R^2=0.76$, F=10.5960, P=<.0001). Incidence of Phytophthora blight of, red pepper was significantly reduced among treatment means on 50th day after treated with DYMC ($R^2=0.82$, P=16.4758, P=<.0001). Disease control value was the highest at 62.5% when DYMC powder at 5 g was applied to mix with pot soil. No significant difference (P=0.05) of effects of plant and root growth showed by treated with DYMC on 60th day, except stem. Mixing the application of DYMC powder with soil to control Phytophthora blight of red pepper was greater than suspension application to dilute with water. DYMC could be used as an effective biocontrol agent to control Phythophthora blight of red pepper.
Gamma irradiation as a new physical treatment was applied to comparative investigates with a conventional ethylene oxide fumigant on the microbiological and physicochemical qualities of selected spices and dry vegetables such as powdered red pepper, black pepper, welsh onion, onion, garlic, carrot, korean cabbage and instant ramyon soup. The microorganisms contaminated in the sample, including total viable count, thermophilic bacteria, aerobic spore and fungi counts between the $10^4\;to\;10^6/g$ range. Coliforms were found only in black pepper and welsh onion powder as the $10^2\;to\;10^3/g$ level. A radiation dose of 7 to 10 KGy were sterilized completely to the contaminated microorganisms, while ehthylene oxide (E.O.) fumigation reduced of them to the $10^3/g$ level. An optimum dose of irradiation was less detrimental than E.O. fumigation to the physicochemical properties of the sample. Sensory evaluation after three months of storage at room temperatures showed that the overall acceptability of irradiated sample was higher than that of the non treated control as well as E.O. fumigated samples. Comparison gamma irradiation with E.O. gas treatment showed that E.O. treatment was less effective than radiation in cotrolling microbial contamination of spices and vegetables.

Mycelial growth of Lentinula edodes on solid or liquid culture media supplemented with differentconcentrations of oak wood vinegar varied depending on the types of wood vinegar or mushroom varieties used.Oak wood vinegar obtained from traditionally carbonizing kiln (TWV) inhibited mycelial growth of L. edodes atthe dilution level of less than $5{\times}10^{-2}$, but stimulated at $10^{-3}$ to $2{\times}10^{-3}$. Wood vinegar from mechanicallycarbonizing kiln (MWV) inhibited at $10^{-3}$, but stimulated at $2{\times}10^{-3}$. In liquid culture media, both wood vinegarinhibited at $5{\times}10^{-2}$, but stimulated at $2{\times}10^{-3}$. Sanjo-302-ho grown in liquid culture media at $2{\times}10^{-3}$, and Sanrim 2and 3-ho grown at $4{\times}10^{-3}$ showed relatively high degree of wood decay (DWD) and growing ability within wood(GAWW) when these isolates were inoculated onto oak wood logs. TWV completely inhibited mycelial growth ofgreen mold fungi, Trichoderma species, tested at $5{\times}10^{-1}$ dilution level, while MWV inhibited at $5{\times}10^{-1}$ to $5{\times}10^{-2}$dilution level. For Diatrype stigma, TWV inhibited mycelial growth at the dilution level of less than $5{\times}10^{-2}$, whileMWV did 80% of mycelial growth at $10^{-2}$, and 100% at $5{\times}10^{-1}$ dilution level. Fresh and dry weight of fruitingbodies harvested after soaking of wood logs into wood vinegar solutions with different concentrations werecompared, and were the highest at $2.5{\times}10^{-2}$ dilution level. Storage test of fruiting bodies at $10^{\circ}C$ for 10 daysshowed that fruiting bodies harvested after soaking in the solution with $2.5{\times}10^{-2}$ dilution level showed the bestfreshness by general test and color changes. In addition, shear force value of L. edodes fruiting bodies measuredby using texture analyzer showed that $2.5{\times}10^{-2}$ dilution level was the best concentration for keeping flesh texture.
Two experiments were conducted to investigate effects of molasses addition to silage materials in ensiling Socheongryongtang meal on the nutritive quality of silage, palatability and various ruminal parameters in Korean native goats. In Experiment 1, Socheongryongtang meal silage was produced by the addition of 0, 0.5, or 1.0% molasses and stored for 40 days at room temperature. There were three replicates per treatment. Lactic acid contents of Socheongryongtang meal silage containing molasses(0.5 and 1.0%) were significantly(p<0.05) higher than that of the control(0%). However, the pH and butyric acid contents of Socheongryongtang meal silage containing molasses(0.5 and 1.0%) were lower than those of the control(0%). In addition, molasses(0.5 and 1.0%) increased the number of lactobacillus, but decreased the number of fungi in Socheongryongtang meal silage. In vitro dry matter disappearance tended to increase by molasses addition. In Experiment 2, three 1.5-yr-old Korean native female goats were employed in cross-over design to measure the palatability and various ruminal parameters of Socheongryongtang meal silage. Feed intake of Socheongryongtang meal silage containing molasses(0.5 and 1.0%) for 30 min significantly higher than that of control(0%). Silmilar trend was found when the feed intake was measured for 6 hr. In various ruminal parameters, molasses addition to silage materials increased propionic acid(P) and total volatile fatty acid contents, but decreased pH, acetic acid (A), and butyric acid. The A/P ratio was the lowest in 1.0% molasses treatment group. It is concluded that molasses addition in ensiling Socheongryongtang meal could enhance its nutritional quality and lengthen storage period. Also the palatability of silage was improved by the addition of molasses.
