2,4,6-Trinitrotoluene (TNT)을 분해할 수 있는 Stenotrophomonas sp. OK-5에서 분리한 NAD(P)H-nitroreductase의 특성을 조사하였다. 먼저 ammonium sulfate precipitation, DEAE-sepharose, 그리고 Q-sepharose 등의 일련의 정제 과정을 통하여 NAD(P)H-nitroreductase을 분리정제하였다. 분취기로부터 얻어진 시료로부터 NAD(P)H-nitroreductase의 효소활성을 가지는 3개의 다른 fractions (I, II 및 III)이 탐침되었다. NAD(P)H-nitroreductase의 fractions I, II, 그리고 III의 비활성(specific activity)은 각각 5.06 unit/mg, 4.95 unit/mg, and 4.86 unit/mg이었으며, crude extract와 비교하여 각각 10.5배, 9.8배, 8.9배 이상 농축되었다. 이 실험에서 이들 3개의 fractions 가운데, fraction I이 가장 높은 비활성을 나타내었다. NAD(P)H-nitroreductase (fractions I, II 및 III)의 효소활성에 영향을 미치는 몇 가지 요인을 조사하였다. 모든 NAD(P)H-nitroreductase (fractions I, II 및 III)의 최적 온도는 30$^{\circ}C$이었으며, 최적 pH는 약 7.5이었다. 4Ag^+, Cu_2^+, Hg_2^+$ 등의 금속이온은 약 80%의 효소활성을 저해하였으나, $Mn_2^+ 이나 Ca_2^+$의 첨가 시에는 약 30~40% 정도의 활성이 감소되었다. 그러나 $Fe_3^+$은 이들 효소의 활성을 증진시켰다. SDS-PAGE에 의해 측정된 NAD(P)H-nitroreductase의 fractions I, II 및 III의 분자량은 모두 약 27 kDa임이 확인되었다.
식물 종자를 비롯한 각종 유전자원을 액체질소에 저장하면 유전형질 특성의 손실 없이 장기간 보존할 수 있다. 본 연구에서 보존조건이 까다로워 단명 종자로 분류되는 들깨 종자를 초저온 동결저장 방법으로 보존할 수 있는지를 조사하였다. 수집한 들깨 품종별 종자의 초기 발아율은 40-95% 수준으로 다양하였는데, 수분함량을 3-8%로 조절한 종자를 액체질소에 처리하여도 발아율은 감소하지 않았다. 종자의 수분함량이 4-5%인 종자의 초저온 처리 후 발아율이 가장 높았으며, 초기 발아율이 낮은 품종에서는 초저온 처리에 의해 발아율이 증가하기도 했다. 인위 노화처리에 따라 종자의 발아율과 ascorbate peroxidase 활성은 감소하였으며, 품종별로 종자의 활력 저하 정도는 크게 달리 나타났다. 대조 처리 종자와 비교하였을 때, 초저온 처리과정에 발생할 수 있는 산화스트레스가 들깨 종자의 활력을 저해하지는 않을 것으로 추정되었다. 따라서 들깨 종자를 4-5% 수분함량으로 건조시켜 초저온 동결 저장하면 활력 손실 없이 장기간 보존할 수 있을 것으로 판단되며, 노화가 급속히 진행되는 품종의 경우에는 고활력 자원을 확보하거나 휴면타파 처리 후 초저온 동결 보존하는 것이 유리할 것으로 생각된다.
삶의 질이 향상됨에 따라 환자의 흉터 치료 수요는 증가하고 있는 반면, 흉터 개선 제품은 대부분 수입 제품에 의존하고 있는 실정이다. 엔케팔린은 신경말단에서 분비하는 신경 펩타이드의 한 종류이다. 피부세포와 신경세포는 배엽 발달 과정 중 공통적으로 외배엽에서 유래하며, 피부세포 막에서 신경세포에서 발현되는 신경펩타이드 수용체 즉, 오피오이드 수용체(Opioid Receptor)가 발현된다. 오피오이드 수용체는 뮤-(${\mu}$), 델타-(${\delta}$), 카파-(${\kappa}$) 세 종류가 있으며, 각각을 MOR, DOR, KOR라고 부르고, 델타-오피오이드 수용체는 피부에서의 상처 치유(Wound-healing)에 직접 관여하는 것으로 보인다. 본 논문에서는 엔케팔린의 Alanine Scan 방법을 이용하여 엔케팔린 유도체를 합성하여, 피부세포내 안전성과 Cell migration assay를 통한 In vitro 상처 치유 촉진 효능, 프로 콜라겐 합성능력 및 보습효과를 실험을 통해 검증하였고, 피부 상처 마우스 모델에서 엔케팔린 유도체의 상처 치료 효과를 확인하였다. 그 결과 엔케팔린 유도체 AGGFL(AS10) 및 YGGAL(AS13)이 상처 치유 효능, 프로 콜라겐 합성증가, 보습 관련 유전자 발현 증가를 확인하였고, 특히 AS13이 피부 상처 마우스 모델에서 뛰어난 상처치료 효과를 확인하였다.
$(Ca^{2+}+Mg^{2+})$-ATPase를 쥐의 근소포체로부터 sucrose density gradient centrifugation의 방법을 사용하여 분리 정제하였다. 정제된 효소를 폴리아크릴 아마이드 젤에서 전기영동한 결과, 토끼와 닭의 경우에서와 같이 분자량 115,000인 단일 단백질 띠로 나타났다. 정제된 이 효소의 활설도는 50 $\\muM$의 $Mg^{2+}, Ca^{2+}, Co^{2+}, Fe^{2+}, Min^{2+}$에 의해서는 증가되었고, 같은 농도의 $Zn^{2+}, Cu^{2+}, Hg^{2+}$에 의해서는 감소되었다. Quinine와 quinacrine 같은 antimalarial drug는 이 효소의 활성도에 큰 영향을 주지 않았으나, p-hydroxymercuric benzoate와 phenylmethylsulfonylfluoride는 이 효소의 활성을 억제하였다. 이 효소는 pH 6과 7 사이에서 가장 높은 활성을 나타내었고, ATP를 기질로 사용하였을 때 Km 값은 98 $\\muM$이었다. $(Ca^{2+}+Mg^{2+})$-ATPase는 microsomal fraction에서 선택적으로 분해되었다. $^{3}H-casein$ 이나 ^{125}I-insulin같은 방사성 동위원소로 표지된 기질을 사용하여 단백질 분해에 대한 활성도를 조사해 본 결과, microsomal preparation에 metalloendoprotease가 존재하였다. 그러나 아직까지는 그 효소가 $(Ca^{2+}+Mg^{2+})$-ATPase를 분해하는지는 확실하지 않다.
이 연구의 목적은 치주인대 섬유아세포에 MTA를 접촉시킨 뒤 성장인자 transforming growth factor-beta1 $(TGF-\beta1)$, fibroblast growth factor-2 (FGF-2) 및 cytokine interleukin-6 (IL-6)의 발현량 변화를 측정하는 것이다. MTA군에서는 100 mg씩의 ProRoot MTA와 증류수를 혼합하고, IRM군은 동량의 IRM 분말을 용액에 혼합하여 이 시료들을 경화반응이 진행되도록 7일간 놓아두었다. 사람의 치주인대 섬유아세포를 배양하여 MTA와 IRM시료 상에 well당 $1\times10^5$개 수준으로 도포한뒤 6, 12, 24, 48시간 동안 배양하였다 (n = 5). 대조군으로는 재료의 접촉 없이 배양한 세포를 사용하였다. 시료에서 상층액을 분리하여 $TGF-\beta1$, FGF-2, IL-6의 발현량을 enzyme-linked immunosorbent assay (ELISA)법으로 측정하였다. MTA군에서, 성장인자인 $TGF-\beta1$과 FCF-2는 대조군에 비해 유의성 있게 발현이 억제되었으며 (p < 0.05), cytokine인 IL-6 발현량은 대조군과 유사한 수준으로 나타났다.

Yoga has been known to have stimulatory or inhibitory effects on the metabolic parameters and to be uncomplicated therapy for obesity. The purpose of the present study was to test the effect of an 8-week of yoga-asana training on body composition, lipid profile, and insulin resistance (IR) in obese adolescent boys. Twenty volunteers with body mass index (BMI) greater than the 95th percentile were randomly assigned to yoga (age $14.7{\pm}0.5$ years, n=10) and control groups (age $14.6{\pm}1.0$ years, n=10). The yoga group performed exercises three times per week at 40~60% of heart-rate reserve (HRR) for 8 weeks. IR was determined with the homeostasis model assessment of insulin resistance (HOMA-IR). After yoga training, body weight, BMI, fat mass (FM), and body fat % (BF %) were significantly decreased, and fat-free mass and basal metabolic rate were significantly increased than baseline values. FM and BF % were significantly improved in the yoga group compared with the control group (p<0.05). Total cholesterol (TC) was significantly decreased in the yoga group (p<0.01). HDL-cholesterol was decreased in both groups (p<0.05). No significant changes were observed between or within groups for triglycerides, LDL-cholesterol, glucose, insulin, and HOMA-IR. Our findings show that an 8-week of yoga training improves body composition and TC levels in obese adolescent boys, suggesting that yoga training may be effective in controlling some metabolic syndrome factors in obese adolescent boys.
Sprague-Dawley 랫트(실험용 쥐)에서 간을 적출하여 Collagen coating된 세포 배양 접시에 간세포 배양을 하여 친환경 재배 배 추출물과 일반 관행 재배 배 추출물의 간세포 기능성을 분석하였다. 3일간 적응 시킨 후 관행 재배 배 추출물과 친환경 재배 배 추출물을 농도 별(동결 건조된 $5\;{\mu}g/ml$, $20\;{\mu}g/ml$, $40\;{\mu}g/ml$) 처리하여 간세포의 에너지원인 ATP assay를 실시한 결과 관행 재배 배 추출물의 경우는 대조군과 유의한 차이는 인정이 되지 않았으나 친환경 재배 배 추출물의 경우 농도 의존적으로 정상군에 비해 180%까지 증가하는 것으로 나타났다. 한편 세포 성장의 경우도 DNA의 특유 염기 구성성분중의 하나인 thymine의 전구체인 $[^3H]$-thymidine을 간세포에 처리하여 DNA 합성을 알아본 결과 친환경 재배 배 추출물 을 농도별로 처리하였을 때 관행재배 배 추출물에 비해 간세포의 세포 성장율이 증가하는 것으로 나타났다($40\;{\mu}g/ml$ 투여 시 관행 재배 배추출물 112% vs. 친환경 재배 배 추출물은 234%; p<0.05). 아울러 세포성장 단백질인 CDK-2, CDK-4의 경우도 친환경 배 추출물 처리 시 현저하게 증가하는 것으로 나타났으며 세포성장 억제 단백질인 p21WAF1/Cip1 및 p27 Kip1의 경우는 억제시키는 것으로 나타났다.
Wound healing is delayed in diabetic patients. Increased apoptosis and endothelial progenitor cell (EPC) dysfunction are implicated in delayed diabetic wound healing. Melatonin, a major secretory product of the pineal gland, promotes diabetic wound healing; however, its mechanism of action remains unclear. Here, EPCs were isolated from the bone marrow of mice. Treatment of EPCs with melatonin alleviated advanced glycation end product (AGE)-induced apoptosis and cellular dysfunction. We further examined autophagy flux after melatonin treatment and found increased light chain 3 (LC3) and p62 protein levels in AGE-treated EPCs. However, lysosome-associated membrane protein 2 expression was decreased, indicating that autophagy flux was impaired in EPCs treated with AGEs. We then evaluated autophagy flux after melatonin treatment and found that melatonin increased the LC3 levels, but attenuated the accumulation of p62, suggesting a stimulatory effect of melatonin on autophagy flux. Blockage of autophagy flux by chloroquine partially abolished the protective effects of melatonin, indicating that autophagy flux is involved in the protective effects of melatonin. Furthermore, we found that the AMPK/mTOR signaling pathway is involved in autophagy flux stimulation by melatonin. An in vivo study also illustrated that melatonin treatment ameliorated impaired wound healing in a streptozotocin-induced diabetic wound healing model. Thus, our study shows that melatonin protects EPCs against apoptosis and dysfunction via autophagy flux stimulation and ameliorates impaired wound healing in vivo, providing insight into its mechanism of action in diabetic wound healing.
Rumex crispus (curled dock), which is a perennial wild plant, has long been used as a laxative, astringent, and medicine to treat blood and skin diseases. We recently reported that the roots of R. crispus are an effective nutraceutical for bone. This study prepared ethanol extracts of the leaves and roots of R. crispus, and analyzed the major constituents using liquid chromatography and mass spectrometry. In addition, their effects on the proliferation and differentiation of human osteoblast-like MG-63 cells, such as cell viability, alkaline phosphatase (ALP) activity, collagen content, and mineralization, were compared. The chromatograms of the chemical constituents of the two extracts exhibited quite different profiles: quercetin and quercitrin were identified as major peaks in the leaf extract, whereas cinnamtannin B1 and procyanidin isomers were the major peaks for the root extract. Neither extract was cytotoxic at concentrations of < $25{\mu}g/ml$. ALP activity and collagen synthesis-which are markers of the early stage of osteogenesis-in MG-63 cells were significantly increased upon the addition of the root extract compared with the addition of the leaf extract. In contrast, the leaf extract had a more stimulatory effect on mineralization-which is marker of the late stage of osteogenesis-in MG-63 cells than did the root extract. In conclusion, extracts of both leaves and roots of R. crispus stimulated the bone-forming activity of osteoblasts; in particular, the root extract was more effective in the early stage of osteoblast differentiation, while the leaf extract was more effective in the late stage. This difference in anabolic activity may be due to differences in the constituents of the leaves and roots. The leaves and roots of R. crispus appear to complement each other as stimulators of bone formation.
