Studies on the Optimal Conditions of Sterilization for Streptococcus pyogenes (Part 1) Effect of Heat Treatment on Typical Death Rate (항암활성을 지닌 Streptococcus pyogenes의 적정 살균조건에 관한 연구 (제 1보) 살균조건에 미치는 열 처리의 영향)

  • Studies were made on the optimal conditions of sterilization for Streptococcus pyogenes treated with heat. The results were as follows: The optimal temperature on growth of Streptococcus pyogenes was 37$^{\circ}C$ and mean generation time was 20 minutes in the logarithmic growth phase. The suspension of Streptococcus pyogenes, adjusted to pH 6-9 and treated with heat at 5$0^{\circ}C$, showed logarithmic death rate. Specific death rate constant(k) values at pH 6-9 were 0.1448, 0.1194. 0.1273 and 0.1707 minute$^{-1}$ , respectively.

Microbial Changes of Salted and Fermented Shrimp by High Hydrostatic Pressure Treatment (초고압처리에 의한 새우젓의 미생물변화)

  • This study was conducted to enhance the storage stability of fermented shrimp with different salt contents using a high hydrostatic pressure. The effects of the magnitude of pressure and treatment time on the microorganisms of the fermented shrimp were investigated. The highest microbial counts with respect to the salt levels were observed at 18% salt, showing $3.4{\times}10^5\;CFU/g$ for general bacteria, $6.4{\times}10^4\;CFU/g$ for halophilic bacteria, $4.2{\times}10^5\;CFU/g$ for yeast and $3.0{\times}10^4\;CFU/g$ for halophilic yeast. The degree of sterilization increased with the magnitude of pressure and treatment time, and the sterilization could be analyzed by the first order reaction kinetics. The sterilization rate constants $(k_p)$ of the halophilic bacteria was lower than that of general bacteria. The $log(k_p)$ increased linearly with pressure and the slope of the regression line of the halophilic bacteria was greater than that of general bacteria, indicating that the sterilization of the halophilic bacteria was more dependent on the pressure. High hydrostatic pressure treatment was an effective non-thermal sterilization method for the salted and fermented shrimp, and the optimum treatment condition was for 10 min at 6,500 atm.

Sterilization of Yakju(Rice Wine) Using a Batch-type High Voltage Pulsed Electric Field System (고전장펄스를 이용한 약주의 회분식 살균)

  • Yakju(rice wine) was sterilized with high-voltage pulses of short time on a batch pulsed electric field(PEF) system. The initial microbial counts of Yakju were 7.52 X $10^4\;CFU/mL$ for total aerobes, 2.20 X $10^4\;CFU/mL$ for lactic acid bacteria and 7.08 X $10^4\;CFU/mL$ for yeasts. The pH, acidity and electric conductivity of Yakju were 3.36, 0.462% and 1.24 mS/cm, respectively. Yakju was treated with 2-250 of pulses exponential-wave formed electric pulses under the field strength of 12.5-25 kV/cm. The critical strengths of the electrical field for the sterilization of Yakju were 7.5 kV/cm for total aerobes, 8.5 kV/cm for lactic acid bacteria and 6.5 kV/cm for yeasts. Logarithmic survival rates decreased linearly at low pulse number, but curvilinearly at high pulse number. The PEF sterilization kinetics of Yakju could be analysed by In s = In A-k In (n) and the sterilization rate constant increased with electric field strength and the size of target microorganisms. No changed in pH, acidity, and the growth of microorganisms were found in the PEF treated Yakju during the storage for 6 weeks at both $4^{\circ}C$ And $30^{\circ}C$.

  • Hot water is used by having a wash, dishes, taking tub and drinking water in residential buildings, and the use objective is to raise comfort of human body sense, washing and sterilization effect and so on. Cold & hot water supply system is understanded simpler than HVAC systems relatively, so it is true that pace of performance improvement is slower than other systems for plan and technical development. In this study, the performance evaluations are conducted under the condition of composition ratio by 1:1 for cold & hot water supply manifold system using functionally complex valves such as constant flow regulating valve and 3-way mixing valve in the area of $105.6m^2$ apartment which consist of the largest part of the whole apartment. Also, flow rate related to simultaneous use of faucets and change of hot water temperature are compared with the existing method.

Application of high voltage pulse for reduction of membrane fouling in membrane bio-reactor and kinetic approach to fouling rate reduction (막결합형 생물반응기(Membrane Bio-Reactor)의 막 오염 저감을 위한 고전압 펄스의 적용과 막 오염 저감 속도론적 해석)

  • Although membrane bio-reactor (MBR) has been widely applied for wastewater treatment plants, the membrane fouling problems are still considered as an obstacle to overcome. Thus, many studies and commercial developments on mitigating membrane fouling in MBR have been carried out. Recently, high voltage impulse (HVI) has gained attention for a possible alternative technique for desalting, non-thermal sterilization, bromate-free disinfection and mitigation of membrane fouling. In this study, it was verified if the HVI could be used for mitigation of membrane fouling, particularly the internal pore fouling in MBR. The HVI was applied to the fouled membrane under different conditions of electric fields (E) and contact time (t) of HVI in order to investigate how much of internal pore fouling was reduced. The internal pore fouling resistance (Rf) after HVI induction was reduced as both E and t increased. For example, Rf decreased by 19% when the applied E was 5 kV/cm and t was 80 min. However, the Rf decreased by 71% as the E increased to 15 kV/cm under the same contact time. The correlation between E and t that needed for 20% of Rf reduction was modeled based on kinetics. The model equation, E1.54t = 1.2 × 103 was obtained by the membrane filtration data that were obtained with and without HVI induction. The equation states the products of En and t is always constant, which means that the required contact time can be reduced in accordance with the increase of E.

Effect of Microbe Control and Water Temperature on Early Growth and Yield of Soybean Sprouts (콩 종실의 미생물 제어방법과 수주온도에 따른 콩나물의 초기 생육 및 수율)

  • High temperature sterilization method (30min. at $40^{\circ}C$longrightarrow30 min. at $70^{\circ}C$longrightarrow30 min. cooling at $20^{\circ}C$) was evaluated better than that of chloride, ion water, $O_3$ water, and salt water for the controling of micro-organisms at the early stage of sprout culture. Due to the improved germination rate and effective control of micro-organisms at early stage, the method resulted in much lower in the number of micro-organisms, higher in sprout yield, and 2 days longer in quality keeping at market than sprouts prepared by conventional method. There are two methods related with water temperature; constant temperature method (18.5$\pm$ $0.5^{\circ}C$) which keeps the same water temperature during the culture and 3-stage temperature method which changes the water temperature depending on the days after culture. Three-stage temperature method set the temperature at 21$\pm$ $1^{\circ}C$) in the second and third days after the initial acceleration of germination (about 4 hours), at 18.5$\pm$0.5$^{\circ}C$ in the fourth and fifth days, and at 17.0$\pm$ $0.5^{\circ}C$) in the sixth and seventh days. Three-stage temperature method could enhance the resistance power to diseases at the early growth stage, control the growth to standard goods (8-9cm in length and 2.15-2.30mm in width), and keep good body color at the middle or final stage. This method also increased the sprout yield by 6% compared with the conventional method, constant temperature method.

Electrochemical Generation of Chlorine Dioxide Using Polymer Ion Exchange Resin (고분자 이온교환수지를 이용한 의료.식품용 멸균제 이산화염소의 전기화학분해 발생)

  • A characteristic study of chlorine dioxide generation by the electrolysis system was performed after chlorite ($ClO_2^-$) is adsorbed from sodium chlorite by a polymer ion exchange resin. A strongly basic anion exchange resin was used and a Ti plate coated with Ru and Ir was used as an electrode. Various parameters such as reaction stirring velocity, reaction temperature, chlorine dioxide product concentration, ion exchange resin content and product maker type for the adsorption quantity in the chlorite adsorption of ion exchange resin were investigated and found the ion exchange resin with the maximum adsorption quantity. A generation trend of chlorine dioxide was observed by the electrolysis system and optimum conditions on the desired value were found using response surface design of DOE (Design of Experiments). The strongly basic anion exchange resin with the maximum adsorption quantity was SAR-20 (TRILITE Gel type II) and the adsorption quantity was around 110 mg/IER (g). Observed generation optimum conditions of chlorine dioxide were constant-current (electrode area base; $A/dm^2$) and flow rate of $N_2$ gas (4.7 L/min) at the desired value of sterilization (900~1000 ppm, 1 h).
