The Correlation of Agronomic Characters and Path Coefficient Analysis in Panax ginseng C.A. Meyer (고려인삼(Panax ginseng C.A. Meyer)의 년차간 형질상관 및 경로계수 분석)

  • This study was carried out to investigate the correlation of agronomic characters, their path coefficients in 2, 3 and 4-year old ginseng plants, and to provide a useful information for ginseng breeding. Correlation coefficients between stem fen변h, number of leaves and number of Iraflets in 2-year age, and stem diameter and leaf length in 3-year age showed highly significant correlation with number of fruits and root weight in 4-year age. The path coefficient analysis indicated that stem length and number of leaflets might give indirect effects on root weight regardless of plant age. On the other hand, stem length and number of leaflets in 2-year age and, stem diameter and leaf length in 3-year age showed direct effects on root weight in 4-year old ginseng. These results may be used for selection of high-yielding ginseng plants. Key words Selection information, correlation and path coefficient analysis.

  • Saponins of ginseng root, leaf and stem were identified by TLC. Eleven unknown spots were detected in ginseng leaf and ten unknown spots in ginseng stem on TLC besides seven ginsenosides such as $ginsenoside-Rg_1,\;-Rf,\;-Re,\;-Rd,\;-Rc,\;-Rb_2,\;and\;-Rb_1$ which are contained in ginseng root. $Ginsenoside-Rg_3\;and\;-Rg_2$ were identified on TLC from mild hydrolysates with 50% acetic acid of total saponins from ginseng root, leaf and stem. Meanwhile, panaxadiol, panaxatriol and oleanolic acid were identified from hydrolysates with 7% ethanolic sulfuric acid of total saponin of ginseng root, while panaxadiol and panaxatriol from those of total saponins of ginseng leaf and stem.

Correlation between Growth Characteristics and Root Yield in Collected Peony Lines (수집작약(蒐集芍藥)의 생육특성(生育特性)과 뿌리 수량(收量)과의 관계(關係))

  • The correlations between growth charateristics and root yield were investigated in 74 peony lines collected from chief producing place in Korea. Among these peony lines, single flower type was 61% and the rest were double type. And pink flower was in majority. Stem length, stem diameter and number of stem per plant were $50{\sim}60cm$, $7{\sim}8mm$ and $5{\sim}6$, respectively. Generally, disease tolerance and root yield were better in single flower type peonies than double type. Among single flower type peonies, the correlation coefficients between each characteristics and root yield were as follows : number of stem $(0.676^{**})$, number of roots $(0.646^{**})$, stem length $(0.617^{**})$, root diameter $(0.590^{**})$ and root length $(0.443^{**})$. Correlation between root yield and paeoniflorin content was highly significant $(0.383^{**})$ among single flower type peonies and significant $(-0.382^*)$ among double flower type peonies. Correlation between stem diameter and root yield was not significant (0.066).

Cadmium and zinc interaction and phytoremediation potential of seven Salix caprea clones

  • We evaluated the interaction between Cd and Zn in the bioaccumulation of seven clones of Salix caprea, which were exposed both to Cd and Zn alone and to a combination of Cd and Zn. Cadmium (Cd) and Zn concentration in the four treatments were administered in the following order: root > leaf > stem, and obvious differences were noted among the treatments and clones. The leaf Cd concentration of clone BH2 and stem Cd concentration of clone BH5 in the combined Cd and Zn treatment group was increased by 62% and 110%, respectively, relative of that of the Cd alone treatment group. On the other hand, the leaf and stem Zn concentration of clone BH8 in the combined Cd and Zn treatment group was reduced by 66% and 61%, respectively, relative to that of the Zn alone treatment group. Translocation of Cd and Zn from the root was higher in the leaf than in the stem, and the combined Cd and Zn treatment stimulated the translocation of Cd from the root to the leaf and stem, whereas it suppressed the translocation of Zn from the root to the leaf and stem. Therefore, the interaction effects were considered strongly synergistic with Cd in the presence of Zn, but proved antagonistic to Zn in the presence of Cd in the combined Cd and Zn treatment group. The phytoremediation potentials of the seven clones, which were estimated from standard indices of Cd and Zn concentration in Cd and Zn alone and the combined Cd and Zn treatment groups, were highest in clone BH3, and lowest in clone BH5. Therefore, we recognize S. caprea as an appropriate material for phytoremediation, and this is particularly the case with clone BH3. However, further research will be required to evaluate the effects of Cd and Zn on the physiological changes as well as tolerance mechanisms against metal toxicity in S. caprea clones.

Propagation by Leafy Stem Cuttings Containing Xylem of Populus alba × P. glandulosa Clone Bongwha1

  • The study was conducted to establish a method for the proliferation of hybrid poplar (P. alba × P. glandulosa) clone Bongwha1, an excellent biomass species. It was found that to collect the cuttings of Bonghwa1, it was necessary to use the main stem rather than the axillary branch. Stem growth by green-wood cuttings showed a tendency to decrease as the length of the collected cuttings increased, but the survival rate was low. Therefore, modified leafy stem cutting was attempted to increase the survival rate of the cuttings. In the modified leafy stem cutting method, 4 leaves were included in the cuttings, and especially, cuttings were performed using cuttings containing 2-4 cm xylem parts. Leafy stem cutting increased root growth and the number of stems, as well as the survival rate of hybrid poplar clone Bongwha1 compared to green-wood cuttings. The root growth of the leafy stem cutting poplar was better as there was more xylem part. Using two-year-old nursery stocks, the leafy stem cutting was used to produce about 66 cuttings. This study is expected to contribute to the mass propagation of high-quality nursery stocks.

Nutrients Contents in Different Parts of Pickly Pear(Opuntia humifusa) and Possible Anti-Breast Cancer Effect (천년초선인장의 부위별 영양성분 분석과 정상 및 암세포에 미치는 효과)

  • In order to examine the nutritional value of prickly pear(Opuntia humifusa), contents of ash, protein, fat, minerals and vitamins were determined on freeze-dried stem, fruit, seed and root from plants harvested in autumn. The average moisture contents for stem, fruit, seed and root were 67~87%. Crude ash content determined on dry weight basis was 2~3%. Crude protein existed mostly in seed(2.95%) and root(2.37%). Crude fat was detected mainly in seed(4.49%). Contents of major minerals(mg/100 mg dry weight) was generally higher in stem. Ca in stem(4,142.30) and fruit(2,790.86) were much higher than in seed(43.37). P in stem, seed and fruit were 448.19, 263.20 and 161.59, respectively. Stem also displayed more abundant Mg(1,110.86), Zn(35.62) and Mn(37.07). However, fruit contained higher amounts of Fe(13.38) and Se(0.15). Vitamin A was negligible in all plant parts. Vitamin E contents in fruit and stem were 1.78 mg and 1.22 mg/mg dry weight, respectively. Vitamin C was detected mostly in fruit(445.40) and stem(260.94). Use of a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide-based microtiter assay of cell viability demonstrated an anti-proliferative effect of O. humifusa extract on the MCF-7 estrogen-dependent human breast cancer cell line.

In vitro Formation of Tuberous Roots from Root Ends in the Rooted Tuberous stem without shoots in Cyclamen persicum MILL.

  • In Japan, propagation of cyclamen is mainly from seedlings. However, seeds are expensive and germination is slow and non..uniform. Therefore, to achieve genetically uniform propagation, multiplication must be vegetative. The rooted tuberous stems without shoots as sources of explants were cultured on the media containing BA and sucrose. After 30 days cultivation, tuberous roots were produced from the root ends attached to a tuberous stem and its capability was dependent on the type of media. The highest percentage of tuberous root formation was observed in Culture on the medium of 1/3 MS containing 0.05mgL$^{-1}$ NAA, 0.5mg L$^{-1}$ BA and 5% sucrose. Growth rates of the tuberous roots were greatly influenced by the cutting positions of a root in explants. The highest growth of was observed if small amount of root end was cut at initiation of tissue culture.

Effects of Antioxidative, DPPH Radical Scavenging Activity and Antithrombogenic by the Extract of Sancho (Zanthoxylum Schinilolium) (산초(Zanthoxylum Schinifolium) 뿌리, 줄기 및 잎 추출물의 항산화, DDPH Radical 소거 작용 및 항혈전 효과)

  • Effects of root, stem and leaf extract of sancho (Zanthoxylum schinifolium) on the inhibition of lipid peroxidation in the hepatic microsome of rat, DPPH radical scavenging activity and activated partial thromboplastin times (APTT) were examined in vitro. The highest inhibition of hepatic microsomal lipid peroxidation was observed by ethyl acetate fraction than that of methylene chloride fraction of the root and stem extracts. The high inhibition of lipid peroxidation was determined in the leaf, the root and the stem in order. The DPPH radical scavenging activity of ethyl acetate fraction was higher than that of n-butanol fraction and it was similar to the root and the steam extract. It was similar to the inhibition of hepatic microsomal lipid peroxidation. The DPPH radical scavenging activity was the highest in 2.500mg/mL of ethyl acetate fraction and it was 4.4 fold higher than that of $\alpha-tocopherol$, as an antioxidant standard. The DPPH radical scavenging activity was dependent on the extract concentration in the range of 0.125-5.000 mg/mL. The throm-boplastin times were higher than that of n-butanol fraction and it was similar to the root and the steam extracts. The leaf extract showed the highest antithrombogenic effect followed by the stem and then the root extract. The activated partial thromboplastin times were ependent on the extract concentration in the range of 0.100-2.000 mg/mL. Consequently, the effects of antioxidative, DPPH radical scavenging activity and antithrombogenic of Z. schinifolium was observed due to the inhibition of lipid peroxidation and the DPPH radical scavenging activity by methylene chloride, n-butanol and ethyl acetate fraction of the leaf extract. Studies on the Root Characteristics of Soybean Varieties in Korea (한국 대두 품종의 뿌리특성 연구)

  • Root is supposed to be important role in growth and yield formation of soybeans and also there is expected to exist varietal differences between soybean root morphology and system. 30 soybean varieties including 10 Korean recommended, 10 Introduced and 10 Korean native varieties were grown at field and pot under standard culture condition. Top and root characteristics were investigated at V4, R1 and R5 stage and T/R relations, correlations with yield were calculated. Top characteristics showed significant difference between 3 variety groups of Korean recommended, Introduced and Korean native variety. Stem length at R1 stage and stem length, stem diameter, number of nodes, number of branches, number of pods and grain yield at R5 stage were significantly different top characteristics. Root characteristics showed significant difference between 3 variety groups were number of thick primary root at V4 stage, primary root angle and root cutting strength at R1 stage, thick root diameter and root dry weight at R5 stage. The correlations between top and root characteristics and soybean yield was greatly different by soybean variety groups, and the top characteristics was more highly correlated then root characteristics. All the top and root characteristics of Korean recommended variety showed no correlations with yield. However, leaf area, stem length, top dry weight in Introduced variety and leaf area, top dry weight, number of nodes, number of thick primary root in Korean native variety were highly correlated with soybean yield.

Efficacy of Soil Amendment with Medicinal Plant Materials for the Control of Root-knot Nematode (Meloidogyne incognita) in Tomato

  • Soil amendments with oriental herbal medicines such as fruit of Anethum graveolens, flower buds of Syzygium aromaticum, rhizome of Cnidium officinale, rhizome of Coptis chinensis, root bark of Paeonia suffructicosa, stem bark of phellodendron amurense, and stem bark of Cinnamomum cassia at the rate of 0.2% (weight by volume of soil) significantly reduced Meloidogyne incognita infection (root gall formation) of tomato seedlings compared with the control. The most effective treatments were root bark of p. suffructicosa and stem bark of C. cassia as they gave minimum numbers of galls on tomato roots (4.7% and 8.9%, respectively, relative to control) as compared to other treatments. Another study with root bark of p. suffructicosa and C. cassia at different application doses also showed consistent results in reducing gall number. The control efficacy decreased as the application doses were lower-ed, indicating their dose-dependent control activities. These treatments significantly enhanced aboveground plant growths (total masses).
