Primer RNA Synthesis by E. coli RNA Polymerase on the SSB-coated 229-nt ssi Signal of Lactococcal Plasmid pGKV21 (Lactococcal plasmid pGKV21의 SSB-coated 229-nt ssi signal 상에서 E. coli RNA polymerase에 의한 시발체 RNA 합성)

  • Plasmid pGKV21 contains a 229-nucleotide (nt) single-strand DNA initiation (ssi) signal. Using asymmetric PCR, we prepared a small single-stranded (ss) DNA fragment of the ssi signal and, using the 229-nt ssDNA fragment, determined the requirements of RNA polymerase for priming and DNA-protein interaction. The ssi fragment prepared was able to generate primer RNAs with almost the same efficiency as the $M13{\Delta}lac182/229$ phage DNA. However, the cssi (complementary strand of the ssi signal) fragment could not synthesize primer RNAs. This result suggests that the 229-nt ssi signal functions in a strand specific manner. Gel retardation and DNase I footprinting demonstrated that the synthesized ssi fragment could interact with both E. coli RNA polymerase and SSB protein to synthesize primer RNA. In Escherichia coli [pWVAp], an addition of rifampicin resulted in an accumulation of ssDNA, indicating that the host-encoded RNA polymerase is involved in the conversion of ssDNA to double-stranded plasmid DNA.

Evaluation of the Response of BRM Analysis with Spring-Damper Absorbing Boundary Condition according to Modeling Extent of FE Region for the Nonlinear SSI Analysis (비선형 SSI 해석을 위해 Spring-Damper 에너지 흡수경계조건을 적용한 BRM의 유한요소 모델링 범위에 따른 응답평가)

  • The boundary reaction method(BRM) is a substructure time domain method, it removes global iterations between frequency and time domain analyses commonly required in the hybrid approaches, so that it operates as a two-step uncoupled method. The BRM offers a two-step method as follows: (1) the calculation of boundary reaction forces in the frequency domain on an interface of linear and nonlinear regions, (2) solving the wave radiation problem subjected to the boundary reaction forces in the time domain. In the time domain analysis, the near-field soil is modeled to simulate the wave radiation problem. This paper evaluates the performance of the BRM according to modeling extent of near-field soil for the nonlinear SSI analysis of base-isolated NPP structure. For this purpose, parametric studies are performed using equivalent linear SSI problems. The accuracy of the BRM solution is evaluated by comparing the BRM solution with that of conventional SSI seismic technique. The numerical results show that the soil condition affects the modeling range of near-field soil for the BRM analysis as well as the size of the basemat. Finally, the BRM is applied for the nonlinear SSI analysis of a base-isolated NPP structure to demonstrate the accuracy and effectiveness of the method.

Efficacy of Single-Dose Antimicrobial Prophylaxis for Preventing Surgical Site Infection in Radical Gastrectomy for Gastric Carcin

  • Purpose: Information regarding antimicrobial prophylaxis (AMP) for gastric cancer surgery is limited. The present study investigated the efficacy of single-dose AMP for the prevention of surgical site infection (SSI) in patients undergoing gastrectomy for gastric carcinoma. Materials and Methods: Between 2011 and 2013, 1,330 gastric carcinoma surgery patients were divided into two AMP administration groups depending on the duration of treatment. Postoperative outcomes including morbidity and SSI were compared between the two groups overall and in matched patients. Risk factors for SSI were analyzed. Results: The extended group (n=1,129) received AMP until postoperative day 1 and the single-dose group (n=201) received single-dose AMP only during an operation. Postoperatively, there were no significant differences between the two groups with respect to overall morbidity, mortality, or length of hospital stay. The SSI rate of the single-dose group was not significantly different from that of the extended group overall (4.5% vs. 5.5%, respectively, P=0.556) or in matched patients (4.5% vs. 4.0%, respectively, P=0.801). There was no increase in the SSI rate of the single-dose group compared to the extended group in subgroups based on different clinicopathological and operative factors. Univariate and multivariate analyses revealed male gender, open surgery, and operating time (${\geq}180$ minutes) as independent risk factors for SSI. Conclusions: Single-dose AMP showed no increase in the postoperative SSI rate compared to postoperative extended use in patients undergoing gastrectomy for gastric carcinoma. The efficacy of single-dose AMP requires further investigation in randomized clinical trials specific to gastric cancer surgery.

A New Hybrid Method for Nonlinear Soil-Structure Interaction Analysis (비선형 지반-구조물 상호작용해석을 위한 새로운 복합법)

  • This paper presents a novel hybrid time-frequency-domain method for nonlinear soil-structure interaction(SSI) analysis. It employs, in a practical manner, a computer code for equivalent linear SSI analysis and a general-purpose nonlinear finite element program. The proposed method first (calculates dynamic responses on a truncated finite element boundary utilizing an equivalent linear SSI program in the frequency domain. Then, a general purpose nonlinear finite element program is employed to analyze the nonlinear SSI problem in the time domain, in which boundary conditions at the truncated boundary are imposed with the responses calculated in the previous frequency domain SSI analysis, In order to validate the proposed method, seismic response analyses are carried out for a 2-D underground subway station in a multi-layered half-space, For the analyses, a equivalent linear SSI code KIESSI-2D is coupled to ANSYS program. The numerical results indicate that the proposed methodology can be a viable solution for nonlinear SSI problems.

Secure De-identification and Data Sovereignty Management of Decentralized SSI using Restructured ZKP (재구성된 영지식 증명을 활용한 탈중앙형 자기 주권 신원의 안전한 비식별화 및 데이터 주권 관리)

  • Decentralized SSI(Self Sovereign Identity) has become an alternative to a new digital identity solution, but an efficient de-identification technique has not been proposed due to the unique algorithmic characteristics of data transactions. In this study, to ensure the decentralized operation of SSI, we propose a de-identification technique that does not remove identifiers by restructuring the verification results of ZKP (Zero Knowledge Proof) into a form that can be provided to the outside by the verifier. In addition, it is possible to provide restructured de-identification data without the consent of data subject by proposing the concept of differential sovereignty management for each entity participating in verification. As a result, the proposed model satisfies the domestic personal information protection law in a decnetralized SSI, in addition provides secure and efficient de-identification processing and sovereignty management.

Korean Middle School Students' Perceptions as Global Citizens of Socioscientific Issues (과학과 관련된 사회.윤리적 문제(SSI)의 맥락에 따른 중학생들의 인성적 태도와 가치관 분석)

  • This study investigates Korean middle school students' perceptions as global citizens (i.e. ecological worldview, social and moral compassion, and socioscientific accountability) of Socioscientific Issues (SSI). We developed questionnaires that consisted of 20 Likert-type items to gauge their preceptions of the three different SSI contexts (i.e. nuclear power generation, bio-technology, climate change), and administered them to 225 9th grade students in Seoul. The results revealed that participants showed relatively high scores for ecological worldview but scored low on social and moral compassion across the SSI contexts. In addition, participants presented much higher scores for ecological worldview and socioscientific accountability regarding the issues of climate change. The participant responses indicated that they perceived more inter-connectedness with the environment and felt the responsibility of promoting sustainable development more to prevent further devastation in the context of climate change compared to nuclear power generation or biotechnology.

Features of Using Smart Devices in Socioscientific Issues Debate Classes (과학관련 사회쟁점(SSI) 토론 수업에서 스마트 기기의 활용 방식과 수업의 특징)

  • In this study, we investigated the features of using smart devices in socioscientific issues (SSI) debate classes. Twenty-seven high school students in Gyeonggi-do participated in this study. The processes of the SSI debate classes with smart devices were classified into five stages: data searching, data sharing, summary and organization of data, debate in the classroom, and online debate. Then some features at each stage were identified. It was found that they collected self-directed and in-depth data while reflecting their knowledge and experience, and selected data which were easy to understand at the stage of data searching. The specificity and validity of the evidences gathered and the clarity of the sources were improved at the stage of data sharing, although they shared more data offline than online. They organized their data by using paper rather than smart devices at the stage of summary and organization of data. Most of them failed to use the summary in debate. They argued and refuted based on concrete grounds at the stage of debate in the classroom, which raised students' interest in debate and led students to participate actively. At the stage of online debate, they were able to rearrange and review the arguments of both sides, and the participation of the students who were passive in classroom debate was enhanced. Based on these results, we suggest some effective teaching methods for SSI debate classes using smart devices.

An Improved Validation Technique for the Temporal Discrepancy when Estimated Solar Surface Insolation Compare with Ground-based Pyranometer: MTSAT-1R Data use (표면도달일사량 검증 시 발생하는 시간 불일치 조정을 통한 정확한 일사량 검증: MTSAT-1R 자료 이용)

  • In this study, we estimate solar surface insolation (SSI) by using physical methods with MTSAT-1R data. SSI is regarded as crucial parameter when interpreting solar-earth energy system, climate change, and agricultural production predict application. Most of SSI estimation model mainly uses ground based-measurement such as pyranometer to tune the constructed model and to validate retrieved SSI data from optical channels. When compared estimated SSI with pyranometer measurements, there are some systemic differences between those instruments. The pyranometer data observed upward-looking hemispherical solid angle and distributed hourly measurements data which are averaged every 2 minute instantaneous observation. Whereas MTSAT-1R channels data are taken instantaneously images at fixed measurement time over scan area, and are pixel-based observation with a much smaller solid angle view. Those temporal discrepancies result from systemic differences can induce validation error. In this study, we adjust hour when estimate SSI to improve the retrieved accurate SSI.

Seismic Soil-Structure Interaction Analyses of LNG Storage Tanks Depending on Foundation Type (기초 형식에 따른 LNG 저장탱크의 지반-구조물 상호작용을 고려한 지진응답 분석)

  • In this study, the soil-structure interaction(SSI) effect on the seismic response of LNG storage tanks was investigated according to the type of foundation. For this purpose, a typical of LNG storage tank with a diameter of 71m, which is constructed on a 30m thick clay layer over bedrock was selected, and nonlinearity of the soil was taken into account by the equivalent linearization method. Four different types of foundations including shallow foundation, piled raft foundation, and pile foundations(surface and floating types) were considered. In addition, the effect of soil compaction in group piles on seismic response of the tank was investigated. The KIESSI-3D, which is a SSI analysis package in the frequency domain, was used for the SSI analysis. Stresses in the outer tank, and base shear and overturning moment in the inner tank were calculated. From the comparisons, the following conclusions could be made: (1) Conventional fixed base seismic responses of outer tank and inner tank can be much larger than those of considering the SSI effect; (2) The influence of SSI on the dynamic response of the inner tank and the outer tank depends on the foundation types; and (3) Change in the seismic response of the structure by soil compaction in the piled raft foundation is about 10% and its effect is not negligible in the seismic design of the structure.

The Impact of Socio-Scientific Issue Debate about Local Environmental Problem on High School Students' Environmental Perception Change (지역환경문제에 관한 사회과학쟁점 토론이 고등학교 학생들의 환경인식 변화에 미치는 영향)

  • This study investigates the effect of SSI debate on first-year high school student's opinions about environmental issue, their judgment grounds, and solutions to regional environmental problems. The SSI debate was about white heron habitats near the village where students live. As the main data of the study, environmental perception questionnaires, and students' workbook including open-ended questions were collected before and after class. The environmental perception questionnaire was analyzed by descriptive statistics, and the response of the open-ended questions was analyzed through inductive qualitative research methods. First, the results of this study shows that the SSI debate has a statistically significant impact on students' environmental attitude. Second, a majority of students agreed on the idea that villagers should drive the birds out of town and they did not change their after the discussion class. However, after the discussion class, students' solutions about the issue were changed in a way that more short-term, feasible, concrete, and less time-consuming solutions to the problem. Based on the results of this study, this study implies that SSI issue debate using local problem should be used more often in science classroom so the students recognize local SSI and improve real world problem solving skills.
