Occurrence of Powdery Mildew on Tomato Caused by Oidiopsis taurica (L v.) Arnaud (=Leveillula taurica) in Korea (Oidiopsis taurica (L v.) Arnaud (=Leveillula taurica)에 의한 토마토 흰가루병 발생)

  • Yellow spot or blotch symptoms on the upper surface of leaf, without the production of velvet-like fungi on the lower surface of leaf as in the gray mold of tomato caused by Cladosporium fulvum, were observed in tomato (cv. Seokwang) plants in May, 1995, in a vinyl-house of the experimental plot of Gyeongnam Provincial Rural Development Administration, Chinju, Gyeongnam, Korea. We identified this disease as powdery mildew of tomato caused by Oidiopsis taurica (L v.) Arnaud (=Leveillula taurica), which was new to Korea. Conidia of the fungus were borne on uni- or bi-septated conidiophores which were developed through the stomata of the tomato leaf. The conidia were slender, clavate and variable in size (31~111.6$\times$13.1 ${\mu}{\textrm}{m}$). The fungal conidia isolated from tomato leaves were inoculated to tomato plants, and the occurrence of the same disease was confirmed based on the symptomatology and the morphology of the pathogen reisolated.

  • 1. The pathogenic fungi which infect to shoot of peony was 7 species. The infected parts were as followed, leaf spot disease was in leaf, leaf blotch was stem, powdery mildew was whole aeri­al part of both leaf and stem. 2. The major diseases were leaf spot and powdery mildew, and begin to occur from late April to mid­dle May, and then rapidly heavy infected. The 'Euiseongpeony' variety was more susceptible to those diseases than the 'Yeongcheonpeony' variety. 3. The blight of top part by seasonal in 1995 was begun at late May and increased rapidly 50.6% in Early August.

Two Unrecorded Species of the Snapper (Perciformes: Lutjanidae) Collected from Jeju Island, Korea

  • Two unrecorded species of the snapper, Lutjanus malabaricus (296.0 mm standard length [SL]) and L. stellatus (350.0 mm SL) belonging to the family Lutjanidae, were first collected from the western coastal waters of Jeju Island, Korea. L. malabaricus is characterized by having a dark marking on the upper half of the caudal peduncle, a band of vomerine teeth that does not protrude posteriorly at the middle, and nine anal soft rays. Compared to its Korean relative, L. malabaricus is distinguishable by having rows of scales that run obliquely in the dorsal-posterior direction above the lateral line (vs. parallel to the lateral line in L. argentimaculatus). L. stellatus can be identified by the absence of wavy blue lines on the head (vs. many blue lines in L. rivulatus) and presence of a white spot above the lateral line (vs. a black blotch on the lateral line in L. rivulatus). We propose new Korean names, "Jin-hong-tung-dom" and "Huin-jeom-tung-dom," for L. malabaricus and L. stellatus, respectively.

Identification of a Cupin Protein Gene Responsible for Pathogenicity, Phage Susceptibility and LPS Synthesis of Acidovorax citrulli

  • Bacteriophages infecting Acidovorax citrulli, the causal agent of bacterial fruit blotch, have been proven to be effective for the prevention and control of this disease. However, the occurrence of bacteriophage-resistant bacteria is one of hurdles in phage biocontrol and the understanding of phage resistance in this bacterium is an essential step. In this study, we aim to investigate possible phage resistance of A. citrulli and relationship between phage resistance and pathogenicity, and to isolate and characterize the genes involved in these phenomena. A phage-resistant and less-virulent mutant named as AC-17-G1 was isolated among 3,264 A. citrulli Tn5 mutants through serial spot assays and plaque assays followed by pathogenicity test using seed coating method. The mutant has the integrated Tn5 in the middle of a cupin protein gene. This mutant recovered its pathogenicity and phage sensitivity by complementation with corresponding wild-type gene. Site-directed mutation of this gene from wild-type by CRISPR/Cas9 system resulted in the loss of pathogenicity and acquisition of phage resistance. The growth of AC-17-G1 in King's B medium was much less than the wild-type, but the growth turned into normal in the medium supplemented with D-mannose 6-phosphate or D-fructose 6-phosphate indicating the cupin protein functions as a phosphomannos isomerase. Sodium dodecyl sulfa analysis of lipopolysaccharide (LPS) extracted from the mutant was smaller than that from wild-type. All these data suggest that the cupin protein is a phosphomannos isomerase involved in LPS synthesis, and LPS is an important determinant of pathogenicity and phage susceptibility of A. citrulli.

Selection of Crabapple Pollinizers for 'Fuji' Apple through Physiological and Genetic Analysis (꽃사과 품종의 생리 및 유전적 분석을 통한 '후지' 사과의 수분수 선발)

  • We investigated characteristics and self-incompatibility genotypes of 11 crabapple cultivars to introduce a new pollinizer of 'Fuji' apple tree in Korea. Flowering dates of eleven crabapples were two to seven days earlier than that of 'Fuji'. The rate of pollen germination in vitro was ranged from 85.6% to 98.0% except 'Virginia'. Controlled pollination treatment with each crabapples to 'Fuji' increased fruit set rate about 20.4% to 34.4%, the number of seed per fruit about 13.8% to 42.3% and fruit weight about 7.4% to 16.7% compared to open pollination. Tested crabapples were resistant to peach fruit moth, brown leaf spot and sooty blotch in general. A PCR amplification method using S-RNase primers was carry out in eleven crabapples. S-alleles, $S_3$, $S_5$, $S_9$, $S_{10}$, $S_{20}$, $S_{26} from six crabapples were determinated. Through sequencing analysis, $S_5$ ('Manchurian', 'Virginia') and $S_9$ ('Yantaishagou') showed 100% homologous to previous result. Based on our results, it was recommended that 'Manchurian', 'Hopa A', 'Hanyaehanakaidou', 'Spectabilis' could be promising pollinzers for 'Fuji' apple cultivar.

A New Short Stem, Lodging Resistance and High Yielding Peanut Variety "Pungan" (단경 내도복 다수성 땅콩 "풍안")

  • A new peanut variety "Pungan"(Arachis hypogaea ssp. hypogaea L.) was developed at the Department of Functional Crop, NICS(National Institute of Crop Science), in Milyang in 2008. It was developed from the cross between the high-yielding cultivar "Saedl" and the very short stem cultivar "Satonoka". "Pungan", a Virginia palnt type, has 20 branches with long ellipseshaped large grains. The 100 grain weight was 88 g, 7 g heavier than the check variety Daekwang. This variety is more resistant to late leaf spot and web blotch compared to Daekwang, a check variety. Moreover, it is lodging resistant owing to its short stem. "Pungsan" outyielded the check variety by 16% with 4.67 MT/ha in regional yield trials.
