Apoptotic Cell Death of Mouse Splenocytes by Polychlorinated Biphenyls and Its Prevention by Serum

  • Cell death induced by polychlorinated biphenyls (PCBs), environmental toxicant, was investigated in mouse splenocytes. The fragmentation of intact DNA, a parameter of apoptotic cell death, was evaluated qualitatively by agarose gel electrophoresis analysis and quantitatively by diphenylamine reaction method. PCBs induced apoptotic cell death of splenocytes in a dose- and time- dependent manner. The effect of serum on the apoptotic cell death induced by PCBs was also investigated. The DNA fragmentation induced by PCB treatment in serum-free medium was clearly inhibited by an addition of serum to the culture medium. The decrease of DNA fragmentation due to serum addition was accompanied with the increase of cell viability.

  • The purpose of this study was to investigate the effects of Kami boatang(KBT) on the immune cells in Balb/c mice. KBT (500mg/kg) was administerd p.o. once a day for 7 days. KBT enhanced the proliferation of thymocytes and splenocytes. KBT enhanced the subpopulation of helper T(Th) cells, but did not affect the subpopulation of Thyl/B220 cells and Th/Tc cells in splenocytes. KBT enhanced the production of ${\gamma}$-interferon and interleukin-2 in thymocytes, but decreased the production of interleukin-4. KBT enhanced the production of ${\gamma}$-interferon, interleukin-2 and interleukin-4 in splenocytes and serum. KBT suppressed the production of nitric oxide, and enhanced the phagocytic activity in peritoneal macrophages. These results suggest that KBT has a potent activity on the immune response via the increase of the production of cytokines and phagocytic activity in vivo.

  • Pumpkin sweet potato (Ipomoea batas L.) has been known as a traditional remedy and food source, not only in South Korea but worldwide. It is rich in fiber, potassium, vitamin C, and other minerals and vitamins, making it a nutritional food loved by many. showed that pumpkin sweet potato had antioxidant biological effects. The in vitro study showed that both splenocytes and cytokine production byactivated peritoneal macrophages increased when water extracts were supplemented at 100 and 250 μL/mL. Notably, the production of IL-1β, TNF-α, and IFN-γ by splenocytes was significantly increased at 100 μL/mL. The results suggest that supplementation with pumpkin sweet potato (Ipomoea batas L.) water extract may enhance immune function by stimulating splenocyte proliferation and improving cytokine production, activating macrophages in vitro.

  • Chaga mushroom (Inonotus obliquus) has been identified to have various biological activities. This study was performed to investigate the potential of chaga mushroom as a immunomodulating functional food. When mouse splenocytes were exposed to various concentration of hot water extracts of chaga mushroom (IOE) with mitogens (Con A, LPS), splenocytes proliferation was significantly increased. Also, IFN-$\gamma$ and IL-4 levels were significantly enhanced. Therefore, our results suggest that chaga mushroom may have the potential of being an immunomodulating functional food.

Immunomodulating Activity of BL18 (Ganshu) Acupuncture on the Experimental Liver Metastasis Model of Mice

  • We investigated that the immunomodulating activity of BL18 (Ganshu) acupuncture on the experimental liver metastasis model of mice. NA (non-acupoint)- and BL18-treatment enhanced the mitogenic activity of BALB/c whole splenocytes induced by various mitogenic stimuli. Acupuncture treatment tended to increase splenocytes differentiation even though did not show significance. Acupuncture treatment caused a marked increase of production of Th1 cytokine (IFN-${\gamma}$) and Th2 cytokine (IL-4) by splenocytes and IL-12 and IFN-Y by macrophages. The increase of cytokine production on BL18-treated group was more pronounced compared to NA-treated group. The liver weight of NA- and BL18-treated group decreased compared to tumor group, but did not showed significant differences.

  • Platycodon grandiflorum have been used as a traditional remedy and food source. This study was performed to investigate the immunomodulating effects of Platycodon grandiflorum in mouse, using ex vivo experiments. Six to seven-week old mice were fed ad libitum on a chow diet, and water extract of Platycodon grandiflorum was orally administrated at two different concentractions (50 and 500 mg/kg B.W./day) every other day for four weeks. In ex vivo experiments, the highest proliferation of splenocytes and levels of cytokine (IL-$1{\beta}$, IL-6, TNF-${\alpha}$) production were observed in 500 mg/kg BW/day supplementation group for all three cytokines stimulated by LPS. In conclusion, this study suggests that Platycodon grandiflorum extracts may enhance the immune function by regulating the splenocytes proliferation and cytokine production capacity by activating macrophages in mice.

  • The purpose of this research was to investigate the effects of Seungyangikkitang (SIT) on the immune cells in BALB/c mice. SIT (500mg/kg) was administerd p.o. once a day for 7 days. SIT enhanced the proliferation of thymocytes, but decreased the proliferation of splenocytes. SIT enhanced the subpopulation of cytotoxic T cells in thymocytes and helper T cells in splenocytes, but did not affect the subpopulation of B220/Thy1 cells. SIT enhanced the production of γ-interferon and interleukin-2 in thymocytes, splenocytes and serum, but did not affect the production of interleukin-4. SIT suppressed the production of nitric oxide, but enhanced the lucigenin chemiluminescence and the engulfment of FITC-conjugated E. coli particles in peritoneal macrophages. These results suggest that SIT has a potent activity on the specific immunity via the cytokine secretion of Th1 cells and the non-specific immunity via the phagocytic activity of macrophages in vivo.

  • Astragalus membranaceus Bunge (黃?), Atractylodes macrocephala Loidz (白朮), Angelica sinensis(Oliv) Kiels (當歸), Lycium chinense Mill (枸杞子), Ligustrum lucidum Ail (女貞子) are Herbs that are frequently used in a lot of prescriptions to reduce the side effects of anti-cancer therapies, especially like chemotherapy and radiotherapy. The radioprotective and hemopoietic effects of these Herbs on BALB/c splenocytes and bone marrow cells are measured. In order to evaluate the Hemopoietic effects, Thymidine uptakes and secretion of colony stimulating factors(CSFs) of splenocytes and myelocytes treated with herbs were measured. Radioprotective effects were accessed by the method of immunocompetence of murine lymphocytes cultured with herbs before irradiation and with LPS, ConA after irradiation. The results are as follows. 1. The orders of Proliferative effects of herbs on splenocytes were Atractylodes macrocephala Loidz (白朮), the higest of all, Astragalus membranaceus Bunge (黃?), Angelica sinensis(Oliv) Kiels (當歸). Lycium chinense Mill (枸杞子) and Ligustrum lucidum Ait (女貞子), the lowest of all. At Optimal concentration, the proliferation ratios of herb-treated splenocytes compared to non-treated ones were like these. Atractylodes macrocephala Loidz (白朮) 44.3, Astragalus membranaceus Bunge (黃?) 17.7, Angelica sinensis(Oliv) Kiels (當歸) 10, Lycium chinense Mill (枸杞子) 6.4, Ligustrum lucidum Ait (女貞子) 2.0.(p<0.05) 2. When splenocytes were cultured during different periods, Atractylodes. macrocephala Loidz (白朮) and Astragalus membranaceus Bunge (黃?) showed the higest proliferation on 3th day, Angelica sinensis(Oliv) Kiels (當歸), Lycium chinense Mill (枸杞子) on 4th day,(p<0.05) and Ligustrum lucidum Ait (女貞子) until 5th day but with no significant increase. 3. The orders of Proliferative effects of herbs on Bone Marrow(BM) cells were Atractylodes macrocephala Loidz (白朮), the higest of all, Astragalus memhranaceus Bunge (黃?), Angelica sinensis(Oliv) Kiels (當歸), Lycium chinense Mill (枸杞子) and Ligustrum lucidum Ait (女貞子), the lowest of all too. At Optimal concentration, the proliferation ratios of herb-treated BM cells compared to non-treated ones were like these. Atractylodes macrocephala Loidz (白朮) 21.7, Astragalus membranaceus Bunge (黃?) 9.9, Angelica sinensis(Oliv) Kiels (當歸) 4.9, Lycium chinense Mill (枸杞子) 2.3, Ligustrum lucidum Ait (女貞子) 1.4(p<0.05). 4. The secretion ratio of colony stimulating factors(CSFs) of each herb-treated group, compared to control, was Atractylodes macrocephala Loidz (白朮) 9.4, Astragalus membranaceus Bunge (黃?) 9.0, Angelica sinensis(Oliv) Kiels (當歸) 4.4, Lycium chinense Mill (枸杞子) 3.8 (p<0.05) but no significant increase in Ligustrum lucidum Ait (女貞子). 5. The mitogen(ConA, LPS) stimulated-lymphocytes cultured with each herb before irradiation of 1-3 Gy showed more enhanced proliferation than control(p<0.05). When compared to each non-irradiated group of all groups, the orders of percentage increase of irradiated group were Atractylodes macrocephala Loidz (白朮), the higest of all, Astragalus membranaceus Bunge (黃?), Angelica sinensis(Oliv) Kiels (當歸), Ligustrum lucidum Ait (女貞子), Lycium chinense Mill (枸杞子). Each percentage showed significant enhancement compared to control group(p<0.05). According to the results, Atractylcdes macrocephala Loidz (白朮), Astragalus membranaceus Bunge (黃?) are suggested to be the most effective hemopoietic and radioprotective herbs, and Angelica sinensis(Oliv) Kiels (當歸), Lycium chinense Mill (枸杞子) the next, but Ligustrum lucidum Ait (女貞子) showed lower effects than expected.

Passive Immunity by Splenocyte Transfer against Amebic Meningoeneephalitis in Mice (세포에 의한 아메바성 수막뇌염에 대한 피동면역의 전달)

  • The role of passive cell-mediated transfer of immunity against primary amoebic meningoen- cephalitis(PAME) in mice was studied. Waegleria fowleri, ITMAP 359, were cultured in CGVS medium. The ICR mice used were six week-old males of average weight of 15 g. Immunization was done by three intraperitoneal injections of $1{\times}10^6$ N. fowleri trophozoites at the interval of one week. Splenocytes were obtained from normal and immune mice spleens, and Ix107 cells were administered intraperitoneally into mice 3 days before challenge infection. Mice were infected intranasally with $7{\times}10^4$ N. fowleri trophozoites in a $3{\;}{\mu}l$ suspension under secobarbiturate anesthesia. Transplants of normal or immune splenocytes seem to alter the pattern of the PAME level- opment. The splenocytcs transferred from immune mice reduced the mortality rate in the JV. fowleri infected mice, as compared with the mice transferred with the same number of normal splenocytes or without splenocyte, The blastogenic response of the splenocytes to both lipopoly- saccharide and concanavalin A was elevated on duty 7 after infection the mice transinoculated with immune splenocytes. The serum antibody titers in the mice transferred with immune spleno- cytes were increased gradually from day 7 up to day 20 after infections by mean of ELISA. It is suggested that the transfer of splenocytes from immuniged mice conferred immunity against N. fowleri infection.

  • Brazilin, the main constitutent of Caesalpinia sappan, was examined for its immunomodulating activities in normal CBA mice. Mitogen induced proliferation and production of ConA induced T-cell growth factors (TCGF) of splenocytes were significantly reduced in brazilin treated group, cmpared to control group. It was also found that suppressor activities of splenocytes in brazilin treated group was significantly increased compared to those in normal control group.
