The purpose of this research was to investigate the effects of Kami Kwiryong Tang (KKT) on the immune cells in BALB/c mice. KKT (500mg/kg) was administerd p.o. once a day for 7 days. KKT decreased the proliferation of thymocytes, but did not affect the proliferation of splenocytes. KKT enhanced the subpopulation of Th (CD4+CD8- single positive cells) cells in splenic T-lymphocytes, but decreased the subpopulation of Th cells in thymocytes. KKT enhanced the production of ${\gamma}$-interferon and interleukin-2, but did not affect the production of interleukin-4 in mice serum. KKT did not affect the production of nitric oxide, but enhanced the phagocytic activity in peritoneal macrophages. These results suggest that KKT is a potent prescription on immune response via the production of cytokines from splenic Th1 cells and the increase of phagocytic activity in vivo.

The purpose of this study is to prove the efficacy of KamiBohuh-tang(KBT) on immunoregulation and the possibility of KBT as an immunoadjuvant. KBT with solid feed was administered orally once a day for 7 days to an experimental group, a solution of salt and solid feed without KBT to a control group. After a week T cell, B cell, cytokines, nitric oxide and phagocytic activity are measured. KBT enhanced the proliferation of splenocytes and the subpopulation of Th cells in splenic T-Iymphocytes, but did not affect the proliferation of thymocytes. KBT decreased the subpopulation of T-Iymphocytes in splenocytes. KBT enhanced the production of interferon-γ. interleukin-2, interleukin-4 in mice serum and the phagocytic activity in peritoneal macrophages but it suppressed the production of nitric oxide. These results suggest that KBT is a potent prescription on immune response via the increase of the proliferation of splenocytes, the production of cytokines from splenic Th cells and the phagocytic activity in vivo.

The purpose of this research was to investigate the effects of the extracts of Glycyrrhiza uralensis (GE) and the combined extracts of Glycine max Merr. and Glycyrrhiza uralensis (GGE) on the activity of murine splenocytes and macrophages. GE and GGE were administered orally twice a day for 7 days at the dose of 500 mg/kg. GE decreased the viability of T- and B-lymphocytes in splenocytes, but GGE increased the viability of B-lymphocytes in splenocytes. GE increased the population of B-lymphocytes in splenocytes, but decreased the population of T-lymphocytes and splenic $CD4^+$ cells. Also, GGE decreased the population of B-lymphocytes in splenocytes, but increased the population of T-lymphocytes and splenic $CD4^+$ cells. Furthermore, GE and GGE enhanced the phagocytic activity of peritoneal macrophages and the production of nitric oxide. These results suggest that the regulative action of immune response of GGE is more potent than their of GE.
Objective : To investigate the effect of Samilshinkihwan(SISKW) on white rats with deteriorated immunity caused by methotrexate. Methods : The test articles were dosed once a day for 14 days by gastric gavage at a dosage 1000, 500 and 250mg/kg/10ml of SISKW from 2 days after last MTX-dosing, and changes in body weight, spleen weight, total blood leukocyte numbers, total lymphocyte numbers, B and T lymphocyte ratio, CD3+CD4+, CD3+/CD8+ and CD4+/CD8+ T lymphocyte ratio in the blood and spleen were measured. In addition, the serum interleukin (IL)-2 levels and the productivity of IL-2 of splenic cells were also demonstrated in this study. Results : The changes on body weight increased significantly in the 1000mg/kgof SISKW group. The changes on the spleen weight, the total blood leukocytenumbers, the total lymphocyte numbers in the blood and spleen, the ratio of T-cell in the blood and spleen and the ratio of CD3+CD4+ T-cell in spleen increased significantly in all SISKW groups as compared with the control group. The ratio of CD4+/CD8+ T-cells in blood increased significantly. The serum IL-2 levels and productivity of IL-2 of splenic cells increased significantly in 1000 and 500mg/kg SISKW groups as compared with the control group. Conclusions : Samilshinkihwanhas an effect of increasing immune responses, especially on cellular immune responses, in white rats with deteriorated immunity caused by methotrexate.
To determine effects of zinc on lipopolysaccharide (LPS)-induced production of proinflammatory cytokines and Iymphokines in tumor-bearing ICR mice, this study has been investigated. Zinc chloride (Zn) at doses of 1 mg/kg was administered orally 30 minutes before i.p. injection of LPS (8 mg/kg) 5 times for 7 days. LPS greatly increased tumor necrosis factor (TNF)-$\alpha$ and interleukin (IL)-1$\beta$, in both serum and splenic supernatants compared with those in controls. However Zn strongly decreased LPS-increased production of TNF-$\alpha$ and IL-1$\beta$ in spleenic supernatants compared with those in controls and insignificantly also reduced in serum. LPS insignificantly decreased IL-2 levels in spleenic supernatants compared with those in controls but significantly increased interferon (IFN)-${\gamma}$ levels. Zn didn't affect IL-2 production in splenic supernatants compared to controls but significantly enhanced the LPS-decreased production of IL-2. Zn significantly increased IFN-${\gamma}$ levels in splenic supernatants compared to controls and did not affect the LPS-increased production of IFN-${\gamma}$. These findings suggest that Zn may strongly attenuate the LPS-induced pathogenesis of proinflammatory cytokines in tumor-bearing state and significantly up-regulate the LPS-induced function of T cells to produce IL-2 with maintaining normally the LPS- increased levels of IFN-${\gamma}$.
In the previous report, we described the in vivo antitumor activity of PJ-4, a protein-polysaccharide fraction prepared from the culture filtrate of an insect-born fungus, Paecilomyces tenuipes DGUM 32001. In the present study, we elucidated the immunomodulating activity of PJ-4 on the BALB/c mouse splenic lymphocytes using flow cytometrical techniques. As a result, PJ-4 was found to stimulate the lymphocytes not only to form lymphoblasts but also to express CD25 (IL-2 receptor $\alpha$ chain) molecule, which is well known as a T cell activation marker. More interestingly, its T cell stimulatory activity was more strongly exerted on CD8$^{+}$ T cells than on CD4$^{+}$ T cells. All these data suggest that PJ-4 exerts its antitumor activity at least partly through stimulation of T cells which play major roles in the cell-mediated immune system.tem.

The purpose of this research was to investigate the effect of Sojagangqi-tang(SJGQT) on the immune cell activity. The addition of SJGQT enhanced the proliferation of cultured-mice splenocytes and thymocytes. Administration of SJGQT(250 mg/kg) accelerated the subpopulation of splenic T lymphocytes especially CD/sup 4+/-TH cells in BALB/c mice. But high concentration(500 mg/kg) of SJGQT decreased the splenic T, B lymphocytes and thymic Tc (CD/sup 8+/) lymphocytes. Oral administration of SJGQT(250 mg/kg) significantly enhanced the production of IFN-γ and IL-4 in mice serum. And also, the addition of SJGQT(100 ㎍/ml) inhibited the proliferation of cultured-Jurkat leukemia cells in vitro. These results suggest that SJGQT have a cellular immuno-modulatory effect and anti-cancer property action
Although there are many reports on the splenic (systemic) T cell response after Toxoptasma gondii infection, little information is available regarding the local T cell responses of peritoneal exudate cells (PEC) and gut intraepithelial Iymphocytes (IEL) following peroral infection with bradyzoites. Mice were infected with 40 cysts of the 76K strain of T. gondii, and then sacrificed at days 0, 1, 4, 7 and 10 postinfection (PI). The cellular composition and T cell responses of PEC and IEL were analyzed. The total number of PEC and IEL per mouse increased after infection, but the ratio of increase was higher in IEL. Lymphocytes were the major component of both PEC and IEL. The relative percentages of PEC macrophages and neutrophils/eosinophils increased signiflcantly at day 1 and 4 PI, whereas those of IEL did not change significantly. The percentage of PEC NK1.1 and ${\gamma\delta}T$ cells peaked at day 4 PI (p < 0.0001), and CD4 and $CD8{\alpha}T$ cells increased continuously after infection. The percentages of IEL $CD8{\alpha}$ and ${\gamma\delta}T$ cells decreased slightly at first, and then increased. CD4 and NK1.1 T cells of IEL did not change significantly after infection. $IFN-{\gamma}-producing$ PEC NK1.1 T cells increased significantly from day 1 PI, but the other T cell subsets produced $IFN-{\gamma}$ abundantly thereafter. The proportion of IEL $IFN-{\gamma}-producing$$CD8{\alpha}$ and ${\gamma\delta}T$ cells increased significantly after infection, while IEL NK1.1 T cells had similar $IFN-{\gamma}$ production patterns. Taken together, CD4 T cells were the major phenotype and the important $IFN-{\gamma}$ producing T cell subsets in PEC after oral infection with T. gondii whereas $CD8{\alpha}T$ cells had these roles in IEL. These results suggest that PEC and IEL comprise different cell differentials and T cell responses, and according to infection route these factors may contribute to the different cellular immune responses.
Toxoplasmn gonnii, an intracellular protozoan infecting many kinds of eukaryotic cells, has been used to experimentally infect macrophages, epithelial cells, fibroblasts, and various cancer cells, but rarely T and B Iymphocytes or granulocytes. The present study was performed to determine the susceptibility of murine (BALB/c or CBA) splenic T and B llrnphocytes, and granulocytes to infection trio T. gondii RH tachyzoites. The ultrastructure of the infected host cells was observed by TEM, and the degree of intracellular parasite proliferation was quantified using 3H-uracil uptake assay. At 24 hrs post-culture, the host cell cytoplasm was found to contain 1 or 2, or a maximum of 7-8 tachyzoites. Infected T Iymphocytes demonstrated a peripherally displaced nucleus, a parasitophorous vacuole enveloping the parasite, and an increased number of mitochondria. In B Iymphocytes infected with tachyzoites, RER was not well developed compared to uninfected B Iymphocytes. Uninfected granulocytes contained many electron dense granules, but T. gondii-infected granulocytes demonstrated a decreased number of granules. Based on the 3H-uracil uptake assay. the susceptibility of T and B Iymphocytes, and granulocytes, to infection with T. gonnii tachyzoites was fairly high irrespective of cell type and strain of mouse. This strongly suggests deterioration in the functioning of infected host immune cells.

To investigate the preventive effects of melatonin (MLT) on the immunotoxicity of cadmium acetate[Cd(AC)2] in ICR mice, Mlt(10,50mg/kg as cadmium) were orally administered to mice once a day (5:00, PM) for 28 consecutive days. Cadmium(Cd) test solution was also administered at 25mg/kg of cadmium through the same route 2hr after administration of MLT daily, Mice were immunized and challenged with sheep red blood cells (SRBC). Immune functions evaluated were delayed type hypersensitivity (DTH) response, mitogenic response, and flow cytometry analysis. The results of these studies were summarized as follows ; DTH response was abnormally increased in mice treated with Cd alone. DTH response was normally depressed in mice treated with Cd plus MLT along with the increase of MLT doses. The mitogenic response of splenic T cell to Con A and that of B cells to LPS was remarkably increased by MLT treatment as compared with treatment of Cd alone In case of CD 8+ cells, the slight increase was observed in MLT treatment. Splenic T cells and B cells were significantly increased by MLT treatment as compared with treatment with Cd alone. These results suggest that MLT has significant preventive effects on the immunotoxic status induced by Cd exposure.
