Influences of Hydrocortisone, DHEA, Estradiol and Testosterone on the Polyamine Metabolism of Mouse Brain, Kidney, Liver and Intestine (Glucocorticoid 및 성(性) Steroid 홀몬에 의한 뇌(腦) 및 복부내(腹部內) 장기(臟器)의 Polyamine 대사(代謝)의 변동(變動)에 관한 연구(硏究))

  • The bilateral castration of male mice was operated under light ether anesthesia, and the sham operated mice were considered as the uncastrated. The treatments of mice with the following steroids were started one hour after operation. Hydrocortisone 50 mg/kg (HC), dehydroepiandrosterone 250 mg/kg (DHEA), ${\beta}-estradiol$ 5 mg/kg (E2), and testosterone 20mg/kg (TS) were subcutaneously injected into male ICR mice at noon for four days. Animals were sacrificed in the next-morning (at 10-12 A.M.) after the last injection. The intestinal putrescine(PT) content was lower and the liver and intestinal spermine(SM) contents were higher in castrated mice(CM), comparing with those of uncastrated mice (UCM). The intestinal PT content of UCM was markedly increased HC. But all brain polyamines of CM were significantly decreased by it. And HC also increased the spermidine(SD) content of kidney and liver and the intestinal PT content in CM. E2 induced the marked increase of liver PT content with the moderate increase of renal SD in UCM. And E2 significantly increased the brain and liver PT contents and the all renal polyamine contents in CM. Both of DHEA and TS induced the increase of renal PT content in UCM, and they also induced the marked increases of all renal polyamines of CM. In addition, TS increased the brain SM of CM. These results suggest that the steroidal regulation mechanism of brain, kidney, liver, and intestine seems to be different from one another, and the renal activity of polyamine synthesis can be markedly enhanced by sex steroids.

Biogenic Amines Content in Commercial Korean Traditional Fermented Soybean Paste (시판 재래식 된장 중의 biogenic amines의 함량 조사)

  • Five commercial Korean fermented soybean pastes were purchased to investigate biogenic amines (BAs) levels, microbiological and chemical qualities in commercial Korean fermented soybean paste. Bacillus spp. were observed about $10^7{\sim}10^9$ CFU/g levels, and lactic acid bacteria were $10^2{\sim}10^3$ CFU/g or not detected in products. The pH values have ranged from 5.05 to 5.75, and amino nitrogen (AN) contents were $252.0{\sim}557.2$ mg% in showing different degrees of aging. Detected Biogenic amines were putrescine (PUT), cadaverine (CAD), tryptamine (TRP), spermidine (SPD), spermine (SPM), histamne (HIS), tyramine (TYR), and agmatine (AGM) in different levels by companies. There are some limitation found to explain the direct relationships between biogenic amines contents and microbiological or chemical qualities in commercial fermented soybean paste because of different manufacturing steps.

HPLC-based Analysis of Biogenic Amines in Aging-Cheese (HPLC를 이용한 숙성치즈로부터 바이오제닉 아민 분석법 개발)

  • Biogenic amines have been used as chemical indicators of fermented foods. So far, several chromatography methods have been developed to detect biogenic amines in foods. However, few methods have identified these compound in domestic cheese. We analyzed the biogenic amines (histamine dihydrochloride, tyramine hydrochloride, ${\beta}$-phenylethylamine hydrochloride, putrescine dihydrochloride, cadaverine, spermidine, tryptamine hydrochloride, ethanolamine hydrochloride and butylamine) in cheese by using HPLC. The calibration curves of the biogenic amines were found to be linear over the concentration range of 10-50 ppm with a correlation coefficient of above 0.99. The limit of detection (LOD) and limit of quantitation (LOQ) of the biogenic amines in the given order were 3.7 and 11.3 ppm, 3.4 and 10.4ppm, 3.4 and 10.3 ppm, 4.0 and 12.2 ppm, 3.4 and 10.4 ppm, 3.4 and 10.5 ppm, 3.5 and 10.7 ppm, 4.1 and 12.5 ppm, and 3.4 and 10.4 ppm, respectively. Recovery rates of the biogenic amines in the given order were 112, 104, 93, 108, 91, 102, 101, and 92%, respectively. The findings of this study suggest that HPLC is a suitable method for the determination of biogenic amines, thereby indicating its potential application in the quality control of aging cheese.

Characterization of Putrescine Uptake in Hamster Amelanocytic Melanoma AMEL-3 Cells

  • The uptake of putrescine, spermidine and spermine by Fortner's hamster amelanocytic melanoma AMEL-3 cells was observed in this study to be time-dependent, temperature-sensitive, pH-dependent and saturable. Metabolic poisons nullified polyamine uptake, an indication that this is an energy-requiring mechanism. The presence of $Na^+$ ions was found to be requisite to full activity. Valinomycin, gramicidin, monensin and the calcium ionophore calcimycin were also observed to inhibit the process substantially. The transporter active site would seem to contain sulfhydryl groups. Other diamines and polyamine analogues, as well as cationic diamidines, suppressed putrescine uptake. The presence of the ornithine decarboxylase inhibitor DFMO in the culture medium induced putrescine inflows. Putrescine, in turn, induced the negative expression of the carrier, thus suggesting that this influx mechanism is governed by up/down regulation. The cationic diamidine CGP 40215A and its analogue CGP039937A competitively inhibited putrescine transport, with Ki values of 1.9 and $15{\mu}M$, respectively. The role of polyamine uptake in these cultures is discussed.

Effect of Polyamines on Indigo Biosynthesis in Hairy Root Cultures of Polygonum tinctorium Lour. (Polyamine이 쪽 모상근배양에서 인디고 생합성에 미치는 영향)

  • We herein studied the effect of polyamines (putrescine, spermidine and spermine) on growth and indigo biosynthesis in hairy root cultures of Polygonum tinctorium Lour. Our results revealed that polyamine treatment increased hairy root growth and indigo biosynthesisat all tested concentrations, with the highest growth rate (4.4 g/ flask) and indigo yield (216 ug/g) induced by 70 mg/L putrescine. These results show far the first time that the growth rates and indigo biosynthesis of Polygonum tinctorium hairy roots may be improved by addition of polyamines to the liquid culture medium.

Putrescine Transport in a Cyanobacterium Synechocystis sp. PCC 6803

  • The transport of putrescine into a moderately salt tolerant cyanobacterium Synechocystis sp. PCC 6803 was characterized by measuring the uptake of radioactively-labeled putrescine. Putrescine transport showed saturation kinetics with an apparent $K_m$ of $92{\pm}10\;{\mu}M$ and $V_{max}$ of $0.33{\pm}0.05\;nmol/min/mg$ protein. The transport of putrescine was pH-dependent with highest activity at pH 7.0. Strong inhibition of putrescine transport was caused by spermine and spermidine whereas only slight inhibition was observed by the addition of various amino acids. These results suggest that the transport system in Synechocystis sp. PCC 6803 is highly specific for polyamines. Putrescine transport is energy-dependent as evidenced by the inhibition by various metabolic inhibitors and ionophores. Slow growth was observed in cells grown under salt stress. Addition of low concentration of putrescine could restore growth almost to the level observed in the absence of salt stress. Upshift of the external osmolality generated by either NaCl or sorbitol caused an increased putrescine transport with an optimum 2-fold increase at 20 mosmol/kg. The stimulation of putrescine transport mediated by osmotic upshift was abolished in chloramphenicol-treated cells, suggesting possible involvement of an inducible transport system.

Development of Immunological Methods for Analysis of 5' -deoxy-5' -methylthioadenosine

  • Studies were undertaken to develop a competitive radioimmunoassay (RIA) and indirect antigen capture enzyme-linked immunosorbent assay (ELISA) for the determination of 5'-deoxy-5'-methylthioadenosine (MTA), which is formed from decarboxylated S-adenosylmethionine by spermidine and spermine synthase. Specific antiserum against MTA was raised in rabbits by immunization with MTA-BSA which was prepared by coupling BSA to oxidized MTA with periodate. Since MTA is oxidized easily to the sulfoxide, the sulfhydryl reagent, DTT. was added to the immunogen. For RIA, immunocomplexes were separated from free MTA by using ammonium sulfate precipitation. The antiserum showed almost no cross-reactivity with a variety of other nucleotides and riboses. But, the level of cross-reactivity of 5'-isobutylthioadenosine (SIBA) was high. These results showed the importance of hydrophobicity adjacent to the 5'-OH for determining antigenicity. The lower limit of detection by this assay was 100 fmol of MTA per tube. Using this assay. MTA levels were more easily and precisely determined in biological samples when compared with HPLC analysis. The RIA procedure is less time consuming. More than 24 analyses can be carried out in 2 h and required only a very small amount of sample ($20{\mu}l$ serum). In ELISA, biotin conjugated MTA-BSA was used as the labelled MTA. The sensitivity limit of this assay was lower than 100 pmol.

Strontium Stimulates IAA Oxidation and Polyamine Synthesis in Germinating Mung Bean Hypocotyls (Vigna radiata L.) (숙주 배축생장과정 중 스트론티움에 의한 오옥신산화 및 폴리아민 생합성 촉진)

  • Mung beans (Vigna radiata L.) were used to investigate the roles of strontium in hypocolyl elongation under IAA regime during the germination. After imbibition in a medium with or without IAA, $Sr^{2+}$ stimulated IAA oxidation. Three to five fold increasing in IAA oxidase activity seems to be direct evidence of growth inhibition through $Sr^{2+}$. Furthermore, the accumulation of spermidinc and spermine by $Sr^{2+}$ in the range of 1 to 10 mM was observed. Spermidine levels were 2 to 3 fold higher than in control seedling grown without strontium. The increase in polyamine levels was observed on a g fresh weight basis. In conclusion, it was demonstrated that the inhibitory action of $Sr^{2+}$ is closely related with the IAA oxidation and polyamine biosynthesis.

Effects of Inhibitors (DFMO, DFMA) on Polyamine Synthetic Enzymes (ADC, ODC) during Ovarian Development of the Tobacco Budworm, Helicoverpa assulta (담배나방의 난소발생시 폴리아민 합성효소에 미치는 저해제의 효과)

  • Effects of $\alpha$-difluoromethylornithine (DFMO) and $\alpha$-difluoromethylarginine (DFMA), inhibitors of polyamine syntheic enzymes (ornithine decarboxylase, arginine decarboxylase), on ovary were investigated during pupal-adult development of Helicoverpa assulta. Two inhibitors (DFMO, DFMA) showed definite inhibition effects on ovarian development. The inhibition effect on ovaries weight was more marked in DFMA-injection than that observed in DFMO-injection. Two inhibitors (DFMO, DFMA) gave rise to a peculiar decrease in ornithine decarboxylase (ODC) or arginine decarboxylase (ADC) activity in ovaries, 72 hrs (5-day old pupa) post injection. However, DFMO clearly exhibited supression of ODC activity after 96 hrs (6-day old pupa). In addition, two inhibitors (DFMO, DFMA) diminished putrescine content in the ovary. The spermidine level was slightly decreased by each injection of the inhibitors. However, two inhibitors (DFMO, DFMA) raised the spermine content at certain developmental stages in the ovary. Although the effect of DFMA was less severe, two inhibitors (DFMO, DFMA) caused not only an overall delay in ovarian development, but also abnormalities in cellular differentiation. Noted effects in the pupal ovary were the appearance of irregular nurse cells and partial destruction of follicle epidermal cells. Adult ovary showed rapid degradation of nurse cells, a reduction in the number of follicle epidermal cells and immature oocytes that had a low yolk content.

Effects of polyamines on hydrogen peroxide-scavenging enzymes in radish seedling plants under paraquat stress (Paraquat 스트레스를 받는 무 (Raphanus sativus L) 유식물에서 H2O2 분해 효소에 대한 폴리아민의 효과)

  • Application of exogenous polyamines (PAs) reduced the paraquat (PQ)-induced cotyledon injuries in radish seedling plants with 1 mM spermidine (Spd) being the most effective protectant. PQ injury symptoms in the cotyledons, e.g., large accumulation of $H_2O_2$, and losses of fresh weight, chlorophyll, and proteins, were significantly alleviated. Likewise, analysis of $H_2O_2$-scavenging enzymes such as catalase (CAT) and guaiacol peroxidase (GPX) showed that pretreatment with Spd among PAs remarkably increased total CAT activity and strongly retarded PQ-induced rapid decline in total GPX activity. In a native gel assay, one CAT isozyme (CAT1) and two GPX isozymes (GPX1 and a newly synthesized GPX isozyme) proved to be more responsible for PQ tolerance, as manifested by the strong increases in their activities by Spd pretreatment. Based on these results, we can suggest that PAs (especially 1 mM Spd) may function as antioxidant protectors by invoking CAT and GPX enzymes which control the endogenous $H_2O_2$ level in radish cotyledons exposed to PQ.
