Study on the Preservation of Liquid Boar Semen at Uncontrolled Room Temperature (조절되지 않은 실온에서의 돼지액상정액 보존에 관한 연구)

  • This study was done to find out the preservation possibility of liquid boar semen at variabel room temperature of 9 to 16$^{\circ}C$. The percentages of sperm motility and NAR acrosome were highest in B tschwiler extender compared to B tschwiler+Hepes, Andro+Hepes and Andro extenders. The extenders with Hepes buffer showed detrimental effect for preservation of liquid boar semen. The pH of ejaculated sperm-rich fraction was 7.5. The pH of B tschwiler+Hepes, B tschwiler, Andro+Hepes and Andro extenders was 6.9, 7.5, 7.1 and 8.1, respectively. The pH of liquid boar semen with B tschwiler+Hepes, B tschwiler, Andro+Hepes and Andro extenders was 6.6, 6.9, 6.7 and 6.9 at 1st day of storage, and 5.5, 5.7, 5.6 and 5.8 at 7th day of storage, respectively. Gilts and sows were inseminated twice with liquid boar semen stored at 9~16$^{\circ}C$ in B tschwiler extender for 3~4 days. Farrowing rate, litter size and average pig weight at birth between AI and natural service did not differ significantly in gilt and sow, respectively. However, sow showed higher farrowing rate and litter size compared to gilt both in AI and in natural service. As a result of this study, we found out that liquid boar semen can be stored for 5~7 days at uncontrolled room temperature of 9~16$^{\circ}C$ in B tschwiler extender.

Cold Storage of Milt from Four Species of Flatfish

  • The proper conditions for cold storage of milt in four species of flatfish were investigated. Various diluents were tested for the preservation of marbled sole, brown sole, starry flounder and olive flounder milt at $1\pm0.5^{\circ}C$ for 7 days. The most effective diluent was $1\%$ NaCl for marbled sole and Stein's extender for brown sole, starry flounder and olive flounder. After 7 days of storage in different dilution ratios (milt: diluent), marbled sole milt diluted with 1:3-10 and brown sole, starry flounder and olive flounder milt diluted 1:1-10 maintained good sperm activity index (SAI). SAI was affected by storage volume and presence of antibiotic. SAI was lower after 7 days storage in the volume of 1.5 mL than in 0.5 or 1.0 mL. SAI after 29 days storage was markedly enhanced by addition of antibiotic in marbled sole (400 ppm gentamicin, 400 ppm neomycin, 200-400 ppm streptomycin), brown sole (600 ppm gentamicin), starry flounder (1,000 ppm gentamicin) and olive flounder (200-1,000 ppm gentamicin, 200-400 ppm neomycin, 200-1,000 ppm streptomycin).

Effect of Thawing Temperature on Sperm Characteristics of Frozen Semen in Miniature Pig (미니 돼지 동결정액의 융해 온도가 정자성상에 미치는 영향)

  • The objective of this study was to investigate the effect of thawing temperature on the sperm viability and acrosomal morphology for semen storage of miniature pig by the 0.5ml straw method. In this present study, sperm viability (SYBR-14/PI staining), membrane integrity (Hypoosmotic Swelling Test), acrosome intactness, intensity and capacitation status (chlorotetracycline staining) in frozen miniature pig sperm were evaluated after thawing at 37, 50 and $70^{\circ}C$ for 5, 10 and 45 sec, respectively. Interestingly, the results indicated that sperm thawed at $70^{\circ}C$ for 5 sec significantly (p<0.05) increased sperm viability, but lower the percentage of AR (acrosome reacted spermatozoa) pattern compared to sperm thawed at $37^{\circ}C$ for 45 sec and $50^{\circ}C$ for 10 sec. In terms of thawing condition, high temperature for a short time using the 0.5ml straw was improved cryosurvival of miniature pig semen. Therefore, appropriate thawing method for cryopreservation of miniature pig is required for increasing post-thawing viability.

  • The gametogenesis of the surf clam, Tresus keenae, were investigated by SEM and TEM. Both the testis and the ovary had follicle tubes surrounded by inter-tubal tissue composed of adipogranular cells that provided storage function. In the vitellogenic oocyte, lipid droplets and lipid yolk granules were found in the vacuoles formed by the Golgi apparatus. Proteid yolk granules were formed by the endoplasmic reticulum and cortical granules in the cytoplasm during vitellogenesis. The mature sperm was primitive and resembled a jar with a cover. The sperm heads were approximately $2.00-2.30 {\cal}um$. The acrosomal rod projected in front of the acrosome. In addition, four large mitochondria were in the midpiece.

  • Artificial insemination technique has been contributed immensely for production of livestock worldwide as a critical assisted reproductive technique to preserve and propagate excellent genes in domestic animal industry. In the past decade, methods for semen preservation have been improved mostly in liquid preservation method for boar semen and freezing method for bull semen. Among many factors affecting semen quality during preservation, reactive oxygen species, produced by aerobic respiration in sperm for survival and motility, are unfavorable to sperm physiology. In mammalian cell as well as in the sperm, antioxidant system plays a role in degradation of reactive oxygen species. Magnetized water forms smaller stabilizing water clusters, resulting in high absorption and permeability of the cell for water, implicating its application for semen preservation. Therefore, this review focuses on preservation methods of boar and bull semen with respect to improvement of extender and reduction of reactive oxygen species by using magnetized water and supplementation of antioxidants.

Effect of Storage in Different Commercial Semen Extenders on the Motility, Viability and Fertility In Vitro of Boar Spermatozoa (수퇘지 정자의 운동성, 생존성 및 체외수정 능력에 대한 시판 액상 정액 보존액과 보존 기간의 영향)

  • The objective of this study was to determine the effect of semen extenders on the motility, viability and fertility in vitro of spermatozoa during storage of fresh boar semen diluted in different commercial extenders used for pig artificial insemination (AI). In this experiment, semen were diluted in Androhep plus, Beltsville Thawing Solution (BTS), Modena, Seminark and Vitasem LD. Five ejaculates were collected from three Duroc boars and sub-samples were diluted ($30{\times}10^6$ spermatozoa/ml) in different extenders. Semen was stored at $170^{\circ}C$ for 10 days. Sperm motility and viability was assessed using Computer-Assisted Semen Analysis (CASA) and flow-cytometry on 1, 3, 5 and 10 day post collection The motility of spermatozoa stored in different extenders was gradually decreased by increasing the duration of storage of semen. However, there was not significant1y different in the sperm motility and viability among other extenders. On the other hand, the in vitro-matured oocytes were fertilized and cultured in vitro to assess the fertility of boar spermatozoa stored for 3 and 10 days in different extenders. The percentage of morula and blastocyst were taken as indicators of fertility in vitro of spermatozoa. Therefore, there were no differences in the rate of embryos developed to the molular and blastocyst stage. There were no differences in the motility and fertility in vitro among 5 kinds of commercial boar semen extenders.

Preservation of Extended Thoroughbred Semen at Low Temperature (Thoroughbred 정액의 액상 보존에 관한 연구)

  • Equine semen was analysed for its survival after storage under different conditions. Total 12 ejaculates from 2 Thoroughbred were analysed for general characterisitcs and preservation at low temperature. The sperm fraction, concentration, nd the rate of motile spermatozoa were 57.91ml per ejaculate, 2.18$\times$108/ml and 74.1%, respectively. The survival rate of spermatozoa was highest when diluted semen with E-Z Mixin was stored at 7~8$^{\circ}C$. The optimum survival rate(>54%) can be obtained upto 24h at 7~8$^{\circ}C$. However only 10% spermatozoa survived after 5h storage at 7~8$^{\circ}C$ without use of E-Z Mixin. Other ranges of temperature(15$^{\circ}C$ and room temperature) gave less survival rates(<25%). These results indicate that the extender could be used as a basic solution for the preservation of equine spermatozoa at low temperature. It also provides a practical method for short-term storage of collected equine semen in a simple manner.

A Study on Development of Boar Semen Extender Kp for Swine AI I. Stabilization of pH Change and In Vitro Survival of Frozen-Thawed Boar Sperm in Kp Extender (돼지인공수정용 정액액상보존제 Kp 의 개발에 관한 연구 I. Kp 의 pH 조절과 냉동정자에 의한 보존성 검정)

  • Boar semen extender Kp (Hankook Life-Science, Korea) was newly formulated by authors. This study was carried out to investigate the optimal concentrations of EDTA, Tris and citrate buffers in the Kp extender (Basic Kp) on the pH change during storage. And then the motility of boar sperm with the Kp pH stabilized (Modified Kp) was compared with those of commercial products imported into Korea such as BTS (Mini-tube. Germany; BTSg), BTS (Tri-bio, USA; BTSa) and Modena (SGI, USA). The pH values of all extenders were increased gradually with the storage days. Especially, the initial pH of Basic Kp was higher than that of BTSg, BTSa and Modena, and also higher than physiological pH of boar sperm (6.8∼7.5). When Basic Kp was added with various concentrations of EDTA (0, 0.63, 1.25 & 2.37g/L), Tris (0, 0.18, 0.35, 0.71 & 1.42g/L) and Citrate (0, 0.75, 0.81, 1.00, 1.25 & 1.50g/L) buffers for pH down-regulation and stabilization of pH, the group added with 1.25g EDTA, 1.42g Tris and 1.00g Citrate well maintained the neutral range of pH during storage (6.88 at day1 to 7.33 at day6 in Modified Kp), Especially, the concentrations of the buffers added in Modified Kp were lower, until 1/2∼1/4 ranges, than those in Modena and other extenders. The motility of frozen-thawed boar sperm diluted with Modified Kp was significantly higher than that of Basic Kp, BTSg, BTSa and Modena (87.0% vs. 55.0∼71.0% at day1; 13.3% vs. 0∼6.3% at day6), Conclusively, Modified Kp in this experiment was kept the favorable physiological conditions in spite of low concentrations of the buffers and motility of frozen-thawed boar sperm was obtained better than that of Basic Kp and other commercial products such as BTSg, BTSa and Modena.

  • Remarkable progress has recently been made in embryo transfer technology, resulting in the birth of IVF and nuclear transfer offsprings in swine. However, further progress of the technology to (I) make a safe, effective and economic estrual-cycle synchronization compound, (2) regulate each step of sperm capacitation (3) induce monospermic fertilization, (4) in vitro grow and mature oocytes, (5) fertilize the oocytes efficently, (6) culture the oocytes to the blastocyst stage in defined media, (7) produce multiply copies of embryos with superior genetic merit, (8) preselect the sex of these superior offsprings, and (9) preserve embryos by freezing and storage in liquid nitrogen is required before this promising technology is applied routinely to swine for practical use.

Antioxidative Effect of Pyruvate, Taurine and Melatonin, and Relationships of the Evaluation Methods on Boar Sperm Viability for In Vitro Storage (돼지 정자의 체외조본에 있어 Pyruvate, Taurine 및 Melatonin의 항산화효과와 평가방법의 상관관계 분석)

  • The objective of this study was to investigate the anti-oxidative effects of pyruvate, taurine and melatonin on sperm characteristics(motility, membrane integrity) and lipid peroxidation(LPO) for in vitro storage of boar semen. Semen was treated with various antioxidants such as pyruvate(1mM), taurine(50mM) and melatonin(100nM) with or without 100uM H2O2. Antioxidant treatments were significantly increased the sperm motility when compare to control group in all incubation periods(P≤0.05). Hypoosmotic swelling test(HOST), membrane integrity was similar to the result of motility. In lipid peroxidation measurement by TBA reactions of spermatozoal plasma membrane, malondialdehyde(MDA) level in control and antioxidant treatments were lower than those of antioxidant plus H2O2 or H2O2 treatment for 3 to 6 h incubation period. Relationships of evaluation methods for sperm viability were investigated by motility, membrane integrity and lipid peroxidation. Among evaluation methods, LPO vs motility and membrane integrity vs LPO were negatively correlated(-0.23~-0.92 and -0.68~-0.85), but membrane integrity vs motility was positively correlated (0.53~0.94) in all treatments. These experiments indicate that supplementation of antioxidant to the semen extender can increase the sperm motility and membrane integrity and decrease the lipid peroxidation of spermatozoal plasma membrane. The HOST might be utilized to evaluate the sperm quality instead of lipid peroxidation or motility.
