The Influence of Glutaraldehyde Concentration on Electron Microscopic Multiple Immunostaining

  • The present study was aimed to evaluate the influence of glutaraldehyde (GA) concentration on multiple electron microscopic (EM) immunostaining using pre-embedding peroxidase and post-embedding immunogold method. Influence of various concentrations of GA included in the fixative on immuoreactivity was assessed in the multiple immunostaining using antisera against anti-transient receptor potential vanilloid 1 (TRPV1) for peroxidase staining and anti-GABA for immunogold labeling in the rat trigeminal caudal nucleus. Anti-TRPV1 antiserum had specificity in pre-embedding peroxidase staining when tissues were fixed with fixative containing paraformaldehyde (PFA) alone. Immunoreactivity for TRPV1 was specific in tissues fixed with fixative containing 0.5% GA at both perfusion and postfixation steps, though the immunoreactivity was weaker than in tissues fixed with fixative containing PFA alone. Tissues fixed with fixative containing 0.5% GA at the perfusion and postfixation steps showed specific immunogold staining for GABA. The results of the present study indicate that GA concentration is critical for immunoreactivity to antigens such as TRPV1 and GABA. This study also suggests that the appropriate GA concentration is 0.5% for multiple immunostaining with peroxidase labeling for TRPV1 and immunogold labeling for GABA.

Customers' perception of the attributes of different formats of menu labeling: a comparison between Korea and the U.S.

  • BACKGROUND/OBJECTIVES: This study compared the perception of customers from Korea and the U.S. on the attributes of different formats of menu labeling The specific objectives were 1) to compare the customers' perceived usefulness, ease-of-understanding, clarity, and attractiveness of different formats of menu labeling between Korea and the U.S.; and 2) to compare the customers' use intention to different formats of menu labeling between Korea and the U.S. SUBJECTS/METHODS: A survey was conducted in Korea and the U.S. The participants were allocated randomly to view 1 of the 7 restaurant menus that varied according to the following types of menu labeling formats: (type 1) kcal format, (type 2) traffic-light format, (type 3) percent daily intake (%DI) format, (type 4) kcal + traffic-light format, (type 5) kcal + %DI format, (type 6) traffic-light + %DI format, and (type 7) kcal + traffic-light + %DI format. A total of 279 Koreans and 347 Americans were entered in the analysis. An independent t-test and 1-way analysis of variance were performed. RESULTS: Koreans rated type 4 format (kcal + traffic light) the highest for usefulness and attractiveness. In contrast, Americans rated type 7 (kcal + traffic light + %DI) the highest for usefulness, ease-of-understanding, attractiveness, and clarity. Significant differences were found in the customers' perceived attributes to menu labeling between Korea and the U.S. Americans perceived higher for all the 4 attributes of menu labeling than Koreans. CONCLUSIONS: The study is unique in identifying the differences in the attributes of different formats of menu labeling between Korea and the U.S. Americans rated the most complicated type of menu labeling as the highest perception for the attributes, and showed a higher use intention of menu labeling than Koreans. This study contributes to academia and industry for practicing menu labeling in different countries using different formats.

A Study on the Recognition of Concrete Cracks using Fuzzy Single Layer Perceptron

  • In this paper, we proposed the recognition method that automatically extracts cracks from a surface image acquired by a digital camera and recognizes the directions (horizontal, vertical, -45 degree, and 45 degree) of cracks using the fuzzy single layer perceptron. We compensate an effect of light on a concrete surface image by applying the closing operation, which is one of the morphological techniques, extract the edges of cracks by Sobel masking, and binarize the image by applying the iterated binarization technique. Two times of noise reduction are applied to the binary image for effective noise elimination. After the specific regions of cracks are automatically extracted from the preprocessed image by applying Glassfire labeling algorithm to the extracted crack image, the cracks of the specific region are enlarged or reduced to $30{\times}30$ pixels and then used as input patterns to the fuzzy single layer perceptron. The experiments using concrete crack images showed that the cracks in the concrete crack images were effectively extracted and the fuzzy single layer perceptron was effective in the recognition of the extracted cracks directions.

  • This research aims to develop the multiple channel electrochemical DNA chip using microfabrication technology. At first, we fabricated a high integration type DNA chip array by lithography technology. Several probe DNAs consisting of thiol group at their 5-end were immobilized on the gold electrodes. Then target DNAs were hybridized and reacted. Cyclic voltammetry showed a difference between target DNA and control DNA in the anodic peak current values. Therefore, it is able to detect a plural genes electrochemically after immobilization of a plural probe DNA and hybridization of non-labeling target DNA on the electrodes simultaneously. It suggested that this DNA chip could recognize the sequence specific genes.

Understanding of Nutrition Labelling Use and Related Factors among Korean Adults

  • This study was conducted to investigate that the nutrition labeling use is associated with demographic and psychosocial factors according to each nutrition information on the nutrition labeling in Korean adults. The study subjects (N=1,140) were individuals who were aged 20 years and more and answered on the question of nutrition label use and who participated in the Korean National Health Examination and Nutrition Survey (KNHANES) in 2010.As age older, there was more interest in information such as sugar, protein, fat, cholesterol than calories. In contrast, as age younger, there was more interest intrans-fat, sodium as well as calories. As higher education level, there were more aware of trans-fat, sodium and calories. From the result that the most interested nutrition items were significantly different by democratic factors, we could understand interested nutrient information on the nutrition labels could change according to individual specific education. Therefore, this can also provide basic data for systematic education program by nutrition label use.

Recommendations for the Selective Labeling of [$^{15}N$]-Labeled Amino Acids without Using Auxotrophic Strains

  • The strategy to incorporate [$^{15}$ N]-labeled amino acids were discussed. Instead of using specific auxotrophic strains for selective labeling, the prototrophic strain, BL2l(DE3), was used with a plasmid, pLysS, and found to be very effective for several amino acids including alanine, lysine, leucine, and threonine. Isoleucine, valine, glutamine, and tyrosine were also found to be effective despite some diffusion into other amino acids. Interesting result was obtained when [$^{15}$ N]-labeled glycine was tried: only glycines were labeled when amino acid mixture was added in the growth medium, and serines were co-labeled when amino acids were omitted. These results can be used as a guideline when selective labeling strategy is considered, and when the resulting data are interpreted.

NTAㆍNi2+-Functionalized Quantum Dots for VAMP2 Labeling in Live Cells

  • An efficient method for labeling individual proteins in live cells is required for investigations into biological mechanisms and cellular processes. Here we describe the preparation of small quantum dots (QDs) that target membrane surface proteins bearing a hexahistidine-tag ($His_6$-tag) via specific binding to an nitrilotriacetic acid complex of nickel(II) ($NTA{\cdot}Ni^{2+}$) on the QD surfaces. We showed that the $NTA{\cdot}Ni^{2+}$-QDs bound to His-tag functionalized beads as a cellular mimic with high specificity and that QDs successfully targeted $His_6$-tagged vesicle-associated membrane proteins (VMAP) on cell surfaces. This strategy provides an efficient approach to monitoring synaptic protein dynamics in spatially restricted and confined biological environments.

  • Semantic role labeling is the research area analyzing the semantic relationship between elements in a sentence and it is considered as one of the most important semantic analysis research areas in natural language processing, such as word sense disambiguation. However, due to the lack of the relative linguistic resources, Korean semantic role labeling research has not been sufficiently developed. We, in this paper, propose an automatic predicate-argument analyzer to begin constructing the Korean PropBank which has been widely utilized in the semantic role labeling. The analyzer has mainly two components: the semantic lexical dictionary and the automatic predicate-argument extractor. The dictionary has the case frame information of verbs and the extractor is a module to decide the semantic class of the argument for a specific predicate existing in the syntactically annotated corpus. The analyzer developed in this research will help the construction of Korean PropBank and will finally play a big role in Korean semantic role labeling.

Food Allergy, a Newly Emerging Food Epidemic: Is the Current Regulation Adequate?

  • Food allergy refers to an immunologically mediated adverse reaction to food, mainly to proteinaceous constituents. Health implications vary between those individuals who experience mild physical discomforts to those with fast-acting, life-threatening anaphylactic reactions. The prevalence of food allergy is higher in children than in adults, estimated around 4-8% and 1-2% respectively in developed countries. Food allergy has no effective cure at the present time and total avoidance of causative foods is the most reliable prophylactic method currently recommended by the medical community. To help food allergic patients to make informed choices of their foods, mandatory labeling of selected food allergens has been introduced in several countries. All food allergen labelling provisions specify a set of allergens common to the regulated countries. Policy divergence, however, exists between countries by inclusion of additional allergens unique to specific countries and enforcement of specific labelling requirements. Such variations in food allergen labelling regulations make it difficult to manage allergen labeling in imported pre-packaged food products. This paper addresses two current issues in food allergen regulation: 1) an urgent need to determine true prevalence of food allergy in the Asia-Pacific region. This will enable refinement to the food allergen regulation to be more country-specific rather than simply adopting CODEX recommendations. 2) There is an urgent need for harmonization of food allergen regulation in order to prevent food allergen regulation becoming a trade barrier.

  • Ontology is a methodology on describing specific concepts and their relationships, and it is being considered important more and more as semantic web and variety of knowledge management systems are being highlighted. Ontology uses the relationships among concerts to represent some concrete semantics of specific concept. When we want to get some useful information from ontology, we severely have to process the transitive relationships because most of relationships among concepts represent transitivity. Technically, it causes recursive calls to process such transitive closure queries with heavy costs. This paper describes the efficient technique for processing transitive closure queries in ontology. To the purpose of it, we examine some approaches of current systems for transitive closure queries, and propose a technique by graph labeling scheme. Basically, we assume large size of ontology, and then we show that our approach gives relative efficiency in processing of transitive closure, queries.
