홍성 인접 사문암 지역 내 토양성분차이

  • This study was for comparisons of transitional element concentrations from the two different soil, serpentinite(SP) and granite area(GR), Kwangcheon and Hongseong area. Soils were collected by soil depths (10, 20 and 30cm) from the sites selected In the plant species(coniferous and deciduous species). In the soils, the SP was high in the Cr, Mn, Fe, Co, Ni and Zn concentrations, while the GR was high in the Cu, As, Sc and V. With the soil depths, the elements had high solubility, such as Cr, Co and Ni concentrations, were high in the SP, while the other elements were not clear. For the both species, the SP was high in the root and bolebark, while in the GR, not clear. Coniferous species in the same soil types, was higher than the deciduous In most elements. The more with increasing ages, the more with element accumulation in most plant parts except leaves. Comparisons between the soils and plant species, in the case of the element contents within the soils, the plant species in the same sites was similar trend. especially, clear in the SP

Development of Efficient Screening Methods for Melon Plants Resistant to Fusarium oxysporum f. sp. melonis (멜론 덩굴쪼김병에 대한 효율적인 저항성 검정법 개발)

  • This study was conducted to establish an efficient screening system to identify melon resistant to Fusarium oxysporum f. sp. melonis. F. oyxsporum f. sp. melonis GR was isolated from infected melon plants collected at Goryeong and identified as F. oxysporum f. sp. melonis based on morphological characteristics, molecular analyses, and host-specificity tests on cucurbits including melon, oriental melon, cucumber, and watermelon. In addition, the GR isolate was determined as race 1 based on resistance responses of melon differentials to the fungus. To select optimized medium for mass production of inoculum of F. oxysporum f. sp. melonis GR, six media were tested. The fungus produced the most spores (microconidia) in V8-juice broth. Resistance degrees to the GR isolate of 22 commercial melon cultivars and 6 rootstocks for melon plants were investigated. All tested rootstocks showed no symptoms of Fusarium wilt. Among the tested melon cultivars, only three cultivars were susceptible and the other cultivars displayed moderate to high resistance to the GR isolate. For further study, six melon cultivars (Redqueen, Summercool, Superseji, Asiapapaya, Eolukpapaya, and Asiahwanggeum) showing different degrees of resistance to the fungus were selected. The development of Fusarium wilt on the cultivars was tested according to several conditions such as plant growth stage, root wounding, dipping period of roots in spore suspension, inoculum concentration, and incubation temperature to develop the disease. On the basis of the test results, we suggest that an efficient screening method for melon plants resistant to F. oxysporum f. sp. melonis is to remove soil from roots of seven-day-old melon seedlings, to dip the seedlings without cutting in s pore s uspension of $3{\times}10^5conidia/mL$ for 30 min, to transplant the inoculated seedlings to plastic pots with horticulture nursery media, and then to cultivate the plants in a growth room at 25 to $28^{\circ}C$ for about 3 weeks with 12-hour light per day.

Rheinheimera aquatica sp. nov., Antimicrobial Activity-Producing Bacterium Isolated from Freshwater Culture Pond

  • A bacterial strain designated GR5$^T$, previously isolated from a freshwater culture pond in Taiwan while screening for bacteria for antimicrobial compounds, was characterized using a polyphasic taxonomic approach. Strain GR5$^T$ was found to be Gram-negative, aerobic, greenish-yellow colored, rod-shaped, and motile by means of a single polar flagellum. Growth occurred at $10-40^{\circ}C$ (optimum, $35^{\circ}C$), pH 7.0-8.0 (optimum pH 8.0), and with 0-2.0% NaCl (optimum, 0.5-1.0%). The major fatty acids were $C_{16:1}{\omega}7c$(36.3%), $C_{16:0}$(16.6%), $C_{12:0}$ 3-OH (12.5%), and $C_{18:1}{\omega}7c$(9.1%). The major respiratory quinone was Q-8, and the DNA G+C content of the genomic DNA was 51.9 mol%. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain GR5$^T$ belongs to the genus Rheinheimera, where its most closely related neighbors are Rheinheimera texasensis A62-14B$^T$ and Rheinheimera tangshanensis JA3-B52$^T$ with sequence similarities of 98.1% and 97.5%, respectively, and the sequence similarities to any other recognized species within Gammaproteobacteria are less than 96.5%. The mean level of DNA-DNA relatedness between strain GR5$^T$ and R. texasensis A62-14B$^T$, the strain most closely related to the isolate, was $26.5{\pm}7.6%$. Therefore, based on the phylogenetic and phenotypic data, strain GR5$^T$ should be classified as a novel species, for which the name Rheinheimera aquatica sp. nov. is proposed. The type strain is GR5$^T$ (=BCRC 80081$^T$=LMG 25379$^T$).

Effete of Ozone Uptake Rate on Photosynthesis and Antioxidant Activity in the Leaves of Betula Species (자작나무류 잎의 오존흡입량이 광합성 및 항산화효소 활성에 미치는 영향)

  • This study was conducted to compare the physiological and biochemical responses of four Betula species in response to ozone, and to find out the relationship between ozone uptake rate and photosynthesis or antioxidant activity. One-year-old seedlings of four Betula sp, B. costata, B. davurica, B. platyphylla var, japonica, and B. ermani, exposed to 100 pub ozone concentration for 8h day$^{-1}$ for 5 weeks in fumigation chamber. Ozone uptake rate, photosynthesis, SOD and GR activity were measured in the leaves of four species once a week. Cumulative ozone uptake rate was largest in the loaves of B. costata(53.8 mmol m$^{-2}$ ), smallest in the leaves of B. davurica(35.5 mmol m$^{-2}$ ). Photosynthesis of four Betula sp. exposed to ozone reduced relative to control, but the photosynthetic responses with changing stomatal conductance were different among species. Ozone exposure increased SOD activities of four species at the early exposing period, but after a critical point SOD activity decreased gradually. GR activity of B. costata was similar to the change of SOD activity, but the others showed the different patterns from B. costata. In conclusion, decreasing both SOD and GR activity at the critical point, B. costata may be sensitive species in response to ozone. In contrast, the others may be resistant species, which gradually increase GR activity following ozone exposure. GR activity was not always in accord with the change of SOD activity against ozone uptake, and the different responses between species were supposed to be affected by the cumulative ozone uptake.

Construction of Expression Vector of Bacillus sp. SSA3 Strain (Bacillus sp. SSA3 균주의 Expression Vector 개발)

  • The promoter regions from chromosomal DNA of Bacillus sp. SSA3 which is responsible for fermentation of Korean traditional soy sauce, were cloned for construction of expression vector of Bacills sp. SSA3. Recombinant plasmids were constructed by insertion of HindIIl-cleaved Bacillus sp. SSA3 chromosomal DNA fragments in front of the CAT gene of pGR71 plasmid and B-galactosidase gene of pUC18 plasmid. 6 recombinant plasmids were isolated from chloramphenicol resistant E. coli JM109 clones. All these plasmids were found to have promoter activity in Bacills sp. SSA3 and E. coli JM109. When these 6 clones of Bacills sp. SSA3 were cultivated in LB agar medium supplemented with 10% NaCI. fused CAT gene expression of 4 clones was significantly decreased in common. But the others were poorly inhibited.

Isolation and Characterization of Aniline-Degrading Bacteria

  • Six isolated strains degrading aniline were selected, identified and designated as pseudomonas putida K6, Pseudomonas acidovorans K82, Achromobacter gr. D. V. K24, Achromobacter xylosocidans K4, Moraxella sp. K21 and Moraxella sp. K22. All of them degraded 1000 ppm aniline completely within 30 to 36 hours. Most of these strains are resistant to antibiotics more than one, but Moraxella sp. has not any antibiotic marker tested. Most strains except for P. acidovorans K82 were shown to have resistance to the heavy metal ions such as Ni, Cu, Li, Ba, Co, etc. but not to Hg to which only P. putida K6 was resistant. M. sp. K21 was capable of degrading aniline to a maximum concentration of 2500 ppm without any repression. The incubation of the cell in limited pH ranges (4-8) had no great effect on aniline degradation. The addition of bactopeptone to the minimal media promoted the speed of aniline degradation, but the addition of glucose rather repressed the rate of aniline degradation. Through enzyme assay, A. gr. D. V. K 24 was shown to degrade aniline through artho-pathway and formed .betha.-ketoadipate as intermediate metabolite.

Control Effects of Some Insecticides on Different Stages of the Stone Leek Leafminer, Liriomyza chinensis Kato (Diptera: Agromyzidae) (파굴파리의 충태별 약제방제 효과)

  • Control effects of some insecticides were evaluated against the stone leek leafminer, Liriomyra chinensis Kato (Diptera: Agromyzidae) with the some different treatment methods. Insecticidal activities effects were estimated on the different development stages of the insects on welsh onion. The insecticides that controlled L. chinensis eggs with over 83% efficacy were spinosad, dimethoate, emamectin, and cartap. The insecticides that showed over 87% of larvicidal activity were dimethoate and cartap. Dimethoate showed 93.3% insecticide residual activity for 3 days a(ter treatment as a foliar spray. For control of pupae, the insecticides that showed over 88% of contact insecticidal activity were terbufos GR and cartap GR. Both dimethoate and cartap had high adulticidal activity with over 95% control efficacy.

Resistance and Susceptibility of Diamondback Moth, Plutella xylostella Strains Collected from Different Region in Korea to Bacillus thuringiensis (국내 지역별 채집계통 및 감수성계통 배추좀나방에 대한 Bacillus thuringiensis 제품의 생물활성 비교)

  • Six populations of the diamondback moth, Plutella xylostella, were collected from the different national areas for resistance and reared in laboratory for two sensitive population. These populations of P. xylostella were examined the developed resistance against commercial products of Bacillus thuringiensis. There were 3 products with B. thuringiensis subsp. kurstaki including Tyuneup$^{(R)}$, Thuricide$^{(R)}$ and Geumulmang$^{(R)}$ and 2 products with B. thuringiensis subsp. aizawai including Tobagi$^{(R)}$ and Scorpion$^{(R)}$. The sensitive population of diamondback moths were provided from National Academy of Agricultural Science (NP) and Highland Agriculture Research Center (GR population) and field populations were caught from 6 different national areas. Resistance against Tyuneup$^{(R)}$ was developed 4.8 and 2.5 times in SP and HS compared with GR population of diamondback moth, respectively. In case of Geumulmang$^{(R)}$, it was developed 9.9 and 6.8 times in SP and NM population compared with NP population, respectively. Otherwise, Tobagi$^{(R)}$ was showed higher resistance in HS than any other population compared with GR population, however, Scorpion$^{(R)}$ that is a same strain with Tobagi$^{(R)}$, was showed only double resistance to SP population. It was supposed that the development of resistance to B. thuringiensis might be caused by the continuous application of the specific commercial product at the specific area. So, we need to use the commercial products of B. thuringiensis in rotation with different B. thuringiensis strains. In the other hand, when HS population with highest resistance were reared in laboratory, their resistance ratio was rapidly dropped to 1.1 times at second generation. We have to examined the resistance mechanism of the diamondback moth to B. thuringiensis strains.

Growth and Soil Chemical Property of Small Apple Trees as Affected by Organic Fertilizers and Mulch Sources (비료원과 멀칭재료에 따른 사과 유묘의 생장 및 토양이화학성 변화)

  • BACKGROUND: This study was conducted to evaluate the effects of the fertilizer sources and ground cover mulches on nutrient release, growth, and photosynthesis in small one-year-old apple (Malus ${\times}$ domestica Borkh.) trees in controlled conditions. METHODS AND RESULTS: Treatments included no fertilizer (NF), commercial organic fertilizer (CF), and poultry litter (PL) for fertilizer treatments, and wood chips (WC), shredded paper (SP), green compost (GC), and grass clippings (GR) for cover mulch treatments. All treatments were applied proportionally based on the volume ratio equivalent to the soil. CF, PL, and GR treatments that had optimum carbon (C) and nitrogen (N) ratios (less than 30:1) for N mineralization through the microbes released the greatest $NH_4^+$ concentrations in the pot media at 90 days after the treatments, but GC mulch with the optimum C:N ratio did not. CF-, PL- and GR-treated plants had the largest leaf area, thickest stem diameter, longest shoot extension, and greater dry matter production. CONCLUSION(s): CF and PL showed an suitable organic nutrient source for improving plant growth in an orchard. Interestingly, GR also could be a nutrient source for tree growth, if vegetation competition is controlled by maintaining vegetation height and recycling enough grass clippings to the soil in an orchard.

Quantitative Analysis of Bacillus amyloliquefaciens GR4-5 in Soil (Bacillus amyloliquefaciens GR4-5 균주의 토양 내 정량 분석)

  • Bacillus amyloliquefaciens GR4-5 was isolated from the rhizosphere soil of Korean ginseng and displayed broad-spectrum suppression of ginseng root rot pathogens. The survivability of B. amyloliquefaciens GR4-5 in soil was investigated under three different conditions; indoor, outdoor - of which soil was put in 14 mL tube after treatment - and field environments. Soil samples were collected over a four-week period from three experimental designs, and assessed for 16S rRNA gene copy number by quantitative polymerase chain reaction (qPCR). In outdoor condition, the 16S rRNA gene copy number of Bacillus spp. was 8.35 log copies g $soil^{-1}$ immediately after the GR4-5 treatment. Two weeks later, the 16S rRNA gene copy number of Bacillus spp. (6.70 log copies g $soil^{-1}$) was similar to that of the control (6.38 log copies g $soil^{-1}$). In indoor condition, the 16S rRNA gene copy number of Bacillus spp. maintained in a certain level for a longer period than those in outdoor and field. The 16S rRNA gene copy number of Bacillus spp. in field experiment was reduced faster than that of outdoor condition. Our results show that B. amyloliquefaciens GR4-5 can survive in bulk soil for 1 week, indicating its potential use as a biocontrol agent following 7 day application intervals. This study presents that outdoor microcosm system design could be a useful method to assess easily the survivability of beneficial microorganisms.
