Effects of Germanium Treatment during Cultivation of Soybean Sprouts (게르마늄 처리가 콩나물의 생장에 미치는 영향)

  • This study was conducted to elucidate the characteristics of soybean sprouts cultured in water containing germanium. In three varieties of Eunhakong, Seomoktae and Subaktae, the yields of germanium soybean sprout were 10~20% more than those of control soybean sprouts after 5 day-cultivation. The hypocotyl of germanium soybean sprouts was thicker than that of control soybean sprouts, but there were no significant differences in the length of hypocotyl and the thickness of cotyledon. Germanium soybean sprouts showed deeper green and yellow color than control soybean sprouts. In addition, the hardness exhibited strongly in germanium soybean sprouts, especially Seomoktae. In the sensory evaluation, germanium soybean sprouts were better than control soybean sprouts in appearance, taste, flavor and preference.

Analyses of Free Amino Acids in Different Parts of Bean Sprouts by Different Cooking Methods and from Different Merchants

  • This study was performed to quantitatively analyze free amino acids in the bean sprouts of three different merchants by the parts and the cooking methods in order to evaluate the nutritional and sensory values of bean sprouts. Three merchant bean sprouts were analyzed from total, cotyledon and hypocotyls. Aspartic acid was the most common acid isolated from bean sprouts from all three merchants and was found more in the hypocotyls than the cotyledon. On the other hand, arginine, the second most common amino acid, was found more in the cotyledon than the hypocotyls while valine, the third or fourth most common amino acid in total bean sprout, occurred in a greater amount in hypocotyls than in cotyledons. After cooking, was the most concentrated amino acid in the liquid portion of both boiled bean sprouts and bean sprout soup was glutamic acid. Total bean sprouts from merchant C showed significantly higher contents of the most abundant amino acids, such as aspartic acid, arginine, alanine, serine, glutamic acid, isoleucine, leucine and tyrosine (p<0.05). After cooking, bean sprouts from merchant B showed less of a decrease in amino acid content in the solid parts than the products from merchants A and C. In conclusion, aspartic acid was the major amino acid in bean sprouts, regardless of the source, but after cooking, glutamic acid became the most abundant amino acid in the liquid part. Additionally, the pattern of release of the amino acids from the solid beans to the liquid portion during cooking was different with each merchant.

Effect of Freezing and Thawing Condition on the Physical Characteristics of Blanched Bean Sprouts as Home Meal Replacement (냉.해동 조건에 따른 간편편이식 콩나물의 물리적 품질 변화)

  • The purpose of this study was to investigate the effect of freezing and thawing rate on the physical properties of soybean sprouts to improve the quality of processed soybean sprouts during distribution and storage. Cooked soybean sprouts were frozen by air-blast freezing (ABF) system at $-45^{\circ}C$ or natural air convection freezing (NCF) system at $-24^{\circ}C$, then thawed using microwave oven by varying output power (0, 400, 800 and 1,000 W) until $75^{\circ}C$. The quality of soybean sprouts was measured by the water content, hardness and springiness. In addition, the internal microstructure of soybean sprouts was observed by optical microscope. For results, water content of soybean sprouts thawed by 1,000 W in a microwave showed the lowest value after natural air convection freezing. Springiness of soybean sprouts thawed by all amounts of output power was decreased in comparison with control. Hardness was increased only in soybean sprouts thawed by 1,000 W after air-blast freezing. However the gaps between springiness and hardness were relatively small with control at 1,000 W thawing, after air-blast freezing. Internal microstructure of the soybean sprouts was more damaged as freezing and thawing time were increased. In conclusion, high freezing and thawing rate might improves the quality of soy bean sprout, and IQF freezing and 1,000 W of microwave thawing appears to be the optimum condition for frozen HMR production. From the results freezing and thawing process parameters might can be use as quality control parameters as various type of sprout products processing.

Manipulating Isoflavone Levels in Mungbean Sprouts by Chemical Treatment (대사유도물질 처리에 의한 발아녹두의 아이소플라본 생합성 양상)

  • We have studied physiological responses of mung bean sprout to the treatment of elicitors. Chemicals such as salicylic acid and methyl jasmonic acid are not only the intermediates found in plant defense system but also could affect plant secondary metabolism. We found that mild treatment of salicylic acid and acetyl salicylic acid (aspirin) increase isoflavone production dramatically in mung bean sprout which has very low level of isoflavones compared with soybean sprout. The isoflavone content in salicylic acid treated- and acetyl salicylic acid treated-mung bean sprout was about 2.3 and 2.2 times higher than that of control, respectively. However, the increasing patterns of isoflavone in cotyledon and hypocotyl and root were not identical. The major increase among isoflavone fractions in cotyledon was led by the increase in malonylglycitin and malonyldaidzin level. Whereas, the increase in hypocotyl and root was led by malonyldaidzin. Methyl jasmonic acid did not show statistically significant increase in mung bean sprout. With this result, we were able to propose the non-transgenic method, which can control the isoflavone production in germinating mung bean.

Experimental Study for the Irradiation Effect of High Energy X-ray on the Growth of Sprout Plants Seeds (고 에너지 X-선이 새싹식물 종자의 생장에 미치는 영향)

  • This study examined the irradiation effect of high energy x-rays on the growth of sprout plants seeds. particularly with regard to acidity, salinity and sugar content change. The experimental groups were one day old sprout plants seeds irradiated with 6MV x-rays at doses of 2, 5, 8, 10 Gy using a linear accelerator. The control group was not irradiated. After two weeks, sprout plants were grown and harvested for component analysis. The acidity of the x-ray irradiated radish and red radish groups were significantly higher than that of the control group and bok choi and buckwheat groups lower than that of the control group. The salinity and sugar content of the x-ray irradiated 5 groups were significantly lower than that of the control group and Soybean and Red radish groups higher than that of the control group. The cause of plants component change were estimated to have occurred by generate basic ions and radiation sensitivity by moisture content. Further genetic studies will be needed to confirm the correlation between high energy x-rays and the cause of plants component change.

Ethanol Extracts of Chungkookjang Stimulate the Proliferation and Migration of Human Umbilical Vascular Endothelial Cells (청국장 에탄올 추출물의 혈관내피세포 증식과 이동 촉진효과)

  • In the fermented soybean product known as "chungkookjang", diverse bioactive compounds are produced when the soybean proteins are degraded during fermentation. Vascular endothelial cells (EC) are crucial in vein function and the formation of new vessels. A treatment to stimulate formation of new blood vessels is needed in cerebrovascular diseases that lead to ischaemic stroke and heart attack, as well as for diabetic ulcers. VEGF (Vascular Endothelial Growth Factor) simulates EC formation. The effect of Chungkookjang ethanol extract (CEE) on the proliferation of EC was studied. CEE (100, $1000{\mu}g/ml$) and boiled CEE were as effective as VEGF (10 ng/ml) for the proliferation of human umbilical vascular endothelial cells (HUVEC). The effect of CEE on the migration of HUVEC was investigated using sprout analysis. CEE ($100{\mu}g/ml$) was as effective as VEGF (10 ng/ml) for the migration of HUVEC. Isolation of specific peptides influencing the growth and migration of EC is needed.

Sprout Characteristics of Improved and Indigenous Soybeans in Korea (한국 재래 나물콩 품종 및 계통의 콩나물 특성)

  • To develop good soybean varieties for sprouts. 285 indigenous lines and 16 recommending varieties were evaluated by the sprout and agronomic characteristics. The range of whole length, hypocotyl length, root length, and yield rate of sprouts was 9.5~23.0cm, 5.9~15.1cm, 3.2~9.2cm, and 121~695%, respectively. Yield rate of sprouts was the highest, 409%, in the lines of mixed seed coat color. At the same time, it was reversely proportional to seed weight : the smaller the seed weight, the higher the yield rate as in all other reports. The number of roots was significantly fewer in the lines of the black and the green seed coat color than others. Based on whole length, rate of root length, and yield rate of sprouts, many indigenous lines were evaluated much better than recommending varieties. In the hypocotyl color of sprouts, the brightness was higher in improved variety than indigenous lines but vice versa in redness and yellowness in general.

Development of the Soybean Sprouting Bucket with Enhanced Air Circulation Function (공기치환기능이 강화된 콩나물 재배통 개발)

  • Traditional soybean sprouting bucket has some problems which are putrefaction and growing inhibition by the high temperature and carbon dioxide in the bucket during culturing. To solve this problems we developed the new soybean sprouting bucket. The new bucket consisted a square shaped bottom which has 5 draining holes (each 10 mm in dia, 2 mm in height) and four side wall which has two rectangular shaped holes (10 cm long, 0.5 cm wide), and the support vessel which 592 tiny draining holes whose center attached a pipe with 36 small holes (each 2 mm in dia). The new one showed lower temperature and carbon dioxide content during culturing, and the putrefaction was lower, whereas growing degree was higher during culturing at 25$\pm$1$^{\circ}C$ than the traditional bucket.

Changes in Postharvest Respiration, Growth, and Vitamin C Content of Soybean Sprouts under Different Storage Temperature Conditions

  • To understand the postharvest characteristics of soybean sprouts, 5-day-old sprouts were harvested, packed in PE film, and stored at 4, 12, and $20^{\circ}C$ for up to 4 days. In addition, the sprout respiration rate was measured after storage at 4, 8, 12, 16, 20, and $24^{\circ}C$ for up to 20h. During the first day of storage at $20^{\circ}C$, the sprouts maintained temperature-dependent longitudinal growth, especially of hypocotyl length; hypocotyl and root grew 0.8cm and 0.2cm, respectively. The hypocotyl thickness decreased by 11, 13, and $18\%$ after 4 days of storage at 4, 12, and $20^{\circ}C$, respectively. No temperature-dependent differences in fresh weight, dry weight, or water content were found, despite decreases of $3\%$ over the 4 days of storage. A significant postharvest decrease of $50\%$ in vitamin C content was observed in the sprouts stored at $20^{\circ}C$ for 3days. Based on the $CO_2$ production rate, the soybean sprouts exhibited an increase in respiration in proportion to the storage temperature; sprouts stored at 8, 12, 16, 20 and $24^{\circ}C$ showed approximately 2, 5, 6, 11, and 17 times, respectively, than the respiration rate of sprouts stored at $4^{\circ}C$. These results indicate the importance of low temperature storage during market circulation for minimizing the postharvest morphological and nutritional degradation of soybean sprouts.

Screening of immunoactive ingredients in frequently consumed food in Korea

  • The objectives of this study were to find out the plant to enhance immune activity among 42 kinds of foods frequently consumed by the Korean elderly consisting of 5 food groups and 5 wild plants. Each sample was assessed the immunoactive effect by measuring $NF-{\kappa}B/AP1$ gene expression, nitric oxide and cytokine production in $RAW-Blue^{TM}$ cell. Soybean sprouts of 47 plants showed the highest $NF-{\kappa}B/AP1$ gene expression at the level of $1.13{\pm}0.03$ (O.D. 650 nm) and Soritae, sweet potato, banana, apple, garlic, crown daisy, cabbage and Ailanthus altissima also had high activity of $NF-{\kappa}B/AP1$ gene in $RAW-Blue^{TM}$ cell stimulated by LPS. NO production of Ailanthus altissima was significantly higher than that of other plants and 16 plants of glutinous sorghum, black rice, Seoritae, Heuktae, sweet potato, banana, apple, garlic, mungbean sprouts, spinach, crown daisy, young pumpkin, cabbage, soybean sprouts, Actinidia arguta and Aster scaber were the next best activity. The above results selected 17 out of 47 plant samples. Moreover, soybean sprouts was significantly shown to increase $TNF-{\alpha}$ ($1,509.55{\pm}1.38pg/mL$) and $IL-1{\beta}$ ($54.56{\pm}1.08pg/mL$) cytokines in comparison with RAW-Blue cell stimulated by LPS. According to the results of in vitro evaluation, the ethanol extract of soybean sprout increased the production of immune-enhancing cytokines by proliferation of macrophages. In addition, $NF-{\kappa}B$ transcription factor activity and NO production ability were excellent, and it was selected as a material having excellent immunological activity.
