초기 육계 사료 내 미세조류(Tetradesmus sp.) 첨가에 따른 장관환경 변화 (Supplementation of Microalgae (Tetradesmus sp.) to Pre-Starter Diet for Broiler Chickens)

  • This study aimed to determine the effects of dietary microalgae (Tetradesmus sp. (TO)) on intestinal immunity and microbiota of pre-starter broilers. One hundred and twenty 1-day-old birds (Ross 308) were allocated to two dietary treatment groups with six blocks in a randomized complete block design. The two experimental diets consisted of a corn-soybean meal-based basal diet and a diet with 0.5% TO powder instead of cornstarch in the basal diet. After feeding the experimental diets for ten days, all birds' body weight and feed intake were measured, and representative eight birds were selected from each treatment group. Small intestinal lamina propria cells were isolated using flow cytometry to examine the frequency of immune cells. Cecal feces were harvested for 16s rRNA gut microbiota analysis and fecal IgA levels. Here, we found that 0.5% TO supplementation increased CD3+CD4+ T cells in the small intestine, but decreased CD3+CD8+ T cells in the small intestine. Gut microbial analysis showed that TO supplementation significantly increased the alpha diversity of the gut microbiome. Taxonomic analysis showed that TO treatment increased the abundance of Firmicutes and decreased that of Bacteroidetes at the phylum level. The distribution of Enterobacteriaceae containing many harmful bacteria at the family level, was lower in the TO group. In the LEfSe analysis, the TO group had a significantly enriched abundance of Agathobaculum at the genus level. Overall, results show that Tetradesmus sp. supplementation influences intestinal T-cell immunity and induces the expansion of beneficial gut microbes in pre-starter broiler chickens.

Effects of substitution of soybean meal with rapeseed meal and glutamine supplementation on growth performance, intestinal morphology, and intestinal mucosa barrier of Qiandongnan Xiaoxiang Chicken

  • Objective: The present study was to evaluate the effects of different rapeseed meal substitution (RSM) and glutamine (Gln) supplementation on growth performance, intestine morphology, and intestinal mucosa barrier of broilers. Methods: Four hundred and twenty Qiandongnan Xiaoxiang Chicken at 1 day of age with similar weight were chosen and were randomly assigned into 7 groups, consisting of 10 replicates per group and 6 broilers per replicate. Three groups were provided with diets separately containing 0%, 10%, and 20% RSM, and the other four groups were fed with diets separately supplemented with 0.5% and 1% Gln based on the inclusion of 10% and 20% RSM. At 21 and 42 days of age, 10 broilers per group were chosen to collect plasma and intestinal samples for further analysis. Results: The results showed that 10% RSM decreased average daily feed intake (ADFI) and average daily weight gain (ADG) of broilers at 21 days of age (p<0.05). Furthermore, both ADFI and ADG of broilers at 21 and 42 days of age were decreased by 20% RSM, while feed conversion ratio (FCR) was increased (p<0.05). Besides, 10% RSM resulted in lower intestinal villus height and the ratio of villus height to crypt depth, deeper crypt depth (p<0.05), combined with the lower mRNA expressions of occludin, claudin-1, and zonula occludens-1 (ZO-1) in broilers at 21 days of age (p<0.05). Similar results were also observed in broilers at 21 and 42 days of age fed with 20% RSM. However, 1% Gln improved the growth performance of broilers fed with 10% and 20% RSM (p<0.05), ameliorated intestine morphology and elevated mRNA expressions of occludin, claudin-1 and ZO-1 (p<0.05). Conclusion: In conclusion, the increasing inclusion of RSM resulted in more serious effects on broilers, however, 1.0% Gln could reverse the negative effects induced by the inclusion of RSM.

Effect of feeding raw potato starch on the composition dynamics of the piglet intestinal microbiome

  • Objective: Raw potato starch (RPS) is resistant to digestion, escapes absorption, and is metabolized by intestinal microflora in the large intestine and acts as their energy source. In this study, we compared the effect of different concentrations of RPS on the intestinal bacterial community of weaned piglets. Methods: Male weaned piglets (25-days-old, 7.03±0.49 kg) were either fed a corn/soybean-based control diet (CON, n = 6) or two treatment diets supplemented with 5% RPS (RPS5, n = 4) or 10% RPS (RPS10, n = 4) for 20 days and their fecal samples were collected. The day 0 and 20 samples were analyzed using a 16S rRNA gene sequencing technology, followed by total genomic DNA extraction, library construction, and high-throughput sequencing. After statistical analysis, five phyla and 45 genera accounting for over 0.5% of the reads in any of the three groups were further analyzed. Furthermore, short-chain fatty acids (SCFAs) in the day 20 fecal samples were analyzed using gas chromatography. Results: Significant changes were not observed in the bacterial composition at the phylum level even after 20 d post feeding (dpf); however, the abundance of Intestinimonas and Barnesiella decreased in both RPS treatment groups compared to the CON group. Consumption of 5% RPS increased the abundance of Roseburia (p<0.05) and decreased the abundance of Clostridium (p<0.01) and Mediterraneibacter (p< 0.05). In contrast, consumption of 10% RPS increased the abundance of Olsenella (p<0.05) and decreased the abundance of Campylobacter (p<0.05), Kineothrix (p<0.05), Paraprevotella (p<0.05), and Vallitalea (p<0.05). Additionally, acetate (p<0.01), butyrate (p<0.05), valerate (p = 0.01), and total SCFAs (p = 0.01) were upregulated in the RPS5 treatment group Conclusion: Feeding 5% RPS altered bacterial community composition and promoted gut health in weaned piglets. Thus, resistant starch as a feed additive may prevent diarrhea in piglets during weaning.

  • Objectives: This study examined regional differences in the intake of dietary total fat and saturated fatty acid (SFA) and their food sources among Korean adults. We also investigated the associations of SFA intake with metabolic diseases by region. Methods: This study included 13,926 adults (≥ 19y) who participated in the 2016 ~ 2019 Korea National Health and Nutrition Examination Surveys. The regions were divided into urban and rural areas according to the administrative districts where the participants lived. Using dietary data obtained from a 24-h recall, intake of total fat and SFA and their food sources were assessed by region. Metabolic diseases included obesity, abdominal obesity, and elevated total cholesterol and their association with SFA intake by region were examined using multiple logistic regression. Results: Of the participants, 19.6% lived in rural areas. In urban areas, the total fat and SFA intakes were higher than in rural areas: 21.2% of energy (%E) came from total fat and 6.9%E from SFA in urban areas, whereas 18.0%E came from total fat and 5.8%E from SFA in rural areas. The percentage of participants who exceeded the dietary reference intakes for total fat and SFA in urban areas was 16.5% and 41.9%, respectively, but 43.4% of participants in rural areas showed lower intake levels for total fat compared to the reference level. Young adults did not show regional differences in fat intake, and the percentage of subjects who exceeded the reference for SFA was high both in urban (58.5%) and rural (55.7%) areas. Among middle-aged and older adults, intake of fatty acids except for n-3 fatty acid was significantly higher in urban areas than in rural areas. About 69% of older adults in rural areas showed a lower intake of total fat compared to the reference level. The food sources for total fat and SFA were meat, soybean oil, eggs, and milk in both areas. The intake of fat from eggs, milk, mayonnaise, and bread was higher in urban areas, but the intake of fat from white rice and coffee mix was higher in rural areas. The SFA intake was positively associated with elevated serum total cholesterol in urban areas (4th quartile vs. 1st quartile, OR: 1.22, 95% CI: 1.06-1.40, P for trend: 0.043), but not in rural areas. Conclusions: Regional differences in total fat and SFA intakes and their food sources were observed among Korean adults. Our findings may help plan nutritional strategies to ameliorate regional health disparities.

COVID-19시대 한국 농촌 지역 노인의 단백질 섭취가 근감소증 예방 및 신체기능에 미치는 영향 (Effects of Protein Intake on Sarcopenia Prevention and Physical Function of the Elderly in a Rural Community of South Korea During the COVID-19 Pandemic)

  • Purpose : We aimed to investigate the effects of nutritional foods on sarcopenia prevention and physical function among the elderly living in rural communities during the COVID-19 pandemic by providing customized nutrition. Methods : This study was conducted in the rural community of SCC. Participants (n=24, over age 65) were randomly assigned into a Protein group (n=12) and a Vitamin group (n=12). The protein group was given 23 g/d of protein (whey, soybean, BCAA) for 8 weeks and the Vitamin group 23 g/d of vitamin (B, C, D, E and mixed minerals such as calcium, magnesium, zinc) for 8 weeks. All participants had their body composition such as height, weight, skeletal muscle mass, body mass index, and body fat percentage, measured using bioelectrical impedance analysis (BIA) and physical function assessed using grip strength and the short physical performance battery (SPPB). Results : At the end of the intervention, there was a significant increase in skeletal muscle mass (p<.01) in the Protein group (p=.002, 4.92 %) compared to the baseline: it increased by 2.33 %. The Vitamin group had a significant increase in body fat percentage after the intervention (p=.001, 15.35 %) compared to the baseline: body fat percentage decreased by 4.49 %. There were no significant differences in left and right Grip strength/Weight, SPPB, 4-m gait speed, chair stand test, and sense of balance in both groups. Conclusion : The findings from this study suggest that 8 weeks of protein intake have a significant effect on skeletal muscle mass and body fat percentage. Protein intake helped promoting the health of the elderly in rural community during the COVID-19 pandemic. It will assist creating a foundation for providing customized nutrition for the elderly in rural community in the future.

Effects of β-glucan with vitamin E supplementation on the physiological response, litter performance, blood profiles, immune response, and milk composition of lactating sows

  • Objective: This study was conducted to evaluate the effects of β-glucan with vitamin E supplementation on the physiological response, litter performance, blood profiles, immune response, and milk composition of lactating sows. Methods: A total of 50 multiparous F1 sows (Yorkshire×Landrace) with an average body weight (BW) of 233.6±4.30 kg and an average parity of 4.00±0.307 and their litters were used in this experiment. All sows were allotted to one of five treatments, taking into consideration BW, backfat thickness, and parity in a completely randomized design with 10 replicates. The experimental diets included a corn-soybean meal-based basal diet with or without 0.1% or 0.2% β-glucan and 110 IU vitamin E/kg diet. Results: All treatments added with β-glucan or vitamin E were statistically higher in the average daily feed intake (ADFI) of lactating sows compared to those of the control (Diet, p<0.01). Additionally, the ADFI of lactating sows was significantly higher in the groups supplemented with 0.1% β-glucan compared to 0.2% β-glucan (BG, p<0.01). There was an increasing trend in piglet weight at weaning (BG, p = 0.07), litter weight at the 21st day of lactation (BG, p = 0.07) and litter weight gain (BG, p = 0.08) in groups supplemented with 0.1% β-glucan. The addition of 110 IU vitamin E/kg diet increased vitamin E concentration significantly in lactating sows (VE, p<0.01) and exhibited a trend for higher concentrations of vitamin E (VE, p = 0.09) in piglets. Adding 0.1% β-glucan compared to 0.2% β-glucan induced a decrease in the concentration of tumor necrosis factor-α in lactating sows (BG, p = 0.06) and in piglets (BG, p = 0.09) on the 21st day of lactation. There were no significant differences in the milk composition of sows. Conclusion: Adding 0.1% β-glucan and 110 IU vitamin E/kg to a lactating sow's diet was beneficial to the growth performance of piglets by leading to an increase in the feed intake of sows and efficiently supplying vitamin E to both the sows and piglets.

담배거세미나방(Spodoptera litura) 성충의 예찰을 위한 최적 성유인제 조성 (Optimal Composition of Sex Attractant for Monitoring Adults of Spodoptera litura (Lepidoptera: Noctuidae) in Korea)

  • 본 연구는 담배거세미나방 국내 개체군의 예찰에 필요한 성유인제의 최적 조성을 찾기 위해 수행하였다. 담배거세미나방 암컷 성충에서 알려진 두 성페로몬 성분인 (Z,E)-9,11-tetradecadienyl acetate, (Z,E)-9,12-tetradecadienyl acetate를 5.5 ~ 10.0 및 9:1 ~ 99.1, 9:1 ~ 39:1 비율 사이에 여러 조성으로 만들어 고무격막 방출제에 담아 펀넬트랩을 이용하여 콩과 땅콩포장에서 수컷 성충에 대한 유인력을 비교한 결과, 두 성분의 19:1 비율이 가장 적절한 유인 조성으로 결정하였다. 그러나 Z9E11-14:Ac 단독 성분 미끼에는 수컷 성충이 거의 유인되지 않았다. 이 결과는 담배거세미나방 성충 예찰용의 성유인제 구성에는 두 성페로몬 화합물이 반드시 필요한 성분임을 나타낸다. 미끼에 담긴 성페로몬은 0.1 ~10 mg 사이에서 함량이 증가할수록 유인된 수가 많았다. 적절한 트랩 설치 높이는 1.5 m로 결정하였다.

Usual intake of dietary isoflavone and its major food sources in Koreans: Korea National Health and Nutrition Examination Survey 2016-2018 data

  • BACKGROUND/OBJECTIVES: Accumulating evidence has shown the beneficial effects of isoflavone on health. There is limited information on the usual isoflavone intake for Koreans. This study examined the usual intake of total isoflavone and its major food sources in Koreans according to age and gender. SUBJECTS/METHODS: The dietary intake data of 21,271 participants aged 1 yrs and older from the Korea National Health and Nutrition Examination Survey (KNHANES) VII 2016-2018 were analyzed. The average isoflavone intake was estimated based on the 24-h dietary recall data in KNHANES and the isoflavone database from the Korea Rural Development Administration (RDA) and literatures. The usual isoflavone intake was estimated by applying the ratio of within- and between-participant variance estimated from the 2009 KNHANES data to the 7th KNHANES (2016-2018) data. The variance of the isoflavone intake was calculated using MIXTRAN macro with intake data for two days in the 2009 KNHANES. Complex sample analysis with stratified variables and integrated weights was conducted. RESULTS: The mean total isoflavone intake in the Korean population aged 1 yrs and older (n = 21,271) was 139.27 mg/d, which was higher than the usual intake of 47.44mg/d. Legumes were a major contributing food group (91%), with arrowroot being a major individual contributor to the isoflavone intake (67.2%), followed by 21.3% of soybean, 5.4% of bean sprouts, and 2.1% of tofu. The usual isoflavone intake was highest in the participants aged 50 to 64 yrs old and increased with age until 50 to 64 yrs and then decreased with further increases in age. The usual isoflavone intake of participants aged 65 yrs and older was higher for men than for women, showing gender differences. CONCLUSIONS: The usual dietary intake of isoflavone varied according to age and gender in the Korean population. This study showed that the usual isoflavone intake was lower than the average isoflavone intake. The difference between percentiles of the usual isoflavone intake was similarly smaller than the average intake. An estimation of average intake can be hindered by the occasional consumption of foods high in isoflavones, suggesting that the usual intake estimation method can be more appropriate. Further research will be needed to establish isoflavone dietary guidelines regarding the effects of isoflavone intake on health outcomes.

Effects of medium chain triglycerides with organic acids on growth performance, fecal score, blood profiles, intestinal morphology, and nutrient digestibility in weaning pigs

  • Objective: This study was conducted to evaluate the effects of medium chain triglycerides (MCT) with organic acids (OA) on growth performance, fecal score, blood profiles, intestinal morphology, and nutrient digestibility in weaning pigs. Methods: A total of 120 weaning pigs ([Yorkshire×Landrace]×Duroc) with an average body weight (BW) of 8.00±0.87 kg were assigned in five treatments considering sex and initial BW in 3 replications with 8 pigs per pen in a randomized complete block design. The experimental diets included a corn-soybean meal based basal diet with or without 0.1% or 0.2% MCT and 0.1% OA. The pigs were fed the diets for 5 weeks (phase 1, 0 to 2 weeks; phase 2, 3 to 5 weeks). A total of 15 barrows with an average BW of 12.48±0.37 kg were used to evaluate the nutrient digestibility by total collection method. The BW and feed intake were measured at the end of each phase. Blood samples and small intestine samples were collected at the end of each phase, too. Results: Supplementing 0.1% MCT with 0.1% OA showed greater BW for week 5 and average daily gain (ADG) for overall period than control diet. Supplementing 0.1% MCT increased (p<0.05) ADG and improved (p<0.05) gain:feed ratio for phase 1. Dietary MCT and OA did not affect the fecal score and blood concentration of cortisol, immunoglobulin G, tumor necrosis factor-α, interleukin-1β (IL-1β), IL-6, and IL-10 in weaning pigs. Pigs fed the diets with 0.1% MCT had greater (p<0.05) villus height of duodenum and ileum for phase 1. Also, pigs fed the diet with 0.1% OA showed greater (p<0.05) villus height and villus height to crypt depth ratio of duodenum for phase 2. There was no significant difference in nutrient digestibility and nitrogen retention of pigs. Conclusion: Addition of 0.1% MCT with 0.1% OA in weaning pig's diet improved growth performance partly by enhancing intestinal morphology in weaning pigs.

Effect of blended protein nutritional support on reducing burn-induced inflammation and organ injury

  • BACKGROUND/OBJECTIVES: Previous studies have reported that protein supplementation contributes to the attenuation of inflammation. Serious trauma such as burn injury usually results in the excessive release of inflammatory factors and organs dysfunction. However, a few reports continued to focus on the function of protein ingestion in regulating burn-induced inflammation and organ dysfunction. MATERIALS/METHODS: This study established the rat model of 30% total body surface area burn injury, and evaluated the function of blended protein (mixture of whey and soybean proteins). Blood routine examination, inflammatory factors, blood biochemistry, and immunohistochemical assays were employed to analyze the samples from different treatment groups. RESULTS: Our results indicated a decrease in the numbers of white blood cells, monocytes, and neutrophils in the burn injury group administered with the blended protein nutritional support (Burn+BP), as compared to the burn injury group administered normal saline supplementation (Burn+S). Expressions of the pro-inflammatory factors (tumor necrosis factor-α and interleukin-6 [IL-6]) and chemokines (macrophage chemoattractant protein-1, regulated upon activation normal T cell expressed and secreted factor, and C-C motif chemokine 11) were dramatically decreased, whereas anti-inflammatory factors (IL-4, IL-10, and IL-13) were significantly increased in the Burn+BP group. Kidney function related markers blood urea nitrogen and serum creatinine, and the liver function related markers alanine transaminase, aspartate aminotransferase, alkaline phosphatase, and lactate dehydrogenase were remarkably reduced, whereas albumin levels were elevated in the Burn+BP group as compared to levels obtained in the Burn+S group. Furthermore, inflammatory cells infiltration of the kidney and liver was also attenuated after burn injury administered with blended protein supplementation. CONCLUSIONS: In summary, nutritional support with blended proteins dramatically attenuates the burn-induced inflammatory reaction and protects organ functions. We believe this is a new insight into a potential therapeutic strategy for nutritional support of burn patients.
