Hydrocolloids have many applications in foods including their use in dysphagia diets. We aimed to evaluate whether hydrocolloids in foods affect the digestibility of starch and protein, and their effects on antioxidant capacity. The thickening hydrocolloids: locust bean gum and carboxymethyl cellulose, and the gel-forming agents: agar agar, konjacglucomannan, and Hot & Soft Plus were blended with corn starch and soy protein, skim milk, or grape juice and were examined for their in vitro-digestability by comparing the reducing sugar and trichloroacetic acid (TCA)-soluble peptide, for antioxidant capacity by total polyphenol contents and the 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity. The hydrocolloids resulted in a decrease in starch digestibility with the gel-forming agents. Hydrocolloids diminished TCA-soluble peptides in skim milk compared to soy protein with the exception of locust bean gum and decreased free radical scavenging capacities and total phenolic contents in grape juice. Our findings may provide evidence for the use of hydrocolloids for people at risk of nutritional deficiencies such as dysphagia patients.
대두가수분해물 분무건조 펩타이드(SoyPep)가 자발성고혈압 횐쥐(spontaneously hypertensive rat, SHR)의 혈중지질수준, 생화학적지표 및 조직병리학적 변화에 미치는 영향을 조사하였다. SHR을 무작위로 대조군, 3% SoyPep 투여군, 10% SoyPep 투여군으로 구분하여 8주동안 실험사육하였다. 혈중지질 및 항산화성 비타민 수준, GOT 및 GPT활성을 측정하였고, 신장, 혈관조직, 혈장중 ACE 활성을 측정하였다. 또한, 각 주요장기의 조직병리학적 변화를 검토하였다. 식이 SoyPep은 SHR의 혈중 중성지질, 총콜레스테롤, LDL-콜레스테롤을 저하시키며, 또한, ACE저해활성 및 혈중 지질 수준의 개선작용을 나타내는 것이 확인되었다. 이들 결과로 보아, 대두 펩타이드는 혈중 지질수준의 증가를 수반하는 질병의 예방을 위한 새로운 기능성 식품소재로서의 활용가치가 매우 높을 것으로 기대된다.
한국 재래식 간장의 독특한 맛을 생성하는 B. licheniformis SSA3-2M1 의 배양액으로부터 proteinase 를 정제한 결과 2 종의 proteinase 의 역가를 확인할 수 있었다. Proteinase I 의 역가는 II 보다 두배가량 되며 최적 pH 가 7-11.5 이었다. Proteinase I 의 역가는 II 보다 두배가량 되며 최적 pH 가 7-11.5 이었다. Proteinase II 의 최적 pH 는7-9 이며 proteinase II 는 I 보다 pH3-5 부근의 상성에서 더 안정하고 활성이 강하였다. Proteinase I 과 II 의 최적온도는 각각 50.deg.C 였으며, proteinase II 의 온도안정성은 30-45 .deg.C 온도범위에서 proteinase I 보다 더 안정하였다. Proteinase I 은 45.deg.C 에서 60% 가 실현되었으며 protein II 는 45.deg.C 에서 5% 가량 실활되었다. 염에 대한 영향은 proteinase I 이 염의 농도 25-35% 범위에서 proteinase I 보다 효소활성이 높은 것으로 나타났다. Proteinase 치는 casein 을 기질로 사용하였을 경우, proteinase I 은 6.89 mg/ml, proteinase II 는 9 mg/ml 의 $K_{m}$ 치를 나타내었다. 이결과는 proteinase I 이 II 보다 기질에 대한 친화력이 높은 것으로 나타났다. 각종 금속이온의 농도 1 mM 에서는 Pb($CH_{3}$CCO)$_{2}$ 는 효소 활성을 증가시켰고 $ZnSO_{4}$, $7H_{2}$O, $K_{2}$$SO_{4}$, $HgSO_{4}$ 등은 저하시켰으며, 5 mM 이상에서는 $HgSO_{4}$ 와 $ZnSO_{4}$ 가 다른 염에 비해서 특히 효소활성을 많이 저히하였다. Proteinase I 과 II 는 대두 단백을 가수분해하여 peptide 와 아미노산을 생성하였으머 peptide 를 다시 아미노산으로 분해하였다. Proteinase I 과 II 는 한국재래식 간장의 중요 맛성분인 아미노산을 생성하는 주효소로 밝혀졌다.
This study was undertaken to examine effects of dietary intake of garcinia cambogia extract, soy peptide and L-carnitine mixture on body weight gain and obesity-related bio-markers in rats fed high-fat diet for 9 weeks with or without regular treadmill exercise. Forty 5-week-old male Sprague-Dawley rats were randomly divided into four groups; sedentary control group (SC), exercised control group (EC), sedentary formula-fed group (SF), and exercised formula-fed group (EF). The SC and EC rats were fed high-fat control diet (fat comprises$40\%$ of total caloris), and SF and EF rats were fed high-fat formula (composed of garcinia cambogia, soy peptide and L-carnitine) supplemented diet. Statistical analyses by two-way ANOVA indicated that the regular treadmill exercise significantly lowered cumulative body weight gain, total visceral fat mass, and epididymal, perirenal and retroperitoneal fat pad weights, and serum concentrations of total cholesterol and LDL + VLDL cholesterol, insulin, c-peptide and leptin. Feeding the formula also resulted in significant reductions in cumulative body weight gain and visceral fat pad weights, along with other related parameters including serum total and LDL + VLDL cholesterol levels, and hepatic enzyme activities involved in fatty acid synthesis. Statistical analyses by one-way ANOVA revealed that the formula consumption significantly improved body weight gain ($18\%$ reduction), total visceral fat weight ($20\%$ reductions), and serum total ($43\%$ reduction) and LDL + VLDL cholesterol ($54\%$ reduction) levels, as well as serum levels of insulin ($49\%$ reduction), and c-peptide ($41\%$ reduction) in sedentary rats, but failed to exhibit significant reductions in these indices in animals under treadmill exercise program. Taken together, these results suggest that the treadmill exercise per n exhibited significant improvements in body fat reduction and other related bio-markers, and so the formula consumption did not achieve a further significant reductions in these bio-markers in exercised rats. Nevertheless, animals fed the formula with regular exercise showed the most efficient weight reduction compared to other groups either fed formula without exercise or received regular exercise without dietary supplementation.
Cytokines play a central role in the mucosal immune response and are involved in regulation of nutrient absorption, metabolism and animal growth. This study investigated the effect of diet manipulation with specialized protein or peptide sources on expression of cytokine (IL-1, IL-6, IL-10, and TNF-${\alpha}$) mRNA abundance in different intestinal regions and at different ages post-weaning in piglets. A total of 48 (17 days of age, $6.16{\pm}0.34kg\;BW$) weanling pigs were fed either a corn-soy/whey protein basal diet, the basal diet supplemented with spray-dried plasma protein (SDPP), or the basal diet supplemented with $Peptiva^{(R)}$, a hydrolyzed marine plant protein. A fourth treatment group was fed the SDPP diet, but the feed intake level was limited (SDPP-LF). Pigs were killed at 3 and 10 d, and intestinal cytokine mRNA was measured by real-time PCR using the relative quantification method. The SDPP-LF group exhibited an increased TNF-${\alpha}$ mRNA abundance compared with the ad libitum SDPP group (p<0.05). The TNF-${\alpha}$ and IL-10 mRNA abundance increased from the proximal to distal part of the intestine, and the mRNA abundance was greater (p<0.01) in the distal intestine as compared with the proximal and middle intestine. The cytokines IL-1-${\beta}$, IL-10 and TNF-${\alpha}$ mRNA abundance also increased from d3 to d10 postweaning (p<0.01). In summary, restricted feeding increased the TNF-${\alpha}$ mRNA abundance in the small intestine, however neither SDPP nor peptide supplementation affected cytokine mRNA expression. Abundance of mRNA for most cytokines examined in this study increased with age post-weaning, suggesting that during 10 d after weaning the mucosal immune system is still under development.
식품단백질의 효율적인 이용을 위하여 neutrase에 의한 glutamine과 asparagine 잔기의 탈아미드화에 관한 연구를 행하였다. Neutrase에 의한 BSA의 최적반응 조건은 $20^{\circ}C$, pH 10에서 3시간 반응시켰을 때로, 이때 BSA는 24%가 탈아미드화되었고. 동시에 2.9%가 가수분해되었다. 또한 pronase, bromelain, ficin 보다는 neutrase에 의한 BSA의 탈아미드화율이 높은 것으로 나타났다. 최적 탈아미드화 조건에서 neutrase에 의한 egg albumin, 분리콩단백 및 casein의 경우 탈아미드화에 수반하여 펩티드결합의 분해가 비교적 높게 관찰되었다.

BACKGROUND/OBJECTIVES: Soy isoflavone (SIF) and soy lecithin (SL) have beneficial effects on many chronic diseases, including neurodegenerative diseases. Regretfully, there is little evidence to show the combined effects of these soy extractives on the impairment of cognition and abnormal cerebral blood flow (CBF). This study examined the optimal combination dose of SIF + SL to provide evidence for improving CBF and protecting cerebrovascular endothelial cells. MATERIALS/METHODS: In vivo study, SIF50 + SL40, SIF50 + SL80 and SIF50 + SL160 groups were obtained. Morris water maze, laser speckle contrast imaging (LSCI), and hematoxylin-eosin staining were used to detect learning and memory impairment, CBF, and damage to the cerebrovascular tissue in rat. The 8-hydroxy-2'-deoxyguanosine (8-OHdG) and the oxidized glutathione (GSSG) were detected. The anti-oxidative damage index of superoxide dismutase (SOD) and glutathione (GSH) in the serum of an animal model was also tested. In vitro study, an immortalized mouse brain endothelial cell line (bEND.3 cells) was used to confirm the cerebrovascular endothelial cell protection of SIF + SL. In this study, 50 µM of Gen were used, while the 25, 50, or 100 µM of SL for different incubation times were selected first. The intracellular levels of 8-OHdG, SOD, GSH, and GSSG were also detected in the cells. RESULTS: In vivo study, SIF + SL could increase the target crossing times significantly and shorten the total swimming distance of rats. The CBF in the rats of the SIF50 + SL40 group and SIF50 + SL160 group was enhanced. Pathological changes, such as attenuation of the endothelium in cerebral vessels were much less in the SIF50 + SL40 group and SIF50 + SL160 group. The 8-OHdG was reduced in the SIF50 + SL40 group. The GSSG showed a significant decrease in all SIF + SL pretreatment groups, but the GSH showed an opposite result. SOD was upregulated by SIF + SL pretreatment. Different combinations of Genistein (Gen)+SL, the secondary proof of health benefits found in vivo study, showed they have effective anti-oxidation and less side reaction on protecting cerebrovascular endothelial cell. SIF50 + SL40 in rats experiment and Gen50 + SL25 in cell test were the optimum joint doses on alleviating cognitive impairment and regulating CBF through protecting cerebrovascular tissue by its antioxidant activity. CONCLUSIONS: SIF+SL could significantly prevent cognitive defect induced by β-Amyloid through regulating CBF. This kind of effect might be attributed to its antioxidant activity on protecting cerebral vessels.
We previously reported that the novel isoflavone-free peptide mixture (black soybean peptide, BSP) had several beneficial effects like antiobesity and hypotriglyceridemic effect. However, there are no reports for BSP on anti-hypertensive activity. BSP induced heme oxygenase-1 (HO-1) in HUVECs, thus investigated the HO-1-induced activity in HUVECs and the anti-hypertensive effects in SHR animal model. BSP significantly induced HO-1 expression both at transcriptional and protein levels in a time- and dose-dependent manner as measured by RT-PCR and Western blot analysis, respectively. These inductions were abolished by pretreatment of N-acetyl-cystein (NAC, 1~10 mM), but not by employing Tempol, a superoxide dismutase (SOD) mimetic (1~5 mM). As expected, enzymatic activity (~2 fold) determined by bilirubin formation assay and cGMP concentration (~6 fold) were significantly increased in BSP-treated cells. Based on the numerous evidences on the beneficial effects of HO-1 and our results, we investigated in vivo effects of BSP on the antihypertensive activity. The administration of BSP (1% in drinking water) significantly decreased mean blood pressure (BP) (from $218.6{\pm}6.99$ to $190.0{\pm}3.42$ mm Hg, p<0.01). This result indicates that BSP is strong inducer of HO-1 expression, which may be triggered by oxidative stress, and has anti-hypertensive activity.
Aspergillus niger CF-34가 생산하는 대두세포벽분해 효소 복합액을 알칼리로 처리하여 조효소액 중에 함유된 protease만을 선택적으로 제거할 수 있었다. 조효소액을 알칼리로 처리하였을 때 세포벽분해활성에는 영향을 적게 주고, 대두 단백질을 분해시켜 쓴 맛의 peptide를 생성하는 protease만을 선택적으로 불활성화시킬 수 있었다. 조효소액의 pH를 $9.0{\pm}0.1$로 조절하여 $20^{\circ}C$에서 약 30분 동안 서서히 교반한 후 다시 pH를 5.0로 조절하는 것이 최적 조건이었다. 이때 조효소액 중 protease 활성은 초기의 약 10% 미만으로 감소하였으며, 각 효소의 잔존 활성을 살펴보면 pectinase는 약 80%, polygalacturonase는 약 85%, xylanase는 약 95%, carboxymethyl cellulase는 약 100% 정도이었고, 대두세포벽분해활성은 초기의 약 90% 정도 유지할 수 있어 protease만이 선택적으로 제거되었다. 이렇게 처리된 효소액을 사용하여 비지 중의 대두 단백질을 추출할 경우 생산효율은 비록 감소하였지만, 대두단백질의 분해를 막고, 쓴맛 생성을 억제하여 품질 및 관능적으로 우수한 제품을 생산할 수 있었다.
대두(Glycine max)는 식물성 단백질이 풍부한 식품 중 하나로, 생리활성물질을 다량 함유하고 있어 심혈관질환, 비만 등에 효능이 있을 뿐 아니라 피부 개선 효과도 있는 것으로 보고되어 있다. 본 연구에서는 대두 단백질을 유산균인 Lactobaillus Rhamnosus 균주에 의해 발효한 대두발효물의 광노화 예방 효과를 조사하기 위해 자외선으로부터 기인한 세포독성에 대한 보호 효과 및 콜라겐 생성, 티로시나아제 활성, 멜라닌 색소 생성에 대한 평가를 실시하였다. 결과로서 대두발효물 처리군은 피부섬유아세포에서 자외선으로 유도된 세포독성을 억제하여 보호 효과를 나타내었으며, 세포 내 콜라겐 합성을 증가시켜 주름 개선의 가능성을 확인하였다. 또 다른 실험에서 티로시나아제 활성 및 멜라닌 색소 생성을 억제하여 피부 개선 효과를 나타내었다. 이러한 결과로 미루어 볼 때 대두발효물이 자외선으로부터 유도되는 주름, 색소 침착 등의 피부 손상을 예방하는 새로운 기능성 소재로서의 활용 가능성을 가지는 것으로 평가된다.
