A Study on the Disintegration of Sewage Sludge using Batch Ultrasonic Pretreatment (회분식 초음파 전처리를 이용한 하수슬러지의 분해에 관한 연구)

  • Anaerobic digestion is the most common process for sewage sludge stabilization and has benefits of VS reduction and biogas production. Many pretreatment methods have been studied to improve hydrolysis rate because the rate of sewage sludge degradation is slow in anaerobic digestion. This study mainly focused upon the effects on disintegration of sewage sludge by ultrasonic pretreatment according to the variation of acoustic density and duration of sonication time. In this study, acoustic density has been changed as follows : 33W/L, 70W/L, 88W/L, 139W/L in case of 40 kHz with the test time changes of 10min, 20min, 25min, 30min and 40min. In the comparison of $SCOD_{Cr}/TCOD_{Cr}$ variation for excess sludge and mixed sludge disintegration, the rates of $SCOD_{Cr}/TCOD_{Cr}$ have been increased in the condition of denser acoustic density and longer sonication time with acoustic frequency of 40kHz. The pH of the excess sewage sludge and mixed sewage sludge has been decreased in the condition of denser acoustic density and longer sonication time with acoustic frequency of 40kHz.

Extraction and Analysis of Astaxanthin from Haematococcus pluvialis Using Sonication (초음파처리를 이용한 Haematococcus pluvialis로부터의 아스타잔틴의 추출 및 분석)

  • The extraction and quantitative analysis conditions for astaxanthin from Haematococcus pluvialis, and the structural characteristics of H. pluvialis extract, H. pluvialis hydrolysate and synthetic astaxanthin were investigated using UV/visible and FT-IR spectrometers. Astaxanthin was dissolved in methanol, and then treated to enhance the solubility by sonication for 45 min. With sonication pretreatment, the solubility of astaxanthin increased up to 1.5 times compared to that without sonication. The extracts were hydrolyzed by cholesterol esterase for the analysis of H. pluvialis extract containing astaxanthin ester. A HPLC method using reverse phase C18 column with methanol-water (95:5, v/v) as mobile phase was developed to analyze astaxanthin. After hydrolysis, the absorption spectrum of H. pluvialis hydrolysat was changed to similar pattern to synthetic astaxanthin, confirming the extraction and analysis condition of astaxanthin from H. pluvialis.

Study on the Changes in Allergen and Allergenicity Originated from Shrimp by Physical Treatments (물리적 처리에 의한 새우유래 Allergen 및 Allergenicity 변화)

  • This research was conducted to evaluate the changes in allergenicity of shrimp by physical treatments using competitive indirect enzyme-linked immunosorbent assay (Ci-ELISA). Shrimp was subjected to physical treatments such as high hydrostatic pressure (HHP), sonication, autoclave and microwave. Heat-stable protein (HSP) purified from raw shrimp was used as a major allergen. The binding ability of monoclonal IgG and shrimp-allergic patients' IgE to HSP treated with HHP decreased, increasing the pressure up to 400 MPa. Especially, it became less than 50% at 400 MPa. The binding ability of mAb to HSP treated with sonication (10, 20, 30 and 60 min) decreased with the treated time. Especially, it became less than 60% with the treatment for 60 min. The allergenicity change of HSP treated with autoclave and microwave little decreased. The binding ability of mAb to HSP during the treatment for 20 min became more than 70%. The results suggest that allergenicity of HSP in raw shrimp be more easily lost by HHP or sonication treatment than by autoclave or microwave treatment.

Changes in Allergenicity of Porcine Serum Albumin by Microwave, Sonication, and High Hydrostatic Pressure (Microwave, 초음파 및 초고압 처리에 의한 돼지 혈청 알부민의 항원성 변화)

  • Even though pork have frequently induecd allergic reactions in Korea, few papers have been published on pork allergy. This study was carried out to investigate the changes in allergenicity of porcine serum albumin (PSA) by microwave, sonication, and high hydrostatic pressure (HHP). The binding ability of p-IgG to PSA treated with microwave (1,5, or 10 min) directly decreased with increasing treatment time. Particularly, the binding ability of PSA treated 10 min was about 30%. Immunoblotting assay with p-IgG showed that band of PSA treated microwave directly disappeared at 5 and 10 min. However, the binding ability of PSA was not changed by the microwave treatment without heat. Also the reduction of allergenicity by sonication or HHP treatment was not found. In conclusion. allergenicity of PSA treated with microwave directly decreased with increasing time, therefore these results may be used for development of hypoallergenic pork.

Monitoring Airborne Nanoparticle Concentrations by Task in a Laboratory Making Carbon Nanotube Films (탄소나노튜브 필름 제조 실험실의 세부작업별 공기 중 나노입자 노출 농도)

  • Airborne nanoparticle concentrations in three metrics (particle surface area concentration, particle number concentration, and particle mass concentrations) were measured by task in a laboratory making carbon nanotubes (CNTs) films using three direct reading instruments. Because of the conducted other researcher's experiment before the tasks, airborne nanoparticle surface area and number concentrations are the highest at the first time conducted weighing and mixing by sonication task, respectively. Because of the mist generated during mixing by sonication, the highest airborne nanoparticle surface area and PM1 concentrations were measured in the task among the total. Nanoparticle surface area concentrations at the researchers' breathing zones had high correlation (r=0.93, p<0.01) with those measured at an area in the laboratory. This result indicates that nanoparticles generated during the experiment contaminated the whole room air. When the experiment performed all the fume hoods weren't operated and making CNTs films task were conducted in the out of the fume hoods. In conclusion, researchers performing making CNTs film experiments were exposed to airborne nanoparticles generated during the experiment without adequate controls. We recommend that adequate controls should be implemented so that workers' exposures to airborne nanoparticle are limited to minimum levels.

Pretreatment Characteristics and Specific Methanogenic Activity of Municipal Sewage Sludge by Dual Frequency Sonication (이중주파수 초음파를 이용한 도시하수슬러지의 전처리 특성 및 비메탄활성도에 관한 연구)

  • The objectives of this study have been carried out to investigate the solubilization of municipal sewage sludge by single and dual frequency ultrasonic pretreatment, and the methane production characteristics of pretreated sewage sludge by specific methanogenic activity test for sewage sludge reduction. The waste activated sludge was collected from thickened tank of Suyoung sewage treatment plant in Busan city, and its concentration was adjusted to 1.0% total solids. Ultrasonic frequency was varied 15, 20, 15+20 kHz, and acoustic density was used a maximum 176W/L. The dual frequency ultrasonic pretreatment was found to be more effective than single frequency ultrasonic in the solubilization rate and methane production. The $SCOD_{Cr}/TCOD_{Cr}$, rate were 15.2%, 13.9%, 17.0% with single frequency of 15 kH2, 20 kHz, dual frequency of 15+20 kHz, respectively. The application of dual frequency ultrasound for sewage sludge pretreatment can be interest for sewage treatment plants having problems in sludge treatment and disposal.

  • Carbon rich fly ash was recently reported to have compositions that are ideal for use as a precursor and catalyst for carbon nanotube growth. This fly ash powder is mostly composed of pure carbon, predominantly present as sp2. In this work, the effect of sonication time on the morphology and structural properties of carbon rich fly ash particles is reported. The obtained results show that ultrasound treatment is an effective tool for producing ultrafine particles/fragments with higher porosity, which might be suitable for the adsorption of gasses. Moreover, carbon nanoparticles (CNPs) of this fly ash were produced in parallel using the ball milling technique, and were evaluated as reinforcements for epoxy based composites. These CNPs have almost spherical shapes with particle sizes of around 30 nm. They were found to have strong C=O carbonyl group bonds, which might be generated during the ball milling process. The tensile testing results of a fly ash CNP reinforced epoxy composite showed significant improvements in the mechanical properties, mainly in the stiffness of the polymer. The stiffness value was increased by around 23% of that of neat epoxy. These CNPs with chemically active groups might also be useful for other applications.
