The Wormicidal Sibstance of Fresh Water Fishes on Clonorchis sinensis(VI) - Struture Identification of Purified Clonorchicidal Sudstance from Epidermal Mucus of Cyprinus carpio nudus (간흡충에 대한 살충성물질에 관한 연구 (VI) - 향어 체표면 점액내 살충성물질의 구조 확인)

  • In order to elucidate the defence mechanism of Cyprinus carpio nudus to Clonorchis sinensis, clonorchicidal substance in the epidermal mucus of this fish was isolated by silica gel column and thin layer chromatography and analyzed for its chemical nature by UV, IR and NMR-spectroscopy. The epidermal mucus of the fish was extracted by ethyl ether and separated into 2 fractions by column chromatography using petroleum ether/chloroform(30/70,v/v) as first solvent and methanol as second solvent. The second fraction with greyish white colour showed clonorchicidal effect. The second was again fractionated into greyish white precipitate and clonorchicidal greenish yellow supernatant fraction, by adding petroleum ether and standing the mixture for 5 days at $5^{\circ}C$. The latter was divided into 7 fractions in column chromatography with acetone/ benzene (10/90, v/v) as carrier. The fraction on equivalent to the spot of Rf. 0.225 value among them disclosed the strongest clonorchicidal effect. By this purification procedure, clonorchicidal substance was purified 154-fold with 0.9% yield from the mucus of the fish, and 10mg of the final fraction killed tne metaceicariae in 22 min. Infra red, nuclear magnetic resonance and ultraviolet spectrometric analysis of the purified substance revealed that the substance is linoleic acid. According to the results of the present studies it seemed that this fish could not be proper intermediate host of Clonorchis sixensis, and that defence mechanism of this fish to the worm seems to be correlated with clonorchicidal substance in epidermal mucus.

Antimicrobial, Antioxidant and Cytotoxic Activities of Dendropanax morbifera Léveille extract for mouthwash and denture cleaning solution

  • PURPOSE. The purpose of this study was to analyze the antimicrobial, antioxidant activity and cytotoxicity of Dendropanax morbifera $L{\acute{e}}veille$ extract for assessing whether Dendropanax morbifera $L{\acute{e}}veille$ can be used for the development of natural mouthwash and denture cleaning solution. MATERIALS AND METHODS. The extract was obtained from branches of Dendropanax morbifera $L{\acute{e}}veille$. The solvent fractions were acquired by fractionating Dendropanax morbifera $L{\acute{e}}veille$ extract using n-hexane, ethyl acetate, chloroform and butanol solvent. Paper disc test was used to evaluate the antimicrobial and antifungal activity of Dendropanax morbifera $L{\acute{e}}veille$ extract and solvent fractions against Streptococcus mutans and Candida albicans. The analysis of antioxidant activity was carried out through DPPH radical scavenging assay. The cytotoxicity of Dendropanax morbifera $L{\acute{e}}veille$ extract was analyzed through MTT assay using normal human oral keratinocytes. RESULTS. Dendropanax morbifera $L{\acute{e}}veille$ extract showed antimicrobial activity against Streptococcus mutans and especially Candida albicans. The solvent fractions of Dendropanax morbifera $L{\acute{e}}veille$ showed strong antimicrobial activity against Streptococcus mutans and Candida albicans in n-hexane and butanol solvent fraction, respectively. Dendropanax morbifera $L{\acute{e}}veille$ extract also showed outstanding antioxidant activity. Butanol, ethyl acetate, and chloroform solvent fraction of Dendropanax morbifera $L{\acute{e}}veille$ tended to have increased antioxidant activity as the concentration increased. Dendropanax morbifera $L{\acute{e}}veille$ extract showed high cell survival rate in cytotoxicity test. CONCLUSION. Dendropanax morbifera $L{\acute{e}}veille$ extract turned out to have antimicrobial, antioxidant activity and cytophilicity. Based on these results, it is expected that Dendropanax morbifera $L{\acute{e}}veille$ is applicable as an ingredient for natural mouthwash and denture cleanser.

Antioxidant Activity of Solvent Fraction from Black Garlic (흑마늘 용매 분획물의 항산화 활성)

  • To confirm antioxidant activity of black garlic, methanol extract of black garlic was fractioned by hexane, chloroform, ethyl acetate, buthanol and water. Antioxidant activities of solvent fractions were assayed in 100, 250, 500 and $1,000\;{\mu}g/mL$ concentrations. The contents of total phenol and flavonoids were significantly higher 5.5~11.6 times in chloroform, ethyl acetate and hexane fraction than other fractions. Antioxidant activities of solvent fractions were increased by higher sample concentrations and their activities were significantly higher in chloroform and ethyl acetate fractions than others. DPPH radical scavenging activity was over 50% in $1,000\;{\mu}g/mL$ concentration, except butanol and water fraction. In the same concentration, reducing power was also significantly lower in butanol and water fraction. ABTS radical scavenging activity was higher in hexane, chloroform and ethyl acetate fractions and was over 70% at $1,000\;{\mu}g/mL$ concentration. In $1,000\;{\mu}g/mL$ concentration, the range of hydroxy radical scavenging activity was 50.27~81.02% and SOD-like ability was 26.73~47.64%. Antioxidant activity in linoleic acid reaction system was significantly higher when storage time was longer and sample concentration was higher in non-polar solvent fractions. Nitrite scavenging activity was relatively higher than antioxidant activity and the activity in $100\;{\mu}g/mL$ concentration was over than 50%, except butanol fraction.

Comparison of Biological Activities of Extracts from Different Parts and Solvent Fractions in Cornus kousa Buerg (산딸나무의 부위별 추출물 및 용매 분획물의 생리활성 비교)

  • Several extracts of different parts and solvent fractions of Cornus kousa were obtained and their functional material contents, antioxidant activities and tyrosinase inhibition effects were determined. Content of total polyphenols and flavonoids contents in flower were 169.638 $mg{\cdot}g^{-1}$ and 25.418 $mg{\cdot}g^{-1}$, respectively, which were much higher than those of other parts. Also, flower extracts showed the strongest effects on DPPH and ABTS radicals scavenging and ferrous ion chelating. In flower, leaf, and stem extracts, inhibition effects on peroxidation of linoleic acid determined by ferric thiocyanate(FTC) method were higher than a synthetic antioxidant, BHT. Tyrosinase inhibition activities were shown only in flower extract. Flower and leaf extracts, showing high biological activities in various system, were successively reextracted with n-hexane, chloroform, ethylacetate and n-butanol. Total polyphenol contents of water fractions were higher than any other solvent fractions in both flower and leaf, 67.006 $mg{\cdot}g^{-1}$ and 67.739 $mg{\cdot}g^{-1}$, respectively. But total flavonoid contents were higher in ethyl acetate fraction for flower extract and butanol fraction for leaf extract. Among the solvent fractions, the highest efficiency of free radical scavenging activities was obtained in ethyl acetate fraction for flower extract and n-butanol fraction for leaf extract. Tyrosinase inhibition activities were higher in water fraction for both flower and leaf extracts, 49.24% and 31.8%, respectively.

Antioxidant Component and Activity of Dropwort (Oenanthe javanica) Ethanol Extracts (미나리 에탄올 추출물의 항산화성분과 항산화활성)

  • This study was conducted to investigate the antioxidant compounds and antioxidant activity on the dropwort (Oenanthe javanica) and its solvent fraction. The dropwort was extracted with 70% (v/v) ethanol, and then partitioned using the solvents of hexane, chloroform, ethyl acetate, butanol, and aqueous. The ethyl acetate fraction contained the highest phenolic and flavonoid of 240.61 mg GA eq/g and 105.57 mg catechin eq/g, followed by ethanol extract of 37.50 mg GA eq/g and 26.50 mg catechin eq/g, respectively. The DPPH radical scavenging activity ($IC_{50}$) on the solvent fractions increased in the order of ethyl acetate, butanol, ethanol extract, chloroform, aqueous, and hexane with 0.08, 0.58, 1.07, 2.43, 2.47, and 3.31 mg/mL, respectively. The ABTS radical scavenging activity was the highest value of 382 mg AA eq/g in ethyl acetate fraction. Reducing power and chelating effect on the ethanol extracts and its solvent fraction were in range of 0.23~0.75 and 0~32.01%, respectively. Hydroxyl radical scavenging activity ($IC_{50}$) was the lowest value of $26.71\;{\mu}g$/mL in ethyl acetate fraction.

Evaluation of Anti-oxidative, Anti-thrombin, Anti-invasive and Pro-apoptotic Activities of Paeonia japonica (백작약(Paeonia japonica)의 항산화, 트롬빈 저해, 암전이 억제 및 암세포사멸 평가)

  • Paeonia japonica is a perennial flowering plant used in traditional medicine therapy. The purpose of this study was to investigate the effect of water extract and solvent fractions obtained from P. japonica on anti-oxidative, anti-thrombin, anti-invasive and pro-apoptotic activities in YD-10B cells, human oral squamous carcinoma cell line. Water fraction revealed the highest extraction yield at 11.44% (w/w). Anti-oxidative activity was the highest in ethyl acetate fraction (85.13%). In the thrombin inhibitory activity test, ethyl fraction was the highest, with a value of 87.54%. Release and activation of MMP-2/pro-MMP-2 ratio in thrombin-treated YD-10B cells were significantly inhibited in the ethyl acetate fraction. At a concentration of $120{\mu}g/m{\ell}$ water extract and solvent fractions of P. japonica inhibited cell proliferation in YD-10B cells except water fraction. Pro-apoptotic effect on human oral squamous carcinoma cell using the Bax/Bcl-2 ratio analysis was higher in water extract than other fractions. These findings suggest that the ethyl acetate fraction of P. japonica potentiates a promising antioxidant, anti-thrombin and anti-invasive agents.

Nitrite-scavenging and Antimutagenic Effects of Various Solvent Extract from Different Parts of Loquat (Eriobotrya japonica, Lindl.) (비파 부위별 용매추출물의 아질산염 소거 및 항돌연변이 효과)

  • The nitrite-scavenging and antimutagenic effects of various solvent extracts from different parts of the loquat. were investigated to develope as source of functional food. The nitrite-scavenging and antimutagenic effects of fractions obtained from methanol extract were most highest in ethyl acetate fraction. Ethyl acetate fraction showed better results in the nitrite scavenging activity of 80% in leaf, 81% in seedless fruit, 73% in peel and 63% in seed. The antimutagenic effect showed strong ethyl acetate fraction of 53.1%, 48.3% in leaf and seedless fruit. The antimutagenic effect increased with increasing concentration of ethyl acetate fraction against mutagenicity induced by direct mutagen of IQ in Salmonella typhimurium TA 100. Addition of 2.5 mg and 5 mg of ethyl acetate fraction showed inhibitory effect of 65.5%, 71.2% in leaf and 63.4%, 69.2% in seedless fruit, respectively.

  • The separation and purification of 2,6-dimethylnaphthalene (2,6-DMN) present in the light cycle oil (LCO) fraction was investigated by a crystallization operation. Solute crystallization (SC) was performed using LCO fraction and iso-propyl alcohol as a raw material and a SC solvent, respectively. Increasing the operation temperature and volume ratio of the solvent to the raw material (S/F) resulted in improving the purity of 2,6-DMN, whereas the yield decreased. As a result of the crystallization operation in three steps containing the SC using LCO fraction (13.9% 2,6-DMN) and isopropyl alcohol, the re-crystallization 1 (RC 1) using the crystals recovered by SC and methyl acetate, and RC 2 using the crystals recovered by RC 1 and methyl acetate, the crystal with 99.9% 2,6-DMN was recovered with 19.5% yield. Furthermore, the separation and purification process of 2,6-DMN present in the LCO fraction was reevaluated by using the experimental results obtained through each operations of SC, RC 1, and RC 2.

Biochemical Components and Physiological Activities of Ice Plant (Mesembryanthemum crystallinum) (아이스플랜트(Mesembryanthemum crystallinum)의 생화학적 성분 및 생리활성 효과)

  • The general components and cyclitol compounds of ice plant (Mesembryanthemum crystallinum) were analyzed to examine the possibility of using extracts as functional food materials, the antioxidant effects and antidiabetic activities of the extracts by solvent fraction were tested. Among the mineral contents, contents of K and Na were the largest ($1,213.33{\pm}2.52$ and $545.53{\pm}12.01mg/100g$, respectively), followed by S, Ca, P, and Mg in order of precedence. Among cyclitol compounds, content of D-pinitol was the largest ($4.04{\pm}0.08mg/g$) while contents of chiro-inositol and myo-inositol were relatively small ($2.82{\pm}0.01$ and $0.25{\pm}0.01mg/g$, respectively). Among total phenol contents by solvent fraction, contents of chloroform and ethyl acetate fractions were large ($35.80{\pm}1.33$ and $23.70{\pm}0.62mg\;GAE/g$, respectively). Among antioxidant activity levels examined by DPPH, ABTS, FRAP, and lipid/MA assays, the chloroform fraction commonly showed the highest level of activity while the ethyl acetate fraction showed relatively high levels of activity. The antioxidant activity levels were proportional to total phenol contents by solvent fraction. As for antidiabetic effects, all solvent fractions showed at least 50% ${\alpha}-glucosidase$ inhibitory activity levels while the ethyl acetate, butanol, and chloroform fractions showed high levels activity of $90.33{\pm}0.40$, $87.98{\pm}0.16$, and $86.38{\pm}0.51%$, respectively. The ${\alpha}-amylase$ inhibitory actively levels were in the range of $25.63{\pm}1.45{\sim}60.34{\pm}2.67%$, which was lower than the ${\alpha}-glucosidase$ inhibitory activity levels, but the inhibitory activity levels by solvent fraction were similar. Given the above study results, ice plant can be utilized as a natural material with antioxidative and antidiabetic functionality.

Antioxidative Activity of Solvent Fraction from Taraxacum officinale (민들레 용매분획물의 항산화 활성)

  • Antioxidant activities and α-glucosidase inhibitory activities of Taraxacum officinale solvent fractions were measured. Extraction yields (relative to raw material) of 50% ethanol, hexane, ethyl acetate, butanol, and water were found to be 10.29, 2.61, 5.54, 2.15, and 0.96%, respectively. Polyphenol and flavonoid contents were high in ethyl acetate extract of Taraxacum officinale at 56.88 mg gallic acid/g and 33.27 mg gallic acid/g, respectively. DPPH, hydroxyl radical scavenging activity, and SOD-like activity measurement (IC50%) of Taraxacum officinale 50% ethanol extract, hexane, butanol, ethyl acetate, and water fractions were 22.64, 18.65, 10.29, 20.81, 20.46 mg/mL, 24.68, 10.69, respectively. It was found to be 9.66, 15.81, 13.77 mg/mL, 32.84, 17.09, 12.73, 33.63, and 33.91 mg/mL, and was high in the ethyl acetate layer. Results showed that α-glucosidase inhibitory activities of Taraxacum officinale solvent fraction were 25.75, 15.93, 35.87, 15.96, and 2.88% for 50% ethanol extract, hexane, butanol, ethyl acetate, and water fractions, respectively.
