The propose of this study was to investigate the antioxidant activity of 80% ethanol extracts and various solvent fractions of dandelion (Taraxacmn officinale) leaves and roots, Total phenolics and phenolic acid contents were also examined. The total phenol content of leaves and roots were $7.9{\pm}0.4%$ and $9.4{\pm}0.3%$ respectively. Eight phenolic acids were separated by GC, among which caffeic acid (113.7 mg%)and m-coumaric acid (152.6 mg) were the dominant phenolic acids in leaves and roots, respectively. Amongst solvent functions of leaves and roots, the ethyl acetate fraction showed the strongest radical scavenging activity. A strong correlation was found between total phenol content and electron-donating ability, and ABTS radical scavenging activity showed a similar trend as electron-donating ability. Hydroxyl-radical-scavenging activity and lipid peroxidation were significantly higher in the ethyl acetate fraction than other factions. In particular, the SOD-like activity was highest (43.6%) in the ethyl acetate fraction of dandelion leaves, and was higher than that of trolox. Thus, the ethyl acetate fraction of dandelion leaves exhibited significant phenol content, antioxidant activity, and free-radical-scavenging effects.

This study aims to confirm the possibility of using the invasive alien plants in Jeju as a functional biomaterial. To achieve this purpose, 70% ethanol extract and solvent fractions were prepared for five invasive alien plants (Hypochaeris radicata, Rumex acetosella, Humulus japonicus Siebold & Zucc., Solanum viarum, Lactuca scariolar) and their antioxidant, antibacterial anti-inflammatory and anti-obesity effects were investigated. The DPPH radical scavenging activity of ethanol extract from invasive alien plants was shown in the order of Rumex acetosella > Hypochaeris radicata > Humulus japonicus. Antimicrobial activity of ethanol extract against food poisoning bacteria (4 species) and oral cavity-induced microorganisms (6 species) was measured. As a result, the extract of Humulus japonicus showed high antibacterial effects against food poisoning bacteria (E. coli, V. parahaemolyticus) and oral microbes (L. casei, S. epidermidis, E. faecalis). In LPS-induced RAW 264.7 cells, the anti-inflammatory effect of ethanol extract from invasive alien plants was investigated. As a result, the NO production inhibition activity was highest in the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract, and the NO production inhibition activity was concentration-dependent. In addition, the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract showed a concentration-dependent inhibitory effect on cytokine (IL-6) production. These extracts also showed inhibitory activity of COX-2, an inflammatory protein. This suggests that NO production inhibition activity by the extract of invasive alien plants is the result of inhibition of iNOS and COX-2 expression. Currently, organic solvent fractions of crude extract are manufactured and the investigation of active ingredients is continuing along with evaluation of biological activity such as anti-inflammatory. These results are expected to be a major data for the study on the separation and utilization of active ingredients with antioxidant, antibacterial and anti-inflammatory effects using foreign plant crude extract and solvent fractions, and are highly likely to be applied to the development of functional food and cosmetics materials.
흑마늘의 기능성을 분석하기 위하여 용매별 계통 분획물을 만들어 100, 250, 500 및 $1,000\;{\mu}g/mL$ 농도에서 항산화 활성을 측정하였다. 총 페놀 및 플라보노이드 화합물의 함량은 chloroform, ethyl acetate 및 hexane 분획물에서 유의적으로 높아 butanol, methanol 및 물 분획물에 비하여 5.5~11.6배 더 많았다. 흑마늘 계통 분획물의 항산화 활성은 시료의 첨가농도가 증가함에 따라 그 활성도 증가하였으며, chloroform과 ethyl acetate 분획물의 활성이 유의적으로 높았다. DPPH 라디칼 소거능은 $1,000\;{\mu}g/mL$ 농도에서는 butanol과 물 분획물을 제외한 모든 시료 모두에서 50% 이상의 활성을 나타내었다. ABTS 라디칼 소거능의 경우 hexane, chloroform, ethyl acetate 분획물에서 비교적 활성이 높아, $1,000\;{\mu}g/mL$ 농도에서는 70% 이상의 소거능을 보였다. $1,000\;{\mu}g/mL$ 농도에서 hydroxyl radical 소거능은 50.27~81.02%의 범위로 높은 반면 SOD 유사활성은 26.73~47.64%로 비교적 그 활성이 낮았다. Linoleic acid 반응계에서 항산화 활성을 측정한 결과 저장기간이 길수록, 시료의 첨가 농도가 높을수록 항산화 활성이 더 높았으며, 또 극성 용매보다는 비극성용매 분획물의 활성이 유의적으로 높았다. 아질산염 소거능의 경우 butanol 분획물을 제외한 모든 시료에서 비교적 그 활성이 높아 $100\;{\mu}g/mL$ 농도에서도 50% 이상의 소거능을 보였다.
고추잎을 기능성식품의 재료로 이용하기 위한 방안을 모색하기 위하여 각 용매 분획물을 이용하여 생리활성 즉, 전자공여효과, 환원력, 아질산염 소거효과, 항균활성 및 tyrosinase 저해활성에 대하여 조사하였다 추출수율을 조사한 결과 물 분획층에서 16.9%로 가장 높은 추출수율을 나타낸 반면 에틸아세테이트 분획층 0.6%로 가장 낮은 추출수율을 나타내었다. 전자공여효과는 전반적으로 농도가 상승함에 따라 증가하였으며, 환원력도 분획물의 농도가 높아짐에 따라 점차적으로 증가하는 것을 볼 수 있었는데, 특히 ethyl acetate 분획물 0∼600 $\mu\textrm{g}$을 첨가하였을 때는 환원력의 증가가 크게 나타나지 않았으나, 900 $\mu\textrm{g}$이상 첨가구에서는 추출물을 첨가하지 않은 대조구와 비교하여 환원력이 매우 증가하는 것으로 나타났다. 고추잎 분획물을 농도별로 첨가하여 아질산염 소거효과를 측정한 결과 pH가 증가함에 따라 아질산염 소거효과가 감소하는 경향을 나타내었으며, ethyl acetate fraction에서 가장 높은 소거효과를 나타내어 500 $\mu\textrm{g}$을 첨가하였을 때 90%이상의 아질산염 소거효과를 나타내었으며, 농도가 증가함에 따라 아질산염 소거효과도 점차적으로 증가하는 경향을 나타내었다. 고추잎 각 분획물의 항균활성은 ethyl acetate 및 butanol 분획물에서 높게 나타났으며, 각 균주에 대한 clear zone의 직경을 확인한 결과 gram 양성균인 Bacillus cereus에서 20 mm로 항균활성이 가장 높게 나타났고, Staphylococcus aureus 18 mm, Streptecoccus mutans 17 mm순으로 주로 gram 양성균에서 높은 활성을 나타내었다. Tyrosinase 저해활성은 에틸 아세테이트 분획물에서 가장 높은 저해 활성을 나타내었다.
참모자반 분획물은 극성에 따라 핵산, 메탄올, 부탄올 및 수층 순으로 분획하여 4개의 분획층을 얻었으며, 각 분획층의 생리활성을 요약하면 다음과 같다. 항산화 활성은 DPPH radical 소거능과 SOD 활성을 측정한 결과 모두 농도 의존적으로 증가하였으며 특히 메탄올 분획 층에서 가장 높은 항산화 효과를 보였다. 아질산염 소거능 실험에서는 부탄올 층이 pH 1.2에서 57.8%로 4개의 분획물중 가장 높은 아질산염 소거능을 나타냈다. 숙취 해소능을 측정하기 위하여 ADH 활성과 ALDH 활성을 측정하였다. 대조구로 사용한 흡광도 값을 100으로 하였을 때 positive control로 사용한 hepos가 150~167%의 활성을 나타내었으며 4가지 분획층 모두 농도 의존적으로 활성이 증가하였다. 특히 ALDH에서는 메탄올 분획층이 10 mg/ml 농도에서 167.4%의 활성을 나타내어 hepos와 비슷한 효과를 보였다. 참모자반 분획물의 혈당강하 효과는 ${\alpha}$-glucosidase 활성억제 효과로 측정하였다. ${\alpha}$-glucosidase 활성억제 효과는 핵산 분획층에서 2 mg/ml의 농도에서 94.1%로 아주 높은 항당뇨 효과를 나타내었다. 그런데 부탄올층, 메탄올층 및 수층은 혈당강하효과가 거의 없는 것으로 나타났다. 피부주름예방을 측정하기 위하여 수행한 elastase 저해효과 실험에서 메탄올 분획층에서 농도 의존적으로 높은 주름개선효과가 나타났으며 2 mg/ml 농도에서 93.2%의 효과를 나타냈으며, positive control로 사용한 Vit. C (1 mg/ml) 보다 더 높은 활성을 나타냈다. 이러한 결과는 모자반 메탄올 추출물 중 특히 메탄올 분획층이 탁월한 주름개선 효과, 숙취 해소능과 항산화 효과가 있는 것으로 사료되며, 핵산층은 혈당강하 효과가 높은 것으로 나타났다. 따라서 모자반 유기용매 분획물은 천연 기능성소재로서의 사용 가능성이 매우 높을 것으로 사료된다.
A series of some novel 2-[5-(substituted phenyl)-[1,3,4]oxadiazol-2-yl]-benzoxazoles were synthesized by using benzoxazole-2-carboxylic acid on reaction with thionyl chloride in presence of ethanol solvent at room temperature gave benzoxazole-2-carbonyl chloride, which is turned into benzoxazole-2-carboxylic acid hydrazide on reaction with hydrazine hydrate in ethanol solvent under reflux. The subsequent treatment of benzoxazole-2-carboxylic acid hydrazide with an appropriate aromatic carboxylic acid in presence of polyphosparic acid under reflux afforded the title compounds. The chemical structures of the newly synthesized compounds were elucidated by their IR, $^1H$ NMR and Mass spectral data analysis. Further the compounds are used to find out their ability towards anti microbial and nematicidal activity.
Water, methanol and ethanol extracts of ginseng leaves were assayed for total phenolics and flavonoids, ascorbic acid, cupric and ferrous ion chelating activities, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, ferric reducing antioxidant power (FRAP) assay and ABTS radical cation decolourization (TEAC) assay for their antioxidant properties. The ethanol extract of ginseng leaves contained significantly (p<0.05) higher amounts of total phenolics and flavonoids (600.57 and 1701 mg/100 g) than methanol (374.43 and 1512.64 mg/100 g) and water extracts (248.30 and 680.05 mg/100 g). Among solvent extracts of ginseng leaves, the ethanol extract showed the most powerful antioxidant activities. However, the ferrous ion chelating activity of ginseng leaf extracts were lower than the cupric ion chelating ability. These differences in concentrations of key antioxidants among various solvent extracts seemed to be responsible for their differences in antioxidant activities. These results suggest that ethanol extract of ginseng leaves has the most effective antioxidant capacity compared to the methanol and water extracts tested in the present study. Thus, it can be applied for the effective extraction of functional material from ginseng leaves for the usage of pharmaceutical and/or food industries.
Water/cosolvent mixture system에 사용된 활성을 잃은 효소를 증발을 통해 재생하였다. model system으로 HRP를 이용하였을 때, 활성을 잃은 효소를 증발과 희석을 통해 용매를 제거하였을 경우, 두 경우 모두 열역학적으로 동일한 상황임에도 불구하고 증발에 의해 재생된 효소의 활성이 더 높게 나타났으며, 이는 증발이 활성을 잃은 효소의 재생에 어떤 영향을 주는 것으로 여겨진다. 형광분석법을 이용하여 효소의 구조를 간접적으로 분석하였을 때, 증발에 의한 경우가 희석에 의한 경우보다 효소의 구조적 변형이 적었음이 관찰되었다.이러한 결과들을 통해 증발에 의한 재생 효과는 구조적으로 비활성화된 효소의 활성을 일부 되살리는 기능을 하는 것으로 여겨진다. 한편 용매의 증발에 의해 재생된 효소는 초기 효소의, 유기용매 내에서의 상대적 안정성을 유지하였다.

The objective of this study was to investigate antioxidant activities of Panax ginseng extracted with various solvents including n-hexane, chloroform, EtOAC, n-butanol and water. Among the various ginseng extracts, ethyl acetate (EtOAC) extracts showed the most powerful scavenging activities against DPPH radicals. Among the other solvent extracts, the butanol extract seemed relatively more effective in scavenging activity, followed by chloroform, water and hexane extracts. Moreover, the highest reducing power and ferrous ion chelating activity were found in the EtOAC extract followed by other extracts of ginseng. EtOAC extracts, which exhibited the best antioxidant activities of all solvent extracts of ginseng, possessed higher concentrations of total phenolics (777.61 mg/100 g) than other extracts. These results suggest that EtOAC extracts of ginseng (P. ginseng C.A. Meyer) have the most effective antioxidant capacity compared to n-hexane, chloroform, n-butanol and water tested in this study, and has important applications for the pharmaceutical and food industries.
