The effects of solvent type and concentration, solid/liquid ratio, extraction time and repeated extraction on recovery of keratinase from solid-state fermentation (SSF) of chicken feather by a local Streptomyces sp. NRC 13S were investigated in order to establish the experimental conditions for keratinase yield. Among solvents tested, 0.5% (v/v) glycerol was the best. Box-Behnken design was used to investigate the effect of relevant variables on keratinase recovery. The factors investigated were solid/liquid ratio (1:1.66-1:6.66 g/mL), glycerol concentration (0.5-5% v/v) and repeated extraction (1-5 cycle). The results showed that the maximum recovery of keratinase (6933.3 U/gfs) was obtained using 0.5 (v/v) glycerol as extracting solvent, in a solid/liquid ratio of 1:5 and three extraction cycles.
지용성 용존 유기물을 해수로부터 효율적으로 추출할 수 있는 고체상 추출 (solid phase extraction)장치를 새로이 고안하였다. 본 장치는 octadecyl silane (ODS) cartridge와 압력 평형 마개, 96 multifolder, 아스피레타 형 진공 펌프로 구성하였다. 본 장치는 기존의 액체-액체 추출법보다 회수 효율은 1.4 배 이상 증대시켰으며, 최소 시료량을 8 배 이상 감소 시켰다. 따라서 용매의 사용을 5 배 이상 감소시킬 수가 있으므로 나중에 사용된 용매를 제거하는 노력도 크게 감소시켰다. 뿐만 아니라 96 개의 시료(각 1ℓ)를 동시에 처리할 수 있으므로 지용성 용존 유기물의 추출 과정을 반자동화시킨 획기적인 장치이다. 본 장치를 통하여 황해 해수 시료에서 추출된 용존 탄화수소의 정성 및 정량 분석 결과 본 장치가 효율성과 신뢰성에서 기존의 액체-액체 추출법을 대체할 수 있을 것으로 사료된다.
An analytical method for the simultaneous measurement of trace Cu, Sn, and Bi in sea water has been investigated by Inductively Coupled Plasma-Mass Spectrometry. Amberlite IRC-718 resin was used as a solid phase in solid-liquid extraction technique for the removal of matrix interferences such as Na, S, P, and other polyatomic ion species. Recoveries of 99.8% for Cu, 99.6% for Sn, and 97.9% for Bi were obtained for the standard spiked sample. The developed method was applied to analysis of trace metals in sea water.
Various parameters such as solvent selection, concentration, solid/liquid ratio, soaking time, temperature, stationary, shaking conditions, and repeated extractions were investigated in order to determine the optimum extraction conditions of ${\beta}$-glucosidase from bagasse fermented by mixed culture of Aspergillus niger NRC 7A and Aspergillus oryzae NRRL 447. Among various solvents tested, non ionic detergents gave the best results than the inorganic or organic salt solutions and distilled water. The optimum conditions for extraction of ${\beta}$-glucosidase were 30 min soaking time at $40^{\circ}C$ under shaking condition at 150 rpm, with solid/liquid ratio 1:15 (w/v), which yielded $2882.74{\pm}95.52U/g$ fermented culture (g fc) of enzyme activity. With repeated washes under the above optimum conditions, the results showed that enzyme extracted in the $1^{st}$ and $2^{nd}$ washes represents about 90% of the total activity.
An ionic liquid-based ultrasonic-assisted extraction method has been successfully applied to the effective extraction of phenolic compounds from Laminaria japonica Aresch. Three kinds of 1-alkyl-3-methyl-imidazolium with different cations and anions were evaluated for extraction efficiency. The results showed that both the characteristics of anions and cations have remarkable effects on the extraction efficiency. In addition, the ionic liquid-based ultrasonic-assisted extraction procedure was also optimized on some extraction parameters, such as ultrasonic power, extraction time and solid-liquid ratio. Compared with the conventional solvent, the optimum approach gained the highest extraction efficiency within the shortest extraction time. Average recoveries of phenolic compounds were from 75.5% to 88.3% at three concentration levels.
A study was carried out to evaluate the solid phase micro-extraction (SPME) for formaldehyde emission analysis of uncoated plywood. In SPME, formaldehyde was on-fiber derivatized through headspace extraction and analyzed by gas chromatography coupled with mass spectrometry (GC/MS). The SPME was compared with desiccators (DC-JAS 233), small-scale chamber (SSC-ASTM D6007) and liquid-liquid extraction (LLE-EPA 556) methods which were performed in accordance with their respective standards. Compared to SSC (RSD 4.3%) and LLE (RSD 5.0%), the SPME method showed better repeatability (RSD 1.8%) and not much difference from DC (RSD 1.4%). The SPME has proven to be highly precise (at 95% confidence level) with better recovery (REC 102%). Validation of the SPME method for formaldehyde quantitative analysis was evidenced. In addition, the SPME by air sampling directly from plywood specimens (SPME-W) correlated best with DC ($r^2$ = 0.983), followed by LLE ($r^2$ = 0.950) and SSC ($r^2$ = 0.935).
The study was carried out to develop and standardize the analytical method of 1,4-dioxane in water. The National Institute of Environmental Research in Korea has monitored for 1,4-dioxane in water since 1998 and it has been detected in environmental water occasionally. But the environmental pollution and analytical method in water is not well known over the world. The trace analytical method at low-${\mu}/L$ level is prerequisite in order to evaluate the residue of 1,4-dioxane in water. Evaluation of the method was demonstrated by comparing and analyzing with liquid-liquid extraction, solid-phase extraction and purge & trap technique. As a result of experiment by standard solutions and raw samples, the precision and accuracy for three methods were obtained within error rate of about 10%. Therefore, three methods were standardized as official monitoring method in Korea. Also, a convenient and simple liquid-liquid extraction method for the analysis of 1,4-dioxane in water was developed by combined with gas chromatography/mass spectrometry and applied to the water samples in Korea
기체크로마토그래피/질량분석법(GC/MS)을 이용하여 물시료 중에 미량으로 함유된 19종의 여러고리방향족탄화수소류에 대한 액체-액체 추출법, disk추출법 및 고체상 추출법을 이용한 시료 매트릭스로부터 분리, 추출하는 방법을 조사하였다. GC/MS의 오븐온도프로그램을 $80{\sim}310^{\circ}C$까지 단계적으로 변화시킨 결과 19종의 PAH류를 분리할 수 있었다. 액체-액체 추출법에 의한 물시료 중 PAH류의 회수율은 71.3~109.5% 범위로 나타났고, filtration에 의한 $C_{18}$ disk추출법의 회수율은 80.7~94.9% 범위로 높게 나타났다. 그러나 $C_{18}$ Sep-Pak을 사용한 고체상 추출법의 회수율은 51.8~77.9%범위이었다. 이상의 추출법에 대한 추출 회수율, 재현성 및 실험의 편리성 등 장단점을 비교한 결과, $C_{18}$ disk를 사용한 거르기법이 PAH류의 추출에 가장 적합한 방법임을 확인하였다. 각 추출법의 검출한계(S/N=5)는 액체-액체 추출법 및 고체상 추출법의 경우 0.25~6.25 ppb, disk추출법은 0.05~1.25 ppb 범위이었다.
미국의 환경청이 지정한 129종의 priority pollutants 중 16종의 유기살충제를 대상물질로 선정하여 수질시료 중의 극미량 분석방법을 연구하였다. 분석방법으로는 기체크로마토그래프-전자포획 및 기체크로마토그래프/질량분석기-SIM(selected ion moitoring)법을 검출방법으로 정하고, 시료용액의 추출 및 농축방법으로는 용매추출법(liquid-liquid extraction)과 고체상 추출법(solid-phase extraction)을 비교 연구하였다. 각 화합물들의 회수율, 상대표준편차 및 검출한계 등을 측정하여 EPA 분석법을 대체할 수 있는지의 가능성을 조사하였다.
수질에서 유해물질을 농축하는 방법으로 액체 추출법과 고체상 추출법을 GC/MS를 검출기로 사용하여 비교하였다. 비휘발성 유해물질 중 11종의 산과 44종의 염기/중성 화합물들을 reagent water에 첨가하여 분석한 결과, 액체추출법은 54종의 화합물에 대해 91%의 평균회수율과 4.6%의 평균상대표준편차를 나타내었으며, 고체상 추출법은 52종의 화합물에 대해 63%의 평균회수율과 8.9%의 평균상대표준편차를 보였다. 두 방법의 검출한계는 $1{\sim}5{\mu}g/l$였다.
