Sclerotinia rot symptoms were frequently found on the stems of Aster yomena in the Gurye region of Korea in April 2016. The symptom, watery soft rot, mainly appeared on the stems, and severely infected plants blighted. White mycelia spread over the stems of the infected plants and the soil surface. Small black sclerotia formed on the plant lesions and inside the diseased stems. Incidence of the disease was as high as 20~80% in the A. yomena fields. Based on the morphological and molecular characteristics of the isolates, the fungi were identified as Sclerotinia minor. This is the first report of Sclerotinia rot caused by Sclerotinia minor on A. yomena in Korea.
Climate in the year of 2000 was characterized as a long severe drought in tile spring, unusually high and low temperature in summer, two times of typoons, and floods by heavy rains in fall. Rice leaf and panicle blast and bacterial grain rot occurred severely comparing with 1999 and Bipolaris leaf spot spread over tile country. Phytophthora blight and anthracnose in red-pepper became epidemic especially in the late season causing severe yield losses. Tomato fusaruim wilt, CGMMV, powdery mildew, and sudden wilt syndrom of cucurbits and strawberry powdery mildew were also severe in 2000. In garlic, sclerotium rot occurred severely mainly due to the frequent rainfalls in planting time and much snowfalls in 1999's winter. Spring potato had severe infection of viruses due to a long spring drought, and fall potato had high incidence of bacterial soft rot and bacterial wilt due to fall floods by heavy rains. In sweet potato fusarium wilt was the most severe as in other year. Disease incidence of apple and pear trees was rotatively mild compared with previous years. In wheat and barley, Gibberella petch rarely occurred because of spring drought.
The year of 2002 was very unusual in climatic condition. Warm winter weather, high temperature and drought in March and April, little precipitation in June and early and mid-July and, nearly continuous rain-falls from late July to whole period of August accompanied with low temperature and insufficient sunshine in 2002 resulted in unfavorable conditions for plant growth in one way or another, In relation to the unusual weather, in general, diseases associated with low temperature, poor radiation and much rainfall occurred severely in this year, In rice, incidence of bacterial grain rot, and false smut increased sharply, whereas other major diseases including blast occurred mildly due mainly to the reduced application of nitrogen fertilizer, In vegetables, potato and sweet potato, and fruit trees, incidence of diseases was largely dependent on climatic condition of growth period in particular crops. However, diseases favored low temperature and much rainfall, such as Phytophthora disease increased markedly compared to those of normal years, while anthracnose and soft rot tended to decrease significantly.
In the 1970s and 1980s, during the nascent phase of ginseng disease research, efforts concentrated on isolating and identifying pathogens. Subsequently, their physiological ecology and pathogenesis characteristics were scrutinized. This led to the establishment of a comprehensive control approach for safeguarding major aerial part diseases like Alternaria blight, anthracnose, and Phytophthora blight, along with underground part diseases such as Rhizoctonia seedling damping-off, Pythium seedling damping-off, and Sclerotinia white rot. In the 1980s, the sunshade was changed from traditional rice straw to polyethylene (PE) net. From 1987 to 1989, focused research aimed at enhancing disease control methods. Notably, the introduction of a four-layer woven P.E. light-shading net minimized rainwater leakage, curbing Alternaria blight occurrence. Since 1990, identification of the bacterial soft stem rot pathogen facilitated the establishment of a flower stem removal method to mitigate outbreaks. Concurrently, efforts were directed towards identifying root rot pathogens causing continuous crop failure, employing soil fumigation and filling methods for sustainable crop land use. In 2000, adapting to rapid climate changes became imperative, prompting modifications and supplements to control methods. New approaches were devised, including a crop protection agent method for Alternaria stem blight triggered by excessive rainfall during sprouting and a control method for gray mold disease. A comprehensive plan to enhance control methods for Rhizoctonia seedling damping-off and Rhizoctonia damping-off was also devised. Over the past 50 years, the initial emphasis was on understanding the causes and control of ginseng diseases, followed by refining established control methods. Drawing on these findings, future ginseng cultivation and disease control methods should be innovatively developed to proactively address evolving factors such as climate fluctuations, diminishing cultivation areas, escalating labor costs, and heightened consumer safety awareness.
Cobweb disease symptoms were observed in a mushroom farm in Buye, Korea during a disease survey in 2008-2011. Five isolates of Cladobotryum sp. were obtained from the infected caps and stipes. These isolates of Cladobotryum sp. were identified as C. mycophilum based on their morphological, cultural characteristics and analysis of the ITS sequences. Early symptoms were noticed as round, fleshy, yellowish brown lesions on mushroom caps. Late symptoms progressed when the parasitic fungus formed white cobweb circular colonies on dead or damaged pinheads, spread on the surface of the casing, and covered entirely fruiting bodies. Optimal temperature and pH for mycelial growth on MEA is $23^{\circ}C$ and 6.0. Microscopically the spores of the fungus are large and most 2~3 celled produced on vertically branched conidiophores. Mushroom caps turned dark brown and shrunk due to soft rot. Testing of sensitivity to selected fungicides showed that isolate was highly resistance to Mancozeb and Thiophanate-methyl, moderately sensitivity to Iprodione, and highly sensitivity to Benomyl, Prochloraz-Mn and Carbendazim.
Pectobacterium, which causes soft rot disease, is divided into 18 species based on the current classification. A total of 225 Pectobacterium strains were isolated from 10 main cultivation regions of potato (Solanum tuberosum), napa cabbage (Brassica rapa subsp. pekinensis), and radish (Raphanus sativus) in South Korea; 202 isolates (90%) were from potato, 18 from napa cabbage, and five from radish. Strains were identified using the Biolog test and phylogenetic analysis. The pathogenicity and swimming motility were tested at four different temperatures. Pectolytic activity and plant cell-wall degrading enzyme (PCWDE) activity were evaluated for six species (P. carotovorum subsp. carotovorum, Pcc; P. odoriferum, Pod; P. brasiliense, Pbr; P. versatile, Pve; P. polaris, Ppo; P. parmentieri, Ppa). Pod, Pcc, Pbr, and Pve were the most prevalent species. Although P. atrosepticum is a widespread pathogen in other countries, it was not found here. This is the first report of Ppo, Ppa, and Pve in South Korea. Pectobacterium species showed stronger activity at 28℃ and 32℃ than at 24℃, and showed weak activity at 37℃. Pectolytic activity decreased with increasing temperature. Activity of pectate lyase was not significantly affected by temperature. Activity of protease, cellulase, and polygalacturonase decreased with increasing temperature. The inability of isolated Pectobacterium to soften host tissues at 37℃ may be a consequence of decreased motility and PCWDE activity. These data suggest that future increases in temperature as a result of climate change may affect the population dynamics of Pectobacterium.

Soft rot is a widespread, catastrophic disease caused by Pectobacterium carotovorum subsp. carotovorum (Pcc) that severely damages the production of Amorphophallus spp. This study evaluated the rhizosphere bacterial and fungal communities in Pcc-infected and uninfected plants of two species of Amorphophallus, A. muelleri and A. konjac. Principal component analysis showed that the samples formed different clusters according to the Pcc infection status, indicating that Pcc infection can cause a large number of changes in the bacterial and fungal communities in the Amorphophallus spp. rhizosphere soil. However, the response mechanisms of A. muelleri and A. konjac are different. There was little difference in the overall microbial species composition among the four treatments, but the relative abundances of core microbiome members were significantly different. The relative abundances of Actinobacteria, Chloroflexi, Acidobacteria, Firmicutes, Bacillus, and Lysobacter were lower in infected A. konjac plants than in healthy plants; in contrast, those of infected A. muelleri plants were higher than those in healthy plants. For fungi, the relative abundances of Ascomycota and Fusarium in the rhizosphere of infected A. konjac plants were significantly higher than those of healthy plants, but those of infected A. muelleri plants were lower than those of healthy plants. The relative abundance of beneficial Penicillium fungi was lower in infected A. konjac plants than in healthy plants, and that of infected A. muelleri plants was higher than that of healthy plants. These findings can provide theoretical references for further functional research and utilization of Amorphophallus spp. rhizosphere microbial communities in the future.

Pseudomonas syringae pv. tomato (Pst) causes a bacterial speck disease in tomato and Arabidopsis. In Chinese cabbage, in which host-pathogen interactions are not well understood, Pst does not cause disease but rather elicits a hypersensitive response. Pst induces localized cell death and $H_2O_2$ accumulation, a typical hypersensitive response, in infiltrated cabbage leaves. Pre-inoculation with Pst was found to induce resistance to Erwinia carotovora subsp. carotovora, a pathogen that causes soft rot disease in Chinese cabbage. An examination of the expression profiles of 12 previously identified Pst-inducible genes revealed that the majority of these genes were activated by salicylic acid or BTH; however, expressions of the genes encoding PR4 and a class IV chitinase were induced by ethephon, an ethylene-releasing compound, but not by salicylic acid, BTH, or methyl jasmonate. This implies that Pst activates both salicylate-dependent and salicylate-independent defense responses in Chinese cabbage.
Erwinia rhapontici causes soft-rot disease in a number of plants such as rhubarb, onion, hyacinth and garlic. Pectate lyase (Pel) depolymerizes pectin and other polygalacturonates, which is though to play a role in bacterial invasion of plants. Pel activity was not detected in E. rhapontici cultured in a minimal salts medium containing glycerol, polygalacturonate, or citrus pectin as a carbon source. However, when sterilized potato tuber and Chinese cabbage slices were added to minimal salts polygalacturonate (0.5%) medium, E. rhapontici produced pectate lyase enzyme. Also Pel activity was consistently detected from macerated potato tubers, Chinese cabbage leaves, lettuce leaves and celery petioles tissue. Pel in the extract of macerated Chinese cabbage caused by E. rhapontici strain 1, resulted in electrolyte loss, tissue maceration and cell death of potato tuber tissue. These results indicate that E. rhapontici produces pectate lyase only in the presence of non-diffusible plant components, and that this enzyme probably contributes to its pathogenicity.
Biocontrol microbes have mainly been screened among large collections of microorganisms $via.$ nutrient-rich $in$$vitro$ assays to identify novel and effective isolates. However, thus far, isolates from only a few genera, mainly spore-forming bacilli, have been commercially developed. In order to isolate field-effective biocontrol microbes, we screened for more than 200 oligotrophic bacterial strains, isolated from rhizospheres of various soil samples in Korea, which induced systemic resistance against the soft-rot disease caused by $Pectobacterium$$carotovorum$ SCC1; we subsequently conducted in $planta$ bioassay screening. Two oligotrophic bacterial strains were selected for induced systemic disease resistance against the $Tobacco$$Mosaic$$Virus$ and the gray mold disease caused by $Botrytis$$cinerea$. The oligotrophic bacterial strains were identified as $Pseudomonas$$manteilii$ B001 and $Bacillus$$cereus$ C003 by biochemical analysis and the phylogenetic analysis of the 16S rRNA sequence. These bacterial strains did not exhibit any antifungal activities against plant pathogenic fungi but evidenced several other beneficial biocontrol traits, including phosphate solubilization and gelatin utilization. Collectively, our results indicate that the isolated oligotrophic bacterial strains possessing induced systemic disease resistance could provide useful tools as effective biopesticides and might be successfully used as cost-effective and preventive biocontrol agents in the field.
