Purification and Some Properties of Peroxidase from the Fruit Malus sieboldii (Regel) Rehder (아그배 Peroxidase의 정제 및 특성)

  • Peroxidase in the fruit of Malus sieboldii (Regel) Rehder was partially purified by DEAE-cellulose column chromatography and Ultro-AcA 54 gel filtration. The optimum pH of peroxidase was 4.5 and optimum temperature was $80^{\circ}C$. The enzyme was stable at pH 5.0 and below $30^{\circ}C$, and inactivated by heat treatment at $80^{\circ}C$ for 15min. In the presence of 30mM $H_{2}O_2$ Km value on o-phenylenediamine as substrate was 1.65mM, and in the presence of 10mM o-phenylenediamine Km value on $H_{2}O_2$ was 7.97mM. L-Ascorbic acid and sodium L-ascorbate greatly inhibited the enzyme activity and among several metal ions $Mn^{2+}$ only increased the activity at 5mM.

  • The synthesis and characterization of silver colloidal nanoparticles by chemical reduction of silver ions in aqueous $AgNO_3$ using sodium borohydride $(NaBH_4)$ as the reducing agent are described. The experimental conditions for aggregation and paricle size of nanosilver particles in water is investigated in terms of concentration of $NaBH_4$, reaction temperature, dropping rate of $AgNO_3$ and concentration of laponite. Stable nanosilver sol is obtained at three molar ratio of $NaBH_4/AgNO_3$ in conditions of without laponite. The size of nanosilver particles is increased as the reaction temperature is increased. The large size of nanosilver sol is obseved as the dropping rate of $AgNO_3$ is increased due to the aggregation of initial high local concentration of nanosilver particles. Stable nanosilver sol at high temperature $(>\;100^{\circ}C)$ can be prepared when laponite is used as protective colloid.

Corrosion Failure Analysis of the Convection Part of District Heating Peak Load Boiler (지역난방 첨두부하보일러 대류부 부식 파손 분석)

  • Corrosion failure in the convection part of peak load boiler (PLB) of the district heating system led to water leakage. Herein, Internal Rotary Inspection System (IRIS) inspection was employed to examine wall thinning and the cause of leakage in the flue tube. The corrosive products of the turbulator and tube were investigated using scanning electron microscope combined with energy dispersive spectroscopy, X-ray diffraction, and inductively coupled plasma (ICP). Majority of the serious corrosion damage was observed near the turbulator located in the upper flue tube. ICP analysis of the boiler water revealed oxide formation of sodium chloride in the lower end part of the flue tube. A cross-sectional view of the turbulator revealed the presence of double-layers of the oxide film, indicating environmental change during operation associated with water leakage. The outer surface of the turbulator consisted of the acid oxides such as $NO_x$ and $SO_x$ along with sodium and chloride ions. Dew-point corrosion is hypothesized as the main cause for the formation of acid oxides in the region of contact of the flue tube and the turbulator.

Effect of gamma irradiation on the size of cellulose nanocrystals with polyethylene glycol and sodium hydroxide/Gd2O3 nanocomposite as contrast agent in magnetic resonance imaging (MRI)

  • The attractive properties of gadolinium-based nanoparticles as a positive contrast agent for magnetic resonance imaging (MRI) have piqued the interest of both researchers and clinicians. Nonetheless, due to the biotoxicity of gadolinium (III) ions' free radicals, there is a need to address this issue. Therefore, this research aimed to develop a biocompatible, dispersible, stable, hydrophilic, and less toxic cellulose nanocrystals/gadolinium oxide nanocomposite as contrast agent properties for MRI purposes. This study aimed to synthesize gadolinium oxide nanoparticles coated with cellulose nanocrystals with polyethylene glycol and sodium hydroxide (CNCs-PEG/NaOH)/Gd2O3 using the gamma irradiation method to reduce the particle size. The results showed that using a gamma irradiation dose of 10 kGy, quasi-spherical morphology with a size of approximately 5.5 ± 0.65 nm could be produced. Furthermore, the cytocompatibility of (CNCs-PEG/NaOH)/Gd2O3 nanocomposite synthesized was assessed through MTT assay tests on Hep G2 cells, which demonstrated good cytocompatibility without any cytotoxic effects within a concentration range of (10 ㎍/mL - 150 ㎍/mL) and had sufficient cellular uptake. Moreover, the T1-weighted MRI of (CNCs-PEG/NaOH)/Gd2O3 nanocomposite revealed promising results as a positive contrast agent. It is envisaged that the gamma irradiation method is promising in synthesizing (CNCs-PEG/NaOH)/Gd2O3 nanocomposite with nanoscale for different applications, especially in the radiotherapy field.

Metabolomic profiling of embryo culture media in patients with repeated implantation failure during assisted reproductive technology cycles

  • Objective: This study investigated the metabolic status of the spent culture media from embryos of patients with repeated implantation failure (RIF) undergoing in vitro fertilization-intracytoplasmic sperm injection cycles in comparison with the embryos from healthy fertile women. Methods: Metabolite levels in spent culture media were assessed and compared between embryos from RIF patients (n=35) and oocyte donors as controls (n=15). Protein levels of insulin-like growth factor 1 (IGF-1) were determined using Western blotting. Concentrations of glucose, pyruvate, and lactate were measured using spectrophotometry. Ionic colorimetric assay kits were utilized to analyze the concentrations of sodium, chloride, calcium, and magnesium ions. High-performance liquid chromatography was employed to measure the concentrations of glutamic acid, aspartic acid, methionine, phenylalanine, and histidine. Results: Glucose consumption and lactate secretion were higher in the control group than in the RIF group. The magnesium concentration was significantly higher in the control group than in the RIF group, but glutamic acid and aspartic acid concentrations were lower in the control group than in the RIF patients (p<0.05). The levels of IGF-1, sodium, calcium, chloride, methionine, histidine, and phenylalanine did not show statistically significant differences between the two groups. Conclusion: The metabolic profile of the culture medium of the embryos in the RIF group differed from that of the control group. These findings suggest potential factors that may affect implantation capacity in RIF patients and provide a new perspective on embryo selection.

Effects of Magnesium and Sulfate Ions on the Sulfate Attack Resistance of Alkali-activated Materials (알칼리 활성화 결합재 모르타르의 황산염 침식 저항성에 미치는 마그네슘 및 황산 이온의 영향)

  • The purpose of this study is to investigate the effect of sulfate (${SO_4}^{2-}$) and magnesium ($Mg^{2+}$) ions on sulfate resistance of Alkali-activated materials using Fly ash and Ground granulated blast furnace slag (GGBFS). In this research, 30%, 50% and 100% of GGBFS was replaced by sodium silicate modules ($Ms(SiO_2/Na_2O)$, molar ratio, 1.0, 1.5 and 2.0). In order to investigate the effects of $Mg^{2+}$ and ${SO_4}^{2-}$, compression strength, weight change, lengh expansion of the samples were measured in 10% sodium sulfate ($Na_2SO_4$), 10%, 5% and 2.5% magnesium sulfate ($MgSO_4$), 10% magnesium nitrate ($Mg(NO_3)_2$), 10% [magnesium chloride ($MgCl_2$) + sodium sulfate ($Na_2SO_4$)] and 10% [magnesium nitrate $(Mg(NO_3)_2$ + sodium sulfate ($Na_2SO_4$)] solution, respectively and X-ray diffraction analysis was conducted after each experiment. As a result, when $Mg^{2+}$ and ${SO_4}^{2-}$ coexist, degradation of compressive strength and expansion of the sample were caused by sulfate erosion. It was found that the reaction of $Mg^{2+}$ with Calcium Silicate Hydrate (C-S-H) occurred and $Ca^{2+}$ was produced. Then the Gypsum ($CaSO_4{\cdot}2H_2O$) was formed due to reaction between $Ca^{2+}$ and ${SO_4}^{2-}$, and also Magnesium hydroxide ($Mg(OH)_2$, Brucite) was produced by the reaction between $Mg^{2+}$ and $OH^-$.

  • In order to evaluate the utility of the anthocyanins of Amaranthys tricolor L. as an edible pigment, the present study was undertaken to investigate the effects of pH. temperature, ascorbic acid, sugars and their degradation products, quercetin, thiourea, sodium pyrophosphate and metal ions on the stability of the anthocyanins in the model systems. The results obtained from this study were as follows. 1. The degradation of total anthocyanins was retarded as the pH levels decreased from 8.0 to 1.0. At pH 1.0, however. the initial degradation reaction proceeded faster than at pH 2.0 to 3.0 2. On heating in buffered aqueous solution at $80^{\circ}C$, the total anthocyanin content was higher at pH 2.0 than other pH levels. Increasing the storage temperature accelerated greatly the pigment degradation. In darkness at $40^{\circ}C$, after 10 days, only 19% of the original amount was left, while at $2^{\circ}C$, under the same conditions of storage, approximately 90% of the pigment was retained. The half-life of the pigment, 63.0 days at $2^{\circ}C$, shortened to 1. 7 days at $40^{\circ}C$. 3. An increase in ascorbic arid concentration from 0. 15 to 0.50 mg/ml lowered the anthocyanin retention. 4. There was no significant difference between glucose and fructose in anthocyanin degradation effect. Furfural was more effective than other sugar degradation products, formic acid or levulinic acid in accelerating anthocyanin breakdown. 5. Neither quercetin nor sodium pyrophosphate had a protective effect on the anthocyanins in the presence of ascorbic acid, while, in the systems 0.5 or 1 mg/ml of thiourea with $150{\;}{\mu}g/ml$ of ascorbic acid, the loss of anthocyanins was significantly reduced. 6. Both mercuric and cupric ions in 30 ppm greatly accelerated anthocyanin degradation.

Sensing NO3-N and K Ions in Hydroponic Solution Using Ion-Selective Membranes (이온선택성 멤브레인을 이용한 양액 내 질산태 질소 및 칼륨 측정)

  • Rapid on-site sensing of nitrate-nitrogen and potassium ions in hydroponic solution would increase the efficiency of nutrient use for greenhouse crops cultivated in closed hydroponic systems while reducing the potential for environmental pollution in water and soil. Ion-selective electrodes (ISEs) are a promising approach because of their small size, rapid response, and the ability to directly measure the analyte. The capabilities of the ISEs for sensing nitrate and potassium in hydroponic solution can be affected by the presence of other ions such as calcium, magnesium, sulfate, sodium, and chloride in the solution itself. This study was conducted to investigate the applicability of two ISEs consisting of TDDA-NPOE and valinomycin-DOS PVC membranes for quantitative determinations of $NO_3$-N and K in hydroponic solution. Nine hydroponic solutions were prepared by diluting highly concentrated paprika hydroponic solution to provide a concentration range of 3 to 400 mg/L for $NO_3$-N and K. Two of the calibration curves relating membrane response and nutrient concentration provided coefficients of determination ($R^2$) > 0.98 and standard errors of calibration (SEC) of < 3.79 mV. The use of the direct potentiometry method, in conjunction with an one-point EMF compensation technique, was feasible for measuring $NO_3$-N and K in paprika hydroponic solution due to almost 1:1 relationships and high coefficients of determination ($R^2$ > 0.97) between the levels of $NO_3$-N and K obtained with the ion-selective electrodes and standard instruments. However, even though there were strong linear relationships ($R^2$ > 0.94) between the $NO_3$-N and K concentrations determined by the Gran's plot-based multiple standard addition method and by standard instruments, hydroponic $NO_3$-N concentrations measured with the ISEs, on average, were about 10% higher than those obtained with the automated analyzer whereas the K ISE predicted about 59% lower K than did the ICP spectrometer, probably due to no compensation for a difference between actual and expected concentrations of standard solutions directly prepared.

Groundwater Quality Characteristics of Pollution Concerned Area in Gyeongnam Using Groundwater Quality Monitoring Data (지하수수질측정망 자료를 활용한 경남 오염우려지역의 지하수 수질 특성)

  • This study analyzed the groundwater quality characteristics according to the main source of pollution and quarter (season) by using data from the pollution exclusive monitoring network in the Gyeongsangnam-do area for five years (2013-2017). The main source of pollution was the industrial complex areas, waste mines, and sewage treatment facilities. The analysis items were field measurement items (water temperature, pH, electrical conductivity, dissolved oxygen, oxide reduction potential), positive ions, and negative ions. Water temperature and pH did not vary significantly according to the main source of pollution. In industrial complex areas, the value of electrical conductivity was the highest, and dissolved oxygen value was the lowest. The mean concentration of positive and negative ions was the largest in industrial complex areas, followed by sewage treatment facilities and waste mines. It was shown that the concentration of sodium ion was the highest in industrial complex areas and calcium ion in waste mines and sewage treatment facilities. The concentration of bicarbonate ion was the highest in all main sources of pollution. Water temperature, pH, and concentrations of cations and anions did not vary significantly from quarter to quarter. Of the water quality types, the Na-HCO3 type accounted for the highest proportion, but the Na-Cl type, which has a high possibility of external contamination, accounted for about 20% of the total data in the pollution exclusive monitoring network.

A Study on Conservation and Desalination for Iron Weapons During the Korean War from DMZ (비무장지대 한국전쟁 전사자 유해발굴 수습 철제 총기류의 보존처리와 탈염처리 방법 고찰)

  • The weapons excavated from the de-militarized zones (DMZ) of Korea are vulnerable to corrosion due to the immediate and drastic environmental change. Especially, the chloride ions (Cl-) in iron weapons cause active corrosion and require removal. In this study, conservation treatment and de-salination was performed for the discovered weapons from excavation sites of soldiers killed in action during the Korean War. Furthermore, an attempt was made to prepare the most stable plan for conservation treatment through the comparative study of soaking weapons in distilled water without chemicals and in a solution of sodium (SSC) at different temperatures. In the preliminarily experiments, the comparison of the eluted Cl- ions according to different conditions of de-salination showed that the highest number of ions were detected from the de-salination with SSC at a temperature of 100℃, and its duration was much smaller, i.e., 1~2 weeks. Accordingly, for the parts from the guns and rifles amongst other objects, a six-time de-salination was conducted in the SSC solution for 8 hours at 100℃ and subsequently, for 16 hours at room temperature during which the distilled water and SSC were exchanged every week. However, in the case of a loaded rifle, the de-salination was not conducted, considering the risk that the high temperature and pressure by impregnation in vacuum could cause an explosion
