  • Inner shell of chestnut (Castanea Mollissima, Fagaceae) is well-known anti-wrinkle agent, which has been used for long time in the treatment of skin aging. In this study, the extract of chestnut inner shell (CMIE) and its major component, ellagic acid (EA), were studied for their protective effects against free radical generation and hydrogen peroxide-induced oxidative DNA damages in the mammalian cells.(omitted)

  • 피부 이미지의 여러 가지 특징들 중 주름은 피부의 상태를 판단하는 중요한 요소이다. 따라서 주름을 추적하기 위해 확대경으로 촬영된 원본 이미지에서 질감 대비 증가, 노이즈 제거 등의 전처리 과정을 수행한 후 Watershed 알고리즘을 이용하여 주름을 선분으로 표현하였다. 이렇게 생성된 주름의 깊이, 너비, 길이 등은 피부 분석 시 특징 정보로 이용할 수 있다. 또한 주름과 주름이 연결되어 이루는 다각형을 논문에서는 셀(Cell)이라고 정의하는데 그것의 크기나 개수 같은 정보도 추출할 수 있게 된다. 그러나 주름으로 만들어진 셀들은 실제와 다르게 과분할 되는 경향을 보인다. 과분할 된 셀들은 잘못된 정보를 제공하기 때문에 피부 상태를 판단하는 결과의 정확도를 떨어뜨린다. 본 논문에서는 이러한 문제점을 인지하고 차후 정확한 셀 정보를 획득하기 위한 확장성 측면에서 각 셀들을 개체화시키고 과분할 된 셀을 검출하는 방법을 제안한다.

Anti-wrinkle Activity of a Curdrania tricuspidata Extract on Ultraviolet-induced Photoaging (Hairless Mice를 이용한 광노화 모델에서 꾸지뽕나무 추출물의 피부주름 형성 억제효과)

  • We investigated the anti-wrinkle activity of an 80% ethanol extract of Curdrania tricuspidata leaves (CTL80) on ultraviolet-induced photoaging in hairless mice. Skin wrinkles were induced by 10 weeks of UVB-irradiation on the back of Skh-1 hairless mice three times a week. Mice were divided into ten groups; normal control (-UVB), UVB irradiated control group (+UVB), dietary groups (UVB+ascorbic acid 0.1%, UVB+CTL80 0.1%, UVB+CTL80 0.25%) and topical application groups (-UVB+base lotion (BL), UVB+BL, UVB+ascorbic acid 1%+BL, UVB+CTL80 1%+BL, UVB+CTL80 2%+BL). Wrinkle formation, histological changes, superoxide dismutase (SOD) activities, glutathione peroxidase (GSH-Px), and the expression of matrix metalloproteinases (MMP-1, MMP-3 and MMP-9) were analyzed. Wrinkles for the +UVB groups formed as a pattern of deep furrows and thick crests. Wrinkles with CTL80 treatment formed as a pattern of shallow furrows and thin crests, with wrinkle areas were lower than the +UVB group. In an antioxidant analysis of mouse blood, SOD and GSH-Px activities were significantly higher in the CTL80 topical application group compared to the +UVB group. The mRNA expression of MMPs in the +UVB group was significantly higher than the normal control group, and significantly lower in the CTL80-treated group. In conclusion, CTL80 exerted anti-wrinkle activity on ultraviolet-induced photoaging by regulating antioxidative defense systems and MMPs expression.

Characterization as Cosmetic Substances of Chestnut Inner Skin Extracts with Antioxidant Activity (항산화 활성을 갖는 율피 추출물의 향장 소재 특성)

  • We investigated characterization as cosmetic substances of chestnut inner skin extracts with in vitro antioxidant activity. Total phenolics of various extracts from chestnut inner skin were the highest 60% methanol (164.82 mg/g), and ethyl acetate fractions (191.14 mg/g). We found that the both samples from chestnut inner skin dose-dependently increased in vitro antioxidant activities (DPPH radical scavenging activity and inhibition of lipid peroxidation). In addition, the both samples also showed a strong UV absorption in the range of UV-B (290-320 nm). Especially the 60% ethanol extracts presented higher inhibitory effect on elastase (46.40% at $100{\mu}g/mL$) than that of the ethyl acetate fractions, so that it showed in vitro anti-wrinkle activity. Finally, the 60% methanol extracts and ethyl acetate fractions showed anti-bacterial activity against skin pathogenic bacteria. Consequently, these results suggest that the chestnut inner skin can be used for cosmetic industry.

Effect of Oral Administration of Pineapple Fruit Extract Containing Glucosylceramide on Skin Barrier Function Improvement in Animal Model of Atopic Dermatitis (글루코실세라마이드 함유 파인애플과실추출물의 경구 투여가 아토피 피부염 동물모델의 피부 장벽기능 개선에 미치는 효과)

  • Glucosylceramides (GluCer) are known to play an important role in both water retention and epidermal permeability barrier function in the mammalian stratum corneum. In this study, we investigated the effects of pineapple fruit extract containing glucosylceramides (PFEG) on the maintenance and recovery of skin barrier function using atopic dermatitis-induced animal models. Five-week-old male Hos:HR-1 mice were divided into four groups fed on standard diet, unsaturated fatty acids-deficient (HR-AD) diet, and HR-AD diet supplemented with 0.01% or 0.1% pineapple-GluCer. Skin barrier function was evaluated by transepidermal water loss (TEWL), dermal moisture content, moisture content of the stratum corneum and wrinkle formation. The control group (HR-AD administration group) showed increased transepidermal water loss (TEWL), while the epidermal moisture content and the moisture content of the stratum corneum slowly decreased. However, in the PFEG groups (with 0.1% or 0.01% glucosylceramide), the TEWL levels were significantly reduced at 2 weeks. The PFEG also helped maintain skin moisturizing function by significantly suppressing the decrease of the epidermal moisture content and the moisture content of the stratum corneum. These results show that the PFEG is effective for maintaining and improving the function of the skin barrier. Therefore, this study suggests that PFEG is a potential candidate material for skin functional foods.

Effect of 640 nm LED Irradiation and Aromatherapy on Ameliorating Neck Skin Condition (640 nm LED 조사와 아로마테라피가 목 피부 상태 개선에 미치는 영향)

  • The purpose of this study was to systematically evaluate and compare the effects of 640 nm LED irradiation and aromatherapy on the skin condition of the neck. Twenty female participants were divided into Group A (640 nm LED irradiation + aromatherapy) and Group B (640 nm LED irradiation only). Total of 8 experiments were conducted on the 2 groups twice a week. As a result of the experiment, moisture content was significantly improved in both group A (p<0.01) and group B (p<0.001), and wrinkle index was significantly improved in both group A (p<0.01) and group B (p<0.05). Additionally, the change in skin density was significantly improved in both group A (p<0.001) and group B (p<0.05), and the change in elasticity was significantly improved in both group A (p<0.001) and group B (p<0.001) as well. However, the change in skin tone was not significant in group A (p>0.05), but the change was drastically improved in group B (p<0.05). Blemishes and pigmentation changes were significantly improved in group A (p<0.05), but was not significant in group B (p>0.05). These results conclude that combining aromatherapy with 640 nm LED irradiation can be highly effective in improving skin condition of the neck.

A Study on functional cosmetic ingredients of the invasive plant Spartina anglica (생태교란종 영국갯끈풀의 기능성화장품 원료로서 효능 연구)

  • The Spartina anglica is recognized as a highly invasive plant and active eradication methods are required. In this study, we aimed to determine the physiologic activities of Spartina anglica extracts as a cosmetic ingredient. Antioxidant properties were investigated by different chemical methods including radical quenching (DPPH and ABTS), reducing power(FRAP) assay and aerial part of S. anglica(SAA) extract presented the strongest antioxidant activities. The significant cytotoxicity was not observed up to a concentration of 0.5 mg/mL in RAW264.7 cells and NHDF cells. The anti-inflammatory activity of S. anglica belowground(SAB) extract had strong effects on cell-based systems, including LPS-induced NO and cytokines(TNF-a and IL-6) production in RAW264.7 cells. Collagen synthesis and skin hydration gene expression of S. anglica extract showed the highest anti-wrinkle and moisturizing effect in NHDF cells. Results presented in this study tend to show that the ethanol extracts of S. anglica could be exploited as useful-bio-resource for bioactive substances in functional cosmetics.

Functional Cosmetic Effect of Sargassum Extracts (모자반추출물의 기능성 화장품소재 특성)

  • There is growing interest in natural cosmetic ingredients as natural cosmetics become popular. As a part of effort to look for natureal cosmetic agent, seaweed Sargassums were tested for functional cosmetic agents. Effective materials were extracted from Sargassum coreanum. Sargassum hemiphyllum, and Sargassum patens by simple hot water extraction. Antioxidation, whitening, anti-wrinkle, UV absorption and anti-inflammation effects were studied for functional cosmetic agents. Sargassum extracts indicated excellent cell viability, strong anti-oxidation effect by DPPH radical scavenging activity and showed significant whitening effect from tyrosinase inhibition. However, effects of antiwrinkle, UV absorption and anti-inflammation were negligible. In conclusion, Sargassum coreanum extracts showed good possibility for anti-oxidation and whitening cosmetic agent.

Antioxidant activity and MMP-1 inhibitory activity of Panax Ginseng Sprout Extracts (새싹인삼 추출물의 항산화 활성 및 MMP-1 저해 활성 )

  • Objectives : As a substitute for high-price ginseng, this study attempted to examine a possibility of the ferment extract of Panax ginseng sprout whether leaves and roots can be used together as a cosmetic ingredient with anti-oxidative and wrinkle-care effects. Methods : In terms of a test method, antioxidant activities were confirmed through total polyphenol contents, total flavonoid contents, DPPH radical scavenging activity and ABTS radical scavenging activity using the Panax ginseng sprout. In addition, to assess wrinkle-care effectiveness, the cytotoxicity of the extract was analyzed through MTT assay, and inhibition of collagenase activity in the cells was tested using the Panax ginseng sprout fermented by Saccharomyces cerevisiae. Resuits : The content of polyphenols and flavonoids in natural plants was highest in Panax Ginseng Sprout Extract at 100℃, which also demonstrated high DPPH, ABTS radical scavenging activity. MTT assay demonstrated that the Panax Ginseng Sprout Ferment Extract did not have a cytotoxic effect in CCD-986SK cell. Also, Panax Ginseng Sprout Ferment Extract was found to inhibit MMP-1 expression by 51.85±6.09% at a concentration of 10%. Conclusions : Therefore, this study has confirmed a possibility of Panax ginseng sprout ferment extract as a cosmetic ingredient with MMP-1-inhibitory effects.

Formulation Design and Evaluation of Ursolic Acid Microemulsion Delivery System for Topical Formulation (마이크로에멀젼을 이용한 우르솔릭산 피부 적용제제의 설계 및 평가)

  • Ursolic acid (UA), a bioactive triterpene acid, has been known to increase collagen content in human skin in addition to other actions such as anti-inflammatory, skin-tumor prevention and anti-invasion. However, it is poorly soluble in water. Therefore, we firstly prepared microemulsion system with benzyl alcohol, ethanol and Cremophor EL, RH 40 and Brij 35 as surfactant in order to increase solubility of UA and then prepared microemulsion was dispersed in o/w cream base for the topical delivery of UA in an effort to improve anti-wrinkle effect. The pseudo-ternary phase diagrams were developed and various microemulsion formulations were prepared using benzyl alcohol as an oil, Cremophor EL, RH 40 and Brij 35 as a surfactant. The droplet size of microemulsions was characterized by dynamic light scattering. The accumulation of VA in the skin from topical cream was evaluated in vitro using hairless mouse skins. The mean droplet size was $26.8{\pm}6.6$ nm for microemulsions II with Cremophor EL. All UA creams showed pseudoplastic flow and hysterisis loop in their rheogram, depending on the type of materials added in topical creams. The in vitro accumulation data demonstrated the UA topical cream prepared with the combination of Poloxamer 407 and Xanthan gum as a copolymer showed higher accumulation percentage than those prepared with either Poloxamer 407 or Xanthan gum. These results suggest that UA topical cream using microemulsion systems may be promising for the topical delivery of UA.
